Abstract Since the first reported case of COVID-19 in Brazil, the public and private educational system started to close. Up to November 2020, scientific discussions about the return of schooling activities have been rarely performed by the national scientific community and police-makers. The great delay of school returning in Brazil contrasts with successful international strategies of school reopening worldwide and seems counterintuitive with the reopening of non-essential activities. Here, important issues to be considered before and during school reopening are reviewed and discussed. COVID-19 testing is essential to avoid disease spreading, but high cost of individual RT-qPCRs impairs an extensive testing strategy for school returning. To reduce costs and increase the speed of diagnosis, we tested the efficiency of a pooled-sample PCR strategy in a cohort of the educational staff in the city of Macaé/RJ, finding five asymptomatic individuals (0,66%) among the 754 people tested. Thus, a polled-sample PCR testing strategy of the educational staff might prevent infection spreading in schools at a reasonable cost. We discuss how our test strategy could be coupled with internationally recognized safety rules to allow for a safe school return and how countries from different world regions are dealing with educational activities during COVID-19 pandemic.

RESUMO Apesar de sua importância em termos biogeográficos, ecológicos e comerciais, a ictiofauna da costa norte do Brasil ainda é pouco conhecida, representando a porção menos amostrada da Zona Econômica Exclusiva Brasileira. Durante extensivo esforço de inventário na costa norte do Brasil, nós obtivemos uma série de espécimes de Cynoscion similis e concluímos que a espécie é comum nesta região. Embora a espécie não tenha sido relatada anteriormente para o estado do Pará, ela pode ter sido registrada em estudos sobre a pesca industrial, sendo identificada apenas como Cynoscion sp. ou pelo nome comum pescada negra. Isso reforça a necessidade de identificação taxonômica confiável de espécies, com objetivo de garantir dados precisos de ecologia e pesca, e apoiar o desenvolvimento de estratégias efetivas de conservação. Aqui, nós fornecemos novos dados morfológicos e moleculares importantes para a diferenciação de Cynoscion similis e C. jamaicensis, a espécie mais próxima morfologicamente, assim como de outros congêneres.
ABSTRACT Despite its importance in biogeographical, ecological, and commercial terms, the fish fauna of the northern Brazilian coast is still poorly known, representing the least sampled portion of the Brazilian Exclusive Economic Zone. We collected Tonkin weakfish, Cynoscion similis specimens during extensive surveys of the northern Brazilian coast and concluded that C. similis is common in this region. While the species had not previously been reported for the northern Brazilian state of Pará, it may have been recorded in studies of industrial fisheries, being identified only as Cynoscion sp. or by the common name pescada negra. This reinforces the need for the reliable taxonomical identification of species, to guarantee the collection of accurate data on ecology and fisheries, and ultimately, support the development of effective conservation strategies. Here we provide additional morphological and molecular data to distinguish Cynoscion similis from the closely related Cynoscion jamaicensis, and other congeners.

Objective: To present the essential guidelines for pharmacological management of patients with psychomotor agitation in Brazil. Methods: This is a systematic review of articles retrieved from the MEDLINE (PubMed), Cochrane Database of Systematic Reviews, and SciELO databases published from 1997 to 2017. Other relevant articles in the literature were also used to develop these guidelines. The search strategy used structured questions formulated using the PICO model, as recommended by the Guidelines Project of the Brazilian Medical Association. Recommendations were summarized according to their level of evidence, which was determined using the Oxford Centre for Evidence-based Medicine system and critical appraisal tools. Results: Of 5,362 articles retrieved, 1,731 abstracts were selected for further reading. The final sample included 74 articles that met all inclusion criteria. The evidence shows that pharmacologic treatment is indicated only after non-pharmacologic approaches have failed. The cause of the agitation, side effects of the medications, and contraindications must guide the medication choice. The oral route should be preferred for drug administration; IV administration must be avoided. All subjects must be monitored before and after medication administration. Conclusion: If non-pharmacological strategies fail, medications are needed to control agitation and violent behavior. Once medicated, the patient should be monitored until a tranquil state is possible without excessive sedation. Systematic review registry number: CRD42017054440.

Other relevant articles identified by searching the reference lists of included studies were also used to develop these guidelines. Results: We initially selected 1,731 abstracts among 5,362 articles. The final sample included 104 articles that fulfilled all the inclusion criteria. The management of agitated patients should always start with the least coercive approach. The initial non-pharmacological measures include a verbal strategy and referral of the patient to the appropriate setting, preferably a facility designed for the care of psychiatric patients with controlled noise, lighting, and safety aspects. Verbal de-escalation techniques have been shown to decrease agitation and reduce the potential for associated violence in the emergency setting. The possibility of underlying medical etiologies must be considered first and foremost. Particular attention should be paid to the patient’s appearance and behavior, physical signs, and mental state. If agitation is severe, rapid tranquilization with medications is recommended. Finally, if verbal measures fail to contain the patient, physical restraint should be performed as the ultimate measure for patient protection, and always be accompanied by rapid tranquilization. Healthcare teams must be thoroughly trained to use these techniques and overcome difficulties if the verbal approach fails. It is important that healthcare professionals be trained in non-pharmacological management of patients with psychomotor agitation as part of the requirements for a degree and graduate degree. Conclusion: The non-pharmacological management of agitated patients should follow the hierarchy of less invasive to more invasive and coercive measures, starting with referral of the patient to an appropriate environment, management by a trained team, use of verbal techniques, performance of physical and mental assessment, use of medications, and, if unavoidable, use of the mechanical restraint. The sexual dimorphic inguinal glands of the frog species Ololygon centralis (Anura: Hylidae) at light and transmission electron microscopy

ABSTRACT The anuran skin characteristically has different types of glands, most of which are microscopic and are spread throughout the skin. Some species have specialized regions where glands agglomerate, forming macroglands. The description of the external morphology of Ololygon centralis (Pombal & Bastos, 1996) revealed the presence of an inguinal gland. Ololygon centralis is the only species of the genus that has a macrogland. The present study found these inguinal macroglands to be present only on male specimens, thus characterizing it as a sexually dimorphic skin gland. Microscopic analysis revealed that these glands are composed of many syncytial units involved by myoepithelial cells. The center of the syncytium is full of a proteinaceous secretion with a basic pH and the absence of sugar residues. Similar glands observed in other anuran species have been associated with pheromone production, suggesting that the inguinal glands described for O. centralis males may have a similar function.

Resumo Os limnaeideos são importantes na epidemiologia de Fasciola hepatica, uma zoonose negligenciada e endêmica. A interação entre o sistema interno de defesa de Pseudosuccinea columella e F. hepatica tem sido pouco estudada. No presente estudo, investigou-se o efeito da infecção por F. hepatica nos hemócitos circulantes de P. columella. Alterações no número médio de hemócitos circulantes foram observadas aos 30 minutos e 1, 7, 10, 14, 21, 28 e 50 dias após a infecção (dpi). Miracídios foram observados na região cefalopodal e manto aos 30 minutos após a infecção. Miracídio/esporocistos foram observados no colar do manto ao 1 dpi, e esporocistos totalmente formados na região cefalopodal aos 7 dpi. Rédias tornam-se evidentes aos 10 dpi entre a hemocele e músculos a partir de 14 dpi; e rédias com cercárias próximas a glândula digestiva aos 50 dpi. A análise estatística dos hemócitos totais de P. columella infectados por F. hepatica demonstrou diferenças significativas nos 30 minutos pós-infecção e 1, 14, 21 e 28 dpi em comparação aos moluscos não infectados (0 dpi). Portanto, a interferência observada no sistema de defesa interna de P. columella pode ter associação direta com o desenvolvimento de F. hepatica.
Abstract The lymnaeids are important in the epidemiology of Fasciola hepatica, a neglected and endemic zoonosis. The interaction between the internal defense system of Pseudosuccinea columella and F. hepatica has been little studied. In the present study the effect of infection by F. hepatica on P. columella circulating haemocytes was investigated. Changes in the average number of total circulating haemocytes have been observed at 30 minutes post-infection and 1, 7, 10, 14, 21, 28 and 50 days post-infection (dpi). Miracidia were observed head-foot and mantle at 30 minutes post-infection. Miracidia/Sporocysts in the mantle skirt 1 dpi, and fully formed sporocysts were observed in the head-foot at 7 dpi. Rediae became evident at 10 dpi and were located between the haemocoel and the muscles from 14 dpi; 50 dpi, the rediae in the digestive gland contained cercariae. The statistical analysis of the total haemocytes of P. columella infected by F. hepatica showed significant differences on the 30 minutes post-infection and 1, 14, 21, and 28 dpi in comparison to uninfected molluscs (0 dpi). Therefore, the interference observed on the internal defence system of P. columella may have direct association with the development of F. hepatica.

RESUMO Os óleos essenciais apresentam possibilidade de serem empregados nas indústrias de alimentos, bebidas, produtos de higiene pessoal e cosméticos para evitar ou reduzir a deterioração lipídica e a contaminação por micro-organismos. Este trabalho teve como objetivos avaliar as propriedades funcionais antimicrobianas e antioxidantes de óleos essenciais de folhas frescas de Coniza bonariensis, Parthenium hysterophorus, Tithonia diversifolia, Ambrosia polystachya, Hedychium coronarium e Baccharis dracunculifolia, extraídos por hidrodestilação. O potencial antioxidante foi avaliado pelas metodologias do consumo do radical DPPH e da inibição da oxidação do sistema β-caroteno/ácido linoleico. A sensibilidade das bactérias Salmonella Cholerasuis, Listeria monocytogenes, Staphylococcus aureus e Escherichia coli frente aos óleos essenciais foi determinada pela utilização do método de difusão em cavidade ágar. Os óleos essenciais destacaram-se pelo elevado conteúdo de terpenoides. Todos os óleos essenciais avaliados pela metodologia do sequestro do radical DPPH não apresentaram CI50 significativos. Pela metodologia do β-caroteno/ácido linoleico, os óleos essenciais de T. diversifolia e H. coronarium não apresentaram atividades significativas e os de C. bonariensis, P. hysterophorus, A. polystachya, e B. dracunculifolia apresentaram CI50 superiores a maior concentração avaliada. Os óleos essenciais das espécies C. bonariensis, T. diversifolia, H. coronarium e de B. dracunculifolia apresentaram atividade antibacteriana para bactérias Gramnegativas e Gram-positivas, com exceção do óleo volátil de P. hysterophorus, que não impediu o crescimento de nenhuma das cepas bacterianas testadas. O óleo essencial de A. polystachya apresentou potencial antibacteriano apenas nas cepas de S. aureus.
ABSTRACT Essential oils can be employed in the food, drinks, toiletry and cosmetics industries to prevent or reduce lipid deterioration and contamination by microorganisms. The aim of this work was to evaluate the functional antimicrobial and antioxidant properties of essential oils of fresh leaves from Coniza bonariensis, Parthenium hysterophorus, Tithonia diversifolia, Ambrosia polystachya, Hedychium coronarium and Baccharis dracunculifolia, extracted by hydrodistillation. The methodologies used for assessing antioxidant potential were consumption of the DPPH radical and oxidation inhibition of the β-carotene/linoleic acid system. Sensitivity of the bacteria Salmonella cholerasuis, Listeria monocytogenes, Staphylococcus aureus and Escherichia coli to the essential oils was determined using the agar-well diffusion method. The essential oils were noteworthy for their high terpenoid content. None of the essential oils assessed by DPPH radical sequestration presented significant IC50. Using β-carotene/linoleic acid methodology, the essential oils of T. diversifolia and H. coronarium showed no significant activity, and C. bonariensis, P. hysterophorus, A. polystachya and B. dracunculifolia showed IC50 greater than the highest concentration evaluated. The essential oils of C. bonariensis, T. diversifolia, H. coronarium and B. dracunculifolia displayed antibacterial activity for gram-negative and grampositive bacteria, with the exception of the volatile oil of P. hysterophorus, which did not inhibit growth in any of the bacterial strains tested. The essential oil of A. polystachya displayed antibacterial activity only for strains of S. aureus.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

IntroductionPurpureocillium lilacinum is emerging as a causal agent of hyalohyphomycosis that is refractory to antifungal drugs; however, the pathogenic mechanisms underlying P. lilacinum infection are not understood. In this study, we investigated the interaction of P. lilacinum conidia with human macrophages and dendritic cells in vitro.MethodsSpores of a P. lilacinum clinical isolate were obtained by chill-heat shock. Mononuclear cells were isolated from eight healthy individuals. Monocytes were separated by cold aggregation and differentiated into macrophages by incubation for 7 to 10 days at 37°C or into dendritic cells by the addition of the cytokines human granulocyte-macrophage colony stimulating factor and interleukin-4. Conidial suspension was added to the human cells at 1:1, 2:1, and 5:1 (conidia:cells) ratios for 1h, 6h, and 24h, and the infection was evaluated by Giemsa staining and light microscopy.ResultsAfter 1h interaction, P. lilacinum conidia were internalized by human cells and after 6h contact, some conidia became inflated. After 24h interaction, the conidia produced germ tubes and hyphae, leading to the disruption of macrophage and dendritic cell membranes. The infection rate analyzed after 6h incubation of P. lilacinumconidia with cells at 2:1 and 1:1 ratios was 76.5% and 25.5%, respectively, for macrophages and 54.3% and 19.5%, respectively, for cultured dendritic cells.ConclusionsP. lilacinum conidia are capable of infecting and destroying both macrophages and dendritic cells, clearly demonstrating the ability of this pathogenic fungus to invade human phagocytic cells.

As importantes propriedades alimentícias e químicas atribuídas a P. angulata fazem dela uma espécie de grande valor para estudos de conservação de sementes. Este trabalho objetivou avaliar o efeito do pré-condicionamento osmótico de sementes de P. angulata submetidas a diferentes condições e períodos de armazenamento. Lotes de sementes foram armazenados em condições de ambiente e refrigerador até 24 meses. Parte dessas sementes foram osmocondicionadas antes ou após o armazenamento. Os resultados apresentados mostram que houve variação no teor de umidade. As sementes mantiveram taxas de germinação e índice de velocidade de germinação elevadas em ambiente quando osmocondicionadas até 24 meses.
The important chemical and food properties conferred to P. angulata make it necessary to conduct studies in seed conservation. This study evaluated the effect of priming of P. angulata seed for varying periods and storage conditions. Lots of seeds were stored in a refrigerator and in ambient conditions for up to 24 months. Some of these seeds were primed before or after storage. The results show that there was variation in moisture content. The germination rate and germination rate index remained high in ambient conditions when primed up to 24 months.

The aim of this work was to analyze terpene oil production and terpene synthases (TPS) gene expression from leaves at different developmental stages of different chemotypes of Lippia alba (Mill.) N.E. Br. ex Britton & P. Wilson, Verbenaceae. Hydro-distilled essential oil were used for chemical analysis and gene expression of three monoterpene synthase genes called LaTPS12, LaTPS23 and LaTPS25 were used for analyses of gene expression associated to oil production. The putative genes were associated to TPS-b gene class. Semi-quantitative PCR and quantitative PCR (qPCR) analysis were used to investigate the expression profile of those three putative genes in different leaf stages and different chemotypes. Additionally, total oil production and gene expression of putative TPS genes cloned from L. alba chemotype linalool were evaluated at different stages of leaf development. The expression level of those three genes was higher when the highest oil production was observed, mainly in young leaves at the fourth nodal segment for all evaluated chemotypes. Total oil production was higher at leaves that had unopened trichomes. We also observed that the 1mM of MeJA treatment increased the gene expression in all chemotypes after 24 h elicitation.

The individual size of Limnoperna fortunei (Dunker, 1857) was evaluated twice a month, for a year, based on samples from Veludo Beach (30º12'35"S, 51º11'68"W) in Guaiba Lake, southern Porto Alegre, Brazil. Physical and chemical data on water quality, such as temperature, transparency, depth, pH, dissolved oxygen, phosphorus, nitrogen, chlorophyll "a", and conductivity were also recorded. Adult samples were collected from underwater "sarandi" branches, Cephalanthus glabratus (Spreng) K. Schum (Rubiaceae), an aquatic macrophyte available at each site. Adults placed on artificial substrates were also checked every six months. Data were analyzed by applying multivariate statistics techniques related to the physical and chemical variables of the water and the density of individuals to estimate the patterns of the individual growth over the year. The adults were separated into three groups according to length classes: G1, G2 and G3 (groups 1 through 3). These three groups are related to the differences in behavior, namely, the ability of locomotion and fixation capacity. Individuals from Group 1 (5 to 7 mm in length) were the most abundant and were present in all the sampled months. Adults from artificial substrates were shown to display significant differences in seasonal growth and faster growth during spring and summer months.

Hemoglobinúria paroxística noturna (HPN) é uma doença clonal adquirida da célula tronco hematopoética em decorrência de uma mutação somática no gene PIG-A, causando inabilidade dos eritrócitos, leucócitos e plaquetas de se protegerem contra lise mediada pelo sistema complemento. Assim, avaliamos a eficiência dos testes de triagem para HPN (teste de Ham e pesquisa dos antígenos CD55 e CD59 em coluna de gel) utilizando a citometria de fluxo (CMF), que é capaz de detectar e quantificar o clone HPN. Inicialmente, selecionamos 63 pacientes que foram testados pelo teste de Ham entre janeiro/2003 e dezembro/2004, na Fundação Hemope. Destes, 15 tiveram seus testes positivos para HPN. O critério de inclusão dos casos para avaliação por CMF foi a obtenção de resultados do teste em gel concordantes com o teste de Ham positivo. Dessa maneira, quatro pacientes foram incluídos no grupo de estudo. Foram adicionados a esse grupo dois casos que exibiam clínica exuberante da doença, mas tiveram os resultados discordantes, explicado pelo fato de que o teste em gel foi realizado após terapia transfusional recente, provocando a suspeita de falso resultado normal. Submetemos esses seis casos à CMF, os quais todos se mostraram verdadeiros portadores da doença através da confirmação da existência do clone HPN em eritrócitos e granulócitos, em expressões variáveis. Os resultados da CMF comprovaram a limitação dos testes de triagem além de demonstrarem a relevância da citometria em identificar variações de intensidade do clone, garantindo inclusive o diagnóstico preciso em pacientes previamente transfundidos.
Paroxysmal nocturnal hemoglobinuria (PNH) is an acquired clonal disease of the hematopoietic stem cell caused by a somatic mutation in the PIG-A gene, resulting in the inability of erythrocytes, granulocytes and platelets to protect themselves against complement system mediated lysis. Thus, PNH screening tests (Ham's test and CD55 e CD59 proteins investigation through gel column agglutination) were evaluated using flow cytometry, a test useful to detect and measure the PNH clone. Initially, 63 patients evaluated using the Ham's test between January 2003 and December 2004 from the Hemope Foundation were selected. From these, 15 cases were positive for PNH. The inclusion criterion for cytometry evaluation was a positive Ham's test. Thus, four patients were included in the study group. Furthermore, two cases with clinical symptoms of the disease but with negative results for PNH were included in this group. Negative results were explained by the gel test being performed after blood transfusion, giving a suspicion of false negative results. These six cases were submitted to flow cytometry with all cases proving to be positive for the disease as the PNH clone was confirmed, to different degrees, in both erythrocytes and granulocytes. The flow cytometry results proved the limitation of screening tests as well as showing the importance of cytometry in the identification of the intense variations of clone guaranteeing precise diagnosis in previously transfused patients.

Desde o final de 1998, um pequeno bivalve invasor, o "mexilhão dourado", Limnoperna fortunei (Dunker, 1857), oriundo do sudeste asiático, está presente no sul do Brasil. Foi provavelmente transportado, não intencionalmente, através de água de lastro para a Bacia do Guaíba, Rio Grande do Sul, Brasil. Em nosso meio, este molusco vem causando problemas de "macrofouling" com grandes prejuízos econômicos e danos à fauna e flora. As coletas foram qualitativas e quantitativas quinzenais no período de um ano, no lago Guaíba, Praia do Veludo (30°12'35"S, 51°11'68"W), ao sul do município de Porto Alegre. Utilizou-se rede de plâncton com abertura de malha equivalente a 36 mm, filtrando-se a quantidade de 30 litros de água. Descrevem-se brevemente as diferentes fases larvais com parâmetros do comprimento. Primeiramente reconhece-se um estágio ciliado, desenvolvendo-se em trocófora (comprimento de 80 µm a 125 µm) com quatro fases distintas, e valvadas com a larva "D" (120 µm a 150 µm), o veliger de charneira reta (150 µm a 190 µm), o veliger umbonado (190 µm a 220 µm) e o pediveliger (220 µm a 250 µm). Quando pós-larvas ou plantígradas (comprimento em torno de 300 µm), começam a secretar o fio de bisso, permitindo a fixação ao substrato. Constatou-se a presença de larvas durante todos os meses amostrados, com picos nos meses de outubro a dezembro.
Since the end of 1998 a small invasive bivalve, the golden mussel Limnoperna fortunei (Dunker, 1857), originating from Southeast Asia, is present in Southern Brazil. It was probably brought not intentionally in ballast water to the Guaíba Basin in Rio Grande do Sul State. In this environment, the golden mussel is causing macrofouling very rapidly with grate financial loss and damages to the native fauna and flora. Quali-quantitative samplings twice a month were performed during one year in the Guaíba lake, at Veludo Beach (30°12'35"S, 51°11'68"W) southward of the municipality of Porto Alegre. Each time, 30 litters of water were filtered through plankton net with a mash opening of 36 mm. The different larval stages with the length parameters were briefly described. Different stages were recognized: five without valves and four valved. The first one recognized as a ciliated stage develops into the trocophora (length from 80 µm to 125 µm) with four distinct stages. The valved stages include: the "D" - shaped (length 120 µm to 150 µm), the straight-hinged veliger (length 150 µm to 190 µm), umbonated-veliger (length 190 µm to 220 µm) and pediveliger (length 220 µm to 250 µm). When pos-larvae or plantigrades (length about 300 µm), they secrete the byssus thread that permit fixation on the subtract. Larvae were present during all the samplings with strong October-December peaks.
