ABSTRACT The increasing demand for using microorganisms in agriculture to improve food production requires constantly assessing microbial diversity. This study aimed to investigate the biochemical properties of individual and combined multifunctional microorganisms, as well as to identify potential applications in biotechnology or agriculture. The experiment comprised 29 treatments, with 7 single and 21 combined microorganisms: M01 (Serratia marcescens), M02 (Bacillus toyonensis), M03 (Phanerochaete australis), M04 (Trichoderma koningiopsis), M05 (Azospirillum brasilense), M06 (Azospirillum sp.), M07 (Bacillus sp.), M08 to M28 (combination among these microorganisms) and M29 (control - no microorganisms). All the single and combined treatments assimilated nitrogen, produced siderophores and indoleacetic acid and solubilized phosphate. Only the treatments M04, M13 and M26 produced HCN. Additionally, all treatments, except for M03, produced biofilm. Only M03, M07, M09, M10, M12 and M13 solubilized potassium.

RESUMO A crescente demanda pelo uso de micro-organismos na agricultura para melhorar a produção de alimentos exige uma avaliação constante da diversidade microbiana. Objetivou-se investigar as propriedades bioquímicas de micro-organismos multifuncionais individuais e combinados, bem como identificar aplicações potenciais em biotecnologia ou agricultura. O experimento compreendeu 29 tratamentos, com 7 micro-organismos isolados e 21 combinações: M01 (Serratia marcescens), M02 (Bacillus toyonensis), M03 (Phanerochaete australis), M04 (Trichoderma koningiopsis), M05 (Azospirillum brasilense), M06 (Azospirillum sp.), M07 (Bacillus sp.), M08 a M28 (combinação entre esses micro-organismos) e M29 (controle - sem micro-organismos). Todos os tratamentos individuais e combinados assimilaram nitrogênio, produziram sideróforos e ácido indolacético e solubilizaram fosfato. Apenas os tratamentos M04, M13 e M26 produziram HCN. Adicionalmente, todos os tratamentos, exceto M03, produziram biofilme. Somente M03, M07, M09, M10, M12 e M13 solubilizaram potássio.

Resumo O ipê-roxo, Handroanthus impetiginosus, é uma espécie arbórea importante na conservação do bioma Cerrado e muito procurada no paisagismo e arborização urbana. No entanto, sua micropropagação é afetada por patógenos, como Oidium sp. O objetivo deste estudo foi avaliar a eficácia dos tratamentos em sementes no controle do oídio do ipê roxo por micropropagação. Os sintomas foram observados durante a germinação in vitro, os postulados de Koch foram realizados para confirmação da patogenicidade, a colonização do patógeno nas folhas foi analisada em microscópios ópticos e de varredura e uma escala para avaliação de severidade foi proposta. Foram realizados dois ensaios para controle do oídio em delineamento inteiramente casualizado, com 30 repetições. Primeiro experimento: sementes foram tratadas com etanol, clorotalonil + tiofanato-metílico (C+TM) e hipoclorito de sódio; segundo experimento: sementes foram tratadas com etanol, NaOCl, C+TM e óleo de neem. A severidade da doença e a área abaixo da curva de progresso da doença foram quantificadas. Os sintomas da doença e estruturas típicas do patógeno foram observados e a patogenicidade foi confirmada. A severidade da doença foi reduzida em 30,78% com 1,5% de óleo de neem por 10 min quando comparado com C + TM por 15 min. Concluímos que o óleo de neem pode ser uma estratégia sustentável para o controle do oídio em ipê roxo na cultura de tecidos.

Abstract Purple ipe (Handroanthus impetiginosus) is an important tree species in Cerrado biome conservation and very popular at the landscaping and urban afforestation. However, its micropropagation is affected by pathogens, such as Oidium sp. The aim this study was evaluate the efficiency of seed treatments in the control of powdery mildew of purple ipe obtained by micropropagation. The symptoms were observed during in vitro germination, a Koch’s postulates were performed for confirm the pathogenicity, colonization of the pathogen on the leaves was analyzed in optical and scanning microscopes and a scale to evaluate severity was proposed. Two experiments were realized to powdery mildew control using a completely randomized design, with 30 replicates. First experiment: Seeds were treated with ethanol (Et), chlorothalonil + thiophanate-methyl (C+TM), and sodium hypochlorite (NaOCl); second experiment: Seeds were treated with Et, NaOCl, C+TM, and neem oil. Disease severity and area under the disease progress curve (AUDPC) were assessed in both experiments. Disease symptoms and typical pathogen structures were observed, and the pathogenicity was confirmed. The disease severity was reduced by 30.78% in 1.5% neem oil for 10 min when compared with C+TM for 15 min. We conclude that neem oil can be a strategy sustainable for the control of powdery mildew in purple ipe in tissue culture.

RESUMO A inoculação e coinoculação da soja por rizobactérias multifuncionais é uma alternativa sustentável que pode contribuir com o crescimento das plantas e com o aumento da produção agrícola, tornando o produto mais competitivo e, ainda, diminuir os custos para o produtor. Objetivou-se avaliar o efeito da inoculação e coinoculação com rizobactérias multifuncionais Serratia sp. e Bacillus sp. no desenvolvimento inicial da soja. O delineamento experimental foi o inteiramente casualizado, com 4 tratamentos e 10 repetições, totalizando 40 parcelas experimentais. Os tratamentos consistiram na microbiolização das sementes de soja e tratamento controle: isolado BRM 32114 (Serratia sp.); isolado BRM 63573 (Bacillus sp.) (antes nomeado 1301); coinoculação com BRM 32114 + BRM 63573; e controle (sem microbiolização). BRM 32114 e BRM 63753 isolados e combinados foram eficientes para melhorar o desenvolvimento inicial de plântulas da soja, proporcionando efeitos significativos para a maioria das variáveis analisadas (comprimento, superfície total, volume de raiz e biomassa de raiz, parte aérea e total), quando comparados ao tratamento controle.

ABSTRACT Soybean inoculation and co-inoculation with multifunctional rhizobacteria is a sustainable alternative that may contribute to plant growth and increased agricultural production, making the product more competitive, as well as reducing costs for the producer. This study aimed to evaluate the effect of inoculation and co-inoculation with multifunctional Serratia sp. and Bacillus sp. rhizobacteria on the early development of soybean. A completely randomized experimental design was used, with four treatments and ten replicates, totaling 40 experimental plots. The treatments consisted of the microbiolization of soybean seeds and a control treatment: BRM 32114 (Serratia sp.) isolate; BRM 63573 (Bacillus sp.) isolate (formerly named 1301); co-inoculation with BRM 32114 + BRM 63573; and control (without microbiolization). BRM 32114 and BRM 63753, both isolated and combined, were eficient to improve the initial development of soybean seedlings, providing significant effects for most of the analyzed variables (length, total surface, root volume and root, shoot and total biomass), when compared to the control treatment.

RESUMO A cultivar de arroz BRS Tropical foi desenvolvida para várzeas tropicais, mas é suscetível à queima da bainha, tornando o bioagente Waitea circinata uma importante ferramenta no manejo da doença. Objetivou-se avaliar os mecanismos de W. circinata envolvidos no parasitismo e na indução de resistência à queima da bainha. O antagonismo in vitro foi avaliado por microscopia óptica e microscopia eletrônica de varredura, seguida da quantificação das enzimas líticas envolvidas no parasitismo, como quitinase, glucanase e protease. Observou-se efeito do micoparasita W. circinata contra Rhizoctonia solani. A aplicação de W. circinata suprimiu a queima da bainha em até 65 % e aumentou a atividade da quitinase, glucanase e lipoxigenase 72 h após a inoculação e a da peroxidase 96 h após a inoculação. Assim, W. circinata mostrou-se efficiente em suprimir a queima da bainha por parasitismo e induziu resistência por meio da ativação de mecanismos bioquímicos.

ABSTRACT The BRS Tropical rice cultivar was developed for tropical foodplains, but is susceptible to sheath blight, making the Waitea circinata bioagent an important tool in managing the disease. This study aimed to assess the W. circinata mechanisms involved in the parasitism and resistance induction to sheath blight. The in vitro antagonism was assessed by optical microscopy and scanning electron microscopy, followed by quantification of the lytic enzymes involved in parasitism, such as chitinase, glucanase and protease. An effect of the W. circinata mycoparasite against Rhizoctonia solani was observed. The W. circinata application suppressed the sheath blight by up to 65 % and increased the chitinase, glucanase and lipoxygenase activity 72 h after the inoculation and that of the peroxidase 96 h after the inoculation. Thus, W. circinata showed to be efficient in suppressing sheath blight by parasitism and induced resistance via the activation of biochemical mechanisms.

RESUMO Waitea circinata (Warcup & Talbot) é um fungo antagonista micorrízico de orquídea, com potencial de biocontrole contra patógenos de arroz. Objetivou-se otimizar o método de extração, obter um novo extrato e avaliar sua eficiência contra patógenos de arroz in vitro e in vivo, bem como compará-lo com outros métodos de extração e W. circinata. Os extratos foram obtidos e testados para inibição de crescimento in vitro contra os patógenos Cochliobolus miyabeanus, Monographella albescens e Sarocladium oryzae, utilizando-se os seguintes extratos: micelial, bruto, liofilizado e de massa micelial. Um ensaio adicional in vitro foi realizado com o principal patógeno do arroz (Magnaporthe oryzae), para avaliar a germinação de conídios e a formação de apressórios. Com base nessa avaliação, os extratos liofilizado e de massa micelial foram testados in vivo contra a brusone do arroz (M. oryzae) e comparados com a suspensão micelial de W. circinata, em diferentes formas de aplicação (simultânea e prévia). O extrato de massa micelial inibiu todos os patógenos, e os extratos bruto e liofilizado inibiram C. miyabeanus e M. albescens, respectivamente. O extrato de massa micelial inibiu a germinação de conídios e a formação de apressórios de M. oryzae em 80 %, e as aplicações simultâneas e prévias suprimiram a brusone em 94 %. Os resultados indicam que o novo extrato pode ser usado para controlar patógenos de arroz.

ABSTRACT Waitea circinata (Warcup & Talbot) is an orchid antagonist mycorrhizal fungus with biocontrol potential against rice pathogens. This study aimed to optimize the extraction method, obtain a new extract and evaluate its efficiency against rice pathogens in vitro and in vivo, as well as to compare it with other extraction methods and W. circinata. The extracts were obtained and screened for in vitro growth inhibition against the pathogens Cochliobolus miyabeanus, Monographella albescens and Sarocladium oryzae, using the following extracts: mycelial, crude, lyophilized and mycelial mass. An additional in vitro assay was performed with the principal rice pathogen (Magnaporthe oryzae), in order to evaluate the conidial germination and appressorium formation. Based on this evaluation, the lyophilized and mycelial mass extracts were tested in vivo against rice blast (M. oryzae) and compared to the W. circinata mycelial suspension, in different application forms (simultaneous and previous). The mycelial mass extract inhibited all the pathogens, and the crude and lyophilized extracts inhibited C. miyabeanus and M. albescens, respectively. The mycelial mass extract inhibited the M. oryzae conidial germination and appressorium formation by 80 %, and the simultaneous and previous applications suppressed the rice blast by 94 %. These results indicate that the new extract can be used to control rice pathogens.

RESUMO Os sistemas tropicais de produção de arroz inundado requerem uma alta entrada de fertilizantes e defensivos químicos. O uso de rizobactérias promotoras de crescimento de plantas (RPCP), um componente sustentável desse sistema, pode aumentar a eficiência do uso de nutrientes e levar a aumentos significativos no rendimento de grãos das culturas tropicais de arroz inundado. Este estudo teve como objetivo determinar o efeito do microrganismo BRM 32110 (Bacillus thuringiensis) em combinação com doses aplicaas de nitrogênio (N), fósforo (P) e potássio (K) no desempenho fisiológico e agronômico de plantas de arroz inundadas no ambiente tropical. Os ensaios foram realizados na safra agrícola 2016/2017 em Formoso do Araguaia, cidade no estado do Tocantins, Brasil. Três experimentos independentes (E1, E2 e E3) foram realizados em delineamento de blocos ao acaso, em esquema fatorial 4 x 2, com três repetições. E1 compreendeu quatro doses aplicadas de N (0, 40, 80 e 120 kg N ha-1) com e sem a adição de BRM 32110, E2 compreendeu quatro doses aplicadas de P2O5 (0, 40, 80 e 120 kg de P2O5 ha-1) com ou sem BRM 32110, e E3 compreendeu quatro taxas de aplicação de K2O (0, 20, 40 e 60 kg K2O ha-1) com e sem BRM 32110. Em solo fértil, não houve interações entre a rizobacteria BRM 32110 e taxas aplicadas de N, P ou K. O BRM 32110 melhorou a captação de nutrientes e, em média, aumentou a matéria seca da parte aérea em 8%, a taxa de fotossíntese em 14% e a produtividade de grãos em 11% nas plantas de arroz inundadas. Nossos resultados sugerem que o uso de microrganismos multifuncionais é uma boa estratégia para melhorar o rendimento de grãos de arroz inundado de forma sustentável.

ABSTRACT Tropical flooded rice production systems require a high input of fertilizers and chemical defensive. The use of plant growth-promoting rhizobacteria (PGPR), a sustainable component of this system, can increase nutrient-use efficiency and lead to significant increases in the grain yield of tropical flooded rice crop. This study aimed to determine the effect of the microorganism BRM 32110 (Bacillus thuringiensis) in combination with nitrogen (N), phosphorus (P) and potassium (K) application rates on the physiological and agronomic performance of tropical flooded rice plants. Trials were performed in the 2016/2017 growing season in Formoso do Araguaia, a city in Tocantins state, Brazil. Three independent experiments (E1, E2, and E3) were performed in a randomized block design in a 4 x 2 factorial scheme with three replications. E1 comprised four N application rates (0, 40, 80 and 120 kg N ha-1) with and without the addition of BRM 32110, E2 comprised four P2O5 application rates (0, 40, 80 and 120 kg P2O5 ha-1) with or without BRM 32110, and E3 comprised four K2O application rates (0, 20, 40 and 60 kg K2O ha-1) with and without BRM 32110. In fertile soil, there were no interactions between the rhizobacterium BRM 32110 and the N, P or K application rates. BRM 32110 improved nutrient uptake and, on average, increased shoot dry matter by 8%, photosynthesis rate by 14% and grain yield by 11% in the flooded rice plants. Our results suggest that the use of multifunctional microorganisms is a good strategy for improving flooded rice grain yield sustainably.

RESUMO A brusone (Magnaporthe oryzae) limita a produção do arroz (Oryza sativa), no Brasil e no mundo. Os objetivos foram caracterizar seis isolados bacterianos morfológica, bioquímica e molecularmente, e determinar seu potencial como antagonistas a M. oryzae. Utilizaram se os isolados bacterianos BRM 32109, BRM 32110, BRM 32111, BRM 32112, BRM 32113 e BRM 32114. Avaliou-se o formato e a cor das colônias, coloração de Gram e produção de pigmentos fluorescentes. Identificou-se a produção de celulase, quitinase, fosfatase, ácido indolacético, sideróforo e formação de biofilme. A identificação molecular foi realizada utilizando-se oligonucleotídeos iniciadores específicos para amplificação por PCR da região 16S rRNA, seguidos de sequenciamento. Os dois ensaios envolvendo antagonismo foram realizados em laboratório, em delineamento inteiramente casualizados, um com cultivo pareado entre isolado de bactérias e M. oryzae, e outro com filtrados bacterianos. Um terceiro ensaio em casa de vegetação para avaliar a supressão da brusone. Identificou-se Pseudomonas sp. (BRM 32111 e BRM 32112), Burkholderia sp. (BRM 32113), Serratia sp. (BRM 32114) e Bacillus spp. (BRM 32110 e BRM 32109 ). Os isolados BRM 32112, BRM 32111 e BRM 32113 inibiram o crescimento da colônia de M. oryzae em 68, 65 e 48%, respectivamente. As suspensões bacterianas BRM 32111, BRM 32112 e seu filtrado suprimiram a brusone foliar em 81,0, 79,2 e 66,3%, respectivamente. BRM 32111 e BRM 32112 são antagonistas ao fungo M. oryzae, solubilizam fosfato, produzem sideróforos, celulase e biofilme e suprimiram a brusone foliar, mostrando-se potenciais agentes biológicos para o controle da brusone foliar.

ABSTRACT Rice blast (Magnaporthe oryzae) limits rice (Oryza sativa) grain yields worldwide. The objective of this investigation was to morphologically, biochemically, and molecularly characterize six bacterial isolates, BRM 32109, BRM 32110, BRM 32111, BRM 32112, BRM 32113, and BRM 32114, and to determine their potential as antagonists to M. oryzae. Morphological characterization was based on colony formation and color, Gram staining, and fluorescent pigment production. Biochemical studies were based on cellulase, chitinase, phosphatase, indoleacetic acid, and siderophore production, as well as biofilm formation. The molecular identification used specific primers for PCR amplification of the 16S rRNA region, followed by sequencing. The antagonism studies involved three experiments, which had randomized designs. Two of them were conducted in laboratory conditions, pairing bacterial colonies and M. oryzae, using bacterial filtrates, and the third was conducted in greenhouse conditions. BRM 32111 and BRM 32112 were identified as Pseudomonas sp., BRM 32113 as Burkholderia sp., BRM 32114 as Serratia sp., and BRM 32110 and BRM 32109 as Bacillus spp. BRM 32112, BRM 32111, and BRM 32113 inhibited the colony of M. oryzae by 68%, 65%, and 48%, respectively. The bacterial suspensions of the BRM 32111, BRM 32112, and BRM 3212 filtrates suppressed leaf blast by 81.0, 79.2, and 66.3%, respectively. BRM 32111 and BRM 32112 were determined to be antagonists of M. oryzae and were found to solubilize phosphate, produce siderophores and cellulose, form biofilms, and suppress leaf blast. These isolates should be further investigated as potential biological control agents for leaf blast control.

RESUMO O uso de microrganismos benéficos nos sistemas de cultivo pode contribuir para a agricultura sustentável, promovendo melhorias no desenvolvimento das culturas e na produção de grãos sem danificar o meio ambiente. No entanto, há muita incerteza quanto aos efeitos do uso de uma combinação de microrganismos para promover o desenvolvimento da planta. O objetivo deste trabalho foi determinar os efeitos das espécies de microrganismos individualmente e em combinação na produção de biomassa, trocas gasosas e conteúdo de nutrientes na parte aérea e raízes das plantas de soja. O delineamento experimental foi inteiramente casualizado, com 30 tratamentos e três repetições. Os tratamentos consistiram na aplicação das rizobactérias BRM 32109, BRM 32110 e 1301 (Bacillus sp.); BRM 32111 e BRM 32112 (Pseudomonas sp.); BRM 32113 (Burkholderia sp.); BRM 32114 (Serratia sp.); Ab-V5 (Azospirillum brasilense) e 1381 (Azospirillum sp.); o fungo Trichoderma asperellum (uma mistura dos isolados UFRA. T06, UFRA. T09, UFRA. T12 e UFRA. T52); 19 combinações desses microrganismos e um controle (sem microrganismos). Na semeadura, o solo foi tratado com Bradyrhizobium e as sementes de soja foram inoculadas. A suspensão de microrganismos foi aplicada em cada tratamento aos 7 dias após o plantio (DAP) no solo e aos 21 DAP nas mudas. Os tratamentos pool de Trichoderma, Ab-V5, 1301 + 32110, 1301 + 32114, 1301 + Ab-V5 e 32110 + Ab-V5 resultaram em acúmulo de biomassa total significativamente maior nas plantas de soja, com valores, em média, 25% superior ao tratamento controle. Nossos resultados sugerem que o uso de microrganismos benéficos em sistemas de cultivo é uma técnica promissora.

ABSTRACT The use of beneficial microorganisms in crop systems can contribute to sustainable agriculture by promoting improvements in crop development and grain yield without damaging the environment. However, there is much uncertainty surrounding the effects of using a combination of microorganisms to promote plant development. The objective of this work was to determine the effects of microorganism species individually and in combination on the biomass production, gas exchange and nutrient contents in the shoots and roots of soybean plants. The experimental design was completely randomized, with 30 treatments and three replicates. The treatments consisted of the application of the rhizobacteria BRM 32109, BRM 32110 and 1301 (Bacillussp.); BRM 32111 and BRM 32112 (Pseudomonas sp.); BRM 32113 (Burkholderia sp.); BRM 32114 (Serratia sp.); Ab-V5 (Azospirillum bras ilense) and 1381 (Azospirillum sp.); the fungus Trichoderma asperellum (a mixture of the isolates UFRA. T06, UFRA. T09, UFRA. T12 and UFRA. T52); 19 combinations of these microorganisms, and a control (no microorganisms). At sowing, the soil was treated with Bradyrhizobium, and then the soybean seeds were inoculated. The microorganism suspension was applied in each treatment at 7 days after planting (DAP) in the soil and at 21 DAP on the seedlings. The Trichoderma pool, Ab-V5, 1301 + 32110, 1301 + 32114, 1301 + Ab-V5 and 32110 + Ab-V5 treatments resulted in significantly higher total biomass accumulation in the soybean plants, with values that were, on average, 25% higher than that in the control treatment. Our results suggest that the use of beneficial microorganisms in cropping systems is a promising technique.

RESUMO Microrganismos multifuncionais podem contribuir significativamente para melhorar o desempenho das plantas, sendo tecnologia inovadora quando se visa o desenvolvimento sustentável. Assim, objetivou-se determinar o efeito de microrganismos multifuncionais aplicados isolados ou em mix sobre o desempenho do arroz de terras altas. O experimento foi conduzido em casa de vegetação, em delineamento inteiramente casualizado, com quatro repetições. Os 26 tratamentos consistiram das rizobactérias Bacillus sp. (BRM 32109, BRM 32110 e 1301), Azospirillum sp. (1381), Azospirillum brasilense (Ab-V5), Pseudomonas sp. (BRM 32112), Pseudomonas fluorescens (BRM 32111), Burkholderia pyrrocinia (BRM 32113), Serratia sp. (BRM 32114), e um gênero fúngico formando um pool de Trichoderma asperellum (T-06, T-09, T-12, e T-52), aplicados isoladamente ou em consórcio, mais o tratamento controle, sem aplicação de microrganismos. Os tratamentos mais efetivos foram os mixes de microrganismos 1301 + Ab-V5 e BRM 32114 + pool de Trichoderma asperellum, uma vez que proporcionaram aumento, em média, de 123% e 88% no número de panículas e de 206 e 167% na produtividade de grão do arroz de terras altas, respectivamente. Os consórcios 1301 + Ab-V5, BRM 32114 + pool de Trichoderma asperellum, BRM 32110 + BRM 32114, BRM 32110 + Ab-V5, 1301 + BRM 32110 e 1381 + pool de Trichoderma asperellum, também proporcionaram maior desempenho morfofisiológico nas plantas de arroz (taxa fotossintética, eficiência de carboxilação, número de perfilhos, biomassa seca de parte aérea e teor de nutrientes na parte aérea e raiz das plantas). Portanto, o uso de microrganismos multifuncionais no manejo do arroz de terras altas foi eficiente em sua capacidade de proporcionar maior desenvolvimento das plantas.

ABSTRACT Multifunctional microorganisms can contribute significantly to improve plant performance and are innovative technology for sustainable development. Thus, objective of this study was to determine the effect of bioagents applied alone or in mix on the performance of upland rice. The experiment was conducted in a greenhouse in a completely randomized design with four replications. The 26 treatments consisted of the rhizobacteria Bacillus sp. (BRM 32109, BRM 32110 and 1301), Azospirillum sp. (1381), Azospirillum brasilense (Ab-V5), Pseudomonas sp. (BRM 32112), Pseudomonas fluorescens (BRM 32111), Burkholderia pyrrocinia (BRM 32113), Serratia sp. (BRM 32114), and a fungal genus formed by a pool of Trichoderma asperellum (T-06, T-09, T-12, and T-52), applied alone or in mix, plus a control treatment without microorganism application. The most effective treatments were the mixes of microorganisms 1301 + Ab-V5 and BRM 32114 + pool of Trichoderma asperellum, as they provided an average increase of 123 and 88% in the number of panicles and 206 and 167% in the grain yield of upland rice plants, respectively. Mixes of 1301 + Ab-V5, BRM 32114 + Trichoderma asperellum pool, BRM 32110 + BRM 32114, BRM 32110 + Ab-V5, 1301 + BRM 32110 and 1381 + Trichoderma asperellum pool also provided better morphophysiological performance in rice plants (photosynthetic rate, carboxylation efficiency, number of tillers, shoot dry biomass and nutrient content in shoot and root). Therefore, the use of multifunctional microorganisms in the management of upland rice was efficient in its ability to provide better development of plants.

Abstract The objective of this research was to identify the best microorganisms, alone or in mixture for total biomass gain (root + shoot), positive change in gas exchange, nutrient uptake (root, shoot and grain) and yield and yield components in the soybean crop. Trial under greenhouse conditions had the experimental design in a completely randomized scheme with 26 treatments and four replicates. The treatments consisted of the rhizobacteria BRM 32109, BRM 32110 and 1301 (Bacillus sp.), BRM 32111 and BRM 32112 (Pseudomonas sp.), BRM 32113 (Burkholderia sp.), BRM 32114 (Serratia sp.), Ab-V5 (Azospirillum brasilense) and 1381 (Azospirillum sp.), and the fungus Trichoderma asperellum (a mix of the isolates UFRA.T06, UFRA.T09, UFRA.T12 and UFRA.T52). Besides, the same isolates were combined in pairs, completing 16 combinations. Control treatments received no microorganism. Microorganisms applied isolated or in combination, provided biomass gain, positive gas exchange, increases in nutrients uptake at the shoot and grain, and improved grain yield and its components than control plants. Stood out the combination Ab-V5 + T. asperellum pool, which provided a 25% improvement in grain yield.

RESUMO Em uma relação planta-endófito, substâncias secretadas por micro-organismos podem ajudar a planta a se defender contra patógenos e promover o aumento de biomassa. Objetivou-se caracterizar bioquimicamente fungos endofíticos de Bambusa oldhamii Munro, visando a futuros estudos sobre promoção de crescimento, bem como indução de resistência. Foram utilizados os seguintes isolados fúngicos de B. oldhamii: 29 (Acrocalymma sp.); 122 (Botryobambusa fusicoccum); 711 (Phoma sp.); e 712 (Phoma sp.). Foi realizada caracterização bioquímica, analisando-se a produção de ácido indolacético, quitinases e glucanases, bem como a presença ou ausência de polifenol oxidases e atividade de solubilização de fosfato in vitro. O isolado 29 produziu maiores quantidades de ácido indolacético e quitinases, rendendo 31,55 mg mL-1 após 96 h e 2,59 U mL-1 após 120 h, respectivamente. O isolado 122 produziu uma quantidade aumentada de glucanases, rendendo 2,65 U mL-1 após 72 h. Os isolados 29, 711 e 712 produziram polifenol oxidases. O isolado 711 apresentou atividade de solubilização de fosfato in vitro. Esses resultados indicam que os isolados em estudo apresentam potencial para indução de resistência em plantas e para futuros estudos de aumento de biomassa vegetal.

ABSTRACT In a plant-endophyte relationship, substances secreted by microorganisms can help the plant to defend itself against pathogens and also to promote an increase in biomass. This study aimed to biochemically characterize endophytic fungi from Bambusa oldhamii Munro, aiming at future studies on plant growth promotion, as well as on resistance induction. The following fungal isolates from B. oldhamii were used: 29 (Acrocalymma sp.); 122 (Botryobambusa fusicoccum); 711 (Phoma sp.); and 712 (Phoma sp.). Biochemical characterization was performed, analyzing the production of indole-acetic acid, chitinases and glucanases, as well as the presence or absence of polyphenol oxidases and phosphate solubilization activity in vitro. The isolate 29 produced larger amounts of indole-acetic acid and chitinases, yielding 31.55 mg mL-1 after 96 h and 2.59 U mL-1 after 120 h, respectively. The isolate 122 produced an increased amount of glucanases, yielding 2.65 U mL-1 after 72 h. The isolates 29, 711 and 712 produced polyphenol oxidases. The isolate 711 showed phosphate solubilization activity in vitro. These results indicate that the isolates under study show a potential for inducing resistance in plants and for future studies on the increase of plant biomass.

Resumo Fungos do complexo Rhizoctonia são micorrízicos de orquídeas e podem germinar suas sementes e desenvolver as plântulas. Neste contexto, nosso objetivo foi selecionar um isolado fúngico para promover a germinação e o desenvolvimento de plântulas de Cyrtopodium saintlegerianum. Foram encontrados pelotons nas raízes desta orquídea e foram identificados três fungos micorrízicos. Nós testamos os isolados obtidos das raízes de C. saintlegerianum e seis outros isolados micorrízicos assim como três isolados patogênicos (de arroz e feijão) para germinar as sementes em meio de aveia-agar. Sementes não inoculadas foram usadas como controle. Os isolados En07 (Waitea circinata), Cs10 (Tulasnella sp.) e Ro88 (Rhizoctonia oryzae) foram eficientes para promover a germinação, mas somente o En07diferiu estatisticamente do controle. O isolado não específico En07 promoveu 81% de germinação e o isolado específico (Cs10) promoveu 60%, evidenciando a associação micorrízica não específica durante a germinação. Plântulas axênicas foram inoculadas com quatro fungos micorrízicos (plântulas não inoculadas - controle). Após seis meses, os isolados En07 e Cs10 foram eficientes na interação com as plântulas, mas não diferiram do controle. Portanto, nossos resultados sugerem que fungos do complexo Rhizoctonia podem ser usados na germinação e no desenvolvimento de plântulas de C. saintlegerianum.

Abstract Fungi of Rhizoctonia complex are mycorrhizal of orchids and may to germinate yours seeds and development the seedlings. In this context, our objective was to select a fungal isolate to promote seed germination and seedling development of Cyrtopodium saintlegerianum. Pelotons were found in the roots and three mycorrhizal fungi were isolated. We tested mycorrhizal isolates obtained from C. saintlegerianum roots and six mycorrhizal fungi from other orchids as well three pathogenic isolates (of rice and bean) to germinate the seeds in oatmeal-agar medium. Seeds not inoculated were used as control. The isolates En07 (Waitea circinata), Cs10 (Tulasnella sp.) and Ro88 (Rhizoctonia oryzae) were efficient to promote seed germination, but only En07 differing statistically of the control. The non-specific isolate En07 promoted germination in 81% of seeds and the specific isolate (Cs10) promoted 60%, evidencing the non-specificity mycorrhizal association in this orchid during germination. Axenic seedlings were inoculated with four mycorrhizal fungi (non-inoculated seedlings - control). After six months, the isolates En07 and Cs10 were efficient in the interaction with the seedlings, but did not differ to the control. Therefore, our results suggested that fungi of the Rhizoctonia complex can be used in the germination and seedling development of C. saintlegerianum.

RESUMO O manejo integrado de brusone (Magnaporthe oryzae) em arroz é realizado, principalmente, com a adoção de controle químico. No entanto, a busca por práticas alternativas tem crescido nos últimos anos. Assim, avaliou-se a variabilidade de 28 isolados de Sarocladium oryzae, quanto à produção de cerulenina, e o seu potencial na redução da severidade de brusone foliar em arroz, quantificando-se a atividade de enzimas ligadas ao mecanismo de defesa das plantas. Mais de 55 % dos isolados de S. oryzae foram antagônicos aos patógenos M. oryzae, Cochliobolus miyabeanus, Thanatephorus cucumeris e Monographella albescens, e 60 % dos isolados produziram cerulenina em níveis detectáveis. Tanto BRM 6461 (296,0 µg mL-1) quanto BRM 6493 (cerulenina não detectável) inibiram a formação de apressórios de M. oryzae em 89,5 % e 85 %, respectivamente. O isolado BRM 6461, aplicado na forma de suspensão de conídios e filtrado, reduziu a severidade da brusone em 68,8 % e 75,5 %, respectivamente. A atividade enzimática, na presença de M. oryzae, foi maior para lipoxigenase após 5 h (filtrado) e 24 h e 72 h (suspensão de conídios) da inoculação do patógeno. Para fenilalanina-amônia liase, a maior expressão ocorreu após 5 h (filtrado) e 72 h (suspensão de conídios). As enzimas quitinase, β-1,3-glucanase e peroxidase e o fitohormônio ácido salicílico não apresentaram diferenças em relação aos controles (água e M. oryzae). O filtrado do isolado BRM 6461, constituído basicamente por cerulenina, reduziu a severidade da brusone e, possivelmente, ativou os mecanismos de defesa da planta de arroz contra M. oryzae.

ABSTRACT The integrated management of rice leaf blast (Magnaporthe oryzae) is carried out mainly with the adoption of chemical control. However, the search for alternative practices has grown in recent years. Thus, the variability of 28 Sarocladium oryzae isolates was evaluated for the cerulenin production, as well as its potential for reducing the severity of rice leaf blast, quantifying the activity of enzymes linked to the plant defense mechanisms. More than 55 % of the S. oryzae isolates were antagonistic to the pathogens M. oryzae, Cochliobolus miyabeanus, Thanatephorus cucumeris and Monographella albescens, and 60 % of the isolates produced cerulenin at detectable levels. Both BRM 6461 (296.0 µg mL-1) and BRM 6493 (undetectable cerulenin) inhibited the formation of M. oryzae appressoria in 89.5 % and 85 %, respectively. The BRM 6461 isolate, applied as conidial suspension and filtered, reduced the severity of rice leaf blast in 68.8 % and 75.5 %, respectively. The enzymatic activity in the presence of M. oryzae was higher for lipoxygenase at 5 h (filtered) and at 24 h and 72 h (conidial suspension) after the pathogen inoculation. For phenylalanine ammonia lyase, the highest expression was at 5 h (filtered) and 72 h (conidial suspension). The enzymes chitinase, β-1,3-glucanase and peroxidase and the salicylic acid phytohormone presented no differences, in relation to the controls (water and M. oryzae). The filtered from the BRM 6461 isolate, basically constituted by cerulenin, reduced the severity of rice leaf blast and possibly activated the defense mechanisms of the rice plants against M. oryzae.

RESUMO: As fitoalexinas são compostos que vêm sendo estudados há alguns anos, apresentando principalmente atividade antimicrobiana. As plantas da família Poaceae são as geograficamente mais difundidas e se destacam por sua importância econômica, uma vez que são os cereais que compõe a base da alimentação, em todos os continentes. Essa família se destaca por apresentar uma variedade de fitoalexinas, sendo que as mais importantes são sintetizadas a partir da via do ácido chiquímico. A caracterização desses compostos com atividade antimicrobiana é realizada por meio de técnicas cromatográficas, sendo que a cromatografia líquida de alta eficiência (HPLC) acoplada com espectrometria de massas é a técnica mais eficiente nesse processo. O trabalho apresenta como objetivo uma abordagem da função, caracterização, biossíntese e aplicações das fitoalexinas em plantas da família Poaceae.

ABSTRACT: Phytoalexins are compounds that have been studied a few years ago, which present mainly antimicrobial activity. The plants of the family Poaceae are the most geographically widespread and stand out for their economic importance, once they are cereals used as staple food. This family stands out for having a variety of phytoalexins, which can be synthesized via the shikimic acid (the phenylpropanoids), or mevalonic acid, being considered terpenoid phytoalexins. The characterization of these compounds with antimicrobial activity is carried out using chromatographic techniques, and the high-performance liquid chromatography (HPLC) coupled with mass spectrometry are the most efficient methods in this process. This research aimed to present an approach of the function, characterization and biosynthesis of phytoalexins in plants of the family Poaceae.

ABSTRACT Blast, caused by the fungus Pyricularia oryzae, is an important disease affecting rice and wheat yield worldwide. This study investigated the cytological aspects of incompatible (non-host resistance) and compatible (host resistance) rice- (R_Po) and wheat- (W_Po) Pyricularia oryzae isolate interactions. Inoculations of rice and wheat with the R_Po and W_Po isolates of P. oryzae, respectively, were expected to be compatible interactions (host resistance), whereas inoculations of rice and wheat with the W_Po and R_Po isolates of P. oryzae, respectively, were considered to be incompatible interactions (non-host resistance). For the compatible interactions (rice-R_Po and wheat-W_Po), fungal hyphae penetrated and colonized the epidermal cells and also invaded many neighboring cells. By contrast, in the case of the incompatible interactions (rice-W_Po and wheat-R_Po), fungal hyphae were not able to penetrate nor colonize the epidermal cells, but when penetration did occur, the hyphae were restricted to the first-invaded epidermal cell. The frequency of appressorial sites exhibiting infection hyphae within the epidermal cell underlying an appressorium was greater in the case of the compatible interactions. By contrast, unsuccessful penetrations with cytoplasmic granulation occurred with high frequency in the incompatible wheat-R_Po and rice-W_Po interactions and the number of necrotic epidermal cells underlying the appressorium was low for the rice-W_Po interaction as well as for the wheat- R_Po interaction, where no symptoms of necrosis were exhibited. However, the opposite was observed for the compatible interactions. The present study presents cytological features associated with incompatible and compatible rice- and wheat-P. oryzae interactions that may be useful to studies involving variability, coevolution, diagnosis, and regulation of quarantine or even in a rice or wheat breeding program whose aim is to transfer genes involved in non-host resistance to host resistance due to similarities in downstream mechanisms.
