Characterization of cowpea cultivars for grain size, color, and biofortification

ABSTRACT Cowpea crops have high socioeconomic importance in the Northeast region of Brazil. These crops generate employment and income; in addition, it is an excellent source of protein, minerals, vitamins, and dietary fiber, and contributes to the food security of thousands of people. Biofortification of cowpea with iron, zinc, and proteins can contribute to prevent the higher hunger. The objective of this work was to assess iron, zinc, and protein contents in cowpea grains of different cultivars and the effect of grain size and color on the contents of these nutrients. Twenty-four cultivars with variation in size and color of the seed coat were evaluated. Iron contents ranged from 7.12 to 8.60 mg 100 g-1, with an overall mean of 7.75 mg 100 g-1. Zinc contents ranged from 4.46 to 4.93 mg 100 g-1, with an overall mean of 4.71 mg 100 g-1. Protein contents ranged from 31.50 to 36.24 g 100 g-1, with an overall mean of 33.57 g 100 g-1. Grain size ranged from 13.55 to 37.88 g, with an overall mean of 21.50 g. The cultivars Caldeirão, BRS-Guariba, and Pingo-de-Ouro-1-2 have higher iron contents; BRS-Aracê and BRS-Imponente have higher zinc contents; BR-14-Mulato, BRS-Guariba, BRS-Aracê, and BRS-Inhuma, have higher protein contents; and BR-3-Tracuateua and BRS-Imponente have larger grain sizes. The size and color of the seed coat do not influence the iron, zinc and protein contents of the evaluated cowpea cultivars.
RESUMO O feijão-caupi é uma cultura de grande importância socioeconômica na região Nordeste do Brasil. Além de ser um gerador de emprego e renda, é uma excelente fonte de proteínas, minerais, vitaminas e fibras alimentares, contribuindo para a segurança alimentar de milhares de pessoas. A biofortificação do grão de feijão-caupi com ferro, zinco e proteínas pode contribuir na prevenção da fome oculta. Este trabalho teve como objetivo caracterizar os teores de ferro, zinco e proteínas nos grãos de cultivares de feijão-caupi e verificar a influência da cor e tamanho do grão nos teores desses nutrientes. Foram avaliadas 24 cultivares com variação no tamanho e cor do grão. O teor de ferro variou de 7,12 a 8,60 mg 100g-1 e média geral de 7,75 mg 100g-1. O teor de zinco apresentou variação de 4,46 a 4,93 mg 100 g-1 e média geral de 4,71 mg 100 g-1. O teor de proteínas variou de 31,50 a 36,24 g 100g-1, com média geral de 33,57 107 g 100g-1. O tamanho do grão variou de 13,55 a 37,88 g e apresentou média geral de 21,50 g. As cultivares Caldeirão, BRS Guariba e Pingo de Ouro 1-2 apresentam maior teor de ferro; BRS Aracê e BRS Imponente, maior teor de zinco; BR 14-Mulato, BRS Guariba, BRS Aracê e BRS-Inhuma, maior teor de proteínas; e BR 3-Tracuateua e BRS Imponente, maior tamanho de grão. O tamanho e a cor do tegumento grão não influenciam nos teores de ferro, zinco e proteínas das cultivares de feijão-caupi avaliadas.

Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group.

Abstract Health professionals working to mitigate the COVID-19 pandemic are one of the main risk groups for the disease, being prioritized for vaccination. Considering this, the aim of this study was to analyze the immune response of these professionals immunized with CoronaVac in the first and second doses. Blood samples were collected after the first and second doses of the vaccine (CoronaVac) and used to investigate hematological and biochemical parameters, analysis of immunoglobulin production, cytokines, and gene expression profile, as well as the identification of subsets of immune cells. Post-first dose immunological phenotypic memory (CD27+) profiles (T CD4+, TCD8+ and CD19+) showed a significant increase, as did Monocyte APCs (CD80+HLA-DR+) in relation to the second dose. The cytokines IL-2, IL-6 and IFN-° showed increased values in relation to the other analyzed cytokines. The Th2/Th17 profile in the second dose was characterized by gene expression analysis. The production of IgM and IgG after vaccination showed statistically significant values in the comparison between doses. CoronaVac showed activation of APCs monocytes, memory response of T and B lymphocytes, with immunoglobulins production. This set of responses is characterized by the Th2/Th17 immunological profile.

OBJECTIVE: Coronavirus disease 2019 (COVID-19) is associated with high mortality among hospitalized patients and incurs high costs. Severe acute respiratory syndrome coronavirus 2 infection can trigger both inflammatory and thrombotic processes, and these complications can lead to a poorer prognosis. This study aimed to evaluate the association and temporal trends of D-dimer and C-reactive protein (CRP) levels with the incidence of venous thromboembolism (VTE), hospital mortality, and costs among inpatients with COVID-19. METHODS: Data were extracted from electronic patient records and laboratory databases. Crude and adjusted associations for age, sex, number of comorbidities, Sequential Organ Failure Assessment score at admission, and D-dimer or CRP logistic regression models were used to evaluate associations. RESULTS: Between March and June 2020, COVID-19 was documented in 3,254 inpatients. The D-dimer level ≥4,000 ng/mL fibrinogen equivalent unit (FEU) mortality odds ratio (OR) was 4.48 (adjusted OR: 1.97). The CRP level ≥220 mg/dL OR for death was 7.73 (adjusted OR: 3.93). The D-dimer level ≥4,000 ng/mL FEU VTE OR was 3.96 (adjusted OR: 3.26). The CRP level ≥220 mg/dL OR for VTE was 2.71 (adjusted OR: 1.92). All these analyses were statistically significant (p<0.001). Stratified hospital costs demonstrated a dose-response pattern. Adjusted D-dimer and CRP levels were associated with higher mortality and doubled hospital costs. In the first week, elevated D-dimer levels predicted VTE occurrence and systemic inflammatory harm, while CRP was a hospital mortality predictor. CONCLUSION: D-dimer and CRP levels were associated with higher hospital mortality and a higher incidence of VTE. D-dimer was more strongly associated with VTE, although its discriminative ability was poor, while CRP was a stronger predictor of hospital mortality. Their use outside the usual indications should not be modified and should be discouraged.

We reported experimental and theoretical investigation of conformers of 1,2,3-triazole derivatives, substances of exclusively synthetic origin, subject of extensive studies, because of several biological properties, such as antiviral, antimicrobial and antileishmaniasis. We reported molecular/supramolecular X-ray structures of antiophidian compounds I and II. For I and II there are two crystallographic different molecules in the unit cell (A and B). To explore the causes of the similarities in the compound’s crystal structures, intermolecular interactions were explored using the Hirshfeld surface as the fingerprint plots. In addition, density functional theory (DFT) calculations were carried out at the ωB97x-D/6-31G(d,p)-PCM-CHCl3 level aiming to contribute to the interpretation of the experimental data and complement the experimental findings. Two structures named 2A and 5B were found in good agreement with the respective X-ray solid state ones (A and B). Theoretical 1H nuclear magnetic resonance (NMR) spectra calculated for 5B rotated structure (torsion angles deviation around 40° to 90°) was in fine agreement with experimental results (in CDCl3) indicating that the solution molecular structure is considerably different from optimized equilibrium geometries and solid-state structure. Therefore, care is needed when using X-ray structures or DFT geometries to model interaction of drugs with biological targets since significant conformational changes may take place in solution.

Alzheimer disease (AD) is a complex disease related to multiple pathogenic mechanisms. A strategy to develop effective drugs is based on the so-called multi-target directed ligands (MTDL) by using hybrid compounds. So, in the present study, we have designed and synthesized two hybrids, containing the indanone-piperidine moiety of donepezil, a drug approved for the treatment of AD, and the lipoic acid scaffold, an antioxidant compound endowed with neuroprotective effects. One hybrid was synthesized in four steps with 42% global yield, and the other hybrid in six steps with 19% global yield. The latter hybrid displayed moderate inhibitory activity against human acetylcholinesterase (hAChE) and greater activity against human butyrylcholinesterases (hBuChE). The selectivity for hBuChE was further rationalized by theoretical study. Importantly, the second hybrid showed a good antioxidant activity, exhibiting better ability in scavenging 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals than lipoic acid.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Enzyme application in biotechnological and environmental processes has had increasing interest due to its efficiency, selectivity and mainly for being environmentally healthful, but these applications require a great volume of enzymes. In this work the effect of different concentrations of ethanol and 2,5 - xylidine on growth and production of laccase by Pycnoporus sanguineus was investigated. In a medium containing 200 mg.L-1 of 2,5 - xylidine or 50 g.L-1 of ethanol, the maximum activity of laccase was 2019 U.L-1 and 1035 U.L-1, respectively. No direct correlation between biomass and activity of laccase was observed for any of the inducers used during the tests. Ethanol concentrations, larger than or equal to 20 g.L-1, inhibited the radial growth of P. sanguineus. This study showed that ethanol, which has less toxicity and cost than the majority of the studied inducers, presents promising perspectives for laccase production by P. sanguineus.

Uma vez que os humanos não podem sintetizar carotenóides, dependem exclusivamente da dieta como fonte desses micronutrientes. Tem sido afirmado que eles podem aliviar doenças crônicas, tais como cânceres. O presente artigo constitui uma revisão global da literatura científica sobre plantas e outros organismos que biossintetizam carotenóides, que incluem as séries alfa-, beta-, gama-, delta-, e epsilon-carotenos. Os resultados das listas de pesquisa da literatura mostram mais de quinhentas fontes.
Since humans cannot synthesize carotenoids, they depend upon the diet exclusively for the source of these micronutrients. It has claimed that they may alleviate chronic diseases such as cancers. The present communication constitutes a global review of the scientific literature on plants and others organisms that biosynthesize carotenoids, which include the series alpha-, beta-, gamma-, delta- and epsilon-carotenes. The results of the literature survey lists more than five hundred sources.

A hanseníase é uma doença crônica infecciosa ocasionada pelo Mycobacterium leprae. Foi considerada incurável por muitos anos. Atualmente a lepra é uma doença em desaparecimento, apesar de podermos encontrá-la principalmente nos países da zona tropical. O Brasil é o país que tem o segundo maior número de casos de lepra ao redor do mundo com quase 30.000 novos casos diagnosticados em 2005. Este trabalho teve como objetivo revisar a literatura dos vegetais e substâncias de origem natural com atividade antileprótica. Foram encontradas 11 plantas e 17 substâncias isoladas de plantas e microrganismos que foram classificados em grupos químicos adequados. Alguns aspectos de pesquisa recente com produtos naturais direcionados à produção de drogas contra a lepra também são discutidos. Foram consultadas 63 referências.
Leprosy is a chronic infectious disease caused by Mycobacterium leprae bacillus. It was considered to be an incurable disease for ages. Nowadays leprosy is a vanishing disease although we can meet it principally in the tropical zone countries. Brazil has the second greatest number of leprosy cases around the world with almost 30,000 new cases diagnosed in 2005. The present work constitutes a literature review on plant extracts and chemically defined molecules of natural origin showing antileprotic activity. The review refers to 11 plants, their families, and geographical distribution, the utilized parts, the type of extract and the tested organism. It also includes 17 compounds isolated from higher plants and microorganisms, classified into appropriate chemical groups. Some aspects of recent antileprotic-activity-directed research on natural products are discussed. For this purpose 63 references were consulted.

Em agosto/1999, em conseqüência de engano na etapa de diluição, um grupo de 14 funcionários de um hospital privado de Contagem -- Minas Gerais, Brasil, foi imunizado com uma dose 25 vezes mais concentrada do que a recomendada da vacina 17-DD contra febre amarela (Biomanguinhos). Todos o pacientes foram acompanhados através de exames clínicos e laboratoriais nos dias 5, 13 e 35 após a imunização. A freqüência de efeitos colaterais e as manifestações clínicas e laboratoriais não diferiram das observadas em indivíduos imunizados com doses convencionais da vacina. Ao tempo da segunda e terceira avaliações, nenhum dos pacientes relatava sintomas e nenhum deles apresentou anormalidades ao exame físico. Apenas um paciente apresentou alteração laboratorial, sendo observada plaquetopenia no momento da primeira e da segunda avaliação, que retornou aos valores normais ao terceiro exame. À primeira avaliação, seis pacientes apresentavam resposta imune ao teste de redução de neutralização em placa. Em cinco pacientes, o título foi 10 vezes maior do que o valor de corte de 2.36 Log10 mUI/ml. As amostras de sangue colhidas aos tempos 2 e 3 (13 e 35 dias) demonstraram que quase todos os indivíduos responderam à vacinação, com a exceção de um que não apresentou positividade em nenhum dos tempos de avaliação. Este episódio confirma a segurança da vacina 17-DD contra febre amarela.
In August/1999, a group of 14 adults from the staff of a private hospital in Contagem -- Minas Gerais State, Brazil, received unintentionally a 25 times concentrated dose of the 17-DD yellow fever vaccine (Bio-Manguinhos), due to a mistake at the reconstitution step. All patients were clinically and laboratorially evaluated at days 5, 13 and 35 post vaccination. Frequency of side effects and clinical observations of this group of individuals were not different from the observed in recipients immunized with normal doses of the vaccine. At the second and third evaluation none of the subjects reported symptoms. None of the patients presented abnormalities at the physical examination at none of the time points and in all cases the blood examination was normal, except for a reduced number of platelets that was detected in one subject at the first and second evaluation and reverted to normal at third evaluation. At the first evaluation point, 8 subjects were serum negative and 6 serum positive for yellow fever at the plaque reduction neutralization test. In 5 subjects the observed titre was 10 times higher as the baseline of 2.36 Log10 mUI/ml. The samples collected at second and third evaluation (13th and 35th days) demonstrated that all subjects responded to the vaccination with the exception of one that did not present a positive result in any of the samples collected. This evaluation confirms the safety of the 17-DD yellow fever vaccine.

Lipoxigenases (linoleato: oxigênio oxido-redutase - EC 1.13.11.12) são dioxigenases que catalisam a adição do oxigênio molecular ao sistema cis, cis, 1,4 - pentadieno dos ácidos graxos polinsaturados, formando hidroperóxidos dos ácidos graxos correspondentes. As lipoxigenases vegetais utilizam o ácido linolênico (C18:3) ou ácido linoléico (C18:2) como substrato e estão associadas a importantes processos fisiológicos, tais como: biossíntese de compostos regulatórios, crescimento e desenvolvimento, senescência, germinação de sementes, resposta a ferimento, proteína de reserva vegetativa e resistência a insetos e patógenos. Quando os tecidos da planta são danificados por patógenos ou mecanicamente, ocorre uma degradação seqüencial de lipídeos, cujo produto inicial são os hidroperóxidos resultante da ação das lipoxigenases. Entre os vários produtos formados, têm-se a traumatina, o ácido jasmônico, os aldeídos voláteis e os oxiácidos. Para avaliar a capacidade de as plantas de soja responderem, por meio da Via das Lipoxigenases, ao ataque do fungo causador da doença cancro-da-haste - Diaphorte phaseolorum (Cke. e Ell) f. sp. meridionalis - foram realizadas a caracterização bioquímico-cinética do pool de lipoxigenases foliares de plantas de soja (Glycine max (L.) Merril ) e a quantificação dos produtos da Via das Lipoxigenases, utilizando-se um cultivar resistente (FT-Cristalina RCH) e um susceptível (FT-Cristalina) ao patógeno. As plantas dos dois cultivares, infectadas com o patógeno ou apenas injuriadas mecanicamente, apresentaram valores de atividade específica de lipoxigenases maiores que os respectivos controles, em diferentes valores de pH e temperatura, e o pool de lipoxigenases dos dois genótipos apresentou cinética de Michaelis-Menten na faixa de concentração de substrato analisada. Ocorreu aumento dos níveis de inibidores de proteases e aldeídos totais, os quais são produtos dessa via bioquímica. Pelos parâmetros bioquímico e cinético, há a sugestão de uma resposta das plantas de soja através das Via das Lipoxigenases .
Lipoxygenases (linoleate: oxygen oxido-reductases, EC 1.13.11.12) are a class of enzymes that catalyze the addition of molecular oxygen to 1,4 - cis,cis - pentadiene motifs of unsaturated fatty acids to form hydroperoxide products. Plant lipoxygenases use linoleic (18:2) or linolenic (18:3) acids as substrates and are involved in important physiological process such as biosynthesis of regulatory molecules, growth and development, senescence, responses to wounding, vegetative storage protein and may directly mediate host resistance to insect, fungal and bacterial pathogens. When plant tissues are damaged, by pathogens or mecanically injured, a series of lipid degradation occurs, via lipoxygenase pathway, giving rise to hydroperoxydes. Among several products formed it can be found traumatin, jasmonic acid, volatile aldehydes and oxyacids. In this work we evaluated the capacity of the soybean plant to respond to the attack of stem canker fungi (Diaporthe phaseolorum (Cke. e Ell.) f. sp. meridionalis), through the lipoxygenase pathway. Leaves from resistant (FT-Cristalina RCH) and susceptible (FT-Cristalina) cultivars were used for kinetic evaluation of lipoxygenase and biochemical quantitation of products from the lipoxygenase pathway. Plants from both cultivars, infected with the pathogen or mecanically injured, showed values of lipoxygenase specific activity higher than the controls in differents pH and temperature values. The lipoxygenase pool of both cultivars showed kinetic of Michaelis-Menten within the range of substrate concentration assayed. There were an increase in protease inhibitors and of total aldehydes contents, which are products of the lipoxygenase pathway. The biochemical and kinetic parameters suggest a response of the soybean plant through the lipoxygenase pathway.
