Herd-level based seroprevalence and associated factors for Toxoplasma gondii in cows in the state of Paraíba, Northeastern Brazil

Abstract We aimed to determine the herd and animal levels seroprevalence and associated factors for Toxoplasma gondii infections in cattle from the state of Paraíba, Northeastern Brazil. Herds (n = 434) and cows aged ≥ 24 months (n = 1,895) were randomly selected, and serum samples were tested with the immunofluorescence antibody test (IFAT) using as cutoff of 64. Of the 434 farms investigated, 197 had at least one seropositive cow (prevalence of 49.0%; 95% CI = 44.3%-53.8%), and the prevalence at animal level was 18.0% (95% CI = 5.3%-21.1%). The antibody titers ranged from 64 to 1024, with the most frequent titers being 64 (10.8%) and 128 (3.7%). The risk factors identified were property located in Sertão region (odds ratio [OR] = 3.07), property located in Agreste/Zona da Mata regions (OR = 2.00), animal purchasing (OR = 2.68), herd size of 34-111 animals (OR = 2.91) and herd size > 111 animals (OR = 6.97). The results suggest the wide distribution of T. gondii infections in cattle throughout the state of Paraíba, and the risk factors identified are not possible to correct.
Resumo Objetivou-se determinar a soroprevalência do rebanhos e de animais e os fatores associados à infecção por Toxoplasma gondii em bovinos do estado da Paraíba, Nordeste do Brasil. Rebanhos (n = 434) e vacas com idade ≥ 24 meses (n = 1.895) foram selecionados aleatoriamente, e as amostras de soro foram testadas com o teste de imunofluorescência (RIFI), usando-se como ponto de corte 64. Das 434 fazendas investigadas, 197 tiveram pelo menos uma vaca soropositiva (prevalência de 49%; IC 95% = 44,3%-53,8%), e a prevalência em nível de animais foi de 18% (IC 95% = 5,3%-21,1%). Os títulos de anticorpos variaram de 64 a 1024, sendo os títulos mais frequentes 64 (10,8%) e 128 (3,7%). Os fatores de risco identificados foram na propriedade localizada na região do Sertão ("odds ratio" [OR] = 3,07), na propriedade localizada na região do Agreste/Zona da Mata (OR = 2,00), na compra de animais (OR = 2,68), no tamanho do rebanho de 34-111 animais (OR = 2,91) e no tamanho do rebanho > 111 animais (OR = 6,97). Os resultados sugerem a ampla distribuição de infecções por T. gondii em bovinos em todo o estado da Paraíba, e os fatores de risco identificados não são passíveis de correção.

Abstract Feeding ecology studies are crucial for understanding energy flow in reef ecosystems. In this study, we used an integrative approach to investigate the diel-feeding ecology of the sergeant-major Abudefduf saxatilis. To examine the possibility of diel-periodicity and size-class differences in foraging behaviour, we tracked fish individuals until their first bite on two subtropical reefs. During each observation, we recorded the substratum that was bitten and estimated the individual’s total length. To assess the diet, we analysed the stomach contents of five individuals from each location. In total, we observed 2,703 individuals biting seven substrates. Our results showed no significant differences in substrate use for diel-periodicity. However, we found significant differences between two size classes. Despite this, both populations tended to forage near the surface in the water column in the morning and on the benthos in the afternoon, although this tendency was not statistically significant. Smaller individuals fed mostly in the water column, while larger individuals foraged on all substrates, likely due to their different energetic demands. Our findings indicate this species has an omnivorous, generalist diet, comprising 12 items from both benthos and plankton. The lack of differences in diel-periodicity is likely due to the sergeant-major’s opportunistic behaviour.
Resumo Estudos de ecologia alimentar são cruciais para entender o fluxo de energia em ecossistemas recifais. Neste estudo, usamos uma abordagem integrativa para investigar a ecologia alimentar do sargentinho Abudefduf saxatilis. Para responder se há periodicidade ou diferenças relacionadas a classes de tamanho no substrato forrageado, acompanhamos indivíduos deste peixe até a primeira mordida, em dois recifes subtropicais. Durante cada observação, registramos o substrato mordido e estimamos o comprimento total do indivíduo. Para estudar a sua dieta, analisamos o conteúdo estomacal de cinco indivíduos em cada recife. No total, observamos 2.703 indivíduos mordendo em sete substratos diferentes. Nossos resultados não mostraram diferenças significativas no uso de substrato para a periodicidade diária. No entanto, encontramos diferenças significativas entre duas classes de tamanho. Apesar disso, ambas as populações tenderam a forragear próximo à superfície na coluna d’água pela manhã e no substrato bentônico à tarde, embora essa tendência não tenha sido estatisticamente significativa. Indivíduos menores forragearam principalmente na coluna d’água, enquanto os indivíduos maiores forragearam em todos os tipos de substratos, provavelmente devido às suas diferentes demandas energéticas. Nossos resultados corroboram que o sargentinho tem uma dieta onívora e generalista, composta por 12 itens de ambos os compartimentos, bentônico e planctônico. A falta de diferenças na periodicidade diária é provavelmente devido ao comportamento oportunista do sargentinho.

RESUMO Este estudo teve como objetivo verificar o impacto de programas comerciais de resfriamento rápido na qualidade físico-química de mangas ‘Rosa’. O estudo foi realizado em um packing house de mangas na região do Vale do São Francisco, Petrolina, PE, Brasil, entre Agosto e Outubro de 2018. O experimento seguiu um delineamento em blocos casualizados com três repetições e um palete por repetição. Os tratamentos foram organizados em esquema fatorial 2 × 2 × 3 (dois tempos de resfriamento rápido [120 e 240 min] × dois tempos de armazenamento x três posições no palete). Sessenta mangas foram distribuídas proporcionalmente em três camadas de diferentes alturas em cada um dos seis paletes. Três paletes foram submetidos ao resfriamento rápido por 120 min e outros três por 240 min. Todas as frutas foram armazenadas por 7 e 14 dias a 6 °C mais três dias de vida de prateleira a 20 °C. Independentemente do programa de resfriamento e da posição da fruta no palete, o tempo de resfriamento de sete oitavos (SECT) não foi alcançado. A acidez titulável do fruto com 240 min de resfriamento rápido diminuiu significativamente após 14 dias de armazenamento refrigerado. A perda de peso das frutas, a firmeza da polpa, a matéria seca e o teor de sólidos solúveis não foram afetados pelos programas de resfriamento rápido, tanto após o armazenamento quanto durante a vida de prateleira. O programa comercial de resfriamento rápido pode ser executado por 120 min.
ABSTRACT This study aimed to verify the impact of commercial fast-cooling programs on physicochemical quality of ‘Rosa’ mangoes. The study was carried out in a mango packing house in the São Francisco Valley region, Petrolina, PE, Brazil, between August and October 2018. The experiment followed a randomized block design, with three replicates and one pallet per replicate. The treatments were organized in a factorial scheme 2 × 2 × 3 (two fast-cooling times [120 and 240 min] × two storage times x three positions in the pallet). Sixty mangoes were proportionally distributed in three layers at different heights on each of the six pallets. Three pallets were subjected to fast cooling for 120 min and the other three for 240 min. All fruits were stored for 7 and 14 days at 6 °C plus three days of shelf-life at 20 °C. Regardless of cooling program and fruit position in the pallet, the seven-eighths cooling time (SECT) was not attained. The titratable acidity of fruits under 240-min fast cooling decreased significantly after 14 days of cold storage. Fruit weight loss, pulp firmness, dry matter, and soluble solids content were not affected by the fast-cooling programs, both after storage and during shelf-life period. Commercial fast-cooling program can be performed for 120 min.

Abstract Mango quality is affected by thermal storage conditions. Therefore, it is important to use cooling techniques and their monitoring to maintain desirable properties in the fruit. The objective of this study was to evaluate the effects of commercial rapid cooling programs on the quality of ‘Rosa’ mango. Mangoes were distributed on different horizontal positions of six pallets. All pallets were put through two rapid cooling times, three pallets for 120 minutes and three pallets for 240 minutes. Subsequently, the fruits were stored in a cold chamber for 7 and 14 days, followed by an evaluation of shelf life for 3 days at 20°C. Experimental design was in randomized blocks with three replicates, with one pallet per replicate. The weight, pH, and skin color of the fruit were assessed. According to the results, fruits closer to the air inlet in the packages showed the highest cooling rates. After storage and shelf life, quality parameters were not significantly affected by the rapid cooling programs. In conclusion, the 120-minute program can be satisfactorily adopted for the rapid cooling process of the ‘Rosa’ mango, considering the reduction of energy costs and the reduction of the dwell time of the fruit in the cold chamber.

Abstract Abstract: Trichiurus lepturus (Actinopterygii, Perciformes) is a commercially and economically important fish. A total of 60 specimens of this cutlassfish were collected of the coast the municipalities of Niterói and Cabo Frio, state of Rio de Janeiro, Brazil. The fish were measured, necropsied, filleted and had their organs investigated for digenetic trematodes. Taxonomic identification was based on morphological and morphometric characters. The specimens of T. lepturus were parasitized with adult specimens of Lecithochirium monticellii. Parasite indices of prevalence, intensity, mean intensity, abundance, mean abundance, range of infection, and site of infection of parasitic species were evaluated. Notes on the taxonomy of the parasite were also included. This is the first report of L. monticellii parasitizing T. lepturus in Brazil.

Resumo A frequência de infecções gastrintestinais foi avaliada em 502 gatos atendidos no Hospital Veterinário da Universidade de São Paulo, SP, Brasil, entre 2005 e 2014. As amostras foram analisadas utilizando-se os métodos de flutuação e sedimentação. Os resultados foram comparados com os de estudos previamente publicados com amostras de fezes oriundas do mesmo hospital, em momentos diferentes. As associações entre a frequência de positividade para cada parasito e idade, raça, sexo, ocorrência de diarreia e uso de anti-helmínticos foram investigadas (teste de qui-quadrado ou teste exato de Fisher). Um teste de qui-quadrado estratificado foi utilizado para comparar os diferentes períodos (1991-1995; 1996-1999; 2000-2004 e 2005-2014). Cryptosporidium spp., Giardia spp., Cystoisospora spp. e Sarcocystis spp. foram os parasitos mais comuns parasitos, seguidos por Toxocara cati e Ancylostoma spp. Cryptosporidium spp. apresentou maior frequência em gatos jovens e Sarcocystis spp. em animais com diarreia (p < 0,05). Comparação das frequências obtidas neste estudo com os de períodos anteriores mostrou que as frequências de Cryptosporidium spp., Cystoisospora spp. e T. cati foram menores (p < 0,05) que as observadas nos períodos anteriores. As frequências de Giardia spp. e Ancylostoma spp. foram semelhantes (p > 0,05) aos resultados encontrados no período de 2000 a 2004 e menores do que os valores encontrados para os outros períodos (p <0,05). Os motivos dessas alterações devem ser investigados.
Abstract The frequency of gastrointestinal infections in 502 cats seen at the Veterinary Hospital of the University of São Paulo, SP, Brazil, between 2005 and 2014, was measured. The samples were analyzed using methods of flotation and sedimentation. The results were compared with those from studies published previously using fecal samples from the same hospital at different times. Associations between the frequency of positivity for each parasite and age, breed, sex, diarrhea and use of anthelmintic were investigated (chi-square or Fisher’s exact tests). A partitioned chi-square test was used to compare different periods. Cryptosporidium spp., Giardia spp., Cystoisospora spp. and Sarcocystis spp. were the most common parasites, followed by Toxocara cati and Ancylostoma spp. Cryptosporidium spp. presented higher frequency in young cats and Sarcocystis spp. with the presence of diarrhea (p < 0.05). Results from this study with previous periods showed that the frequencies of Cryptosporidium spp., Cystoisospora spp. and T. cati were lower (p < 0.05) than those observed in previous periods. The frequencies of Giardia spp. and Ancylostoma spp. were similar to the results found in the preceding period and lower than the values found for the other periods (p < 0.05). The reasons for these changes should be investigated.

A 20-week program of resistance or concurrent exercise improves symptoms of schizophrenia: results of a blind, randomized controlled trial

Objective: To evaluate the effects of 20 weeks of resistance and concurrent training on psychotic and depressive symptoms, quality of life outcomes, and serum IGF-1, IGFBP-3, and brain-derived neurotrophic factor (BDNF) concentrations in patients with schizophrenia. Methods: In this blind, randomized controlled clinical trial, 34 patients with schizophrenia were assigned to one of three groups: control (CTRL, n=13), resistance exercise (RESEX, n=12), or concurrent exercise (CONCEX, n=9). Symptoms, quality of life, strength, and other variables were assessed. Results: A significant time-by-group interaction was found for the RESEX and CONCEX groups on the Positive and Negative Syndrome Scale (PANSS) total score for disease symptoms (p = 0.007), positive symptoms (p = 0.003), and on the arm extension one-repetition maximum (1RM) test (p = 0.016). In addition, significant improvements on negative symptoms (p = 0.027), on the role-physical domain of the Short Form-36 Health Survey (p = 0.019), and on the chest press 1RM test (p = 0.040) were observed in the RESEX group. No changes were observed for the other variables investigated. Conclusions: In this sample of patients with schizophrenia, 20 weeks of resistance or concurrent exercise program improved disease symptoms, strength, and quality of life. Resumo Os objetivos deste trabalho foram determinar a frequência de anticorpos anti-Toxoplasma gondii e anti-Neospora caninum e identificar os fatores de risco associados com as soropositividades em búfalos do Estado da Paraíba, Brasil. Foram utilizados 136 búfalos oriundos de 14 propriedades. Para a detecção de anticorpos anti-T. gondii e anti-N. caninum, foi empregada a reação de imunofluorescência indireta (RIFI). Das 136 amostras analisadas, 17 (12,5%) foram positivas para anticorpos anti-T. gondii, com títulos variando de 64 a 1.024; e 26 (19,1%) para anticorpos anti-N. caninum, com títulos de 200 a 1.600. Das 14 propriedades, em 10 (71,4%) houve animais soropositivos tanto para T. gondii como para N. caninum. O manejo semi-intensivo (odds ratio = 2,99) e a presença de suínos (odds ratio = 4,33) foram identificados como fatores de risco para T. gondii e N. caninum, respectivamente. Sugere-se que T. gondii e N. caninum estão disseminados em búfalos do Estado da Paraíba, bem como a necessidade de mais estudos acerca da importância desses agentes nessa espécie e em suínos, e da influência do tipo de criação na ocorrência de animais soropositivos.
Abstract The aims of this survey were to determine the frequency of anti-Toxoplasma gondii and anti-Neospora caninum antibodies and to identify the risk factors associated with seropositivity among buffaloes in the state of Paraíba, Brazil. This survey included 136 buffaloes belonging to 14 herds. To detect anti-T. gondii and anti-N. caninum antibodies, the indirect fluorescent antibody test (IFAT) was used. Among the 136 samples analyzed, 17 (12.5%) were positive for anti-T. gondii antibodies with titers ranging from 64 to 1,024, and 26 (19.1%) for anti-N. caninum with titers from 200 to 1,600. Animals seropositive for both T. gondii and N. caninum were found in 10 of the 14 herds (71.4%). Semi-intensive management systems (odds ratio = 2.99) and presence of pigs (odds ratio = 4.33) were identified as risk factors for T. gondii and N. caninum, respectively. It can be suggested that T. gondii and N. caninum are widespread in buffaloes in Paraíba, and that additional surveys are needed in order to ascertain the importance of these agents for this species and for pigs, and the influence of the farming type on occurrences of seropositive animals.

Os objetivos deste trabalho foram determinar a prevalência de rebanhos positivos e de animais soropositivos para Toxoplasma gondii em ovinos do Estado da Paraíba, Nordeste do Brasil, bem como identificar fatores de risco. Foram colhidas amostras de sangue de 540 ovinos de 63 rebanhos em 14 municípios da mesorregião do Sertão. Para o diagnóstico sorológico da infecção por T. gondii, foi utilizado o teste de imunofluorescência indireta (RIFI). Rebanhos com pelo menos um animal soropositivo foram considerados positivos. Dos 63 rebanhos, 28 (44,44%) foram positivos, e 60 (11,11%) dos 540 animais foram soropositivos. A variável tamanho do rebanho > 25 animais foi identificada como fator de risco (odds ratio = 3,2; IC 95% = 1,09 – 9,34; P = 0,033). Os resultados deste trabalho sugerem a disseminação de T. gondii em ovinos no Estado da Paraíba.

Physical characteristics of oily spraying liquids and droplets formed on coffee leaves and glass surfaces

As características físicas da calda de pulverização são importantes para que haja boa formação de gotas e eficiência de controle sobre determinado alvo. Em função dessas características, pode-se decidir sobre qual o melhor adjuvante, em sua respectiva concentração, poderá ser utilizado durante a pulverização. Para tanto, foram preparadas dez caldas fitossanitárias em que variaram as concentrações do inseticida lufenuron + profenofós, óleo mineral, água e sulfato de manganês. Gotas pendentes formadas a partir dessas misturas foram medidas em sua tensão superficial. Aplicaram-se gotas sobre a superfície de folhas de café e foram medidos a tensão superficial, o ângulo de contato formado e a área da folha molhada pela gota. Uma superfície lisa de vidro foi tomada como comparativo às folhas de café. As maiores concentrações de óleo resultaram em menores valores de tensão superficial, menores ângulos de contato de gotas com a superfície foliar e maiores áreas molhadas pelas gotas. Ambas as superfícies mostraram-se hidrofílicas.
The physical characteristics of a spray liquid are important in getting a good droplet formation and control efficiency over a particular target. As a function of these characteristics, it is possible to decipher which is the best adjuvant based on the respective concentration used during the spray. Therefore, ten spraying liquids were prepared, which varied in concentrations of pesticide lufenuron + profenofos, mineral oil, water and manganese sulfate. Pendant droplets formed from these mixtures were measured to examine their impact on surface tension. Droplets were applied to the surface of coffee leaves and the surface tension, contact angle formed and the leaf area wetted by the droplet, were measured. A smooth glass surface was taken as a comparative to the coffee leaves. The highest concentrations of oil resulted in lower surface tension, smaller contact angles of droplets on leaf surfaces and larger areas wetted by the droplets. Both surfaces showed hydrophilic behavior.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Tachia sp. are used as antimalarials in the Amazon Region and in vivo antimalarial activity of a Tachia sp. has been previously reported. Tachia grandiflora Maguire and Weaver is an Amazonian antimalarial plant and herein its cytotoxicity and antimalarial activity were investigated. Spectral analysis of the tetraoxygenated xanthone decussatin and the iridoid aglyone amplexine isolated, respectively, from the chloroform fractions of root methanol and leaf ethanol extracts was performed. In vitro inhibition of the growth of Plasmodium falciparum Welch was evaluated using optical microscopy on blood smears. Crude extracts of leaves and roots were inactive in vitro. However, chloroform fractions of the root and leaf extracts [half-maximal inhibitory concentration (IC50) = 10.5 and 35.8 µg/mL, respectively] and amplexine (IC50= 7.1 µg/mL) were active in vitro. Extracts and fractions were not toxic to type MRC-5 human fibroblasts (IC50> 50 µg/mL). Water extracts of the roots of T. grandiflora administered by mouth were the most active extracts in the Peters 4-day suppression test in Plasmodium berghei-infected mice. At 500 mg/kg/day, these extracts exhibited 45-59% inhibition five to seven days after infection. T. grandiflora infusions, fractions and isolated substance have potential as antimalarials.

Brazilian version of the foot health status questionnaire (FHSQ-BR): cross-cultural adaptation and evaluation of measurement properties

OBJECTIVE: To conduct a cross-cultural adaptation of the Foot Health Status Questionnaire into Brazilian-Portuguese and to assess its measurement properties. INTRODUCTION: This instrument is an outcome measure with 10 domains with scores ranging from 0-100, worst to best, respectively. The translated instrument will improve the examinations and foot care of rheumatoid arthritis patients. METHODS: The questions were translated, back-translated, evaluated by a multidisciplinary committee and pre-tested (n = 40 rheumatoid arthritis subjects). The new version was submitted to a field test (n = 65) to evaluate measurement properties such as test-retest reliability, internal consistency and construct validity. The Health Assessment Questionnaire, Numeric Rating Scale for foot pain and Sharp/van der Heijde scores for foot X-rays were used to test the construct validity. RESULTS: The cross-cultural adaptation was completed with minor wording adaptations from the original instrument. The evaluation of measurement properties showed high reliability with low variation coefficients between interviews. The a-Cronbach coefficients varied from 0.468 to 0.855, while correlation to the Health Assessment Questionnaire and Numeric Rating Scale was statistically significant for five out of eight domains. DISCUSSION: Intra- and inter-observer correlations showed high reliability. Internal consistency coefficients were high for all domains, revealing higher values for less subjective domains. As for construct validity, each domain revealed correlations with a specific group of parameters according to what the domains intended to measure. CONCLUSION: The FHSQ was cross-culturally adapted, generating a reliable, consistent, and valid instrument that is useful for evaluating foot health in patients with rheumatoid arthritis.
