Ambuic acid (AA) is a highly-modified cyclohexenone and known as a promising inhibitor of quorum sensing in methicillin-resistant Staphylococcus aureus, and is thus a candidate as an antivirulence drug. This molecule is mainly produced by the species of Pestalotiopsis and, since its discovery twenty years ago, only a restricted amount of AA-derivatives have been described. Despite being a promising subject, methods for the analysis of modified AA-analogues via mass spectrometry remain unexplored. In order to adress this question, the marine fungus Pestalotiopsis sp. 4A11 associated with the ascidian Didemnum perlucidum was grown in a solid rice medium and its crude extract was chemically studied. From this extract, AA and 10-hydroxy ambuic acid (10-HAA) were isolated and identified using NMR spectroscopy with the aim of obtaining model compounds for the MS analysis. These served as reference compounds (seeds) to guide the dereplication of other AA-analogues via LC-MS/MS-based molecular networking. Based on the manual interpretation of the fragmentation pathways of the seeds and related compounds observed in the networks, six AA-derivatives were dereplicated in the extract. Furthermore, three analogues with unprecedented chemical formulas were proposed as putative unprecedented AA-derivatives. The fragmentation annotation proposed represents a fast and feasible method for characterizing AA-derivatives.

Thallium (Tl) is a rare metal with toxic effects notably harmful to human health and the environment. In this article, the natural occurrence of Tl was evaluated in a deactivated manganese mine located in the western region of Bahia, Brazil. The extraction procedure with aqua regia was used to dissolve the soil, sediment, and tailings samples. The Tl content was determined by differential pulse anodic stripping voltammetry (DPASV) and manganese (Mn) and iron (Fe) by flame atomic absorption spectroscopy (FAAS). The Tl levels ranged from 0.64 to 473 mg kg-1, triggering an alert since most of the samples (soil and sediment) analyzed presented Tl concentrations above 1.0 mg kg-1, recognized as the maximum limit recommended by environmental agencies in North America and Europe.

RESUMEN Objetivos: este estudio investigó la bioactividad del extracto de hoja en bruto (EHB) y las fracciones hexano (HX) y acetato de etilo (AcOEt) de Talinum paniculatum solo y en asociación con fluconazol (FLC) contra cepas de referencia y aislados clínicos de Candida albicans resistente a FLC. Además, evaluó la capacidad antioxidante, la composición química de esta planta y los mecanismos subyacentes del efecto fungicida. Métodos: la actividad antifúngica se evaluó mediante microdilución en caldo para establecer las concentraciones inhibitorias mínimas (CIM) y microbicidas mínimas (CMM). Durante el tratamiento con FLC y productos vegetales se detectó la generación de especies reactivas de oxígeno (ERO) (radicales hidroxilo [HO●]) en células de C. albicans utilizando las sondas fluorescentes permeables a la membrana APF y HPF. El perfil de cromatografía líquida de alta resolución (CLAR), el análisis cuantitativo de compuestos antioxidantes y el ensayo DPPH fueron evaluados. Resultados: el EHB y las fracciones presentaron una excelente actividad antifúngica y selectividad contra las células de C. albicans, pero no tuvieron efectos sinérgicos con FLC. Los valores de CIM para EHB y sus fracciones contra la cepa referencia de C. albicans fueron del orden de: HX (31,25 μg ml-1) < AcOEt (62,5 μg ml-1) < EHB (500 μg ml-1), y contra C. albicans resistente a FLC: HX (125 μg ml-1)= AcOEt < EHB (500 µg ml-1). EHB y sus fracciones fueron más potentes antifúngicos que FLC contra los aislados clínicos. Además, estos productos vegetales tienen efectos fungicidas contra C. albicans resistentes a FLC, esto conirmó el potencial antifúngico. Por el contrario, durante la asociación se demostró que los productos vegetales causan un aumento en la CIM de FLC de 2 a 16 veces. La exposición a FLC aumentó los niveles de ERO (HO●) en las células de C. albicans. Los aumentos en las CIM de FLC se debieron a la acción de los antioxidantes presentes en EHB y sus fracciones para prevenir la inhibición del crecimiento mediada por ERO inducida por FLC en C. albicans. Conclusión: T. paniculatum puede ser una fuente de compuestos bioactivos con potencial antifúngico. Sin embargo, debido al uso común de su hoja comestible, se recomienda usarla con precaución durante la terapia con FLC (ya que puede disminuir la susceptibilidad a FLC).
SUMMARY Aims: This study investigated the bioactivity of the crude leaf extract (CLE) and fractions hexane (HX) and ethyl acetate (EtOAc) from Talinum paniculatum alone and in association with fluconazole (FLC) against reference strain and clinical isolates of FLC-resistant Candida albicans. Furthermore, the antioxidant capability, chemical composition of this plant, and the effect's underlying mechanisms were evaluated. Methods: The antifungal activity was evaluated using checkerboard assay to establish the minimum inhibitory (MIC) and minimum microbicidal concen trations (MMC). During FLC and plant products challenges, the reactive oxygen species (ROS) generation (hydroxyl radicals [HO●]) were detected in C. albicans cells using the membrane-permeable fluorescent probes APF and HPF. High-performance liquid chromatography (HPLC) profile, quantitative analysis of antioxidant compounds, and free radical scavenging activity (DPPH assay) tests were performed. Results: The CLE and fractions presented outstanding antifungal activity and selectivity against C. albicans cells but had no synergistic effect’s with FLC. The MIC values for CLE and its fractions against C. albicans reference strain were in the order of HX (31.25 µg ml-1) < EtOAc (62.5 μg ml-1) < CLE (500 μg ml-1), and against FLC-resistant C. albicans HX (125 μg ml-1) = EtOAc < CLE (500 μg ml-1). CLE and its fractions had more potent antifungal activities than FLC against the clinical isolates. Moreover, fungicidal effect’s for these plant products were demonstrated against FLC-resistant C. albicans, which further conirmed an antifungal potential. Conversely, during association, plant products were shown to cause an increase in FLC MIC anywhere from 2- to 16-fold. FLC exposure led to an increase in the steady-state levels of ROS (HO●) in C. albicans cells. Next, we found that the increases in FLC MICs were owing to action of antioxidants containing-CLE and its fractions in preventing FLC-induced ROS-mediated growth inhibition of C. albicans. Conclusion: T. paniculatum can be a source of bioactive compounds with antifungal potential. However, because of the common use of its edible leaf, caution is advised during therapy with FLC (since it can decrease FLC susceptibility).

RESUMO A busca de novas alternativas para reutilizar resíduos é uma maneira de minimizar impactos ambientais promovendo práticas sustentáveis. Este trabalho analisou a viabilidade técnica da produção de briquetes a partir de resíduos de poda urbana e bagaço de cana-de-açúcar, transformando-os em um produto com maior valor agregado, além de direcionar seu uso e reduzir a disposição inadequada. Foram estudados cinco tratamentos a partir de diferentes proporções dos dois resíduos: sendo T1, T2, T3, T4 e T5, respectivamente, 100% de resíduo de poda urbana; 50% de resíduo de poda urbana e 50% de bagaço de cana; 24% de resíduo de poda urbana e 75% de bagaço de cana; 10% de resíduo de poda urbana e 90% de bagaço de cana; e 100% de bagaço de cana. Os materiais foram caracterizados física, química e energeticamente. Foram produzidos briquetes e avaliados quanto ao desempenho mecânico, com testes de expansão, resistência mecânica e friabilidade para verificar a qualidade do produto final. Os tratamentos 1 e 2 apresentaram os melhores resultados, com menor friabilidade e valor médio de resistência mecânica de 167,14 e 107,56 kgf, respectivamente. Conclui-se que ambos os resíduos (bagaço de cana e poda urbana) apresentaram potencial para produção de briquete, bem como a mistura entre os dois materiais.
ABSTRACT The search for new alternatives in order to reuse residues is one way to minimize environmental impacts by promoting sustainable practices. This study analyzed the technical feasibility of briquettes production from urban pruning residue and sugarcane bagasse, turning them into a product with higher added value besides directing their use and reducing their improper disposal. Five treatments were studied from different ratios of the two residues: T1, T2, T3, T4 and T5, respectively made of 100% of urban pruning; 50% urban pruning and 50% sugarcane bagasse; 25% urban pruning and 75% sugarcane bagasse; 10% urban pruning and 90% sugarcane bagasse; 100% sugarcane bagasse. The materials were characterized physically, chemically and energetically. Briquettes were produced and evaluated for mechanical performance, with expansion tests, mechanical resistance and friability to verify the quality of the final product. The treatments 1 and 2 showed the best results, with lower friability and average values of mechanical resistance of 167.14 and 107.56 kgf, respectively. It is concluded that both residues (sugarcane bagasse and urban pruning) had potential for briquette production, as well as the mixture between both materials.

RESUMO Os painéis de madeira reconstituída são muito empregados em vários ramos da indústria da madeira, como nos setores moveleiros e vários segmentos da construção civil urbana e rural. No Brasil são gerados resíduos agrícolas e florestais abundantemente, tornando viável o estudo, desenvolvimento e aplicação de materiais alternativos e sustentáveis, produzidos a partir do aproveitamento destes resíduos. O objetivo deste trabalho foi produzir painéis de partículas homogêneos de alta densidade, com resíduos de madeira de Pinus elliottii e casca de aveia (Avena sativa), prensados com dois tipos de adesivo, poliuretano à base de óleo de mamona e melamina formaldeído, e avaliar o desempenho físico destes painéis. O desempenho físico dos painéis foi avaliado com base nas normas ABNT NBR 14810:2006, ANSI A208.1:1999 e BS EN 312:2003. Foi realizado uma análise de variância (ANOVA) para testar a influência dos fatores percentual de casca de aveia, percentual de adesivo e tipo de adesivo, e as interações entre estes fatores nas propriedades físicas dos painéis. Os resultados apontaram propriedades físicas, em alguns tratamentos superiores aos requisitos estipulados por normas consultadas. Conclui-se que os painéis produzidos com o adesivo poliuretano à base de óleo de mamona apresentaram melhor desempenho físico, se comparado aos painéis produzidos com o adesivo melamina formaldeído.
ABSTRACT Reconstituted wood panels are widely used in various branches of the wood industry, such as in the furniture sectors and various segments of urban and rural construction. In Brazil, agricultural and forestry residues are abundantly generated, making feasible the study, development and application of alternative and sustainable materials, produced from the use of these wastes. The aim of this study was to produce high-density homogenous panels with wood residues of Pinus elliottii and oat hulls (Avena sativa), pressed with two types of adhesive, castor oil-based polyurethane and melamine formaldehyde, and to evaluate the physical performance of these panels. The physical performance of the panels was evaluated based on the ABNT NBR 14810: 2006, ANSI A208.1: 1999 and BS EN 312: 2003 standards. Analysis of variance (ANOVA) was performed to test the influence of the factors percentage of oat hull, percentage of adhesive and type of adhesive, and the interactions between these factors on the physical properties of the panels. The results indicated physical properties, in some treatments superior to the requirements stipulated by the consulted standards. It was concluded that the panels produced with the castor oil-based polyurethane adhesive showed better physical performance when compared with the panels produced with the melamine formaldehyde adhesive.

ABSTRACT The use of animals from dairy farms is an alternative to meat production since it provides an increment of total income for farmers. This study aims to evaluate the performance of Holstein calves finished in two feeding systems (feedlot or pasture). Forty-three animals with 58 days old and 57 kg were divided in two treatments: 23 animals finished in feedlot with corn silage plus concentrate based on corn and soybean meal (40:60); 20 animals kept in cultivated pastures according to the period of the year: Italian ryegrass (Lolium multiflorum) and pearl millet (Pennisetum americanum) with supplementation with the same feedlot-concentrate at 1% body weight. Animals were slaughtered with 200 kg. Dry matter and nutrient intake were determined, with the use of chromium oxide for estimating pasture intake. Feedlot animals had greater total intake and total digestible nutrients, resulting in higher average daily gain (0.949 vs 0.694 kg day-1). Crude protein intake, neutral detergent fiber and feed conversion did not show significant differences. Holstein calves have improved performance when finished in feedlot.

Resumo A Bacia Hidrográfica do rio Bacanga (BHRB) apresenta problemas ambientais relacionados a urbanização e lançamentos de esgoto in natura que comprometem a qualidade desse ecossistema. Devido ao reduzido número de estudos associados à avaliação da qualidade da água no local, este estudo teve como objetivo avaliar a situação atual desse ecossistema por meio do Índice de Proteção da Vida Aquática. Seis amostragens bimestrais foram realizadas em seis pontos ao longo da bacia, coletando parâmetros físico-químicos para a aplicação do Índice de Estado Trófico e Índice de Parâmetros Mínimos para a Proteção da Vida Aquática, relacionando-os com a resolução Conselho Nacional do Meio Ambiente - CONAMA/357. Os resultados revelaram alterações significativas na qualidade da água da BHRB, o aumento de nutrientes e das concentrações de clorofila-a conduziram ao um estado geral de eutrofização. Os valores de surfactantes foram altos colocando em risco a biota aquática e as taxas de oxigênio dissolvido estiveram abaixo do permitido pela resolução na maioria dos pontos amostrados. A situação atual da qualidade da água para proteção da vida aquática é péssima em 61,92% dos pontos amostrados.
Abstract Bacanga River Basin faces environmental problems related to urbanization and discharge of untreated domestic sewage, which compromise its ecosystem health. Due to the small number of studies that assessed its water quality, the present study aimed to assess the current status of this ecosystem based on the aquatic life protection index. Samples were carried out every two months, in a total of six events, in six sites along the basin, where the water samples were collected to assess physicochemical parameters and calculate the trophic state index and the index of minimum parameters for the protection of aquatic communities. The data were also compared with values determined by the resolution National Environment Council - CONAMA 357/05. Our results reveal significant changes in the water quality of Bacanga River Basin. An increase in nutrients and chlorophyll-a concentration led it to eutrophication. The surfactant values were high and put in danger the aquatic biota. Dissolved oxygen rates were below the values allowed by the resolution in most sites sampled. The current water quality is terrible for the protection of aquatic life in 61.92% of the sites sampled.

<title>RESUMO</title><p>A emissão de dióxido de carbono é, atualmente, a principal causa do aquecimento global, devido ao efeito estufa. Ao redor do planeta pesquisadores têm-se esforçado na busca de soluções viáveis para o problema; com isto, o biocarvão está entre as mais recentes propostas de mitigação das mudanças climáticas em razão da sua simplicidade e eficiência. Desta forma, este trabalho teve, por objetivo, maximizar o teor de carbono fixo na produção de biocarvão. Para isso, analisaram-se sete tratamentos, temperaturas de pirólise: 200, 250, 300, 350, 400, 450 e 500 °C. A partir do rendimento gravimétrico de produção e do teor de carbono fixo calcularam-se os respectivos fatores de rendimento gravimétrico para cada tratamento, os quais indicam em qual temperatura se tem a maior quantidade de carbono fixo em função do biocarvão produzido. A pirólise com temperatura máxima de 300 °C indicou o maior fator de rendimento gravimétrico, atingindo 0,22672 C/biocarvão.</p>
<title>ABSTRACT</title><p>Currently, the emission of carbon dioxide is the main cause of global warming due to greenhouse effect. Researchers around the world have been studying to find viable solutions to the problem. Biochar is among the latest proposals for mitigating climate change due to its simplicity and efficiency. Therefore, this study aimed to maximize the fixed carbon content in the production of biochar. Seven treatments were analysed with pyrolysis temperature of 200, 250, 300, 350, 400, 450 and 500 °C. From the production yield and fixed carbon content, the gravimetric yield factor was calculated for each treatment, which indicates the amount of fixed carbon. The pyrolysis temperature of 300 °C showed the greatest gravimetric yield factor, reaching 0.22672 C/biochar.</p>

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

O objetivo deste estudo foi determinar a prevalência de helmintos gastrintestinais e renais de Zenaida auriculata naturalmente infectados capturados em Londrina, Paraná. Duzentos e um pombos-de-bando foram capturados e os helmintos gastrintestinais e renais foram coletados e identificados de acordo com as estruturas morfológicas. Cento e dezesseis (57,71%) pombos estavam parasitados por helmintos com as seguintes prevalências:Ornithostrongylus quadriradiatus em 50 pombos (24,88%), seguido por Ascaridia columbae em 47 (23,38%),Paratanaisia bragai e P. confusa em 34 (16,92%), Tetrameres fissispina em 17 (8,46%),Synhimantus nasuta em 14 (6,47%), Brachylaima mazzantii em 4 (1,99%) e Raillietina allomyodes em 2 pombos (1,00%). Setenta e quatro/201 (37,00%) aves apresentaram-se infectadas por apenas uma espécie, e 96/201 (48,00%) pombos com nematodas. A associação entre diferentes classes de helmintos ocorreu em 40/201 (20,00%) animais. Os resultados mostraram diferenças estatísticas entre a presença de nematodas (p = 0,00001) e trematodas (p ≤ 0,05) em pombos, e houve associação entre o local de captura e a presença de trematodas e A. columbae (p ≤ 0,05). Este trabalho é o primeiro a relatar a infecção de Z. auriculata no Brasil com O. quadriradiatus,A. columbae, T. fissispina, S. nasuta, R. allomyodes, P. bragaie P. confusa.
This study aimed to determine the prevalence of gastrointestinal and renal helminths from naturally infected Zenaida auriculatacaptured in Londrina, Paraná State. Two hundred and one Eared doves were trapped and the gastrointestinal and renal helminths were collected and identified according to morphological structures. One hundred and sixteen (57.71%) doves were parasitized by helminths with specific prevalences forOrnithostrongylus quadriradiatus in 50 doves (24.88%),Ascaridia columbae in 47 (23.38%), Paratanaisia bragai and P. confusa in 34 (16.92%),Tetrameres fissispina in 17 (8.46%), Synhimantus nasuta in 14 (6.47%), Brachylaima mazzantii in 4 (1.99%) and Raillietina allomyodes in 2 doves (1.00%). Seventy four/201 (37.00%) birds were infected with only one species, and 96/201 (48.00%) pigeons were infected with nematodes. The association between different classes of helminths occurred in 40/201 (20.00%) animals. The results showed statistically differences between the presence of nematode (p = 0.00001) and trematode species (p ≤ 0.05) in the doves, and there was an association between the local of capture and the presence of trematodes and A. columbae (p ≤ 0.05). This study is the first to report the infection of Z. auriculata from Brazil with O. quadriradiatus, A. columbae, T. fissispina, S. nasuta, R. allomyodes, P. bragai and P. confusa.

In the present investigation we evaluate methods for the isolation and growth of marine-derived fungal strains in artificial media for the production of secondary metabolites. Inoculation of marine macroorganisms fragments in Petri dishes proved to be the most convenient procedure for the isolation of the largest number of strains. Among the growth media used, 3% malt extract showed the best result for strains isolation and growth, and yielded the largest number of strains from marine macroorganisms. The percentage of strains isolated using each of the growth media which yielded cytotoxic and/or antibiotic extracts was in the range of 23-35%, regardless of the growth media used. Further investigation of extracts obtained from different marine-derived fungal strains yielded several bioactive secondary metabolites, among which (E)-4-methoxy-5-(3-methoxybut-1-enyl)-6-methyl-2H-pyran-2-one is a new metabolite isolated from the Penicillium paxilli strain Ma(G)K.

Avaliação da solarização do solo para o controle de Ralstonia solanacearum

O presente trabalho avaliou o emprego da solarização como uma alternativa para o controle da murcha bacteriana, causada por Ralstonia solanacearum, em amostras de solo infestado com o patógeno, dispostas em bolsas de náilon e enterradas em parcelas solarizadas ou não. Dois experimentos foram instalados, um em Campinas (SP), de fevereiro a abril de 2001, e o outro em Piracicaba (SP), de dezembro de 2001 a janeiro de 2002. Os ensaios foram efetuados em delineamento inteiramente casualizado, esquema fatorial, com quatro repetições, tendo cada parcela 4 x 4 m. Os fatores avaliados foram a solarização (com ou sem), efetuada com filme plástico transparente de 100 µm de espessura, o período de tratamento (30 e 60 dias e 37 e 60 dias para o primeiro e o segundo experimentos, respectivamente) e a profundidade de colocação das amostras (10 e 20 cm), fator verificado apenas no segundo ensaio. Após os períodos estipulados de solarização, o solo de cada bolsa foi colocado em vasos, para os quais foram transplantadas mudas de tomateiro (Lycopersicon esculentum). No solo não solarizado, em ambos os experimentos, 43 a 100% dos tomateiros murcharam. No segundo experimento, 6 a 22% dos tomateiros murcharam no solo solarizado por 37 dias. Entretanto não foram detectadas plantas murchas nas parcelas solarizadas do primeiro experimento e no segundo ensaio nenhum tomateiro murchou solo solarizado por 60 dias, nas duas profundidades estudadas. Os resultados indicam que a solarização é uma técnica promissora para o controle de R. solanacearum.
The use of soil solarization for the control of bacterial wilt, caused by Ralstonia solanacearum, was evaluated by burrowing nylon bags containing soil infested with the bacteria in plots solarized or not. Two experiments were carried out in Campinas (SP), from February to April/2001, and Piracicaba (SP), from December/2001 to February/2002. The experiments were set up in a completely randomized factorial design, with four replications, in 4 x 4 meter plots. The factors evaluated were soil solarization with a transparent plastic film 100 µm thick, period of treatment (30 and 60 days and 37 and 60 days for the first and second experiments, respectively) and, only for the second experiment, soil depth (10 and 20 cm). The soil of each nylon bag collected after the previously established solarization periods, was placed in pots where tomato (Lycopersicon esculentum) seedlings were transplanted. In the non-solarized soil, in both experiments, 43 to 100% of the tomato plants wilted. In the second experiment, 6 to 22% of the plants grown on the soil solarized for 37 days wilted. No wilted tomato plants were observed in the solarized plots of the first experiment nor in the soil solarized for 60 days, in both soil depths, of the second experiment. The results obtained indicate that soil solarization has potential for the control of R. solanacearum.
