ABSTRACT The objective of this study was to evaluate the effect of irrigation with low- and high-salinity water on the increment of salts in the soil, the production components of sour passion fruit and the leaching of salts by rainfall and by leaching fraction of 10%. The treatments were arranged in randomized blocks, in a split-plot scheme, corresponding to three irrigation management practices [evaluation of soil chemical attributes before irrigation; and after irrigation with water of low electrical conductivity (0.35 dS m-1), and high electrical conductivity (4.00 dS m-1) in the main plot], and in the subplot two soil depths of evaluation (0-20 and 20-40 cm) and three soil sampling times (at 115 days after transplanting of seedlings (DAT) - beginning of flowering; at 199 DAT - end of the dry season; and at 379 DAT - end of the rainy season). Irrigation increased the electrical conductivity of the soil saturation extract, with higher values in the surface layer and at the end of the dry season. Rainfall during the rainy season reduced the saline character of soil from moderately saline and strongly saline, in treatments irrigated with water of low and high electrical conductivity, to non-saline and slightly saline, respectively. Irrigation with high-electrical conductivity water negatively affected the production components and yield of sour passion fruit.

Abstract Soybean seeds are usually infested by fungi, and disinfestation has been carried out mainly with synthetic fungicides, however, alternative controls have been sought, including essential oils. Thus, the effect of essential oils extracted from the leaves of the hybrid Eucalyptus tereticornis x E. urophylla (HEtu) and E. grandis x E. urophylla (HEgu) in the sanitary and physiological quality of soybean seeds was evaluated. The sanity test was conducted out in completely randomizes design (CRD), factorial scheme 2x5 (two essential oils, in five concentrations: 0, 0.5, 0.75, 1.0 and 2.0 µL.mL-1), the seeds were placed in gerbox boxes in an incubator at 20 ºC for seven days, the fungi were identified according to their morphology. The physiological quality was evaluated in CRD, factorial scheme 2x4+1, being: two essential oils, in four concentrations (0.5, 0.75, 1.0 and 2.0 µL.mL-1), plus an additional control treated with autoclaved distilled water, after eight days, the percentage of germination, percentage of abnormal seedlings and percentage of dead seeds were evaluated. The oils were efficient in controlling the Fusarium and Cladosporium genera, especially at concentrations of 1.0 and 2.0 µL.mL-1. HEgu oil was more efficient in sanitary control, while HEtu oil was superior in all physiological variables. Despite the greater efficiency in sanitary control, HEgu oil negatively alters the physiological quality of the seeds, while HEtu oil at a concentration of 2.0 µL.mL-1, in addition to the efficiency in phytosanitary control, does not affect the physiological quality of the seeds.

This study was performed to evaluate the effects of a functional oil blend (castor oil plus cashew nutshell oil) on complete blood count, health parameters, and intestinal structure in nursery pigs. A total of 128 crossbreed piglets (6.79 ± 1.76 kg of body weight) were allocated in a randomized complete block design with two dietary treatments: a functional oil-free diet or a diet based on added functional oil (1500 mg functional oil blend/kg of diet). Both diets had 400-500 mg colistin sulphate/kg. Pigs fed functional oil had higher erythrocytes and monocytes on days 11 and 23, respectively. Lymphocyte concentration was reduced, and ileum pH was increased in pigs fed functional oil on day 23. On day 37, lower jejunal pH was observed. However, no treatment effect on diarrhoea occurrence was observed. The lactic acid bacteria count was greater in jejunum and ileum of pigs fed functional oil on day 23. Pigs fed functional oil had more jejunal lesions on day 37. Based on the criteria of this study, dietary supplementation to piglets with functional oil blend changed the concentration of erythrocytes and monocytes as an important role in the defence of the organism; however, it showed a reduced capacity to modify the pH in small intestinal contents, which was reflected in an absence in the incidence of diarrhoea. In addition, piglets fed the functional oil blend had improvements in lactic acid bacterial counts, without the ability to attenuate lesions in the jejunum.

Abstract Staphylococcus spp. represents the main mastitis agents in ruminants and contaminants of milk due to their expressive capacity to make biofilms. The aims in this study was evaluate evaluated the antimicrobial activity of Mauritia flexuosa L. extracts against Staphylococcus spp. adhered to a stainless steel surface. Two isolates from cows with clinical mastitis were evaluated; one was identified as Staphylococcus aureus, and the other Staphylococcus haemolyticus. Additionally the ATCC 25923 strain, S. aureus from human was evaluated. The chemical profile obtained from gas chromatography revealed the presence of carbohydrates, organic acids, and flavonoids. The minimum bactericidal concentrations of the ethanolic extract (EE) and aqueous extract (AE) were 4.4 and 5.82 mg/mL, respectively. After EE treatment at 4.4 mg/mL for 2.5 min, total removal of mature biofilms grown on stainless steel coupons was observed (reduction by 3.85-4.81 log units). This extract from M. flexuosa shows potential as an effective sanitizer and may represent a natural alternative against Staphylococcus spp.

This study aimed to develop a reliable stability-indicating method (SIM) for amoxicillin 500 mg capsules (DP-drug product). A literature review addressing amoxicillin (AMX; DS-drug substance) forced degradation studies and the existing SIMs was conducted to verify the most significant outcomes. Subsequently, the forced degradation of DP and DS was carried out following the Brazilian Health Surveillance Agency (ANVISA) guidelines, including thermal degradation (dry and wet heat), acidic and alkaline hydrolyses, hydrogen peroxide oxidation, reaction with copper, and photodegradation. Both DS and DP were more susceptible to 0.015 M NaOH, resulting in approximately 50% degradation. AMX DS and DP were not significantly photodegraded, but some degradation products (PDegs) showed susceptibility to light exposure. Thermodegraded samples showing ≥ 10% degradation exhibited modified profiles in thermogravimetric (TG) and differential scanning calorimetric (DSC) analyses. The X-ray powder diffraction patterns (XRPD) of DS samples exposed to dry and wet heat displayed complete amorphization of AMX, attesting to the occurrence of physical degradation concomitantly with chemical degradation, which can alter the drug’s bioavailability. In contrast, the thermodegraded DP samples exhibited intact AMX crystals interspersed with the amorphous form, perhaps partly protected by the excipient. The validated SIM was able to detect and quantify about 80 PDegs.

Abstract Light is considered a factor that influences the seed germination of many weed species, and it can signal whether the environmental conditions are favorable or are not favorable for germination. We aimed to study if there is an influence of light quality and dormancy overcoming in seed germination of Echium plantagineum L. We carried out a 2 x 6 factorial experiment, with and without dormancy overcoming with potassium nitrate followed by immersion in gibberellic acid; six light qualities, obtained through the light filters: blue, green, red, far-red, white light and absence of light. The evaluations performed were germination speed index (GSI), average germination time (AGT), germination at the four and 14 days after seeding (DAS), accumulated germination and relative frequency of germination. We observed significant interaction among the light qualities and seed dormancy overcoming or not for the studied variables. There was no significant effect of light qualities, in the evaluated variables, when performing dormancy overcoming, presenting germination above 90% in all the light qualities. However, without dormancy overcoming, we observed greater GSI, germination at four and 14 DAS for the red light filter with 5, 4, 29 and 45%, respectively. When the seeds were submitted to the absence of light, and without dormancy overcoming, there was only 7% of germination at 14 DAS. The seeds of E. plantagineum presented greater germination under incidence of red light, without dormancy overcoming, being classified as preferably positively photoblastics, provided that the dormancy is not overcome.

ABSTRACT The objective of this study was to evaluate the production efficiency of cowpea (Vigna unguiculata L. Walp.), ‘BRS Pujante’, from the weather-based irrigation scheduling using single and dual Kc. The study was carried out in randomized block design with two treatments (single and dual crop coefficient) and 12 replicates. Chlorophyll a, chlorophyll b and total chlorophyll contents, soil matric tension, number of pods per plant, number of grains per pod, average 100 grains weight, total grain yield and water use efficiency were evaluated. Scheduling with single and dual Kc did not interfere with chlorophyll pigments. Single Kc promoted tension values closer to zero during cowpea development stages III and IV. Significant difference was observed only for the average 100 grains weight, with higher value for dual Kc. The irrigation depth with the single Kc was 360.8 mm and there was a 9.3% water saving when compared to dual Kc, without significantly affecting cowpea yield. It is possible to use irrigation scheduling with single Kc for the stages I and II and dual Kc for the stages III and IV of cowpea, ‘BRS Pujante’.

ABSTRACTThe cardiopulmonary changes in propofol- or thiopental-anesthetized dogs induced to pulmonary hypertension (PH) were evaluated. Twenty adult animals were randomly assigned to two groups: propofol group (PG) and thiopental group (TG). In PG, propofol was used for induction (8(0.03mg.kg-1) and anesthesia maintenance (0.8mg.kg-1.minute-1), while, in TG, thiopental was used (22±2.92mg.kg-1; 0.5mg.kg-1.minute-1, respectively). Mechanical ventilation using time cycle was started. PH was induced by administration of serotonin (5HT) (10µg.kg-1 and 1mg.kg-1.hour-1) through a thermodilution catheter positioned in the pulmonary artery. The measurements were performed before administration of 5HT (T0), after 30 minutes (T30), then at 15-minute intervals (T45, T60, T75 and T90). No differences between groups were registered for systolic (sPAP) and mean pulmonary arterial pressure (mPAP), mean arterial pressure (MAP), total peripheral resistance index (TPRI) and pulmonary vascular resistance index (PVRI). In PG, sPAP and mPAP increased from T30. While in TG, sPAP and mPAP increased from T75. In PG, heart rate (HR) increased from T30, in which PG was higher than TG. The TPRI values decreased from T30 in PG, and in TG, at T45, T60 and T90. In PG, at T0, PVRI was lower than at other times. In PG, arterial partial pressures of oxygen (PaO2) decreased from T60 and alveolar-arterial oxygen gradient (PA-aO2) increased at T60. In TG, at T0 PaO2 was higher than at T30, T45, T60 and T90, while PA-aO2 at T0 was lower than at T90. From T30 to T90, TG showed higher PaO2 means and lower arterial partial pressures of carbon dioxide (PaCO2) values when compared to PG. In PG, from T30, PaCO2 increased, while in TG this parameter was stable. In conclusion, thiopental anesthesia attenuated the cardiopulmonary changes resulting from serotonin-induced PH, probably by attenuation of vasoconstriction and bronchoconstriction.

ABSTRACTThe aim of this study was to determine the effects of various dilution ratios on thaw sperm motility, velocity and presence of spermatic abnormalities in the common carp. 20 males with three years of age, raised at the Departamento Nacional de Obras Contra as Secas (DNOCS) were used. From the collected semen a total of nine pools were formed. The semen from each pool was evaluated for motility, velocity and spermatic morphology before and after cryopreservation. The sperm cryopreservation was performed in medium ACP-104 + DMSO 10% diluted in 1:1 or 1:3 (sperm:extender). The samples were loaded into 0.25mL straws, frozen in liquid nitrogen vapor (Styrofoam) and stored in liquid nitrogen (-196 °C). There was no significant difference (P>0.05) between the ratios of dilution on the kinetic and morphological parameters of thawed semen from common carp. However, these parameters were significantly different (P<0.05) compared to fresh semen (control). Low velocity values were recorded in thawed semen (VCL 42.6-46.5μm/s; VSL 33.2-37.1μm/s; VAP 38.9-43.2μm/s), however, observed motility above 59% (59.8-67.8%), and low levels of spermatic abnormalities (19.3-19.5%) suggested that the ACP-104 can be used as a suitable medium for cryopreservation of common carp semen in any of the tested dilution ratios.

The sector of ornamental rocks produces significant volume of waste during the sawing of the blocks and demand to find ways to recycle, given its environmental impact. Considering the possibilities of use of industrial by-products as mineral admixtures, aiming at sustainable development in the construction industry, this paper aims to study the performance of the processing sludge of ornamental rocks and grinding after heat treatment, based on their potential application as partial substitute for cement. The residue was characterized, cast and milled to produce glassy material. Was analyzed the mechanical performance and pozzolanic activity with partial replacement of cement by waste in natural condition and after heat treatment in mortars for comparison. The results were promising, so it was possible to verify that after heat treatment, the treated waste is presented as a material with pozzolanic characteristics.

Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Asparaginases are the cornerstone therapy of many successful combination regimens for the treatment of acute lymphoblastic leukemia (ALL), the most common malignancy in children and adolescents. The aim of this work was to produce recombinant Erwinia carotovora L-asparaginase II in Escherichia coli fed-batch cultures. Using a robust fed-batch technique with pre-determined exponential feeding rates, our bioreactor culture system yielded 30.7 grams of dry cell weight and 0.9 grams of soluble rErAII protein per liter of culture broth. The homogeneous rErAII activity was determined by isothermal titration calorimetry (ITC). The enzyme Km values for the main substrates L-Asn and L-Gln were 33x10-6 M and 10x10-3 M, respectively. Our work shows that it is possible to produce an active homogeneous rErAII enzyme in the soluble cell fraction through IPTG-induced E. coli fed-batch cultivation.

In this work, transparent ferroelectric ceramics of lanthanum modified lead zirconate titanate (PLZT, with La/Zr/Ti=9/65/35), doped with rare-earth ions (RE = Nd3+, Er3+, Yb3+ Tm3+ and Ho3+), were processed by hot uniaxial pressing aiming the investigation of their potentiality as near-infrared laser active media. Their structural, microstructural, electric and spectroscopic characterizations were performed. For 1.0 wt.% RE2O3 doping, a strong influence of the rare-earth type was verified in the PLZT ferroelectric and optical characteristics. It was also observed that the Nd3+ substitution for Pb2+/La3+ reduces the ferro-paraelectric phase transition temperature and favors the transparency of the ceramic bodies. Spectroscopic analysis indicates that PLZT:Nd3+ is a potential diode-pumped laser active media in the near infrared region, and with some improvements, PLZT:Er3+ and PLZT:Tm3+ might also become so .
