ABSTRACT Toxoplasmosis is an infection of vast worldwide distribution whose etiologic agent is Toxoplasma gondii. This disease can cause problems ranging from mild symptoms to serious conditions, such as encephalitis, miscarriage and blindness. Therefore, it is of utmost importance to perform a diagnosis with reproducible techniques in order to obtain a good prognosis. The aim of this review was to analyze the efficiency of toxoplasmosis diagnostic techniques based on sensitivity and specificity results. Five research platforms in English language were used (Eric, Elsevier, Google Scholar, PubMed and SciELO), which contained data on the diagnosis of toxoplasmosis. The search and selection were performed for studies published prior to June 2021. The search resulted in the inclusion of 13 articles published from 2005 to 2020. The data revealed the use of different samples in the standardization of techniques such as serum, total blood, colostrum and amniotic fluid. The flow cytometry, lateral flow immunoassay and qPCR techniques showed 100% sensitivity, whereas the ELISA, western blotting, qPCR and RE-LAMP techniques achieved 100% specificity. Significantly, the qPCR and LAMP techniques were more accurate when the likelihood ratio was assessed. The meta-analysis identified that ISAGA and western blotting have low sensitivity values and LIASON, ELFA and ELISA, using a silica bioconjugate, also have low specificity values. It was noted that a wide range of methods have high values of sensitivity and specificity. Therefore, the choice of the method will be based on the conditions and its financial viability.

Abstract Cnesterodon hypselurus is a small fish that has a restricted distribution in southern Brazil, including headwaters of the Tibagi and Itararé river basins (Upper Paraná River). This study reported C. hypselurus in a headwater of Cinzas River basin, where there were no previous records of this species, and employed microsatellite loci and mitochondrial haplotypes in a population genetic analysis. A total of 57 specimens was analyzed, including 30 from Cinzas River basin, 25 from Itararé River basin and two from Tibagi River basin. Results indicated low genetic diversity levels (HE = 0.334 and h = 0.246) for the sample from Cinzas River, suggesting reflections of a founder effect after the species had dispersed from one watershed to another, possibly by headwater captures. Since different populations were detected between the Cinzas and Itararé rivers (DEST = 0.248, P-value < 0.05) and other occurrence sites are still unknown in the Cinzas River basin, the data herein have great relevance and should be taken into account in future management and conservation actions, as well as in evolutionary studies of C. hypselurus.

Abstract Background Eugenol shows both antibacterial and antiparasitic activities, suggesting that it might be evaluated as an option for the treatment of praziquantel-resistant schistosome. Methods The in vitro activities of three eugenol derivatives (FB1, FB4 and FB9) on adult worms from Schistosoma mansoni were examined by fluorescence and scanning electron microscopy to analyze effects on the excretory system and integument damage, respectively. Biochemical tests with verapamil (a calcium channel antagonist) and ouabain (a Na+/K+-ATPase pump inhibitor) were used to characterize eugenol derivative interactions with calcium channels and the Na+/K+-ATPase, while in silico analysis identified potential Na+/K+-ATPase binding sites. Results The compounds showed effective doses (ED50) of 0.324 mM (FB1), 0.167 mM (FB4), and 0.340 mM (FB9). In addition, FB4 (0.322 mM), which showed the lowest ED50, ED90 and ED100 (p < 0.05), caused the most damage to the excretory system and integument, according to both fluorescence and scanning electron microscopy analysis. The death of adult worms was delayed by ouabain treatment plus FB1 (192 versus 72 hours) and FB9 (192 versus 168 hours), but the response to FB4 was the same in the presence or absence of ouabain. Besides, no changes were noted when all of the eugenol derivatives were combined with verapamil. Moreover, FB1 and FB9 inhibited Na+/K+-ATPase activity according to in silico analysis but FB4 did not show a time-dependent relationship and may act on targets other than the parasite Na+/K+-ATPase. Conclusion Eugenol derivatives, mainly FB4 when compared to FB1 and FB9, seem to act more effectively on the integument of adult S. mansoni worms.

ABSTRACT Introduction: Squamous cell carcinoma (SCC) is the most common tumor among all cancers in the oral cavity. Despite advances, the prognosis of this neoplasm remains a challenge for professionals. Faced with this situation, several studies try to associate the histopathological analysis with prognosis, so that therapeutic planning becomes more accurate. Objectives: This research aimed to conduct an epidemiological study of oral SCC and classify them histopathological assessment according to the World Health Organization (WHO) and the Budding and Depth of Invasion (BD) model. A retrospective research was conducted. Methodology: Data from medical records filed at UOPECCAN Hospital between 2009 and 2015 were analyzed. The sample consisted of 57 patients. Epidemiological data were collected and the blocks were rescued and cut for histopathological analysis. Associations were performed using the chi-square test with a significance level of 5% (p = 0.05) by the GraphPad Prism program. The two histopathological analyzes were correlated using Spearman’s statistical test. Results: After analyzing the samples, we found a higher prevalence of oral SCC in male smokers aged above 40 years. There was no correlation between the BD and WHO methods. The WHO classification was significantly associated with age (p = 0.03), and follow-up care (p = 0.05). However, the BD model associated lymph node involvement (p = 0.005) and clinical staging (p = 0.005). Conclusion: The BD classification was more objective for histopathological analysis and may be an important tool for analyzing patient prognosis, assisting in the treatment decision.

Objective: To evaluate body dissatisfaction and distorted body self-image in women with prolactinoma. Methods: Body dissatisfaction and distorted body self-image were evaluated in 80 women with prolactinoma. All patients were in menacme, 34% had normal body mass index (BMI), and 66% were overweight. Most patients (56.2%) had normal prolactin (PRL) levels and no hyperprolactinemia symptoms (52.5%). The Body Shape Questionnaire (BSQ) was used to assess the patients’ dissatisfaction with and concern about their physical form, and the Stunkard Figure Rating Scale (FRS) was used to assess body dissatisfaction and distorted body self-image. The patients were divided according to PRL level (normal vs. elevated) and the presence or absence of prolactinoma symptoms. Results: The normal and elevated PRL groups had similar incidences of body dissatisfaction and distorted body self-image. However, symptomatic patients reported a higher incidence of dissatisfaction than asymptomatic patients. Distorted body self-image was less common among symptomatic patients. Conclusion: Symptomatic patients showed higher body dissatisfaction, but lower body self-image distortion. The presence of symptoms may have been responsible for increased body awareness. The perception of body shape could have triggered feelings of dissatisfaction compared to an ideal lean body. Therefore, a distorted body self-image might not necessarily result in body dissatisfaction in women with prolactinomas.

A novel method for the synthesis of nonsymmetrical 3,3’-(aryl/alkyl-methylene)bis-2-hydroxy-1,4-naphthoquinones was developed by using the Mannich adduct of naphthoquinone and the reaction with another moiety of 2-hydroxy-1,4-naphthoquinone. This novel method produces for the first time nonsymmetrical 3,3’-(aryl/alkyl-methylene)bis-2-hydroxy-1,4-naphthoquinones. In a preliminary study, these compounds (15c, 15f and 15h) were evaluated regarding their effect over the viability of PC3 metastatic prostate cancer cell using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays at 100 µM. Three of these compounds presented relevant cytotoxic effects at 72 h posttreatment.

Abstract: The objective of this work was to evaluate the protein profiles of natural and semidry fermented Coffea arabica, either subjected to treatments with different yeast inoculation methods with starter culture or to an uninoculated control. Saccharomyces cerevisiae CCMA 0543 and Candida parapsilosis CCMA 0544 were separately inoculated into coffee by directly spraying the cherries on a terrace or in buckets, for 16 hours before sun drying. Protein quantification showed a significant difference between the protein profiles of the samples collected after natural dry fermentation. The MALDI-TOF MS analysis generated a list of 96 peaks with different mass-to-charge ratios (m/z) in the samples collected at the beginning and the end of fermentation. The highest number of peaks in the natural dry coffee was observed at the end of fermentation in the samples inoculated with S. cerevisiae CCMA 0543, in bucket, and in C. parapsilosis CCMA 0544 sprayed on the terrace. However, in the semidry processed coffee, the highest number of peaks was observed in the initial fermentation, with a decrease in the peptide peaks after fermentation. The fermentation with different microorganisms, processing types, and inoculation methods affects m/z profiles, influencing the types of proteins found in coffee.

Abstract In order to inhibit natural gas hydrate formation, monoethyleneglycol (MEG) is usually injected into producing well heads. The MEG regeneration process is continuously performed at the platform. Scaling problems usually occur due to the presence of chlorides and carbonates. This work presents salt solubility data for the aqueous system with strontium carbonate, MEG and carbon dioxide. A specific analytical method was developed. Thus, experimental data for strontium carbonate (SrCO3) solubility at various carbon dioxide pressures are reported. Solubilities of SrCO3 in water were measured from 760 to 1610 mmHg at 278.15. 288.15. 298.15 and 323.15 K. For the mixed solvent (ms) conditions, solubilities were measured at 298.15 K and four isobars, i.e., 760, 1210, 1410 and 1520 mmHg. Experimental data were correlated and demonstrated to be accurate for thermodynamic modeling and process simulation.

ABSTRACT: The use of microalgae as a source of raw material for biofuel production has been highly targeted. Compared to other biological options for the capture and use of CO2, microalgae crops have the following advantages: high productivity, lack of competition with feed and food-based products, use of unproductive and non-arable land, and allowance of the use of wastewater for its cultivation. The use of wastewater from industrial, agricultural, and domestic sewage has been indicated as an alternative means to reduce the cost of crops, since such waste is generally released into the environment without previous treatment. This work addresses the use of petrochemical wastewater (ARP) from effluent produced at the Gabriel Passos Refinery (Petrobrás refinery unit) for the cultivation of a strain of Scenedesmus obliquus BR003. Twelve experimental units of 20 L each were constructed in the experimental area of the Biofuel Laboratory of the Federal University of Viçosa, Minas Gerais, Brazil in an uncontrolled environment, and the cultures were evaluated by quantification of total carbohydrates, total lipids, dry biomass, and growth evaluated by absorbance. The results showed that the wastewater from petrochemical effluent is an efficient growth medium for the growth of Scenedesmus obliquus BR003 microalgae for large-scale cultivation on land that is not suitable for other agricultural crops.

Abstract INTRODUCTION: The Kato-Katz technique is the standard diagnostic test for Schistosoma mansoni infection in rural areas. However, the utility of this method is severely limited by the day-to-day variability in host egg excretion in the stool. In high-transmission areas, the point-of-care circulating cathodic antigen (POC-CCA) urine assay has proven to be a reliable test. However, investigations of the reliability of the POC-CCA assay in low-transmission regions are under way. This study aimed to evaluate the sensitivity and specificity of the POC-CCA assay and the morbidity of schistosomiasis in a low-endemic area in Brazil. METHODS: Pains City is a low-transmission zone for schistosomiasis. A total of 300 subjects aged 7-76 years were randomly selected for the POC-CCA cassette test. For S. mansoni diagnosis, three stool samples on six slides were compared with one urine sample for each subject. The sensitivity and specificity in the absence of a gold standard were calculated using latent class analysis. Clinical examinations and abdominal ultrasounds were performed in 181 volunteers to evaluate morbidity associated with schistosomiasis. RESULTS: The sensitivity and specificity of the Kato-Katz technique were 25.6% and 94.6%, respectively. By contrast, the sensitivity and specificity of the POC-CCA assay were 68.1% and 72.8%, respectively. Hepatosplenic schistosomiasis was diagnosed in two patients (1.1%). CONCLUSIONS: Overall, the POC-CCA urine assay proved to be a useful test for diagnosing S. mansoni in a low-endemic area in Brazil. Severe clinical forms of schistosomiasis can be present even in such low-endemic areas.

ABSTRACT: An analysis of the pre-surgical electrocardiographic profile was made of 124 dogs submitted to different surgical interventions to correlate the variables age, sex, size, absence or presence of cardiovascular abnormalities detected during physical examination and of the surgical cases with electrocardiographic findings. No association between these variables and electrocardiographic changes were observed. A total of 79 cães (63.7%) showed some type of change in the electrocardiogram. The study suggests that pre-surgical electrocardiographic examination is of great importance, regardless of sex, age, weight, historic or clinical signs associated with heart disease.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

OBJECTIVE: After acute myocardial infarction, during the cardiac repair phase, periostin is released into the infarct and activates signaling pathways that are essential for the reparative process. However, the role of periostin in chronic cardiac remodeling after myocardial infarction remains to be elucidated. Therefore, the objective of this study was to investigate the relationship between tissue periostin and cardiac variables in the chronic cardiac remodeling induced by myocardial infarction. METHODS: Male Wistar rats were assigned to 2 groups: a simulated surgery group (SHAM; n = 8) and a myocardial infarction group (myocardial infarction; n = 13). After 3 months, morphological, functional and biochemical analyses were performed. The data are expressed as means±SD or medians (including the lower and upper quartiles). RESULTS: Myocardial infarctions induced increased left ventricular diastolic and systolic areas associated with a decreased fractional area change and a posterior wall shortening velocity. With regard to the extracellular matrix variables, the myocardial infarction group presented with higher values of periostin and types I and III collagen and higher interstitial collagen volume fractions and myocardial hydroxyproline concentrations. In addition, periostin was positively correlated with type III collagen levels (r = 0.673, p = 0.029) and diastolic (r = 0.678, p = 0.036) and systolic (r = 0.795, p = 0.006) left ventricular areas. Considering the relationship between periostin and the cardiac function variables, periostin was inversely correlated with both the fractional area change (r = -0.783, p = 0.008) and the posterior wall shortening velocity (r = -0.767, p = 0.012). CONCLUSIONS: Periostin might be a modulator of deleterious cardiac remodeling in the chronic phase after myocardial infarction in rats.

Comprehensive two-dimensional gas chromatography with time of flight mass spectrometry (GC×GC-TOFMS) is an appropriate technique for the elucidation of molecular composition of petrochemical samples, such as biodegraded oils. Biomarkers were separated and identified, and conventional biomarker ratios were determined via gas chromatography-mass spectrometry (GC-MS) and GC×GC-TOFMS. In the extracted ion chromatogram m/z 123 + 177 + 191, coelutions between tricyclic terpanes, hopanes and 25-nor-hopanes with secohopanes were resolved by GC×GC-TOFMS. GC×GC-TOFMS allowed the identification of complete series of 25-nor-hopanes, nor-gammacerane, C29 28-nor-spergulanes and oleanane not identified by using GC-MS. The biomarker ratios from the studied oils indicated that they derived from marine source rock deposited under anoxic conditions. The higher chromatographic resolution and sensitivity achieved by using GC×GC-TOFMS allowed for three new parameters to characterize biodegraded oils. These results indicated the superiority of GC×GC-TOFMS for separation and identification of individual and non-target compounds in severely biodegraded oils.
