Crotalaria ochroleuca (Fabaceae) is rich in bioactive compounds used for nematode control. Transformed root culture allows the production of a large amount of hairy root biomass, conditioned by auxin biosynthesis-related genes integrated via the process of Agrobacterium rhizogenes infection, which is known to cause a hairy root phenotype. We transformed A. rhizogenes to increase the biomass of hairy roots and optimized the process of production of bioactive compounds. The transformed nature of hairy roots was confirmed by polymerase chain reaction, which confers kanamycin resistance. The transformed roots were cultured in full-strength in liquid media to realize the growth of hairy roots and the production of bioactive compounds. The transformed roots were grown in a culture medium supplemented with elicitors to produce total phenols, and flavonoids. Chromatographic analysis of transformed roots revealed the presence of flavonoids apigenin-6,8-C-diglucoside and luteolin-6-C-glucoside. The results were obtained by conducting physiological and biochemical studies with the Flavonoids and studying the pathways that led to the production of large amounts of bioactive compounds from the hairy roots of C. ochroleuca. It was observed that the extraction of the compounds significantly affected nematodes and insect larvae, resulting in significantly high levels of economic damage to crops.

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

Abstract Fibrinolytic proteases are a promising alternative in the pharmaceutical industry, they are used in the treatment of cardiovascular diseases, especially thrombosis. Microorganisms are the most interesting source of fibrinolytic proteases. The aim of this study was the production of fibrinolytic protease from Streptomyces parvulus DPUA 1573, the recovery of the protease by aqueous two-phase system and partial biochemical characterization of the enzyme. The aqueous two-phase system was performed according to a 24-full factorial design using polyethylene glycol molar mass, polyethylene glycol concentration, citrate concentration and pH as independent variables. It was analyzed the effect of different ions, surfactants, inhibitors, pH and temperature on enzyme activity. The best conditions for purifying the enzyme were 17.5% polyethylene glycol 8,000, 15% Phosphate and pH 8.0, it was obtained a partition coefficient of 7.33, a yield of 57.49% and a purification factor of 2.10-fold. There was an increase in enzyme activity in the presence of Fe2+ and a decrease in the presence of $\beta$-Mercaptoethanol, phenylmethylsulfonyl fluoride and Iodoacetic acid. The optimum pH was 7.0 and the optimum temperature was 40 ºC. The purified protease exhibited a molecular mass of 41 kDa. The fibrinolytic protease from Streptomyces parvulus proved to be a viable option for the development of a possible drug with fibrinolytic action.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

ABSTRACT: Poisonous plants are a significant cause of death among adult cattle in Brazil. Plants that affect the central nervous system are widely spread throughout the Brazilian territory and comprise over 30 toxic species, including the genus Ipomoea, commonly associated with a lysosomal storage disease and a tremorgenic syndrome in livestock. We describe natural and experimental Ipomoea pes caprae poisoning in cattle from a herd in the Northside of Rio de Janeiro, Brazil. Affected cattle presented episodes of severe ataxia, abnormal posture followed by falling, muscular tremor, contraction, and spasticity, more prominent in the limbs, intensified by movement and forthcoming, and recumbence. Grossly, a substantial amount of leaves and petioles were found in the rumen. Histopathological examination showed degenerative neuronal changes, mostly in cerebellar Purkinje cells, which were confirmed with Bielschowsky silver. The characteristic clinical changes and mild histological lesion strongly suggested a tremorgenic syndrome. Lectin- immunohistochemistry evaluation reinforced this hypothesis; all lectins tested failed to react with affect neurons and Purkinje cells, which ruled out an underlying lysosomal storage disease. One calf given I. pes caprae leaves experimentally developed clinical signs similar to natural cases. On the 28th day of the experiment, the plant administration was suspended, and the calf recovered within four days. I. pes caprae’s spontaneous tremorgenic syndrome in cattle is conditioned to exclusive feeding for several months. We were able to experimentally reproduce toxic clinical signs 12 days following the ingestion.

ABSTRACT The aim of this work was to determine the persistence of auxinic herbicides applied on tropical pasture and toxicity for succeeding crops. The herbicides were applied in an area of dystrophic red‒yellow latosol with pasture infested of weeds. At 40, 80, and 280 days after application of herbicide, the soil samples were collected at depths of 0 to 20 cm. Soil with residues of 2,4-D, 2,4-D + picloram, triclopyr, and a soil without herbicide application were analyzed with six replicates. Seven crops were cultivated in these soils: cucumber (Cucumis sativus L.), velvet bean [Mucuna pruriens (L.) DC.], pigeon pea [Cajanus cajan (L.) Millsp.], alfalfa (Medicago sativa L.), lablab bean [Lablab purpureus (L.) Sweet], corn (Zea mays L.), and sorghum [Sorghum bicolor (L.) Moench]. The plants of cucumber, pigeon pea, and alfalfa were the most susceptible to the auxinic herbicide residues. However, the lablab bean was the only one among the dicot evaluated that showed tolerance to the 2,4-D + picloram residual when cultivated in soils at 280 days after application of herbicide. Corn and sorghum showed lower chlorophyll content in soils with 2,4-D + picloram residual up to 80 days after application of herbicide.

ABSTRACT Much of the Brazilian semiarid region faces a considerable process of degradation of natural resources, and ethnobotanical studies have collaborated with important information about the use and traditional knowledge, serving as a tool to design conservation strategies of native plant species. Thus, this study aimed to determine medicinal species meriting conservation priorities in a “Caatinga” area in the northeastern of Brazilian territory. The ethnobotanical data were collected through semi-structured interviews with key subjects selected through the “snowball” technique. The availability and species conservation priority was verified by relative density, risk of collection, local use and use of diversity in the forest fragment sampled. It was recorded 42 native medicinal plants and conservation priority score was calculated for seven species, including Mimosa tenuiflora, Hymenaea courbaril, Ximenia americana and Amburana cearensis need immediate conservation and attention, since their collection does not occur in a sustainable way. In order to ensure the perpetuation of the species and the sustainability of traditional therapeutic practice there needs to be a development of conservation practices of caatinga remaining to better conserve the species of the biome.

ABSTRACT: The working equid population in Corumbá, Southern Pantanal, is very large and has a crucial role in the main economic activity of the State of Mato Grosso do Sul, the beef cattle industry. The aim of the present study was to estimate the prevalence of equine infectious anaemia (EIA) in working equids of ranches in the municipality of Corumbá, by the official agar gel immunodiffusion (AGID) test, and evaluate the adoption of the Programme for the Prevention and Control of Equine Infectious Anaemia proposed by Embrapa Pantanal and official entities in the 1990s. From September to November 2009, forty ranches distributed through the area of the municipality were visited, and serum samples were obtained from 721 equines and 232 mules. According to previous publications and the present data, it was concluded that the prevalence of EIA in this population has increased from 18.17% to 38.60%, which represents at this time approximately 13,000 infected animals. There was no significant difference between the apparent prevalence of equines and mules. It was also verified that the control programme was not known by the greater part of the interviewed ranch owners, managers and foremen and, in their perception, EIA is not a primary threat to address. Among the studied variables, the serologic testing practice significantly reduced the risk for the presence of EIA seropositivity, as well as the separation of riding equipment and segregation of seropositives.

ABSTRACT: Blood supply of mammary gland is extremely important, since it is a crucial factor in the mammary tissue development. The objective of this study was to describe and assess main mammary artery hemodynamic characteristics of lambs with two growth rates, using Doppler ultrasound. Two experimental groups, A and B, (n=10) of female Brazilian Bergamasca lambs were kept under the same management conditions and facilities. There were two phases: Phase 1, from 90 days of age until onset of puberty; Phase 2, from puberty until 1 year of age. During phase 1, a specific diet was formulated to each experimental group to obtain two different average daily gains: 150g, for treatment A, and 250g, for treatment B. During phase 2, there was no difference on the diet offered to both groups. Every 14 days, all animals were weighed and their body condition score was evaluated. Every 60 days, ultrasound exams (Doppler) of mammary artery were performed to obtain resistance and pulsatility indexes. Treatment B lambs showed higher weight gain and higher body condition score (P<0.05). However, for resistance and pulsatility indexes, there was no difference between treatments (P>0.05). Hemodynamic values remained constant, independent of diet or body development presented by the animals. Obtained data is valuable information on Bergamasca ewe lambs development.

Abstract Aim: To analyze temporal distribution of larvae and juveniles and the early development and of Moenkhausia cf. gracilima. Methods Samples were taken quarterly in twenty-five sites in the upper Paraná River floodplain between August 2013 and May 2015. The samples were taken under the water surface at night using 0.5 mm mesh plankton nets. In the laboratory, samples were sorted, identified and separated into larval (preflexion, flexion and postflexion) and juvenile periods. Results A total of 248 individuals was collected, mainly in the Saraiva Lagoon, suggesting that the entire life cycle of this species occurs in this environment. The reproductive period takes place between December and April, since the postflexion larvae were found until May. However, the occurrence of juveniles between February and May indicates probable batch spawning. Among the 95 individuals used for ontogenic description, 82 were larvae and 13 juveniles. Larvae may be characterized by irregular pigmentation in the upper region of the head, mouth, and body, increasing throughout development; upper lobe of the caudal fin more pigmented than the lower lobe, only visible in postflexion larvae; terminal mouth; anal opening located anterior to the median region of the body and total number of myomers ranging from 34 to 40 (15 to 20 pre and 16 to 23 postanal), while juveniles have characteristics similar to adults. The total number of fin rays is: P. 11-16, V. 7-11, D, 9-11 and A. 21-23. Conclusions According to the distribution of developmental periods it is possible to conclude that this species reproduces in the summer, preferably in lagoons. Growth analysis indicated important alterations in larval morphology (metamorphosis) that may be associated with the ecomorphological characteristics of the species. The morphological separation of larvae of M. cf. gracilima from other larvae of small characids, especially at preflexion and flexion stages may be complicated by the overlap of traits, suggesting the use of other variables, mainly morphometric, for the separation of the species.

As cultivares constituíram as parcelas, o fósforo as sub-parcelas e o azoto as sub-sub-parcelas.
An experiment was conducted without irrigation in randomized block design, in a split-split plot design under five levels of nitrogenand four levels of phosphorus with three replicates for each treatment, in Alegre (ES), to estimate fertilization levels of cultivars. A function of production was used to obtain the levels of physical maximun and optimal economics. The cultivars were integrated in the plots, phosphorus in the subplots and nitrogen were integrated in the subsubplots. There was no significance for phosphorus and their interactions. The other factors showed significance. Therefore, we performed another analysis involving Genotype and Nitrogen factors through statistical model split split plot in space. The estimated productivity (t ha-1) obtained a significant statistical adjustment (p < 0,05) to the second-order model for the surveyed cultivars. The results indicated positive responses to physical productivity depending on nitrogen fertilization. The net operating revenue was positive for Guaçu/IZ.2, Cameroon-Piracicaba and Cana D’África, whose values were 326.80 R$; 219.54 R$ and 281.49 R$, respectively, without nitrogen fertilization, because of the low grass price (41.82 R$ per ton) and the high nitrogen quote (2,670.00 R$ per ton). Thus, it can be concluded that there is a possibility to manage the production of elephant grass for energy purposes without the use of nitrogen fertilization.

Plants have the ability to undergo morphophysiological changes based on availability of light. The present study evaluated biomass accumulation, leaf morphoanatomy and physiology of Neonotonia wightii andPueraria phaseoloides grown in full sunlight, as well as in 30% and 50% shade. Two assays were performed, one for each species, using a randomized block design with 10 replicates. A higher accumulation of fresh mass in the shoot of the plants was observed for both species under cultivation in 50% shade, while no differences were detected between the full sunlight and 30% shade. N. wightii and P. phaseoloides showed increase in area and reduction in thickness leaf when cultivated in 50% shade. There were no changes in photosynthetic rate, stomatal conductance, water use efficiency and evapotranspiration of P. phaseoloidesplants because growth environment. However, the shade treatments caused alterations in physiological parameters of N. wightii. In both species, structural changes in the mesophyll occurred depending on the availability of light; however, the amount of leaf blade tissue remained unaltered. Despite the influence of light intensity variation on the morphophysiological plasticity ofN. wightiiand P. phaseoloides, no effects on biomass accumulation were observed in response to light.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
