Abstract Studies about the viability and vigor of seeds during storage, especially on the characterization of enzymatic changes, may increase the harvesting efficiency of immature seeds and guarantee their viability for a prolonged period. Therefore, this study aimed to evaluate the behavior of immature seeds of Tabebuia caraiba during storage. For this, thirty selected mother plants at the anthesis stage located in São João do Cariri and Sumé, Paraíba, Brazil were selected. Flowers of each tree were marked, and the fruits were harvested at 35, 42, and 49 days after anthesis (DAA) and characterized according to their maturation stages. The seeds from each maturation stage and city were packed in paper bags and stored in a refrigerator (6 ± 2 °C) for 360 days. The seed quality, seedling vigor, and enzymatic activity of superoxide dismutase (SOD), catalase (CAT), and peroxidase (POX) were evaluated at the beginning of the storage period and intervals of 90 days. The seeds of T. caraiba harvested at 35, 42, and 49 DAA maintained high viability for 360 days of storage. T. caraiba seeds stored for 360 days withstood with high vigor when harvested at 42 DAA. The CAT, SOD, and POX activity was correlated to the deterioration process of the seeds under storage conditions.

Abstract The purpose of this study was to isolate Toxoplasma gondii from tissues of free-range chickens in the southwestern region of Goiás, to detect and molecularly characterize the genetic material of the parasite, and to determine the seroprevalence of the protozoan parasite in these animals. A seroprevalence of T. gondii antibodies of 76% (19/25) was found among the chickens, while genetic material from their tissues was detected in 56% (14/25). A total of 14 isolates was obtained in the bioassay, ten of which were considered acute, eight were considered isolates of high virulence lethal to mice, and four of low virulence, considered non-lethal but with the ability to chronify the infection. Seven of the ten isolates showed significant morphometric differences from the RH strain, in terms of nucleus-complex-apical distance, length and width. Genotyping of the acute isolates was performed by RFLP-PCR, using 11 genetic markers: SAG1, SAG2 (3’SAG2 and 5’SAG2), alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and APICO. The results were compared and classified according to the genotypes listed on the ToxoDB Platform, where different profiles were observed indicating the presence of two known genotypes (#7 and #63) and five new genotypes (NEW 3, NEW4, NEW5, NEW6, NEW 7). The results showed high seroprevalence, isolation rate, molecular detection and genotypic variations of T. gondii in free-range chickens in the southwestern region of Goiás.

ABSTRACT Soybean foliar diseases negatively affect leaf area index (LAI) and compromise the main structure for photosynthesis and grain yield. The objective of this study was to propose a new methodology to determine the commencement of fungicide application based on the soybean LAI, spray volumes, and the inclusion of a multisite fungicide to control Asian soybean rust (ASR). The study was implemented in a randomized block design at two locations, with four replicates in a factorial scheme plus one additional treatment (4 × 2 × 2 + 1) - four soybean LAI (0.9, 1.8, 3.6, and 6.1) indicating the initiation stages of fungicide applications, two spray volumes (70 and 120 L ha-1), with or without multisite fungicide (mancozeb), and a control treatment without fungicide. The spray deposition on plants did not differ between the treatments for 1.8 and 3.6 LAI. Treatments including the multisite fungicide for LAI between 3.6 and 6.1 for both spray volumes presented superior ASR control results. The highest soybean yields were observed when fungicide application started at 3.6 LAI for both spray volumes. The best correlation between disease control and grain yield occurred when the applications started at 3.6 LAI with 70 L ha-1 spray volume and the multisite fungicide.

ABSTRACT: Spontaneous and experimental poisoning by Tephrosia cinerea in the northeastern semiarid region of Brazil has only been described in sheep. Pathologically, such poisoning leads to ascites and centrilobular liver fibrosis. However, these effects require an experimental study in goats. This study aimed to determine the goats’ susceptibility to the ingestion of T. cinerea and the minimum toxic dose, describing the main clinical and anatomopathological findings. Poisoning was reproduced experimentally in one sheep that received 10g/kg of the ground plant and in two goats, the first receiving a dose of 5g/kg and the second receiving 10g/kg of the ground plant. The sheep presented abdominal distension 34 days after beginning the ingestion of the plant, developing sternal decubitus, breathing difficulty, opisthotonos, mandibular trismus, salivation, dysphagia, vocalization, and pedaling movements on the 50th day of the experiment. Fluid accumulation was observed in the abdominal cavity and liver via necropsy, with an irregular, slightly whitish capsular surface. Histologically, the main lesions observed in the liver were moderate fibrosis, marked sinusoidal distension, accompanied by marked hemorrhage, sometimes forming bridges between the centrilobular regions, associated with a dissociation of hepatocyte cords. There were discrete Alzheimer’s type II astrocytes in the gray matter in the region of the occipital cortex in the nervous system. Goat 2 showed apathy, drowsiness, and weight loss; on the 62th day, lateral decubitus evolved to sternal decubitus, with a rotation of the neck towards the flank. At necropsy, marked edema was observed on the face and dewlap, and a slight accumulation of liquid; slightly yellowish material was observed in the abdominal cavity. There were discrete blackened areas on the capsular surface in the liver. Histologically, the liver showed mild centrilobular fibrosis associated with mild dissociation of hepatocyte cords and mild vacuolar degeneration of the hepatocyte cytoplasm. Goat 1 showed no clinical signs; at necropsy, discrete multifocal areas were observed in the liver on the capsular surface. Histologically, diffuse intracytoplasmic vacuolar degeneration of hepatocytes was detected. The clinical picture and anatomopathological findings differ between the species, proving the lower susceptibility of goats to Tephrosia cinerea ingestion (compared to sheep), with differences in the pathogenesis and epidemiological aspects of poisoning.

Abstract The aim of this study was to evaluate the seroprevalence and factors associated with the presence of anti-Toxoplasma gondii and anti-Neospora caninum antibodies in sheep from Paraná state. The detection of antibodies for T. gondii and N. caninum was performed by homemade and commercial indirect ELISA, respectively. Multiple logistic regression analysis was used to verify the factors associated with the seroprevalence. Antibodies anti-T. gondii and anti-N. caninum were observed in 42.7% and in 17.6% of the animals, respectively. The protective factors associated to seropositive were “some level of confinement” (full or semi-extensive confinement) (OR=0.53) for T. gondii and “use of skilled labor” (OR=0.64) for N. caninum. The risk factors were “presence of cats” (OR=1.75) for T. gondii and “feeding of dogs with sheep placental remains” (OR=1.79) for N. caninum. In addition, to presenting a significant and simultaneous seroprevalence for both agents (9.9% of the animals), the results also indicate that deficiencies in management and environmental sanitation, the presence of reservoirs, and types of exploitation enhance the seropositivity. Thus, studies like this might support sanitary programs and public policies for the prevention of T. gondii and N. caninum in the sheep herds of Paraná state.

The secondary metabolites of the aerial parts of Phyllanthus acuminatus Vahl (Phyllanthaceae) and the biological activity of one of them were characterized in this study. This chemical investigation led to isolation of two lignans, luclaricin (1), a new arylnaphtalene-typelignan, and justicidin B (2). The structures of isolated compounds were determined based on 1D and 2D-NMR, HRESIMS and IR. The cytotoxic activity of justicidin B was assessed against tumor cell lines (MCF-7, U251, NCI-ADR/RES, NCI-H460, OVCAR-3, HT29 and K562) and a non-tumor cell line (HaCat) using sulforhodamine B assay. Justicidin B showed strong cytotoxic activity against the tumor cell lines HT29, K562 and U251 with GI50 values of 2.28, 3.29 and 5.76 mol L-1, respectively.

Abstract: INTRODUCTION: To characterize Trypanosoma cruzi (TcI) isolated from a Panstrongylus megistus specimen found in one of the biggest metropolitan areas of Latin America, the relationship between the TcI group of T. cruzi and the transmission cycle in the urban environment was studied. METHODS: The T. cruzi strain, Pm, was isolated in a culture medium from the evolutionary forms present in the hindgut of a live male specimen of P. megistus found in the Jabaquara subway in São Paulo City. The sample from the triatomine showed trypomastigote forms of Trypanosomatidae, which were inoculated in the peritoneum of Balb/c mice. The sample was then inoculated in Liver Infusion Tryptose medium and J774 cells for the molecular identification and characterization of the parasite. The Pm strain of T. cruzi was identified by isolation in axenic culture medium, and based on the morphology, cell infection, growth kinetics, and molecular characterization. RESULTS: After isolation, the protozoan was identified as T. cruzi. No parasites were detected in the peripheral blood of the animal, which can be a characteristic inherent to the strain of T. cruzi that was isolated. Cell invasion assays were performed in triplicate in the J774 cell line to confirm the invasive ability of the Pm strain and revealed amastigote forms of the parasite within macrophages. CONCLUSIONS: Our biological and molecular characterizations helped understand parasite-host interactions and their evolutionary history in context of the associations between vectors, ecotopes, hosts, and groups of the parasite.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Myocardial scintigraphy with meta-iodo-benzyl-guanidine (123I cMIBG) has been studied in Parkinson's disease (PD), especially in Asian countries, but not in Latin America. Most of these studies include individuals with PD associated to a defined dysautonomia. Our goal is to report the cardiac sympathetic neurotransmission in de novo Brazilian patients with sporadic PD, without clinically defined dysautonomia. We evaluated retrospectively a series of 21 consecutive cases with PD without symptoms or signs of dysautonomia assessed by the standard bedside tests. This number was reduced to 14 with the application of exclusion criteria. 123I cMIBG SPECT up-take was low or absent in all of them and the heart/mediastinum ratio was low in 12 of 14. We concluded that 123I cMIBG has been able to identify cardiac sympathetic neurotransmission disorder in Brazilian de novo PD patients without clinically defined dysautonomia.

The aim of this study was to evaluate the residual effect of fertilization with sewage sludge compost and rock phosphate on the chemical properties of the soil, nutrient content in plant and corn yield. The study was conducted on Haplic Cambisol. The treatments, in a factorial 2 x 4, corresponded to two doses of Gafsa phosphate (0 and 90 kg P2O5 ha-1) and four doses of sewage sludge compost (0, 25, 50 and 75 Mg ha-1 dry weight basis). The experimental design was in randomized blocks with three replications. In general, the corn yield and content of macronutrients in soil and plant on the second crop of corn were not influenced by the residual effects of fertilization with phosphate rock. However, corn yield and nutrient content in soil and leaves increased with the dose of sewage sludge compost applied to the soil, and the dose of 75 Mg ha-1 was the one with the highest residual effect. The yield of corn was lower in the second successive cultivation, due to the impoverishment of the soil phosphorus and potassium, recommending new fertilization with sewage sludge in every cultivation.

The objetive of this work was to determine physiological and productive parameters and the thermal comfort indices in "Santa Inês" breed of sheep created in two different shelters, one being covered with clay tile (TBA) and the other covered with cement fiber tiles (TFC), in the municipality of São João do Cariri, Paraíba State, Brazil, during the months of August to October 2003. Twenty castrated male sheeps were distributed in individual bays. The environmental indices analyzed were the temperature and humidity of the black globe index and the thermal charge of radiation index. The data used were the air temperature, black globe temperature, relative humidity and wind velocity. The physiological indices reffered were the rectal temperature (TR) and the respiratory frequency (FR) and the production indices refered were the weight gain, food conversion and the dry matter consumption. The shelters with TBA and TFC did not present significant differences in indices of thermal comfort, neither in the morning nor in the evening periods. There was a little increase in the indices during the evening period. The animals could maintain a TR within the normal limits and during the evening period the FR was higher than that in the morning. The production indices did not vary between the systems analyzed and were considered satisfactory.

The combination effect of cattle manure and mineral fertilization on palm heart yield in peach palm plants was evaluated. The experiment was conducted from April 2000 to May 2002 in Quartz Psamment at the Universidade Federal da Paraíba, in Areia, Brazil. The experiment design was a randomized block with treatments arranged in a 4 x 2 factorial scheme with four cattle manure rates (0; 10; 20 and 30 t ha-1) in the presence or absence of NPK fertilizer in four replications. Each plot consisted of 20 plants spaced 2 x 1 m. Estimates of maximum yield for first class (0.84 t ha-1) and second class (1.10 t ha-1) palm heart were obtained when using cattle manure at the rates of 15.4 and 14.0 t ha-1 in combination with mineral fertilization. In the absence of mineral fertilization the maximum yield of first class (0.76 t ha-1) and second class (1.0 t ha-1) palm heart was attained with cattle manure rates of 15.8 and 16.0 t ha-1. First class palm heart presented the maximum yield estimated at 1.94 t ha-1 with cattle manure applied at the rate of 15.0 t ha-1. In the absence of N-P-K fertilization the maximum yield of 1.85 t ha-1 was due to the application of 16.6 t ha-1 of cattle manure.
