Abstract Samanea tubulosa Benth. it has been widely used in traditional medicine to treat inflammatory processes. The present study aimed to investigate the antinociceptive effect and mechanism of action of the fractions obtained from the Samanea tubulosa pods in mice. The antinociceptive activity was evaluated in formalin, capsaicin and glutamate tests and the. The possible mechanisms of action involved in the antinociceptive effect of the hexane and ethyl acetate fraction in the opioid system, also the the K + ATP channels and the L-arigine pathways of nitric oxide were evaluated. The chemical characterization analysis revealed in the hexane fraction the presence of triterpenes such as lupenone and lupeol. In the glutamate test, the hexane and ethyl acetate fractions showed antinociceptive activity at the dose of 12.5 and 25 mg kg-1. The antinociception produced by the hexane and ethyl acetate fractions was significantly reversed by naloxone, indicating that the fractions act through the opioid pathway. Antinociceptive response of the ethyl acetate fraction was blocked by glibenclamide, indicating that this fraction acts via the K + ATP channels activation. It is concluded that the fractions under study exert antinociceptive activity possibly related to the opioid route and through K+ ATP channels activation.

ABSTRACT Infrared thermography (IR) is a non-invasive tool with potential to indicate changes in the animal’s thermal conditions in response to the thermally stressful environment. The objective of this study was to evaluate the application of IR to estimate the rectal temperature of crossbred goats of the Boer breed. Six male crossbred goats of the Boer breed were distributed in a completely randomized design and submitted to temperatures of 26, 30 and 34 °C. Rectal temperature (RT) and thermograms data were collected from animals at each air temperature evaluated. In the thermograms, the temperatures of the ocular globe (PT), head (HT), shoulder (ST), hindquarter (HQ) and maximum infrared (IRMax) of the animals’ surfaces were collected, the latter being observed in the lower region of the animals’ eyes, at all air temperatures evaluated. The correlation of PT, HT, ST, HQ and IRMax data with the RT was evaluated through the Pearson coefficient analysis and the concordance using Bland-Altman diagrams. With the exception of the IRMax surface temperature, the others were adequate for the accurate estimation of RT, with PT standing out for presenting the highest correlation coefficient with RT (r = 0.951) and estimation errors varying in the range of ± 0.27 °C.

Currently, the pharmaceutical industry devotes great attention to drug degradation products because these compounds can offer risks to patients. A previous degradation study of betahistine (N-α-methyl-2-pyridylethylamine) conducted under different stress conditions detected three main impurities named A, B and C. Degradation products were analyzed by high-resolution mass spectrometry in electrospray source and time of flight analyzer (ESI-TOF) and nuclear magnetic resonance (NMR). Impurity mutagenicity was evaluated by Derek Nexus and Sarah Nexus softwares. Liquid chromatography hyphenate with tandem mass spectrometry (LC-MS/MS) analysis of the betahistine forced degradation sample indicated the presence of a new impurity, which was named impurity C1. 2D NMR experiments allowed the complete structural characterization of the new entity. The active pharmaceutical ingredient and degradation impurities were classified as inactive in the in silico mutagenic studies. Systematic investigation of a forced degradation sample led to the characterization of a new betahistine impurity. The in silico mutagenicity study of the betahistine degradation impurities may be useful in the risk assessment of the drug products.

ABSTRACT The aim is to characterize and to compare the black (TBG) and gray (TGG) globe temperatures in the production of sheep in the Brazilian semi-arid, using Santa Inês and Doper breeds, distributed in two thermal environments (exposed to the sun and shaded), containing 10 animals in each picket. The experimental design was completely random, 2 x 2 factorial scheme (shaded environment and sun x black and gray globe) in 46 replicates (evaluation days). There was an increase of the TBG in relation to the TGG during the evaluation times, with maximum temperature (48.1°C) at 1p.m. in the environment exposed to the sun, concomitantly with elevation of the black globe temperature and humidity index - BGHI (96.7). The result was distinct for TGG, with a maximum temperature of 37.6°C and BGHI of 86.2 in the same environment. Regardless of the thermometer used, the BGHI values exceeded the considerable comfortable limit for sheep. Under shaded conditions, the globe temperatures showed similar behavior. In sunny conditions, these temperatures diverged, higher in the black, because of the absorption conditions of the thermal radiation of the different colors.

RESUMO Os caprinos são animais homeotérmicos considerados rústicos do ponto de vista bioclimático. Porém, em condições térmicas desfavoráveis, pode ocorrer a redução de sua eficiência produtiva. Realizou-se o presente estudo com o objetivo de avaliar a dilatação pupilar como indicador do estresse térmico em caprinos mestiços Boer, mantidos em condições ambientais controladas. Seis caprinos machos mestiços da raça Boer foram distribuídos em delineamento inteiramente casualizado e submetidos às temperaturas de 26, 29 e 33 °C. Foram monitoradas a dilatação e temperatura pupilar, frequência respiratória, frequência cardíaca, temperatura superficial e retal. Ocorreu a elevação nas respostas fisiológicas em razão do aumento da temperatura do ar. Constatou-se aumento da dilatação pupilar dos animais com a elevação da temperatura do ar, observando-se um acréscimo de 26,96 mm2 de área pupilar quando comparadas às temperaturas de 26 e 33 °C, sendo possível estabelecer uma correlação entre temperatura do ar e aumento do diâmetro pupilar chamado de razão de estresse pupilar (REP).
ABSTRACT Goats are homeothermic animals considered as rustic from the bioclimatic point of view, but the reduction of their productive efficiency may occur in unfavorable thermal conditions. The present study was carried out to evaluate pupil dilation as an indicator of thermal stress in Boer crossbred goats maintained under controlled environmental conditions. Six male crossbred goats of the Boer breed were distributed in a completely randomized design submitted to temperatures of 26, 29 and 33 °C. The pupillary dilation and pupillary temperature, respiratory frequency, heart rate, surface and rectal temperatures were monitored. The significant increase in physiological responses occurred in response to the increase in air temperature. An increase of pupillary dilation was observed in the animals with the elevation of the air temperature, with an increase of 26.96 mm2 in pupillary area when comparing the temperatures 26 and 33 °C, and it was possible to establish a correlation between air temperature and pupillary diameter increase, named pupillary stress ratio (PSR).

A cana-de-açúcar pode ser cultivada como matéria-prima para a produção de cachaça artesanal e forragem para alimentação animal, porém normalmente não se utiliza um bom aporte tecnológico, pois são plantadas variedades usadas para produção de açúcar e álcool. Este trabalho teve como objetivo avaliar o desempenho agroindustrial e a estimativa de parâmetros fenotípicos em variedades de cana-de-açúcar avaliadas em duas safras. O delineamento experimental utilizado foi o látice triplo 4 x 4, sendo avaliadas dezasseis variedades de três diferentes instituições. Foram avaliados caracteres agronómicos e tecnológicos com importância agroindustrial. Houve diferença significativa entre as variedades e as safras para os caracteres avaliados na análise conjunta. A análise dos parâmetros fenotípicos contribuiu para o entendimento do desempenho dos caracteres ao longo das duas safras. As variedades possuem comportamento dessemelhante, porém, apresentam regularidade no desempenho, ao longo do tempo, para a maioria dos caracteres agronómicos e industriais avaliados. As características fenotípicas POL e ATR, denotam boa confiabilidade em prever o valor genotípico para estas variáveis. O alto número de medições necessárias para os caracteres TCH, PUREZA e MMC, dificultam a seleção de variedades potenciais. A correlação fenotípica avaliada em duas safras não é suficiente para realizar a recomendação de variedades.
Sugarcane can be cultivated as raw material for craft cachaça production and forage for animal feed, but usually a good technological input is not used, because varieties for sugar and ethanol production are planted. The aim of this study was to evaluate the agroindustrial performance and the estimation of phenotypic parameters of sugarcane varieties of the first and second crop seasons. The experimental design was a triple lattice 4 x 4, being assessed sixteen varieties of three different institutions. Agronomic and technological characters with agroindustrial importance were evaluated. There was a significant difference between the varieties and the crop seasons for the traits evaluated in the joint analysis. The phenotypic parameters analysis contributed to the understanding of the traits performance throughout two crop seasons. Varieties do not present a similar behavior, however, they have regularity in the performance, over time, for most agronomic and industrial characters evaluated. Phenotypic characteristics POL and TRS, show good reliability in predicting the genotypic value for these variables. The high number of measurements required for the TSH, Purity and SM characters make it difficult to select potential varieties. Phenotypic correlation evaluated in two harvests is not sufficient to carry out the recommendation of varieties.

Lupeol, a triterpene frequently found in Asteraceae plant species, showed moderate to low activity in different strains of Plasmodium falciparum, the most virulent malaria etiological agents. In this work, lupeol was isolated from Parahancornia fasciculata, a plant that is used to treat malaria in the Amazonia region. In the search of more activity lupeol derivatives, five new 1,2,3-triazole hybrid molecules were synthetized by copper-catalyzed azide-alkyne cycloaddition. The antiplasmodial activity of the semi-synthetic compounds were evaluated by the lactate dehydrogenase assay; the lupeol propargyl ether was the only one to disclosing increased activity (half maximal inhibitory concentration-IC50-62.0 ± 1.92 µmol L-1) in relation to lupeol (IC50 117.00 µmol L-1). Therefore, this work revealed a new class of interesting lupeol derivatives that can be obtained by linking electron donors to the hydroxy group at C-3.

ABSTRACT With the demand in the production at large-scale food, confinement of animals has become a necessity of the productive process because of the increase in production capacity and optimization of the spaces reserved for creations. In this context, the aim of this study was the development and validation of models using fuzzy logic for predicting climate indices and productive performance of European quails kept in a climatic chamber. The model developed was analyzed from two points of view; the first one took into account the prediction of climate indexes where the input variables were the temperature (°C) and relative humidity (%) of the air. In the second, related to the prediction of productive performance, the input variables were air temperature (°C) and age of the birds (weeks) while the output variables were the food intake (FI, g), water consumption (WC, g), weight gain (WG, g) and food conversion (FC g g-1) of the birds. The Mamdani method was used for the preparation of the rules, and in the defuzzification was applied the center of gravity method. Based on the results generated by the models and compared with the experimental data it was obtained coefficients of determination (R2) of: 0.9771; 0.9897; 0.9955; 0.9995; 0.9993 and 0.9788, for BGTHI, RTL, WC, FI, WG and FC, respectively.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Intraspecific variability of Holostylis reniformis: concentration of lignans, as determined by maceration and supercritical fluid extraction (SFE-CO2), as a function of plant provenance and plant parts

Maceration and supercritical fluid extraction were used to prepare extracts from parts of plants (Holostylis reniformis) collected in two different regions of Brazil. ¹H NMR, HPLC-DAD-ESI/MS, HPLC-DAD, GC-MS, and chemometric techniques were used to analyse lignans in the extracts and showed that yields of SFE-CO2 were less than or equal to those of hexane maceration extracts. These analyses, in conjunction with the concentrations of aliphatic hydrocarbons, fatty acids and their methyl and ethyl derivatives in the extracts, also allowed the chemical composition of parts and provenance of the plant to be differentiated.

Several Lippia species have been used in folk medicine mainly for gastrointestinal and respiratory diseases. Their biological properties have been partially associated to the terpenoids found in their essential oils. According to the World Health Organization, cancer is the leading cause of death worldwide and is described as a complex group of diseases with several hallmarks. One of its acceptable defining features is the cell proliferation beyond their boundaries forming the tumors. Importantly, some drugs currently available were discovered by the investigation of plant secondary metabolites. Thus, this study aimed to evaluate in vitro cytotoxic effect of the essential oils extracted from five Lippia species against tumor cell lines. The results indicated that mouse colon carcinoma CT26.WT cell line viability was significantly reduced showing an IC50 of 19.05, 30.20 and 36.30 µg/ml when treated with the essential oils of L. sidoides, L. salviifolia and L. rotundifolia, respectively. Human lung carcinoma A549 cell line also had a compromised viability to the action of L. alba carvone chemotype essential oil. The tested essential oils did not compromise viability of the normal cell line CHO. These finds suggest that the studied Lippia essential oils might be good candidates for further in-depth studies.

The genus Eremanthus is recognized by the predominance of sesquiterpene lactones from the furanoheliangolide type, a class of substances extensively tested against cancer cell lines. Thus, the species E. crotonoides (DC.) Sch. Bip., Asteraceae, obtained on "restinga" vegetation was evaluated against U251 and U87-MG glioma cell lines using the MTT colorimetric assay. Dichloromethane fraction was cytotoxic to both glioblastoma multiforme cell lines. We then conducted UPLC-PDA-ESI-MS/MS analysis of the dichloromethane fraction, which allowed the identification of the sesquiterpene lactones centratherin and goyazensolide. The isolation of centratherin was performed using chromatographic techniques and the identification of this substance was confirmed according to NMR data. Cytotoxic activity of centratherin alone was also evaluated against both U251 and U87-MG cells, which showed IC50 values comparable with those obtained for the commercial anticancer drug doxorubicin. All the tested samples showed cytotoxic activity against glioblastoma multiforme cells which suggests that E. crotonoides extracts may be important sources of antiproliferative substances and that the centratherin may serve as prototype for developing new antiglioblastoma drugs.

This study aimed to evaluate the safety of the hydroalcoholic extract (HE) of Syzygium cumini (L.) Skeels, Myrtaceae, leaves in rodents. Acute toxicity was evaluated through the determination of a LD50 in mice and rats (up to 14 days). In mice, the oral administration (p.o.) of the HE (0.1 at 6 g/kg) did not cause any death. When administered by intraperitoneal route (i.p.) the HE (0.1 at 1 g/kg) caused death of the animals (LD50 of 0.489 g/kg). In rats, the HE (0.5, 1 and 2 g/kg, p.o.) did not cause any death, while by i.p., only the 2 g/kg dose was lethal to 67% of the animals. To evaluate chronic toxicity, groups of rats daily received the HE (0.05, 0.1 and 0.25 g/kg) through p.o., during 30, 90 or 180 days and the effects on behavior, body weight, feed consumed were measured. Histology, hematology and biochemical parameters were measured at the end of the treatment. After a 30-day treatment, the HE caused changes in some biochemical parameters. Histological examination of the liver, kidneys, lungs, heart, stomach, intestine and pancreas showed normal architecture suggesting no morphological disturbances. These data may mean that the HE of S. cumini does not exert acute or chronic toxic effects by oral administration.

The safety of babassu mesocarp (Orbignya phalerata Mart., Arecaceae), which exhibited anti-inflammatory and antithrombotic activities, was evaluated by determining the potential acute toxicity in mice. A lyophilized ethanol extract of babassu mesocarp (BME) was administered to C3H/HePas mice (10/group) in a single dose of 1000, 3000 and 5000 mg/kg, by gavage. General behavior adverse effects and mortality were determined for up to fourteen days. Selected biochemical parameters including glucose, triacylglyceride, cholesterol, urea, alkaline phosphatase and creatinine were determined by colorimetric assay. The heart, liver, spleen, kidneys and brain were weighted and evaluated macro and microscopically. The median lethal dose (LD50) of BME was greater than 5000 mg/kg. No behavior or body weight alterations were detected after the treatment. The acute treatment with BME has no effect on macroscopic and microscopic aspect of examined organs. Instead, BME increased the alkaline phosphatase and reduced the urea concentration in all groups. A significant increase on triacylglyceride was detected in the group BME1000. In conclusion, the acute treatment with high doses of BME can affect some biochemical parameters with a long lasting effect, although any change was detected at tissue level or body and organ weight.

Nos compósitos cimentíceos as fibras vegetais podem sofrer vários graus de degradação quando expostas ao ambiente alcalino. O meio alcalino degrada os componentes macromoleculares das fibras, principalmente pela solubilização da lignina e a desfibrilação da celulose, comprometendo suas propriedades físicas e mecânicas. A resistência das fibras vegetais ao ataque alcalino pode ser aumentada por meio de tratamentos químicos superficiais como a acetilação que apresenta capacidade de reduzir a natureza higroscópica de compostos lignocelulósicos pela introdução de grupos funcionais hidrofóbicos em sua estrutura química. Objetivou-se, então neste trabalho, a aplicação do tratamento de acetilação em diferentes níveis, visando melhorias na durabilidade das fibras. O tratamento de acetilação nas fibras de curauá foi aplicado em diferentes temperaturas e tempos reacionais e sua eficiência, considerando-se a redução da hidrofilicidade e a manutenção do desempenho físico-mecânico das fibras, foi avaliada pelos ensaios de absorção de água por imersão, resistência mecânica, espectroscopia de infravermelho, microscopia eletrônica de varredura, análise termogravimétrica e difratometria de raios-x. Quando acetiladas, as fibras de curauá apresentaram absorções até 55% menores que nas fibras não tratadas. As perdas na resistência mecânica foram próximas aos 52%. O tratamento de acetilação agregou grupos químicos hidrofóbicos na composição e alterou o caráter cristalino das fibras.
In cementitious composite the vegetable fibers may suffer various degrees of degradation when exposed to alkaline medium. The alkaline degradation of macromolecular components of fibers mainly by lignin solubilization and cellulose defibrillation, compromising the physical and mechanical properties. The resistance of plant fiber to alkali attack can be improved by chemical surface treatments such as acetylation that can reduce the hygroscopic nature of lignocellulosic introducing hydrophobic functional groups in its chemical structure. Thus, the aim of this study was the application of the acetylation treatment at different levels to improve the durability of the fibers.The acetylation treatment in curauá fiber was applied at different temperatures and reaction times, and its efficiency, considering the reduction of hydrophilicity and maintenance of the physical-mechanical performance of fibers was evaluated by water absorption, mechanical resistance, infrared spectroscopy, scanning electron microscopy, thermogravimetric analysis and diffraction of x-rays. When acetylated curauá fibers absorbed up to 55% less water than untreated fibers. In strength, losses were up 52%. The acetylation treatment introduced hydrophobic chemical groups in the composition and changed crystalline character of fibers.
