Brain-derived neurotrophic factor serum levels following ketamine and esketamine intervention for treatment-resistant depression: secondary analysis from a randomized trial

Abstract Objectives Evidence suggests that ketamine’s influence on brain-derived neurotrophic factor (BDNF) might be involved in its mechanism of rapid antidepressant action. We aimed to evaluate the differential impact of ketamine and esketamine on serum BDNF levels and its association with response patterns in treatment-resistant depression (TRD). Methods Participants (n = 53) are from a randomized, double-blind clinical trial comparing the efficacy of single-dose ketamine (0.5mg/kg, n = 27) and esketamine (0.25mg/kg, n = 26) in TRD. Depression severity was assessed before and 24 hours, 72 hours, and 7 days after the intervention, using the Montgomery-Åsberg Depression Rating Scale (MADRS). Blood samples were collected before infusion, 24 hours, and 7 days afterwards. Results There were no significant changes in BDNF levels at post-infusion evaluation points, and no difference in BDNF levels comparing ketamine and esketamine. Both drugs exhibited similar therapeutic effect. There was no association between BDNF levels and response to treatment or severity of depressive symptoms. Conclusion There was no significant treatment impact on BDNF serum levels – neither with ketamine nor esketamine – despite therapeutic response. These results suggest that ketamine or esketamine intervention for TRD has no impact on BDNF levels measured at 24 hours and 7 days after the infusion. Abstract Scientific diving (SD) is defined as any diving activity that applies scientific procedures to produce subsidies for studies and technical works in underwater environments. The first report of an underwater scientific study in Brazil dates to the 19th century, in the Abrolhos reefs. Currently, in Brazil, scientific diving has been performed in various areas, from shallow coastal regions to remote and sometimes hard-to-reach places, such as oceanic islands, flooded caves, and icy areas like Antarctica. However, the regulation of SD in Brazil still lacks more concrete actions towards an effective and efficient self-regulation that offers physical safety to practitioners and institutional safeguards for organizations that use it in their research projects. Thus, this article aims to contribute to a better understanding of this critical issue in Brazil and to serve as a reference and incentive for the training of professionals and the development of these activities in the country. It includes: 1) a historical review of SD; 2) a diagnosis of the training and application of SD in Brazil; 3) the evolution of marine sciences in Brazil from the perspective of SD; 4) a review of the use of environmental assessment and underwater conservation techniques in oceans and internal waters; 5) an analysis of the evolution of scientific diver training in Brazil, including a diagnosis on training; 6) the history and updates of the rules, regulations, and safety of SD. Given all the potential of diving combined with specific techniques for research, monitoring, and marine and limnic science in Brazil, we aim to understand the evolution of scientific diving teaching and to outline perspectives in the country, as it is crucial for the training of qualified scientists capable of performing these underwater tasks. Finally, we present future plans for the development of this activity in Brazil from the point of view of research and the labor market.

Polyphasic characterization of forage legumes root nodule bacteria isolated from semiarid region in Brazil

Leguminosas forrageiras são importantes recursos na região semi-árida, devido a sua capacidade de adaptação a solos de baixa fertilidade e fixação de nitrogênio quando associados a bactérias diazotróficas. Aqui, aplicamos uma abordagem polifásica com o objetivo de caracterizar bactérias isoladas de nódulos obtidos de Clitoria ternatea e Stylosanthes capitata cultivadas em solos do semiárido brasileiro. O isolamento das bactérias produziu uma coleção de 126 isolados: 45 isolados de C. ternatea e 81 de S. capitata. Os testes de nodulação resultaram em apenas dez isolados que nodularam seu hospedeiro original: seis de C. ternatea e quatro de S. capitata. Esses dez isolados de nódulos de leguminosas tiveram suas características fenotípicas e genotípicos avaliadas. Todos os isolados cresceram em frutose, glicose, glutamato de sódio, maltose, xilose e sacarose. Testes metabólicos mostraram uma relação entre a tolerância ao sal e altas temperaturas; onde o isolado que tolerou a maior concentração de sal também tolerou a temperatura mais alta. Três isolados apresentaram atividade amilolítica e quatro foram capazes de produzir carboximetil celulolítico. A estreptomicina foi o antibiótico mais limitante para o crescimento bacteriano, e o ácido nalidíxico foi o menos limitante. Sete dos dez isolados eram produtores de IAA. O sequenciamento parcial do gene 16S rRNA permitiu identificar isolados como membros dos gêneros Bacillus (1), Bradyrhizobium (4), Leifsonia (3), Microvirga (1) e Rhizobium (1). Esses dados revelam diversidade fenotípicas e genotípicas de bactérias que habitam nódulos das forrageiras C. ternatea e S. capitata, e representam uma importante fonte microbiana para a prospecção de novos produtos biotecnológicos para promover melhor desenvolvimento das leguminosas forrageiras na região semiárida.
Forage legumes are important resources in semiarid regions due to their abilities to adapt to soils with low fertility and fix nitrogen when associated with diazotrophic bacteria. Here, we applied a polyphasic approach to characterize a set of legume nodule isolates obtained from Clitoria ternatea and Stylosanthes capitata cultivated in the soils of a semiarid region of Brazil. A total of 126 bacterial isolates were obtained: 45 isolates from C. ternatea and 81 isolates from S. capitata. Nodulation tests revealed only ten isolates that nodulated their original host: six isolates from C. ternatea and four isolates from S. capitate. These ten legume nodule isolates were phenotypically and genotypically characterized. All isolates grew in fructose, glucose, sodium glutamate, maltose, xylose, and sucrose. Metabolic tests showed a relationship between tolerance to salt and high temperatures, where isolates that tolerated the highest salt concentration also tolerated the highest temperature. Three isolates showed amylolytic activity, and four isolates showed carboxymethyl cellulolytic activity. Streptomycin was the most limiting and nalidixic acid was the least limiting antibiotic to bacterial growth. Seven out of ten isolates were indol-acetic acid producers. Additionally, 16S rRNA gene partial sequencing enabled the identification of members of the genera Bacillus (1), Bradyrhizobium (4), Leifsonia (3), Microvirga (1), and Rhizobium (1). These data reveal phenotypically and genotypically diverse bacteria inhabiting the nodules of the forage legumes C. ternatea and S. capitata represent an important microbial source to protect new biotechnological products and improve forage legumes in semiarid regions.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The mineral waste studied was host rock from a chromite mine located in Andorinha/Bahia, extracted and stockpiled in yards without specific application. Host rock was submitted to chemical analysis, XRD, SEM-EDS, IR and TGA and classified according to ABNT standards for solid waste classification. Analyses confirmed that this host rock, classified as ultrabasic, consists mainly of dolomite, calcite and diopside. Hazard assessment results showed this host rock should be classified as class II B - inert waste, important for its potential application in agriculture as a soil acidity correction agent.

This work presents the first data on incubation temperature of Phrynops geoffroanus (Schweigger, 1812) in a natural environment, and provides information on nest predation, hatching success and size of offspring born in the nests on sandy beaches along the Upper Xingu River. Thirty-one P. geoffroanus nests were found, of which eleven were completely predated, mainly by Cerdocyon thous (Linnaeus, 1766). Incubation was completed in nine out of the 17 nests protected by netting. The nests presented an average of 13.1 eggs and were distributed over the various geomorphological sectors of the nine sampled beaches. The size and weight of the hatchlings varied significantly between nests, and the incubation period in protected nests lasted for 76.5 days, less than reported for controlled incubation in the laboratory. Daily variation in incubation temperature in the three nests monitored for temperature was lower in those situated in fine sand sediments. Incubation temperature varied from 22 to 39 C and may have affected hatching success, which reached 60.8% in protected nests. Nest distribution in different geomorphological sectors indicated the plasticity of P. geoffroanus in terms of variation in nesting environment, which partly explains the species' broad geographical distribution.

Oysters (Ostreidae) manifest a high degree of phenotypic plasticity, whereby morphology is of limited value for species identification and taxonomy. By using molecular data, the aim was to genetically characterize the species of Crassostrea occurring along the Brazilian coast, and phylogenetically relate these to other Crassostrea from different parts of the world. Sequencing of the partial cytochrome oxidase c subunit I gene (COI), revealed a total of three species of Crassostrea at 16 locations along the Brazilian coast. C. gasar was found from Curuçá (Pará state) to Santos (São Paulo state), and C. rhizophorae from Fortim (Ceará state) to Florianópolis (Santa Catarina state), although small individuals of the latter species were also found at Ajuruteua beach (municipality of Bragança, Pará state). An unidentified Crassostrea species was found only on Canela Island, Bragança. Crassostrea gasar and C. rhizophorae grouped with C. virginica, thereby forming a monophyletic Atlantic group, whereas Crassostrea sp. from Canela Island was shown to be more similar to Indo-Pacific oysters, and either arrived in the Atlantic Ocean before the convergence of the Isthmus of Panama or was accidentally brought to Brazil by ship.

OBJETIVO: Buscar estudos que avaliem a comorbidade entre transtorno de estresse pós-traumático e transtornos do humor, bem como entre transtorno de estresse pós-traumático e outros transtornos de ansiedade. MÉTODO: Revisamos a base de dados do Medline em busca de estudos publicados em inglês até abril de 2009, com as seguintes palavras-chave: "transtorno de estresse pós-traumático", "TEPT", "transtorno de humor", "transtorno depressivo maior", "depressão maior", "transtorno bipolar", "distimia", "transtorno de ansiedade", "transtorno de ansiedade generalizada", agorafobia", "transtorno obsessivo-compulsivo", "transtorno de pânico", "fobia social" e "comorbidade". RESULTADOS: Depressão maior é uma das condições comórbidas mais frequentes em indivíduos com transtorno de estresse pós-traumático, mas eles também apresentam transtorno bipolar e outros transtornos ansiosos. Essas comorbidades impõem um prejuízo clínico adicional e comprometem a qualidade de vida desses indivíduos. Comportamento suicida em pacientes com transtorno de estresse pós-traumático, com ou sem depressão maior comórbida, é também uma questão relevante, e sintomas depressivos mediam a gravidade da dor em sujeitos com transtorno de estresse pós-traumático e dor crônica. CONCLUSÃO: Os estudos disponíveis sugerem que pacientes com transtorno de estresse pós-traumático têm um risco maior de desenvolver transtornos afetivos e, por outro lado, transtornos afetivos pré-existentes aumentam a propensão ao transtorno de estresse pós-traumático após eventos traumáticos. Além disso, vulnerabilidades genéticas em comum podem ajudar a explicar esse padrão de comorbidades. No entanto, diante dos poucos estudos encontrados, mais trabalhos são necessários para avaliar adequadamente essas comorbidades e suas implicações clínicas e terapêuticas.
OBJECTIVE: To review studies that have evaluated the comorbidity between posttraumatic stress disorder and mood disorders, as well as between posttraumatic stress disorder and other anxiety disorders. METHOD: We searched Medline for studies, published in English through April, 2009, using the following keywords: "posttraumatic stress disorder", "PTSD", "mood disorder", "major depressive disorder", "major depression", "bipolar disorder", "dysthymia", "anxiety disorder", "generalized anxiety disorder", "agoraphobia", "obsessive-compulsive disorder", "panic disorder", "social phobia", and "comorbidity". RESULTS: Major depression is one of the most frequent comorbid conditions in posttraumatic stress disorder individuals, but individuals with posttraumatic stress disorder are also more likely to present with bipolar disorder, other anxiety disorders and suicidal behaviors. These comorbid conditions are associated with greater clinical severity, functional impairment, and impaired quality of life in already compromised individuals with posttraumatic stress disorder. Depression symptoms also mediate the association between posttraumatic stress disorder and severity of pain among patients with chronic pain. CONCLUSION: Available studies suggest that individuals with posttraumatic stress disorder are at increased risk of developing affective disorders compared with trauma-exposed individuals who do not develop posttraumatic stress disorder. Conversely, pre-existing affective disorders increase a person's vulnerability to the posttraumatic stress disorder--inducing effects of traumatic events. Also, common genetic vulnerabilities can help to explain these comorbidity patterns. However, because the studies addressing this issue are few in number, heterogeneous and based on a limited sample, more studies are needed in order to adequately evaluate these comorbidities, as well as their clinical and therapeutic implications.

This article reviews the literature on obsessive-compulsive disorders with the objective of discussing the current state of the scientific research on this topic. The authors refer to the international literature and stress Brazilian research, especially that carried out by their own group, PROTOC (Obsessive-Compulsive Spectrum Disorders Project).

The Barbatimão is a tree which bark is rich in tannin. It is used on popular medicine as a wound healing agent, in the treatment of gastric lesions, as anti-leishmanial agent and as anti-inflammatory. Red blood cells (RBC) are labeled with technetium-99m (Tc-99m) and are utilized in many procedures in nuclear medicine. Some authors have reported that drugs (natural and synthetic) can alter the labeling of RBC with Tc-99m. This study evaluates the effect of barbatimão infusion on the labeling of red blood cells (RBC) and plasma (P) proteins with Tc-99m. Heparinized blood from Wistar rats was incubated with NaCl 0.9% as control and different concentrations of barbatimão infusion. Following the addition of stannous chloride (SnCl2), as reducing agent, and Tc-99m, as sodium pertechnetate, the blood samples were centrifuged. P and RBC were separated and were also precipitated with trichloroacetic acid 5%. Insoluble (IF-P and IF-RBC) fractions were isolated. The percentage of radioactivity in all the samples was determined. All the barbatimão infusion concentrations decreased the labeling of RBC, IF-P and IF-RBC. We can speculate that the barbatimão infusion interfered on the labeling of RBC due to the redox properties and/or it can also act as a chelator of the stannous ion.
