Resumo A doença de Parkinson (PD) promove sobrecarga entre pacientes e cuidadores. Objetivo: Analisar se a severidade da doença (UPDRS e Karnofsky index), o tempo total de doença, as condições cognitivas do paciente (MEEM), a presença de outras doenças, a idade do paciente, as condições socioeconómicas (ABEP2015), o fato de se ambos vivem juntos, o tempo total que o cuidador exerce esta função, as horas semanais de cuidados e a presença de assistência de outros cuidadores poderiam ser correlacionados e influenciar estatisticamente o grau de sobrecarga dos cuidadores, medido pela entrevista de Zarit (ZBI). Métodos: Após a aprovação do comitê de ética, o recrutamento de pacientes e respectivos cuidadores foram feitos e após a avaliação com as escalas adequadas, todos os dados foram submetidos ao método de correlação de Pearson e análise de regressão linear multivariada (ANOVA). Resultados: Avaliou-se um total de 21 pacientes e respectivos cuidadores. 72% (N=15) de cuidadores reportaram ter sobrecarga. Um terço deles relatou nível moderado ou severo de sobrecarga. Não foi possível estabelecer relação causa-efeito pelo método estatístico adotado, porém a severidade da doença, medida pelo UPDRS, foi a única variável que alcançou correlação de Pearson positiva moderada com significância estatística com a ZBI (r=0,48, com p<0,05), mas na ANOVA, nenhuma variável independente teve um impacto estatisticamente significante nas pontuações de ZBI. Conclusão: Apesar dos nossos resultados conflitantes, a otimização do tratamento disponível, com melhor controle da severidade PD, poderia ser considerada como um elemento importante e que poderia efetivamente atingir o objetivo de reduzir a carga entre os cuidadores.

Abstract Parkinson’s disease (PD) promotes burden among patients and caregivers. Objective: To analyze whether disease severity (UPDRS and Karnofsky index), total disease duration, patient cognitive status (MMSE), presence of other diseases, patient age, socioeconomic conditions (ABEP2015), living together with patient, total time caregiving, weekly hours of care and presence of assistance from other caregivers are correlated with, and influence statistically, the degree of caregiver burden measured by the Zarit Burden Interview (ZBI). Methods: After ethics Committee approval, patients and respective caregivers were recruited. Following evaluation with the proper scales, all data were submitted to Pearson’s correlation method and multivariate linear regression analysis (ANOVA). Results: A total of 21 patients and respective caregivers were evaluated. 72% (N=15) of caregivers reported burden. One third of caregivers reported a moderate or severe level of burden. A cause-effect relationship could not be established by the statistical method adopted, but disease severity measured by the UPDRS was the sole variable showing statistically significant moderate positive Pearson’s correlation with ZBI (r=0.48, for p<0.05). On ANOVA, however, no independent variable had a statistically significant impact on ZBI scores. Conclusion: Despite our conflicting results, optimization of the available treatment, with better control of PD severity, can be considered an important element to effectively achieve the goal of reducing burden among caregivers.

RESUMO Introdução: O câncer é considerado uma doença genética, por isso identificar e caracterizar os genes envolvidos em sua origem e progressão é fundamental para compreender suas bases moleculares. Objetivos: Nosso objetivo foi verificar se os indivíduos de Macapá com diagnóstico de câncer apresentavam os polimorfismos genéticos relacionados com o gene XRCC1. Materiais e métodos: Foram analisadas 30 amostras de ácido desoxirribonucleico (DNA) de indivíduos com câncer e 30 amostras controle. Todas elas foram amplificadas e analisadas pela técnica de reação em cadeia da polimerase (PCR)-polimorfismo de tamanho de fragmentos de restrição (RFLP), com a utilização da enzima de restrição MspI. Resultados: Com relação ao polimorfismo 194T, observamos que todas as amostras dos casos apresentaram o alelo polimórfico Trp (Arg/Trp). Nas amostras controle, em 96,6% também identificamos o alelo polimórfico Trp e, entre essas, uma foi homozigota para o mesmo alelo (Trp/Trp). Quanto ao polimorfismo 399A, 83,3% das amostras dos indivíduos com câncer e 23,3% das amostras controle apresentaram o genótipo Arg/Gln. Verificamos que 73,3% dos controles e 16,6% dos casos apresentaram genótipo Arg/Arg. Encontramos apenas uma amostra, entre os controles, homozigota para o alelo polimórfico Trp/Trp. Conclusão: Nossos resultados demonstraram a frequência do alelo polimórfico 194Trp nos dois grupos amostrais analisados. Encontramos também um número significativo do alelo polimórfico 399A em indivíduos com câncer. Desse modo, podemos destacar o polimorfismo 399Gln como possível marcador genético para ser usado no prognóstico do câncer nessa população.

ABSTRACT Introduction: Cancer is considered a genetic disease. For this reason, identification and characterization of the genes involved in its origin and progression are of fundamental importance in understanding its molecular basis. Objective: Our objective was to determine whether people from Macapá with a diagnosis of cancer have genetic polymorphisms related to the XRCC1 gene. Materials and methods: We analyzed 30 samples of deoxyribonucleic acid (DNA) of cases with cancer and 30 control samples. All samples were amplified and analyzed by the polymerase chain reaction-restriction fragment length polymorphisms (PCR-RFLP) method, with the use of restriction enzyme MspI. Results: Regarding the 194T polymorphism, we found that all samples of the cases presented the polymorphic allele Trp (Arg/Trp). In control samples, 96.6% also identify the polymorphic allele Trp and, among these, one was homozygous for the same allele (Trp/Trp). Regarding the 399A polymorphism, 83.3% of the cases and 23.3% of the controls had the Arg/ Gln genotype, respectively. We found that 73.3% of controls and 16.6% of cases had the Arg/Arg genotype. Among the controls, we found only a sample that was homozygous for the polymorphic allele Trp/Trp. Conclusion: Our results demonstrated the allele frequency of 194Trp polymorphism in both sample groups analyzed. We also found a significant number of polymorphic allele 399A in people with cancer. Thus, we can highlight 399Gln polymorphism as a genetic marker of cancer risk in this population.

Introdução: A anemia falciforme (AF) é a forma mais grave da doença falciforme; apresenta formas variantes que são chamadas de haplótipos βS. Existem cinco principais haplótipos desse gene: árabe-indiano/saudi, Senegal, Benin, Bantu e Camarões. Objetivos: Caracterizar a presença dos haplótipos em pacientes com AF no Amapá. Métodos: Foram estudadas 46 amostras, todas amplificadas e analisadas pela técnica de reação em cadeia da polimerase-polimorfismo de tamanho de fragmentos de restrição (PCR-RFLP). Resultados: Bantu (61,2%) seguido de Benin (26,5%) e Senegal (12,2%). Conclusão: Foram identificados três haplótipos característicos da etnia africana, com a presença do Senegal. Em nosso estudo não encontramos a presença de haplótipos atípicos, sugerindo a concentração e semi-isolamento de grupos fundadores com pouca miscigenação.

Introduction: Sickle cell anemia (SCA) is the most severe form of sickle cell disease; it presents variants that are called haplotypes βS. There are five major haplotypes βS gene: Arab-Indian/Saudi, Senegal, Benin, Bantu, and Camaroon. Objective: Characterize the presence of haplotypes in patients with SCA in Amapá. Methods: 46 sample were studied, all samples were amplified and analyzed by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). Results: Bantu (61.2%), followed by Benin (26.5%) and Senegal (12.2%). Conclusion: We identified three haplotypes characteristic of African ethnicity, with the presence of Senegal. In our study we found the presence of atypical haplotype, suggesting concentration and semi-isolation of the founding groups with little mixing.

Introdução:A doença falciforme é uma doença hereditária, hematológica, de caráter multifatorial, com alta prevalência mundial; sua causa é a mutação no sexto códon do gene da globina beta (βs).Objetivo:Identificar os haplótipos presentes em indivíduos com doença falciforme no Amapá e relacioná-los com a origem afrodescendente.Método:Foram analisadas por meio de técnicas moleculares 46 amostras de sangue de indivíduos com doença falciforme de Macapá, capital do Amapá, com a finalidade de fornecer informações sobre a distribuição das frequências dos haplótipos, contribuindo para o entendimento da formação étnica da população amapaense.Resultados:Nosso estudo revelou que o mais frequente é o haplótipo Bantu (61,2%), seguido de Benin (26,6%) e Senegal (12,2%). Nossos resultados apresentaram diferenças estatísticas em relação a estudos realizados em outras regiões, destacando-se que o presente estudo mostra uma frequência elevada do haplótipo Senegal quando comparado com alguns estudos brasileiros.Conclusão:Os resultados amapaenses apresentam características únicas quando relacionados com os haplótipos de outras regiões, com alta frequência de Senegal e Benin, ausência de atípicos, Camarões e Saudi, confirmando que o Brasil apresenta diversidade de origens étnicas, bem como diferentes frequências de haplótipos.

Introduction:Sickle cell disease (SCD) is a hereditary, hematologic, multifactorial disease, with high prevalence worldwide; its cause is a mutation in the sixth codon of the beta globin gene (βs).Objective:To identify the haplotypes present in people with SCD in Amapá, and relate them to African descent.Methods:We analyzed, by molecular techniques, 46 blood samples from people with SCD in Macapá, the capital of Amapá, with the purpose of obtaining information about haplotype frequency distribution, which helps understand the ethnic background of Amapá's population.Results:Our study revealed that the most frequent haplotype is Bantu (61.2%), followed by Benin (26.6%) and Senegal (12.2%). Results showed statistical differences from studies conducted in other regions. A high frequency of the Senegal haplotype stands out, in comparison with some Brazilian studies.Conclusion:Amapá's results exhibit unique characteristics when compared to haplotypes in other regions, with high frequency of Senegal and Benin haplotypes, absence of atypical, Cameroon and Saudi, confirming that Brazil shows ethnic background diversity, as well as different haplotype frequencies.
