Toads of the Rhinella genus have a pair of paratoid glands that store biological secretions of high toxicity and varied chemical composition, rich in biologically active compounds. The present work aimed to carry out the investigation of the metabolomic profile and evaluation of the biological potential of the secretion paratoid glands (PGS) from Rhinella granulosa. The paratoid secretion was collected in the Piauí state (Brazil), extracted with methanol and the extract was analyzed by ultra-performance liquid chromatography with quadrupole time-of-flight mass spectrometry. Fifty chemical constituents were identified. The extract showed cytotoxicity against tumor cell lines of the central nervous system (half maximal inhibitory concentration (IC50) = 1.9 µg mL-1) and prostate (IC50 = 1.6 µg mL-1), unsatisfactory antimicrobial potential (minimal inhibitory concentration (MIC) > 312 µg mL-1) and inhibited the enzyme acetylcholinesterase (IC50 = 5.119 mg mL-1). The results presented relevant information about the PGS and contributed to the understanding of the metabolomic and biological potential of R. granulosa.

Abstract This work describes the circulation over the continental slope and the São Paulo Plateau in the Santos Basin during the SANSED winter 2019 survey. The cruise consisted of four legs in the period between June, 11 and August, 03 2019. The observed circulation is dominated by the Atlantic southwestern boundary current regime and remotely-generated anticyclones and cyclones. The former is composed by the Brazil Current, the Intermediate Western Boundary Current and their mesoscale meanders; the latter are 300km vortical rings with origin in the eastern side of the South Atlantic Basin. A Lagrangian scheme applied over satellite altimeter maps indicate that the origin of these rings is primarily the Cape Basin of South Africa. The interaction between the boundary currents, their cyclonic meanders, and the anticyclonic rings is complex, and varies widely. During the SANSED winter 2019 survey period, three anticyclones interacted with the Brazil Current, instabilizing it, forming dipoles with the current cyclonic meanders, leading to their downstream propagation. Ancienter cyclonic eddies within Santos Basin may interfere with the propagation of the large anticyclones further south. In addition, the continuous arrival of remotely-originated anticyclones, the larger portion over the São Paulo Plateau presented a tendency of counter-clockwise circulation during the whole cruise period.

ABSTRACT The expression ‘you need to know to conserve’ is a well-known cliche among biologists. Documenting the richness of a group of organisms is the first step towards understanding biodiversity and preparing efficient conservation plans. In this context, many efforts have been made to quantify the number of species on Earth and estimate the number of species still unknown to science. A few countries have complete and integrated databases estimating the approximate number of species recorded for their territory, particularly in the Global South. In Brazil, a country of continental dimensions, revealing the richness of the second most diverse clade of invertebrates (=Mollusca) has been a goal of taxonomists. Recently, in an unprecedented, collective, and integrated effort among Brazilian malacologists, it was possible to estimate how many valid species of molluscs are there in Brazil. In this effort, more than 30 mollusc experts joined together to update the Taxonomic Catalogue of the Brazilian Fauna (TCBF), a governmental website that allows a quick and real-time updating of all Metazoan. So far, more than 5,000 updates have been made in TCBF, indicating the presence of 3,552 valid species of molluscs in Brazil, distributed among the main clades as follows: Caudofoveata (10 spp.), Solenogastres (6 spp.), Polyplacophora (35 spp.), Scaphopoda (43 spp.), Cephalopoda (92 spp.), Bivalvia (629 spp.) and Gastropoda (2,737 spp.). The present study, in addition to demonstrating for the first time the richness of Brazilian molluscs, also presents the state of the art of this important phylum of invertebrates highlighting its most representative and neglected groups.

ABSTRACT Semi-mechanized and mechanized harvesting use machines that promote the transference of vibrational energy and impact to achieve the detachment of coffee fruits. The aim of this study was to evaluate the vibration transmissibility in coffee fruit-peduncle systems, using high-speed cameras, submitted to high frequency harmonic excitation in different combinations between frequency and amplitude of vibration, identifying working ranges suitable to perform selective harvesting. Vibration transmissibility was determined for the coffee fruit-peduncle systems, for the maturation stages unripe and ripe that were subjected to a sinusoidal harmonic displacement, in which the input parameters were frequency (35, 45 and 55 Hz) and peak-to-peak amplitude (3.5, 5.0 and 6.5 mm). An experiment was used to study the effect of frequency and amplitude on vibration transmissibility in a completely randomized design in a factorial scheme 3 x 3 x 2, with three replications. The frequency of 35 Hz, associated with the amplitudes 3.5-6.5 mm, was the one that most influenced the results of vibration transmissibility. For the frequency of 55 Hz and amplitude of 6.5 mm, in the ripe maturation stage, the vibration transmissibility was higher than 1.0, which could be a suitable combination for selective coffee harvesting.

ABSTRACT Sorghum ( Sorghum bicolor L. Moench) is a crop of paramount importance for Brazil, used in human and animal food. Considering the reduced number of studies on the influence of using a densimetric table in seed processing and its respective reprocessing to the equipment, this study aimed to evaluate the physiological quality of processed sorghum seeds reprocessed in a densimetric table and stored for three months. The treatments consisted of four collection points in the terminal part of the densimetric table, representing the processing material: upper part (P1), upper middle part (P2), middle part (P3), and lower part (D). Subsequently, the samples that were reprocessed in the table came from P3 and were divided into the upper part, which was followed to bagging (P3R1), and the lower part (P3R2), which followed for disposal, totaling six treatments. Storage was performed in a refrigerated environment, using a completely randomized design, with the means being compared by Tukey’s test at 5%. We concluded that the reprocessing of sorghum seeds in the densimetric table can improve the physical quality of a lot due to its standardization at the end of the stages, contributing to the improvement of the physiological quality and use of the material that would be discarded. The equipment performance allows the removal of seed fractions with lower apparent specific mass and promotes positive stratifications in the physiological quality of sorghum seed lots.

Abstract Introduction Burnout syndrome (BS) in healthcare professionals (HCP) has been a major concern, and even more so during the coronavirus disease 2019 (COVID-19) pandemic. The need for adequate tools to assess BS is urgent. The objective of this study was to validate the Brazilian Portuguese version of the Copenhagen Burnout Inventory (CBI) in HCP. Methods The sample comprised 1,054 Brazilian HCP. Data were collected for 1 month (May-2020 to June-2020) using an online self-administered questionnaire. Results All three CBI dimensions demonstrated optimal reliability. All consistency measures attained values > 0.90. Split-half correlation values with Spearman-Brown reliability were higher than 0.8. The parallel analysis suggested two factors: personal burnout (PB) and work-related burnout (WB) items were associated with factor 1, and client-related burnout (CB) items were associated with factor 2. Conclusion Our study corroborates the validity of the Brazilian Portuguese version of the CBI, pointing to a close relation between PB and WB in HCP. A public domain tool with evidence quality to ensure sufficient content validity can aid in burnout evaluation and encourage both expansion of the research field and accurate detection and treatment of this syndrome in Brazilian HCP.

ABSTRACT: We report a case of a free-ranging five-month wildcat with bilateral hind limbs paralysis since birth due to a segmental lumbar spinal cord aplasia. The species confirmation of the southern tiger cat (Leopardus guttulus) was determined by genetic sequencing. This southern tiger cat native to Brazil had autophagy in both pelvic limbs during the initial phase of hospitalization, followed by a right tibial fracture with bone exposition. Euthanasia was chosen due to animal welfare and submitted for postmortem examination. Grossly, there was an 8.5cm in-length segmental interruption of the spinal cord between the third and fifth lumbar vertebrae, with a lack of spinal cord tissue and collapsed associated dura mater. Microscopically, the representative sections of the L3 to L5 spinal cord had only .an irregular trace of gray matter adhered to the meninges (lumbar spinal cord aplasia) In the region of L6, a focally extensive, cystic, and well-defined tubular cavitation was noted dorsally to the central canal, replacing and compressing the adjacent nervous tissue (syringomyelia). Metagenomics examination did not detect any virus responsible for the presented spinal cord malformations. This seems to be the first description of segmental spinal cord aplasia reported in a wild feline.

RESUMO: O objetivo do presente relato foi descrever um caso de aplasia segmentar da medula espinhal lombar em um felino silvestre, com aproximadamente cinco meses, resgatado de seu ambiente natural, apresentando paralisia bilateral dos membros posteriores. A espécie gato-do-mato-pequeno (Leopardus guttulus) foi determinada por sequenciamento genético. Após curto período de hospitalização, iniciou autofagia de ambos os membros pélvicos, seguido de fratura com exposição óssea. Optou-se pela eutanásia e o cadáver foi encaminhado ao Setor de Anatomia Patológica da Universidade Federal Rural do Rio de Janeiro para necropsia. Macroscopicamente, havia uma interrupção segmentar grave da medula espinhal entre a terceira e a quinta vértebras lombares, medindo 8,5cm de comprimento, com resquícios de tecido nervoso e com meninges colapsadas. Ao exame histológico, em seções da medula espinhal na região de L3 a L5, .havia apenas vestígio de tecido nervoso aderido às meninges, morfologicamente compatível com substância cinzenta (aplasia de medula espinhal lombar). Na região de L6 notou-se áreas multifocais com cavitações tubulares, císticas e bem delimitadas, dorsalmente ao canal central substituindo e comprimindo o tecido nervoso adjacente (siringomielia). O exame de metagenômica não detectou qualquer vírus responsável pelas malformações da medula espinhal. Com base no histórico, sinais clínicos, necropsia e achados histológicos, o diagnóstico de aplasia segmentar grave com siringomielia foi estabelecido em um L. guttulus.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Abstract Mucus, produced by Palythoa caribaeorum has been popularly reported due to healing, anti-inflammatory, and analgesic effects. However, biochemical and pharmacological properties of this mucus remains unexplored. Therefore, the present study aimed to study its proteome profile by 2DE electrophoresis and MALDI-TOF. Furthermore, it was evaluated the cytotoxic, antibacterial, and antioxidant activities of the mucus and from its protein extract (PE). Proteomics study identified14 proteins including proteins involved in the process of tissue regeneration and death of tumor cells. The PE exhibited cell viability below 50% in the MCF-7 and S-180 strains. It showed IC50 of 6.9 μg/mL for the J774 lineage, and also, favored the cellular growth of fibroblasts. Furthermore, PE revealed activity against Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, and Staphylococcus epidermidis (MIC of 250 μg/mL). These findings revealed the mucus produced by Palythoa caribaeorum with biological activities, offering alternative therapies for the treatment of cancer and as a potential antibacterial agent.

The cariogenic processes are mainly caused by the bacterium Streptococcus mutans (S. mutans) and consist of the demineralization of the tooth that occurs when the acid production overcomes the natural repair or if a problem occurs in the last one. In this work, we performed the synthesis of twenty-one 1,4-naphthoquinones tethered to 1,2,3-1H-triazoles (8a-8k and 9a-9j), antibacterial evaluation against the S. mutans in vitro and the acute toxicity of the better ones in vivo. We observed strong inhibition results in the disc diffusion test ranging, the halos of inhibitions, from 18.66 (± 0.57) to 29 (± 2.64) mm, and good values in the minimum inhibitory concentration (5 to 50 μg), for the compounds 9e, 9h, 9i and 9j: Furthermore, they do not have a cytotoxic effect at the concentrations tested. Besides that, in the in vivo test, they show some slight alteration in the histopathological analyses and the biochemistry. Thus, we found four potential candidates to become instruments for the treatment of cavities.

Abstract Microbial biomass is considered a renewable and environmentally friendly resource. Thus, the research conducted a kinetic study and thermodynamic equilibrium modeling of the cobalt (Co) and manganese (Mn) bioadsorption process using the Rhodococcus opacus (RO) strain as a biosorbent. The inactive biomass subjected to 0.1 M NaOH pretreatment was brought into contact with synthetic solutions of Co and Mn. The experimental data for the Co(II) and Mn(II) bioadsorption process were fit to the Langmuir model with kads of 0.65 and 0.11 L.mg-1, respectively. A better statistical fit was also obtained for the pseudo-second order kinetic model (R2Co(II) = 0.994 and R2Mn(II) = 0.995), with 72.3% Co(II) and 80% Mn(II) removals during the first 10 min. In addition, a higher affinity of RO for the Co(II) ion was observed, with maximum uptake values of 13.42 mg.g-1; however, a higher adsorption rate was observed for Mn(II) ion (k = 0.21 g.mg-1.min-1 at 318 K). The bioadsorption process was spontaneous and dependent on temperature, being endothermic and irreversible for the Co(II) ion (∆H = 2951.91 J.mol-1) and exothermic and reversible for the Mn(II) ion (∆H = -2974.8 J.mol-1). The kinetic and thermodynamic equilibrium modeling allowed to identify the main mechanisms involved in the biosorption process of both metals.

ABSTRACT Objectives: Currently, not much is known about the interactions between voice and growth hormone (GH). We have described large kindred with isolated GH deficiency (IGHD) due to a GHRH receptor mutation, resulting in severe short stature and high-pitched voice. These IGHD individuals have little interest in GH treatment, as they consider themselves “short long-lived people”, rather than patients. Interestingly, they report normal general quality of life, but they rate their Voice-Related Quality of Life (V-RQOL) as low. Here, we assessed the social and auditory-perceptual impacts of artistic-intervention voice therapy with semioccluded vocal tract exercises (SOVTE) and choral singing, on their voices. Material and methods: Seventeen GH-naïve adult IGHD individuals were enrolled in a single-arm interventional pre-post study with 13 weekly sessions of choir singing over 90 days. Outcome measures were V-RQOL scores, self-assessment of voice, and auditory-perceptual analysis (GRBAS scale, G: grade of the severity of dysphonia; R: roughness; B: breathiness; A: asthenia; and S: strain). Results: Marked improvements in total (p = 0.0001), physical (p = 0.0002), and socioemotional (p = 0.0001) V-RQOL scores and in self-assessment of voice (p = 0.004) were found. The general grades of vocal deviation (p = 0.0001), roughness (p = 0.0001), breathiness (p = 0.0001) and strain (p = 0.0001) exhibited accentuated reductions. Conclusions: Voice therapy with semioccluded vocal tract exercises and choral training improved social impact and perceptual voice assessments in IGHD subjects and markedly improved their voice-related quality of life. This is particularly important in a setting where GH replacement therapy is not widely accepted.

Abstract Background Endogenous phospholipases A2 (PLA2) play a fundamental role in inflammation, neurodegenerative diseases, apoptosis and cellular senescence. Neurotoxins with PLA2 activity are found in snake venoms from the Elapidae and Viperidae families. The mechanism of action of these neurotoxins have been studied using hippocampal and cerebellar neuronal cultures showing [Ca2+]i increase, mitochondrial depolarization and cell death. Astrocytes are rarely used as a model, despite being modulators at the synapses and responsible for homeostasis and defense in the central nervous system. Preserving the cell division ability, they can be utilized to study the cell proliferation process. In the present work cultured astrocytes and glioblastoma cells were employed to characterize the action of β-micrustoxin (previously named Mlx-9), a PLA2 isolated from Micrurus lemniscatus snake venom. The β-micrustoxin structure was determined and the cell proliferation, cell cycle phases and the regulatory proteins p53, p21 and p27 were investigated. Methods β-micrustoxin was characterized biochemically by a proteomic approach. Astrocytes were obtained by dissociation of pineal glands from Wistar rats; glioblastoma tumor cells were purchased from ATCC and Sigma and cultured in DMEM medium. Cell viability was evaluated by MTT assay; cell proliferation and cell cycle phases were analyzed by flow cytometry; p53, p21 and p27 proteins were studied by western blotting and immunocytochemistry. Results Proteomic analysis revealed fragments on β-micrustoxin that aligned with a PLA2 from Micrurus lemniscatus lemniscatus previously identified as transcript ID DN112835_C3_g9_i1/m.9019. β-micrustoxin impaired the viability of astrocytes and glioblastoma tumor cells. There was a reduction in cell proliferation, an increase in G2/M phase and activation of p53, p21 and p27 proteins in astrocytes. Conclusion These findings indicate that β-micrustoxin from Micrurus lemniscatus venom could inhibit cell proliferation through p53, p21 and p27 activation thus imposing cell cycle arrest at the checkpoint G2/M.

Abstract: COVID-19 rapidly spread across the world in an unprecedented outbreak with a massive number of infected and fatalities. The pandemic was heavily discussed and searched on the internet, which generated big amounts of data related to it. This led to the possibility of attempting to forecast coronavirus indicators using the internet data. For this study, Google Trends statistics for 124 selected search terms related to pandemic were used in an attempt to find which keywords had the best Spearman correlations with a lag, as well as a forecasting model. It was found that keywords related to coronavirus testing among some others, such as “I have contracted covid”, had high correlations (≥0.7) with few weeks of lag (≤4 weeks). Besides that, the ARIMAX model using those keywords had promising results in predicting the increase or decrease of epidemiological indicators, although it was not able to predict their exact values. Thus, we found that Google Trends data may be useful for predicting outbreaks of coronavirus a few weeks before they happen, and may be used as an auxiliary tool in monitoring and forecasting the disease in Brazil.
