Objective: This study evaluated the effect of three metal conditioners on the shear bond strength (SBS) of a prosthetic composite material to cpTi grade I having three surface treatments. Material and Methods: One hundred sixty eight rivet-shaped specimens (8.0x2.0 mm) were cast and subjected to polishing (P) or sandblasting with either 50 mm (50SB) or 250 mm (250SB) Al2O3. The metal conditioners Metal Photo Primer (MPP), Cesead II Opaque Primer (OP), Targis Link (TL), and one surface modification system Siloc (S), were applied to the specimen surfaces, which were covered with four 1-mm thick layers of resin composite. The resin layers were exposed to curing light for 90 s separately. Seven specimens from each experimental group were stored in water at 37ºC for 24 h while the other 7 specimens were subjected to 5,000 thermal cycles consisting of water baths at 4ºC and 60ºC (n=7). All specimens were subjected to SBS test (0.5 mm/min) until failure occurred, and further 28 specimens were analyzed using scanning electron microscope (SEM) and X-ray energy-dispersive spectroscopy (EDS). Data were analyzed by 3-way ANOVA followed by post-hoc Tukey's test (α=0.05). Results: On 50SB surfaces, OP groups showed higher SBS means than MPP (P<0.05), while no significant difference was found among OP, S, and TL groups. On 250SB surfaces, OP and TL groups exhibited higher SBS than MPP and S (P<0.05). No significant difference in SBS was found between OP and TL groups nor between MPP and S groups. The use of conditioners on 250SB surfaces resulted in higher SBS means than the use of the same products on 50SB surfaces (P<0.05). Conclusion: Sandblasting associated with the use of metal conditioners improves SBS of resin composites to cpTi.

Staphylococcus aureus resistente à meticilina (MRSA, do inglês methicillin-resistant Staphylococcus aureus) pode crescer como biofilme estruturado em diferentes superfícies, incluindo mucosa bucal e superfícies de próteses. Estes biofilmes podem se dispersar nos fluidos orais e ser aspirados, causando infecções sistêmicas, como a pneumonia aspirativa. Este estudo avaliou a eficácia de duas soluções desinfetantes e irradiação por microondas na desinfecção de corpos-de-prova acrílicos contaminados com biofilme de MRSA. Trinta e seis espécimes de resina acrílica foram fabricados, esterilizados e contaminados com MRSA (107 ufc/mL). Após a incubação (37 °C/48 h), os espécimes foram divididos em quatro grupos: não desinfetados (controle positivo); imersos em hipoclorito de sódio 1% por 10 min; imersos em gluconato de clorexidina 2% por 10 min e irradiados por microondas durante 3 min a 650 W. A viabilidade das células foi avaliada pelo método de redução de XTT. Todos os espécimes do grupo controle apresentaram formação de biofilme após 48 h de incubação. O valor médio de absorbância destes espécimes foi de 1.58 (OD a 492 nm). Nenhuma evidência de formação de biofilme foi observada em todas as amostras desinfetadas. A desinfecção em hipoclorito de sódio 1%, gluconato de clorexidina 2% e irradiação em microondas resultou em 100% de redução do metabolismo do biofilme de MRSA. O uso de soluções químicas e irradiação em microondas mostrou-se eficaz na eliminação do biofilme maduro de MRSA sobre corpos-de-prova de resina acrílica.

Methicillin-resistant Staphylococcus aureus (MRSA) can grow as structured biofilm in different surfaces, including oral mucosa and denture surfaces. Such biofilms can be released into the oral fluids and aspirated, causing systemic infections such as aspiration pneumonia. This study evaluated the efficacy of two disinfectant solutions and microwave irradiation in disinfecting acrylic specimens contaminated with MRSA biofilm. Thirty-six acrylic specimens were made, sterilized and contaminated with MRSA (107 cfu/mL). After incubation (37 °C/48 h), the specimens were divided into 4 groups: not disinfected (positive control); soaking in 1% sodium hypochlorite for 10 min; soaking in 2% chlorhexidine gluconate for 10 min; and irradiating by microwave for 3 min at 650 W. The viability of cells was evaluated by XTT reduction method. All specimens from the positive control group showed biofilm formation after 48 h incubation. The mean absorbance value of the control specimens was 1.58 (OD at 492 nm). No evidence of biofilm formation was observed on specimens after the disinfection methods. Disinfection by soaking in 1% sodium hypochlorite and 2% chlorhexidine gluconate and irradiating by microwaves resulted in 100% reduction of MRSA biofilm metabolism. The use of chemical solutions and microwave irradiation was shown to be effective for eradicating mature MRSA biofilms on acrylic resin specimens.

A adesão de Candida albicans às superfícies é o primeiro passo para o desenvolvimento da estomatite protética, uma infecção frequente diagnosticada entre os usuários de próteses. Uma adequada higienização é essencial para prevenir a formação de biofilme microbiano e o início desta infecção. O objetivo deste estudo foi avaliar a efetividade da escovação com diferentes soluções na eliminação de biofilme de C. albicans. Para isso, discos de resina acrílica foram confeccionados, esterilizados e inoculados com uma suspensão de 10(7) células/mL de C. albicans. Após incubação (37°C/48 h), os espécimes foram aleatoriamente divididos em 10 grupos experimentais (n=9): 5 submetidos à escovação com água ou agentes de limpeza (água destilada, dentifrício, digluconato de clorexidina (CHX) a 2%, hipoclorito de sódio (NaOCl) a 1% e Polident fresh cleanse®) e 4 apenas imersos nos agentes de limpeza. Espécimes não submetidos à higienização foram utilizados como controle positivo. A viabilidade celular foi verificada pelo teste de redução do XTT. Os resultados obtidos foram analisados pelos testes de Mann-Whitney e Kruskal-Wallis (α=0,05). A escovação com todos os agentes de limpeza apresentou redução significativamente superior (p<0,0001) na viabilidade do biofilme quando comparada à exposição dos espécimes às soluções. Escovação com CHX a 2% e NaOCl a 1% resultaram em 100% de inativação do biofilme. A exposição aos agentes de limpeza resultou em redução significativa (p<0,0001) na viabilidade celular, com CHX a 2% sendo o mais efetivo (p<0,0001). A utilização de agentes de limpeza em associação ao método de escovação provou ser efetivo para reduzir biofilme C. albicans, sendo as soluções de CHX e NaOCl as mais efetivas.

The adhesion of Candida albicans to surfaces is the prerequisite for occurrence of denture stomatitis, a common disease diagnosed among denture wearers. A routine of denture cleansing is essential to prevent biofilm formation and the onset of this infection. The aim of this study was to investigate the effectiveness of combining brushing and cleansing agents in killing C. albicans biofilm. Disks of acrylic resin were made, sterilized, and inoculated with C. albicans (10(7) cfu/mL). After incubation (37°C/48 h), specimens were randomly assigned to 10 experimental groups (n=9): 5 subjected to brushing with distilled water or cleansing agents - dentifrice slurry, 2% chlorhexidine gluconate (CHX), 1% sodium hypochlorite (NaOCl), and Polident fresh cleanse® (combined method) - and 4 exposed to the cleansing agents without brushing (immersion). Non-cleansed specimens were used as positive controls. The viability of cells was evaluated by XTT reduction method. Results were analyzed by Mann-Whitney and Kruskal-Wallis tests (α=0.05). The combined method was significantly more effective (p<0.0001) in reducing biofilm viability than the immersion. Brushing with CHX and NaOCl resulted in 100% removal of the biofilm. Immersion in the agents reduced significantly (p<0.0001) the biofilm viability, with CHX being the most effective (p<0.0001). The use of the combined method of brushing with cleansing agents is an effective method to reduce C. albicans biofilm, being CHX and NaOCl the most effective solutions.

OBJECTIVE: The effect of disinfection by immersion in sodium perborate solution and microwave irradiation on surface roughness of one denture base resin (Lucitone 550 -L), 3 hard chairside reline resins (Tokuyama Rebase II-TR, New Truliner-NT, Ufi Gel hard-UH) and 3 resilient reline materials (Trusoft-T; Sofreliner-S, Dentusil-D) was evaluated. MATERIAL AND METHODS: Thirty specimens of each material were made and divided into 3 groups: Control - not disinfected; P - daily disinfection by immersing in sodium perborate solution (3.8%); MW - microwave disinfection (6 min/650 W). Roughness measurements were made after polymerization (baseline) and after 1, 3 and 28 days. Roughness differences relative to the baseline readings were analyzed by Student's t-test (P=0.05). RESULTS: At baseline, Trusoft showed the highest (P<0.001) mean surface roughness (3.54 µm), and its surface roughness was significantly reduced after 28 days of disinfection by immersion in sodium perborate (P=0.013). Roughness measurements of material Trusoft were not performed after microwave disinfection due to the severe alterations on the surface. In the 3 groups evaluated, changes in roughness were significant for materials Ufi Gel hard (from 0.11 to 0.26 µm; P<0.041) and New Truliner (0.19 to 0.76 µm; P<0.019). The roughness of materials Lucitone 550 (0.37 µm), Tokuyama Rebase II (0.37 µm), Sofreliner (0.49 µm) and Dentusil (0.38 µm) remained unaffected (P>0.05). CONCLUSIONS: The roughness of the hard reline materials Ufi Gel hard and New Truliner was adversely affected by microwave disinfection, immersion in water or in sodium perborate. Microwave disinfection caused severe alterations on the surface of the resilient liner Trusoft.

A água pode influenciar as propriedades mecânicas das resinas acrílicas. Assim, o efeito do armazenamento em água na resistência ao impacto (RI) de uma resina para base de prótese (Lucitone 550-L) e quatro materiais reembasadores (Tokuyama Rebase II-T; UfiGel Hard-U; Kooliner-K; New Truliner-NT) foi avaliado. Barras da resina L foram confeccionadas (60 x 6 x 2 mm) e reembasadas (2 mm) com T, U, K, NT e L. Amostras não reembasadas de cada material também foram confeccionadas (60 x 6 x 4 mm) para fins comparativos. As amostras foram submetidas aos testes de impacto tipo Charpy sem armazenamento (controle) e após imersão em água por 7, 90 e 180 dias. Os resultados (kJ/m²) analisados por meio de ANOVA dois fatores e teste de Tukey (p=0,05) revelaram que, após 90 dias, U exibiu aumento na RI (0,93) comparado com 7 dias (0,58). K (1,48) e L/K (7,21) exibiram diminuição no período de 7 dias (1,01 e 3,23, respectivamente). NT (0,60) apresentou aumento na RI após 180 dias (1,52), enquanto L/NT (7,70) apresentou redução (3,17). A imersão em água melhorou a RI de U e NT e diminuiu a RI de K, L/K, e L/NT. A água pode afetar de maneira diversa a RI das resinas acrílicas e, consequentemente, a resistência à fratura das bases de próteses reembasadas.

Water may influence the mechanical properties of the acrylic resins. Thus, the effect of water storage on the impact strength (IS) of one denture base (Lucitone 550 - L) and four reline resins (Tokuyama Rebase II - T; UfiGel Hard - U; Kooliner - K; New Truliner - NT) was evaluated. Bars of L were made (60 x 6 x 2 mm) and relined (2 mm) with T, U, K, NT and L. Intact specimens of each material (60 x 6 x 4 mm) were also fabricated for comparative purposes. Specimens were submitted to Charpy impact tests without water storage (control) and after immersion in water for 7, 90 and 180 days. Data (kJ/m²) analyzed by two-way ANOVA and Tukey's test (p=0.05) revealed that after 90 days, U exhibited an increase in the IS (0.93) compared to 7 days (0.58). K (1.48) and L/K (7.21) exhibited a decrease at the 7-day period (1.01 and 3.23, respectively). NT (0.60) showed an increase in the IS after 180 days (1.52), whereas L/NT (7.70) showed a decrease (3.17). Water immersion improved the IS of U and NT, and decreased the IS of K, L/K, and L/NT. Water may affect differently the IS of acrylic resins and, consequently, the resistance to fracture of relined denture bases.

This study evaluated the effect of water-bath and microwave post-polymerization treatments on the flexural strength and Vickers hardness of four autopolymerizing reline resins (Duraliner II-D, Kooliner-K, Tokuso Rebase Fast-TR and Ufi Gel Hard C-UGH) and one heat-polymerized acrylic resin (Lucitone 550-L), processed using two polymerization cycles (short cycle - 90 minutes at 73ºC and 100ºC for 30 minutes; and long cycle - 9 hours at 71ºC). For each material, thirty specimens (64 x 10 x 3.3 mm) were made and divided into 3 groups (n=10). Specimens were tested after: processing (control group); water-bath at 55ºC for 10 minutes (reline materials) or 60 minutes (L); and microwave irradiation. Flexural strength tests were performed at a crosshead speed of 5 mm/min using a three-point bending device with a span of 50 mm. The flexural strengths values were calculated in MPa. One fragment of each specimen was submitted to Vickers hardness test. Data were analyzed by 2-way ANOVA followed by Tukey's HSD test (alpha=0.05). L microwaved specimens (short cycle) exhibited significantly higher flexural strength means than its respective control group (p<0.05). Water-bath promoted a significant increase (p<0.05) in flexural strength of K and L (long cycle). The hardness of the tested materials was not influenced by the post-polymerization treatments. Post-polymerization treatments could be used to improve the flexural strength of some materials tested.

INTRODUÇÃO: A maioria das resinas acrílicas utilizadas para confecção de bases de próteses é composta pelo polimetacilato de metila. Muitos autores têm discutido o processo de polimerização dessas resinas em relação à conversão do monômero em polímero devido a sua importância na melhora da biocompatibilidade e das propriedades físicas. Para assegurar a utilização desses materiais, testes preliminares de citotoxicidade in vitro têm sido desenvolvidos para avaliação da biocompatibilidade. Um dos ensaios biológicos sugeridos para a análise da citotoxicidade é o teste de incorporação de ³H-timidina, o qual mede o número de células por meio da síntese de DNA. OBJETIVO: O objetivo do presente estudo foi avaliar, por meio do teste de incorporação de ³H-timidina, a citotoxicidade de duas resinas acrílicas para base de próteses submetidas aos tratamentos em microondas e em banho de água após a polimerização. MATERIAL E MÉTODO: Nove corpos-de-prova em forma de discos (10 x 1 mm) foram confeccionados com as resinas acrílicas Lucitone 550 e QC 20 de acordo com as instruções dos fabricantes, e foram armazenados em água destilada a 37ºC por 48 h. Os corpos-de-prova foram divididos em três grupos: 1) tratamento em forno de microondas por 3 min a 500 W; 2) tratamento em banho de água a 55ºC por 60 min; e 3) sem tratamento térmico. Extratos foram preparados pela colocação de 3 discos em tubos de ensaio estéreis com 9 mL de meio de cultura Eagle e incubação a 37ºC por 24 h. O efeito citotóxico dos extratos foi avaliado utilizando o teste de incorporação de ³H-timidina. RESULTADOS: Os resultados indicaram que os componentes liberados pelas resinas foram citotóxicos para as células L929 exceto para as amostras tratadas em banho de água (p<0.05). Em comparação com o grupo sem tratamento, o banho de água diminuiu a citotoxicidade das resinas acrílicas. CONCLUSÃO: O tratamento em microondas não influenciou a citotoxicidade das resinas acrílicas para bases de próteses.

INTRODUCTION: Most denture base acrylic resins have polymethylmethacrylate in their composition. Several authors have discussed the polymerization process involved in converting monomer into polymer because adequate polymerization is a crucial factor in optimizing the physical properties and biocompatibility of denture base acrylic resins. To ensure the safety of these materials, in vitro cytotoxicity assays have been developed as preliminary screening tests to evaluate material biocompatibility. ³H-thymidine incorporation test, which measures the number of cells synthesizing DNA, is one of the biological assays suggested for cytotoxicity testing. AIM: The purpose of this study was to investigate, using ³H-thymidine incorporation test, the effect of microwave and water-bath post-polymerization heat treatments on the cytotoxicity of two denture base acrylic resins. MATERIALS AND METHODS: Nine disc-shaped specimens (10 x 1 mm) of each denture base resin (Lucitone 550 and QC 20) were prepared according to the manufacturers' recommendations and stored in distilled water at 37ºC for 48 h. The specimens were assigned to 3 groups: 1) post-polymerization in a microwave oven for 3 min at 500 W; 2) post-polymerization in water-bath at 55º C for 60 min; and 3) without post-polymerization. For preparation of eluates, 3 discs were placed into a sterile glass vial with 9 mL of Eagle's medium and incubated at 37ºC for 24 h. The cytotoxic effect of the eluates was evaluated by ³H-thymidine incorporation. RESULTS: The results showed that the components leached from the resins were cytotoxic to L929 cells, except for the specimens heat treated in water bath (p<0.05). Compared to the group with no heat treatment, water-bath decreased the cytotoxicity of the denture base acrylic resins. CONCLUSION: The in vitro cytotoxicity of the tested denture base materials was not influenced by microwave post-polymerization heat treatment.

INTRODUÇÃO: A dureza das resinas para base de prótese e para reembasamento imediato pode apresentar alterações devido à polimerização continuada e absorção de água. Entretanto, a magnitude do efeito de cada um desses processos ainda não foi definida. OBJETIVO: Neste estudo, o grau de conversão de duas resinas autopolimerizáveis para reembasamento (Duraliner II-D and Kooliner-K) e de uma resina termopolimerizável para base de prótese (Lucitone 550-L) foi avaliado, indiretamente, por meio da mensuração da dureza. O efeito da imersão em água sobre essa propriedade também foi analisado. MATERIAL E MÉTODOS: Após a polimerização, amostras (diâmetro - 5 mm; espessura - 2 mm) foram armazenadas a seco em temperatura ambiente e a dureza Vickers (VHN) foi mensurada após 0, 2, 7, 30 e 90 dias. As amostras foram, então, imersas em água a 37º C e a dureza foi avaliada nos períodos citados. Cinco amostras foram preparadas para cada material. Os resultados foram analisados utilizando-se o teste de Kruskal-Wallis (P=.01). RESULTADOS: Para o armazenamento a seco, o material L apresentou aumento significativo na dureza (P<.01) de 0 (VHN=23.2) para 30 dias (VHN=27.1), após o qual nenhuma diferença significativa foi observada. Aumento contínuo na dureza foi observado para o material D de 0 (VHN=4.6) para 90-dias (VHN=7.3). Para o material K, aumento na dureza foi observado até 7 dias (VHN=9.2) quando ocorreu a estabilização (P>.01). Após 2 dias de armazenamento em água, todos os materiais apresentaram redução significativa na dureza (P<.01). CONCLUSÕES: A dureza dos materiais avaliados aumentou durante o armazenamento a seco e diminuiu após a imersão em água.

INTRODUCTION: The hardness of denture base materials may undergo changes due to continued polymerization reaction and water uptake. However, the extent to which these processes affect the hardness of materials is still unclear. OBJECTIVE: In this study, the degree of conversion of two hard chair-side reline resins (Duraliner II-D and Kooliner-K) and one heat-cured acrylic resin (Lucitone 550-L) was evaluated indirectly by measuring the surface hardness. The effect of immersion in water on this property was also analyzed. MATERIALS AND METHODS: After processing following the manufacturers' instructions, specimens (5mm diameter and 2mm thickness) were dry stored at room temperature and the Vickers hardness (VHN) was measured with a hardness tester after 0, 2, 7, 30 and 90 days. Specimens were then immersed in water at 37ºC and hardness was evaluated after the same time intervals. Five specimens were prepared for each material. Data were analyzed by Kruskal-Wallis test (P=.01). RESULTS: When dry stored, material L showed an increase in hardness (P<.01) from 0-day (VHN=23.2) to 30-day (VHN=27.1), after which no significant change was observed. A continuous increase in hardness was observed for material D from 0-day (VHN=4.6) to 90-day dry storage (VHN=7.3). For material K, an increase in hardness was observed up to 7-day (VHN=9.2), and then leveled off (P>.01). After 2-day water storage, all materials showed a significant reduction in hardness (P<.01). CONCLUSION: In general, the hardness of the materials evaluated increased during dry storage and decreased after immersion in water.
