Ursolic acid (UA) is being investigated due to its anti-inflammatory potential, and structural modifications can enhance its biological activities. The aim of this study was to evaluate the immunomodulatory effect of the ursolic acid derivatives (UAD) in macrophages and in the carrageenan-induced paw edema model. RAW264.7 cells were cultured in the presence or absence of UA or UAD (1-18). Nitric oxide (NO), nuclear factor kappa B (NF-κB), tumor necrosis factor (TNF), and cellular viability were measured. 30 min before the carrageenan-induced paw edema, the UAD1 and UAD2 (200 mg kg-1) were administered intraperitoneally. The results showed that UAD2-4, UAD7, UAD9-11 had half maximal inhibitory concentration (IC50) greater than 90 µM and were able to reduce NO, NF-κB and TNF production. Moreover, UAD1 and UAD2 reduced paw edema and IL-6 production. In conclusion, the results obtained demonstrated a variation in the response between the derivatives due to the chemical modifications, showing potential to reduce the inflammatory mediators, deserving further investigations.

Abstract The iAtlantic Project has established an international collaborative strategy to improve mapping and characterization of deep and open ocean ecosystems in understudied regions of the Atlantic and evaluate their health. In December 2022, the first iAtlantic expedition in the South Atlantic set off to map and explore seafloor ecosystems in the Santos Basin slope (200-1,000 m depths) in collaboration with the Petrobras ‘Santos Basin - Regional Characterization Project.’ The 17-day ‘iAtlantic_BR10-Petrobras’ cruise was conducted on board the research vessel NPqHOc Vital de Oliveira (Brazilian Navy) and performed (a) water column structure characterization, (b) seafloor morphology mapping, (c) description of benthic habitats and communities by seafloor imagery and biological/ geological sampling, and (d) ex-situ experiments to assess the functioning of sedimentary ecosystems and their responses to climate-related environmental changes. This study describes the rationale behind the iAtlantic_BR10-Petrobras cruise science plan, reports its sampling strategy and methods, and summarizes its collected data and preliminary results.

In this study, the interaction between benzothiazole (BTA, concentration of a drug required for 50% inhibition in vitro (IC50) = 0.77 mM) and benzimidazole (BIA, IC50 = 2.14 mM) with urease was quantitatively assessed, using UV-Vis, molecular fluorescence, and circular dichroism. The results showed that both compounds interact with urease by a static fluorescence quenching mechanism with a non-fluorescent complex formation. The main forces responsible for stabilizing the supramolecular complex between BTA and urease were hydrophobic while, for BIA, van der Waals interactions and hydrogen bonds were the main ones. Urease conformation changes due to the interaction process were analyzed by circular dichroism and synchronous fluorescence. Besides, a competitive assay with substrate and inhibitors was used to evaluate the preferential urease site of interaction with BTA and BIA. Our experimental and theoretical studies supported that both, BTA and BIA, are mixed-inhibitors of ureases with a slight preference to the active site of such enzymes. Finally, both BTA and BIA showed to possess anti-Helicobacter pylori (one reference strain and six clinical isolates) activity, presenting minimal inhibitory concentration (MIC) values ranging from 38-150 and 20-164 µM, respectively. The urease inhibitors omeprazole and hydroxyurea showed MIC values in the range of 46-185 µM and 1683-> 3366 µM, respectively.

Abstract During the last quarter of 2019, the beaches, mangroves, and estuaries of Northeast Brazil received an unprecedented volume of crude oil from the sea, which became the worst environmental disaster ever to reach the Brazilian coast. The oil, having reached the shores completely unnoticed, left both society and government agents completely clueless on (i) where the oil was coming from; (ii) how much oil was still in the ocean to reach the shorelines; and (iii) which beaches were going to be affected next! By exploring remote sensing data and ocean numerical modeling, along with oil dispersion chemistry on sea water, this study investigates the possible origin and path of the spill and whether it could have been detected from space. The oil dispersion modeling simulations performed for this investigation revealed a possible region and timing of the oil spill, also indicating the likelihood of it being advected toward the shoreline under the ocean surface.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

RESUMO Introdução: Em função da urgência e demanda de uma resposta à pandemia do novo coronavírus (Covid-19), vários testes de detecção de anticorpos para a síndrome respiratória aguda grave do coronavírus 2 (Sars-CoV-2) têm sido desenvolvidos. Objetivo: Este estudo teve como objetivo avaliar o desempenho do teste rápido utilizado em um inquérito epidemiológico para Sars-CoV-2 em comparação com outros ensaios sorológicos. Métodos: Foram avaliadas 86 amostras de soro em três ensaios sorológicos: um imunoensaio de fluxo lateral - Wondfo Sars-CoV-2 Antibody Test (TRW) - e dois imunoensaios de quimioluminescência: Elecsys anti-Sars-CoV-2 (ECLIA) e Sars-CoV-2 IgG (CMIA-IgG). Resultados: As sensibilidades diagnósticas estimadas dos testes sorológicos na avaliação dessas amostras foram: TRW 59% [95% intervalo de confiança (IC) 43,4%-72,9%], ECLIA 66,7% (51%-79,4%) e CMIA-IgG 61,5% (47,1%-73%). Enquanto isso, a especificidade diagnóstica estimada para TRW foi 78,7% (95% CI 65,1%-88%), ECLIA 72,3% (58,2%-83,1%) e CMIA-IgG 76,6% (74%-95,5%). Os valores de sensibilidade e especificidade foram inferiores aos afirmados pelos fabricantes. Embora 16,2% (14/86) dos resultados tenham sido discordantes entre os três ensaios serológicos para Sars-CoV-2, o grau de concordância pelo índice Kappa foi adequado: TRW/CMIA-IgG [0,757 (95% IC 0,615-0,899)], TRW/ECLIA [0,715 (0,565-0,864)] e ECLIA/CMIA-IgG [0,858 (0,748-0,968)]. Conclusão: O teste sorológico pode ser uma ferramenta diagnóstica útil, o que reforça sua avaliação criteriosa, bem como o momento correto de sua utilização.
ABSTRACT Introduction: Due to urgency and demand of a response to the Covid-19 pandemic, numerous Sars-CoV-2 immunoassays have been rapidly developed. Objective: This study aimed at assessing the performance of rapid Sars-CoV-2 antibody test in comparison to high-throughput serological assays. Methods: A total of 86 serum samples were evaluated in the three assays: a lateral flow immunoassay - Wondfo Sars-CoV-2 Antibody Test (WRT) - and two chemiluminescence immunoassays: Elecsys Anti-Sars-CoV-2 (ECLIA), and Sars-CoV-2 IgG (CMIA-IgG). Results: The estimated diagnostic sensitivities of serological tests in the evaluation of serum samples from the epidemiological survey were: WRT 59% [95% confidence interval (CI) 43.4%-72.9%], ECLIA 66.7% (51%-79.4%), and CMIA-IgG 61.5% (47.1%-73%). Meanwhile, the estimated diagnostic specificity was for WRT 78.7% (95% CI 65.1%-88%), ECLIA 72.3% (58.2%-83.1%), and CMIA-IgG 76.6% (74%-95.5%). The sensitivity and specificity values were lower than manufacturers’ claimed. Although 16.2% (14/86) of serological results were discordant among the three Sars-CoV-2 serological assays, the degree of agreement by the kappa index was adequate: WRT/CMIA-IgG [0.757 (95% CI 0.615-0.899)], WRT/ECLIA [0.715 (0.565-0.864)], and ECLIA/CMIA-IgG [0.858 (0.748-0.968)]. Conclusion: The serological testing may be a useful diagnostic tool, which reinforces its careful evaluation, and, as well as the correct time to use it.
RESUMEN Introducción: Debido a la urgencia y la demanda de una respuesta a la pandemia de Covid-19, se han desarrollado rápidamente numerosos inmunoensayos del Sars-CoV-2. Objetivo: Este estudio tuvo como objetivo evaluar el rendimiento de la prueba rápida de anticuerpos contra el Sars-CoV-2 en comparación con los ensayos serológicos de alto rendimiento. Métodos: Se evaluaron un total de 86 muestras de suero en los tres ensayos: un inmunoensayo de flujo lateral - Wondfo Sars-CoV-2 Antibody Test (TRW) - y dos inmunoensayos de quimioluminiscencia: Elecsys Anti-Sars-CoV-2 (ECLIA) y Sars-CoV-2 IgG (CMIA-IgG). Resultados: Las sensibilidades diagnósticas estimadas de las pruebas serológicas en la evaluación de muestras de suero de la encuesta epidemiológica fueron: WRT 59% [intervalo de confianza (IC) del 95%: 43,4%-72,9%], ECLIA 66,7% (51%-79,4%) y CMIA-IgG 61,5% (47,1%-73%). Mientras tanto, la especificidad diagnóstica estimada fue para WRT 78,7% (95% CI 65,1%-88%), ECLIA 72,3% (58,2%-83,1%) y CMIA-IgG 76,6% (74%-95,5%). Los valores de sensibilidad y especificidad fueron más bajos que los declarados por los fabricantes. Aunque el 16,2% (14/86) de los resultados fueron discordantes entre los tres ensayos serológicos del Sars-CoV-2, el grado de concordancia del índice kappa fue adecuado: WRT/CMIA-IgG [0,757 (IC del 95%: 0,615-0,899)], WRT/ECLIA [0,715 (0,565-0,864)] y ECLIA/CMIA-IgG [0,858 (0,748-0,968)]. Conclusión: La prueba serológica puede ser una herramienta diagnóstica útil, lo que refuerza su evaluación cuidadosa, así como el momento adecuado para usarla.

The ofloxacin (OFLOX) is a second-generation synthetic antibiotic that can be classified as a multifunctional drug, but is a poorly soluble drug, which influences its efficiency. The inclusion complexes of OFLOX with β-cyclodextrin (βCD) or hydroxypropyl-β-cyclodextrin (HPβCD) can improve the chemical characteristics of the drug; however, studies showing the biological activity of these inclusion complexes are still scarce. The present work aimed to investigate the multifunctionality of the OFLOX and their inclusion complexes. Thus, the 1:1 βCD/OFLOX or HPβCD/OFLOX were prepared and analyzed by Fourier transform infrared spectroscopy (FTIR), thermogravimetric and differential thermal analysis (TGA and DTA), 1H nuclear magnetic resonance (NMR) and isothermal titration calorimetry (ITC). The antitumor and antibacterial effects were assessed. The results confirm the formation of the inclusion complexes, which had lower minimum inhibitory concentration (MIC) values, higher cytotoxicity and promoted the apoptosis. The present study showed, for the first time, the promising effects of the inclusion complexes as antitumor, improving the biological activities of the uncomplexed ofloxacin.

RESUMO A interleucina-4 (IL-4) tem grande importância nos processos inflamatórios em casos de acidente vascular cerebral (AVC), uma vez que ela é capaz de polarizar micróglias para o fenótipo anti-inflamatório chamado M2. Este estudo analisou se a variação entre o genótipo TT e os demais genótipos (CT e CC), no polimorfismo -589 (rs2243250) do gene IL4, possui associação com o prognóstico de AVC hemorrágico e com aspectos clínicos que são fatores de risco para doenças cerebrovasculares. O resultado deste estudo mostra que não há associação estatística do polimorfismo do IL4 nem com prognóstico nem com os aspectos clínicos dos pacientes com AVC hemorrágico.
ABSTRACT Interleukin-4 (IL-4) has great significance in inflammatory processes in cases of stroke, since it is able to polarize microglia to the antiinflammatory phenotype called M2. This study analyzed if the variation between TT genotype and the other genotypes (CT and CC), in -589 (rs2243250) polymorphism of IL4 gene, has association with the prognosis of hemorrhagic stroke (HS) and with clinical aspects which are risk factors for cerebrovascular diseases. The result of this study shows that there is no statistical association of the IL4 polymorphism with either prognosis or clinical aspects in HS patients.
RESUMEN La interleucina-4 (IL-4) tiene gran importancia en los procesos inflamatorios en casos de accidente cerebrovascular (ACV), puesto que hace que las microglías sean polarizadas hacia el fenotipo antiinflamatorio M2. Este estudio analizó si la variación entre el genotipo TT y los demás genotipos (CT y CC), en el polimorfismo -589 (rs2243250) del gen IL4, posee asociación con el pronóstico de ACV hemorrágico y con aspectos clínicos que son factores de riesgo para enfermedades cerebrovasculares. El resultado de este estudio enseña que no hay asociación estadística del polimorfismo del IL4 ni con el pronóstico ni con los aspectos clínicos de pacientes con ACV hemorrágico.

Xanthenones were synthesized via one-pot tricomponent reaction, under solvent-free conditions, using aldehydes, phenolic and cyclic 1,3-dicarbonyl compounds. Natural organic acids (NOAs), compounds present in many living metabolisms, were used as potential green catalysts. NOA are considered to be more eco-friendly and user-friendly alternative to traditional methodologies. Optimization studies showed that oxalic acid was the best NOA catalyst for such reaction furnishing the xanthenones with up to 93% of yield. Theoretical calculations were performed to evaluate this reaction mechanism and regioselectivity. The results showed that the regiospecificity of this three-component reaction is kinetically and thermodynamically controlled by the addition of β-naphthol C2, instead of C10, to the aldehyde. Our results also disclosed two xanthenones as novel calcium channels blockers. Eco-friendly reaction conditions, easy workup procedure, short reaction times and good yields are some of the advantages of our methodology.

Morphological traits, allometric relationship and competition of two seed-feeding species of beetles in infested pods

Abstract Pair-wise competition produces asymmetric consequences for the interacting species, resulting in reduction of species fitness at the individual scale; however, little is known of the effects of competition on the allometric patterns of insects. In this study, we explored how competition, by means of pod infestation, affects the development of female and male individuals in the co-occurring bruchine beetles Merobruchus terani and Stator maculatopygus. We found differences between M. terani and S. maculatopygus in all morphometric traits, but no significant differences between males and females in either species. We also found, with an increasing degree of pod infestation, a positive trend in the pronotum, elytron and body weight of M. terani and a negative trend in morphological traits and body weight of S. maculatopygus. A negative allometry was maintained, suggesting that with increasing body weight, the body structures did not increase proportionally. On the other hand, we found that increasing the degree of pod infestation produced a wider variation in the individuals' body size than in low levels of infestation. Finally, we discuss how pod infestation can trigger competition between species, with both positive and negative impacts, even though the species function similarly in resource exploitation.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Low cost driver device for microclimate maintenance in the pre-milking of dairy cattle

Devido à importância do ambiente na produção animal e, portanto, do controle ambiental adequado, objetivou-se com este trabalho a construção de um sistema de monitoramento e controle das variáveis meteorológicas, temperatura e umidade relativa do ar, de baixo custo, associado a um sistema de resfriamento adiabático evaporativo (SRAE). O desenvolvimento do sistema controlador compreendeu as etapas de montagem, teste e aferição em laboratório e, posteriormente, a etapa de validação do equipamento realizada a campo. A etapa de validação apresentou resultados que permitiram concluir que o sistema pode ser utilizado com segurança no monitoramento e controle dessas variáveis por meio dos atuadores. O controlador mostrou-se eficiente no manejo do microclima no curral de espera e permitiu a manutenção da temperatura do ar dentro da faixa de conforto para bovinos leiteiros na pré-ordenha, apresentando média de 25,09 ºC durante o turno da tarde. O equipamento apresentou custo inferior (R$ 325,76) quando comparado à média de outros no mercado (R$ 450,00).
Due to the importance of the environment on animal production and thus environmental control, the study aims to build a system for monitoring and control the meteorological variables, temperature and relative humidity, low cost, which can be associated with an evaporative cooling system (ECS). The system development included all the stages of assembly, test and laboratory calibration, and later the validation of the equipment carried in the field. The validation step showed results which allowed concluding that the system can be safely used in the monitoring of these variables. The controller was efficient in management of the microclimate in the waiting corral and allowed the maintenance of the air temperature within the comfort range for dairy cattle in pre-milking with averaged 25.09 ºC during the afternoon. The equipment showed the lower cost (R$ 325.76) when compared to other middle market (R$ 450.00).

Por meio de técnicas cromatográficas, um novo peptídeo aniônico, denominado otacidina, foi isolado das secreções cutâneas de três anuros brasileiros Osteocephalus taurinus. O sequenciamento de novo dos aminoácidos foi realizado por meio de análises por espectrometria de massas por tempo de vôo acoplada à ionização dessortiva de matriz por laser (MALDI-TOF-MS/MS). O potencial antimicrobiano deste novo metabólito foi avaliado contra cepas de Escherichia coli e Pseudomonas aeruginosa, sendo observadas concentrações inibitórias mínimas (MICs) de 37,4 e 78,4 mM, respectivamente. Este trabalho é apenas o segundo relato de um peptídeo aniônico com propriedades antimicrobianas, isolado da pele de um anfíbio.
By means of chromatographic techniques, a novel anionic peptide, named otacidin, was isolated from the skin secretions of tree Brazilian anurans Osteocephalus taurinus. The de novo sequencing of the amino acids was performed by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS/MS) analyses. The antimicrobial potential of this novel metabolite was evaluated against Escherichia coli and Pseudomonas aeruginosa, being observed minimum inhibitory concentration (MICs) of 37.4 and 78.4 mM, respectively. This work is just the second report of an antimicrobial anionic peptide from an amphibian.

Essential oil composition and variability in Hyptis fruticosa

The composition of six samples of essential oil (EO) extracted from leaves, flowers and seeds of several plants of Hyptis fruticosa Salzm. ex Benth., Lamiaceae, was investigated by GC/MS and GC/FID. 1,8-Cineole, spathulenol, α-pinene, β-pinene were the major constituents. Ten constituents that have not been previously described in the composition of the oil of H. fruticosa were identified. Hydrocarbons sesquiterpenes represented the main group, followed by hydrocarbons monoterpenes. The results were submitted to Cluster Analysis which allowed three groups of EO to be distinguished with respect to the content of α-pinene/β-pinene, 1,8-cineole and spathulenol. Growth stages of the plants and geographical parameters seem to be important factors determining the variability of the oil. Sesquiterpenes were mainly produced in the seeds.

This article describes the preparation of 5-nitroisatin and of 5-chloroisatin from isonitrosoacetanilide in a single step, using readily available and inexpensive reagents.
