Immunostimulating activity of Uncaria tomentosa in RAW 264.7 macrophages

Abstract Uncaria tomentosa is a plant native to the Amazon that has immunomodulatory and antitumor properties due to the alkaloids found in the plant, being able to modify the immune response by potentiating or suspending the action of cytokines secreted by macrophages that induce the immune response, either by the classical route (M1) or through the alternative route (M2). Macrophages activated by M1 convert L-arginine into L-citrulline and nitric oxide (NO), whereas macrophages activated by the M2 pathway use the enzymatic activity of arginase to convert the same substrate into L-ornithine and urea. The aim of this work was to evaluate the immunostimulating activity of the crude hydroalcoholic extract from the bark of the U. tomentosa stem in RAW 264.7 macrophages. Concentrations of 0.2, 0.1 and 0.05 mg/mL of U. tomentosa extract associated with LPS, INF-γ and IL-4 inducers were tested by determining NO production and arginase enzyme activity. Nitric oxide production was enhanced by the extract when associated with LPS and LPS + INF-γ inducers. In the activity of the arginase enzyme, the extract decreased the stimulation of IL-4 on the enzyme, mainly at 0.2 mg/mL concentration. Therefore, it is concluded that the crude hydroalcoholic extract of the stem bark of U. tomentosa in RAW 264.7 cells, at a concentration of 0.2 mg/mL, showed considerable pro-inflammatory activity.
Resumo A Uncaria tomentosa é uma planta nativa da Amazônia que possui propriedades imunomoduladoras e antitumorais devido aos alcaloides encontrados no vegetal, sendo capaz de modificar a resposta imune ao potencializar ou suspender a ação de citocinas secretadas por macrófagos que induzem a resposta imune, seja pela via clássica (M1) ou pela via alternativa (M2). Macrófagos ativados pela M1 converte L-arginina em L-citrulina e óxido nítrico (NO), já os macrófagos ativados pela via M2 utilizam a atividade enzimática da arginase para converter o mesmo substrato em L-ornitina e ureia. O trabalho teve como objetivo avaliar a atividade imunoestimuladora do extrato hidroalcoólico bruto da casca do caule da U. tomentosa em macrófagos RAW 264.7. Foram testadas as concentrações 0.2, 0.1 e 0.05 mg/mL do extrato de U. tomentosa associadas aos indutores LPS, INF-γ e IL-4, por meio da determinação da produção de NO e atividade da enzima arginase. A produção de óxido nítrico foi potencializada pelo extrato quando associado aos indutores LPS e LPS + INF-γ. Na atividade da enzima arginase, o extrato diminuiu a estimulação da IL-4 sobre a enzima, principalmente na concentração 0.2 mg/mL. Portanto, conclui-se que o extrato hidroalcoólico bruto da casca do caule de U. tomentosa em células RAW 264.7, na concentração 0.2 mg/mL, demostrou atividade pró-inflamatória considerável.

Fish diversity in the rivers that drain the Baixada Maranhense and the Mearim basin in northeastern Brazil

Abstract The Baixada Maranhense Environmental Protection Area of the Brazilian state of Maranhão encompasses a biologically rich region that includes the basins of four principal rivers, the Mearim, Pericumã, Pindaré, and Turiaçu, which form a complex of wetlands. The present study provides a comprehensive inventory of the fish fauna of the rivers that drain the Baixada Maranhense based on the identification of voucher specimens that were deposited in a scientific collection. Expeditions were conducted between 2014 and 2020. The inventory identified a total of 100 fish species representing 79 genera, 34 families, and 11 orders, which are found in both freshwater and estuarine environments. Six (Potamotrygon orbignyi, Leporinus piau, Moenkhausia loweae, Serrasalmus spilopleura, Pachypops fourcroi and Peckoltia greedoi) of the 100 taxa identified here are considered to be new records for one or more of the basins surveyed during this study, based on the existing data, while four – Colossoma macropomum, Megaleporinus macrocephalus, Cichla sp. and Cichla kelberi – are not native. The most speciose orders were the Siluriformes and Characiformes, while the most diverse families were the Loricariidae, Characidae, Auchenipteridae, Cichlidae, and Serrasalmidae. These data are consistent with the findings of most of the previous studies of freshwater ecosystems in the Neotropical region, in particular those of the rivers of Maranhão. The results of the present study represent an important advance in the understanding of the diversity of the fish fauna of the Mearim, Pericumã, Pindaré, and Turiaçu rivers, which are still poorly studied, and have likely had their diversity underestimated up to now.
Resumo A Área de Proteção Ambiental da Baixada Maranhense do estado brasileiro do Maranhão abrange uma região biologicamente rica que engloba as bacias de quatro rios principais, Mearim, Pericumã, Pindaré e Turiaçu, formando um complexo de áreas inundáveis. O presente estudo fornece um inventário abrangente da ictiofauna de rios que drenam a Baixada Maranhense a partir da identificação de espécimes com material testemunho depositados em coleção científica. As expedições foram realizadas entre 2014 e 2020. O inventário identificou um total de 100 espécies de peixes representando 79 gêneros, 34 famílias e 11 ordens que são encontradas em ambientes de água doce e estuarinos. Dos 100 táxons identificados, seis (Potamotrygon orbignyi, Leporinus piau, Moenkhausia loweae, Serrasalmus spilopleura, Pachypops fourcroi e Peckoltia greedoi) são considerados novo registros para uma ou mais das drenagens aqui levantadas, em comparação com os dados existentes, enquanto quatro: Colossoma macropomum, Megaleporinus macrocephalus, Cichla sp. e Cichla kelberi não são nativos. As ordens mais especiosas foram Siluriformes e Characiformes, enquanto as famílias mais diversas foram Loricariidae, Characidae, Auchenipteridae, Cichlidae e Serrasalmidae. Esses dados corroboram a maioria dos outros estudos de ecossistemas de água doce da região Neotropical, em particular o dos rios do Maranhão. Os resultados do presente estudo representam um avanço importante no conhecimento da diversidade da fauna de peixes dos rios Mearim, Pericumã, Pindaré e Turiaçu, que ainda são pouco estudados e provavelmente com uma diversidade subestimada.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

During the pneumatic conveying of plastic pellets, it has been observed that materials with similar physical characteristics may develop a substantial difference in pressure drop. In this work, the pressure drop in a particle-laden 2.7 meter long horizontal channel with circular cross-section is presented from an experimental perspective. Experiments are carried out for cylindrical polystyrene beads with an average diameter of 3.2 mm and mass loadings of 0.06 to 0.11 (kg particles/kg gas). The air mass flow rate was studied in the range from 0.085 kg/s to 0.170 kg/s. The pressure drop curve is shown as a function of air velocity and particle load. Response surface methodology showed high statistical significance for air velocity, particle load and their cross-relation.

An undergraduate physical chemistry experiment based on the drop counting method for surface tension measurements is proposed to demonstrate adsorption isotherms of binary aqueous solutions of ethanol, n-propanol, and n-butanol. Excess surface is obtained by the derivative of surface tension taken with respect to alcohol activity, after this activity calculation using van Laar equation. Laboratory class contents are surface tension, excess surface, percolation of hydrogen bonds, micelle, activity, and ideal solution.

A pigmentação de olhos pretos (forma selvagem) e vermelhos (forma mutante) de Triatoma infestans foi estudada por espectrofotometria e comparada à de olhos vermelhos (selvagem) e brancos (mutante) de Drosophila melanogaster. Os perfis do espectro de absorção dos pigmentos de olho preto e vermelho de T. infestans foram semelhantes entre si e ao dos olhos de tipo selvagem de D. melanogaster. A similaridade com a forma selvagem de D. melanogaster indicou que ambos os tipos de olho de T. infestans continham omocromos do tipo xantomatina, o que foi confirmado por cromatografia ascendente em papel. Não foram detectadas pteridinas, melaninas e ominas como pigmentos de olho em T. infestans. A diferença na cor de olho em T. infestans foi considerada uma função da concentração de xantomatina, sendo menor o conteúdo de omocromo nos olhos vermelhos, embora isso provavelmente não afete a acuidade visual do inseto. Esses resultados estão de acordo com dados de outros autores quanto a semelhanças envolvendo outras características entre espécimes de olho preto e vermelho de T. infestans.
The pigmentation of black (wild) and red (mutant) eyes of Triatoma infestans was studied spectrophotometrically and compared with red-eyed (wild) and white-eyed (mutant) forms of Drosophila melanogaster. The spectral absorption profiles of the black and red eye pigments of T. infestans were similar to each other and to that of the wild-type eyes of D. melanogaster. The similarity to the wild form of D. melanogaster indicated that both eye forms of T. infestans contained ommochromes of the xanthommatin type, a finding confirmed by ascending paper chromatography. Pteridines, melanins, and ommins were not detected as eye pigments in T. infestans. The eye color difference in T. infestans was assumed to be a function of the xanthommatin concentration, with a smaller content of ommochrome in red eyes, although this probably did not affect the insect's visual acuity. These data support other findings regarding the similarities between black- and red-eyed specimens of T. infestans for other characteristics.

Preliminary notes on yeasts associated with necrotic cactus stems from different localities in Brazil

As leveduras encontradas em cladódios necrosados de três cactos colunares (Pilosocereus machrisii, Pilosocereus vilaboensis and Praecereus euchlorus) em oito localidades no Brasil foram identificadas e uma análise de similaridade baseada na distância Sorensen foi usada para comparar as comunidades nas diferentes localidades. Dos 56 cladódios necrosados amostrados, 32 produziram colônias de leveduras. Dez espécies de leveduras foram identificadas a partir de 53 isolados, com as espécies Pichia cactophila, Candida sonorensis, Geotrichum sp. e Sporopachydermia cereana sendo as mais comuns. As espécies restantes ocorreram em baixas freqüências. O dendograma (UPGMA) construído a partir da análise de similaridade agrupou comunidades de leveduras obtidas em diferentes espécies de cactos e indicou que as espécies de cactos hospedeiros não foram determinantes nesse agrupamento.
The yeast species found in necrotic stems of three columnar cacti (Pilosocereus machrisii, Pilosocereus vilaboensis, and Praecereus euchlorus)at eight localities in Brazil were described and a similarity analysis using Sorensen distances was used to compare the composition of yeast species at these localities. Of 56 necrotic cactus stems sampled,32 produced yeast colonies. Ten species of yeast or yeast-like microorganisms were identified from 53 isolates, with Pichia cactophila, Candida sonorensis, Geotrichum sp., and Sporopachydermia cereana being the most common. The remaining species occurred in low proportions in the cacti surveyed. The similarity analysis provided a dendogram (UPGMA) that clustered the yeast communities from different cactus species and indicated that host cactus species was unimportant in this clustering.

In this paper data on catch and effort by gear are analysed, for weakfish (Macrodon ancylodon) landed at Santos from January 1959 to December 1963. The region studied was a narrow band about 30 nautical miles wide, along the southern coast of Brazil, between 23º S and 34º S. From the analysis of monthly data of production per unit-effort by squares of 1º on a side, we divided the studied region in two areas: area I (from 23º S to 27º S) and area II (from 28º S to 34º S). The index of unit-effort production and the effort concentration index were computed grouping the data by quarters. From the results we may drow the following conclusions: 1 - In a general way, the effort for catch weak-fish has been applied satisfactorily because the medium and large parejas have operated almost in areas with average density on¹ higher than average density; 2 - The production per unit-effort in the area I is smaller than the production in area II, which suggests that the index of abundance is higher in area II; 3 - The decrease in production per unit-effort, in area II, suggests a decrease in the abundance or availability of weak-fish in this area; 4 - In the area I, the index of production per unit-effort remained approximately at the same level.
