Abstract Although there are reports that polyphenol resveratrol (Rsv) may cause muscle hypertrophy in basal conditions and attenuate muscle wasting in catabolic situations, its mechanism of action is still unclear. Our study evaluated the ex vivo effects of Rsv on protein metabolism and intracellular signaling in innervated (sham-operated; Sham) and 3-day sciatic denervated (Den) rat skeletal muscles. Rsv (10-4 M) reduced total proteolysis (40%) in sham muscles. Den increased total proteolysis (~40%) in muscle, which was accompanied by an increase in the activities of ubiquitin-proteasome (~3-fold) and lysosomal (100%) proteolytic systems. Rsv reduced total proteolysis (59%) in Den muscles by inhibiting the hyperactivation of ubiquitin-proteasome (50%) and lysosomal (~70%) systems. Neither Rsv nor Den altered calcium-dependent proteolysis in muscles. Mechanistically, Rsv stimulated PKA/CREB signaling in Den muscles, and PKA blockage by H89 (50μM) abolished the antiproteolytic action of the polyphenol. Rsv reduced FoxO4 phosphorylation (~60%) in both Sham and Den muscles and Akt phosphorylation (36%) in Den muscles. Rsv also caused a homeostatic effect in Den muscles by returning their protein synthesis rates to levels similar to Sham muscles. These data indicate that Rsv directly inhibits the proteolytic activity of lysosomal and ubiquitin-proteasome systems, mainly in Den muscles through, at least in part, the activation of PKA/CREB signaling.

ABSTRACT: Dental and periodontal affections consist of health concerns of great importance in sheep flocks, as they affect the health and welfare of animals and represent one of the main causes of premature slaughter. Studies on the occurrence of these conditions in sheep are scarce in Brazil. The present study aimed to evaluate the prevalence of periodontal diseases and tooth wear in sheep flocks in the State of Goiás. For this, a clinical oral examination was conducted on 325 sheep from seven farms in six municipalities of the State. Periodontal lesions, characterized by gingival recession in at least one incisor tooth, were observed in 48.3% (157/325) of the sheep, while in cheek teeth it occurred in 13.2% (43/325) of the evaluated sheep. Dental wear on cheek teeth was identified in 69.2% (255/325) of the sheep, and 10.7% (35/325) of the ovines showed dental wear on incisor teeth. Sheep older than 36 months had an increase in the frequency of tooth wear. A total of 171 (52.62%) out of the 325 examined sheep had signs of bleeding on probing, which is compatible with gingivitis, and 69 (40.36%) were older than 36 months. Thirty-eight (11.69%) out of the 325 evaluated ovines had gingivitis in all teeth, among which 21 (55.26%) were older than 36 months. Logistic regression analysis allowed the observation that age was associated with the occurrence of gingivitis. Less frequent changes such as periodontal pockets and loss of at least one tooth were also observed in 4.9% (16/325) and 17.8% (58/325) of the sheep, respectively. It indicates that the presence of periodontal diseases and tooth wear is a common occurrence in the evaluated flocks. Therefore, there is a need to include the oral health of sheep in the routine of the farm as an essential preventive measure to improve the quality of life, animal welfare, and zootechnical indices.
RESUMO: As afecções dentárias e periodontais constituem uma preocupação sanitária de grande importância em rebanhos ovinos, uma vez que afetam a saúde e o bem-estar dos animais, e representam uma das principais causas para o abate prematuro. No Brasil, estudos sobre a ocorrência dessas afecções em ovinos são escassos. Assim, o presente estudo teve como objetivo avaliar a prevalência de doenças periodontais e desgaste dentário em rebanhos ovinos no estado de Goiás. Para isso, realizou-se o exame clínico bucal em 325 ovinos em sete propriedades rurais de seis municípios do estado. As lesões periodontais, caracterizadas por recessão gengival em, pelo menos, um dente incisivo, foram observadas em 48,3% (157/325) dos ovinos, enquanto nos mastigatórios ocorreu em 13,2% (43/325) dos ovinos avaliados. O desgaste dentário em dentes mastigatórios foi identificado em 69,2% (255/325) dos ovinos e 10,7% (35/325) dos ovinos apresentaram desgaste dentário em dentes incisivos. Ovinos com idade superior a 36 meses tiveram um aumento na frequência do desgaste dentário. Dos 325 ovinos examinados, 171 (52,62%) apresentaram sinais de sangramento à sondagem, compatíveis com o quadro de gengivite, sendo que 69 (40,36%) apresentaram idade superior a 36 meses. Dos 325 ovinos avaliados, 38 (11,69%) apresentaram gengivite em todos os dentes, dos quais 21 (55,26%) apresentaram idade superior a 36 meses. Pela análise de regressão logística, foi possível observar que a idade mostrou associação com a ocorrência de gengivite. Alterações menos frequentes como a bolsa periodontal e a perda de, pelo menos, um elemento dentário também foram observadas em 4,9% (16/325) e 17,8% (58/325) dos ovinos, respectivamente. Os resultados do presente estudo indicam que a presença doenças periodontais e desgaste dentário é uma ocorrência comum nos rebanhos avaliados. Portanto, enfatiza-se a necessidade de incluir a saúde bucal dos ovinos na rotina da propriedade como uma medida preventiva essencial para melhorar a qualidade de vida e o bem-estar dos animais, bem como os índices zootécnicos.

Various beneficial biological activities have been reported for passion fruit (Passiflora edulis), grown in tropical and subtropical regions. However, no study has yet demonstrated its action against the Zika virus (ZIKV) infection. The present study aimed to investigate the anti-ZIKV of the ethanolic passion fruit seed extract (PFSE), from which, liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis allowed to putatively annotate the occurrence of piceatannol, astringin, scirpusin A, scirpusin B, isookanin-7-O-glucoside and naringenin-7-O-glucoside, two quadranguloside derivatives, tyrosine, and phenylalanine. PFSE is well tolerated by cell lines (Vero E6 and HTR-8/SVneo) and tissue explants from the human placenta. With 1- and 24-h treatments, PFSE consistently reduced the viral load and NS1 protein expression of the two strains of ZIKV (MR766 and PE243) in placental explants and cell lines. Thus, PFSE has a promising potential for developing anti-ZIKV treatments, protecting the placenta against infection caused by ZIKV, along with relevant antioxidant potential, represented by ferric reducing antioxidant power (FRAP) of 390.5 Trolox eq per g dry extract and half maximal inhibitory concentration (IC50) for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) of 12.1 µg mL-1, and more than 90% protection from lipid peroxidation. Although further studies need to be conducted, PFSE treatment was safe and effective in trophoblast cells and placental explants, thus representing a promising class of all-accessible products based on the reuse of industrial by-products.

Abstract Since the first reported case of COVID-19 in Brazil, the public and private educational system started to close. Up to November 2020, scientific discussions about the return of schooling activities have been rarely performed by the national scientific community and police-makers. The great delay of school returning in Brazil contrasts with successful international strategies of school reopening worldwide and seems counterintuitive with the reopening of non-essential activities. Here, important issues to be considered before and during school reopening are reviewed and discussed. COVID-19 testing is essential to avoid disease spreading, but high cost of individual RT-qPCRs impairs an extensive testing strategy for school returning. To reduce costs and increase the speed of diagnosis, we tested the efficiency of a pooled-sample PCR strategy in a cohort of the educational staff in the city of Macaé/RJ, finding five asymptomatic individuals (0,66%) among the 754 people tested. Thus, a polled-sample PCR testing strategy of the educational staff might prevent infection spreading in schools at a reasonable cost. We discuss how our test strategy could be coupled with internationally recognized safety rules to allow for a safe school return and how countries from different world regions are dealing with educational activities during COVID-19 pandemic.

Electrochemical procedures have emerged as a powerful tool for the synthesis of complex organic molecules including transformation of natural products. In this study, the electrosynthesis of biseugenol in MeOH afforded one new oxidized derivative, which was characterized as methoxyl-oxo-biseugenol (MOB) by nuclear magnetic resonance (NMR) and electrospray ionization-high resolution mass spectrometry (ESI-HRMS) analysis. Since biseugenol was previously described as a cytotoxic natural product, MOB was also tested against tumoral cells B16F10-Nex2 (murine metastatic melanoma) and SKMEL-25 (human metastatic melanoma) as well as against non-tumoral cells MØ Raw 264.7 (murine macrophage immortalized) and HUVEC (human umbilical endothelium). As results, MOB showed inhibitory concentration that affects 50% of cells (IC50) values of 9.5 ± 1.6 mg mL-1(B16F10Nex2), 13.2 ± 2.5 mg mL-1 (SKMEL-25), 19.9 ± 3.5 mg mL-1 (MØ Raw 264.7) and 36.3 ± 7.4 mg mL-1 (HUVEC). Therefore, selectivity index (SI) values of MOB to tumoral cells B16F10Nex2 and SKMEL-25 were calculated as 2.1 and 2.8, respectively, higher than biseugenol (1.4 and 1.7, respectively). Based on these results, the superior cytotoxic activity of MOB in comparison to biseugenol could be associated, at least in part, to the presence of a non-aromatic ring and a conjugated carbonyl system instead of a phenol moiety.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

The use of sorghum in human nutrition has been expanded due to its nutritional composition and its functional potential. Extrusion has been used to improve the quality of sorghum. The aim of this work was to assess the effect of extrusion in the nutritional composition, phenolic compounds content of two sorghum, and characterize the profile of chemical constituents by paper spray mass spectrometry (PS-MS). Extrusion increased the carbohydrate and fiber contents and reduced moisture, lipids and ashes, and enabling a higher degree of liberation of phenolic compounds and antioxidant activity. The fingerprint obtained in both ionization modes had been influenced by extrusion, allowing for a greater compounds’ identification on the extruded samples. The analysis of sorghum samples’ main components made distinguishing of the main constituents possible, as well as highlighted the extrusion effect and the influence of the genotypes. This study demonstrated that extrusion enabling a higher liberation of phenolic compounds, which were identified by the PS-MS technique.

Abstract Given the scarcity of studies with elderly and the existence of studies investigating the effect of vitamin D supplementation in PEH (post exercise hypotension), this study evaluated the effect of a single megadose of vitamin D on resting blood pressure (RBP) and post-exercise hypotension (PEH) in the elderly. 11 hypertensive elderly women (70.3 ± 1.7 years) received a single megadose of 200.000 IU of cholecalciferol or a placebo, orally, through capsules. On day 7, the subjects performed 30 minutes of aerobic exercise with blood pressure measurement before exercise and every 10 minutes after exercise during 60 minutes, besides cardiac autonomic modulation. RBP did not significantly change. Exercise promoted significant systolic PEH only in one moment post exercise in treated group and in the placebo group promoted significant systolic PEH at four moments. Significant diastolic PEH did not occur in any of the groups. Sympathovagal activity increased at post exercise balance in supplemented subjects at 20 min, 40 min, 50 min and 60 min when compared to rest; this increase was not observed in the placebo. A megadose of vitamin D did not reduce RBP, promoted partial inhibition of systolic PEH and increased sympathovagal balance.

Este estudo teve como objectivo avaliar o desenvolvimento e produtividade da cultura do feijão (Phaseolus vulgaris L.), quando submetida à inoculação e aplicação de fertilizante nitrogenado (N) à sementeira e/ou cobertura. O delineamento experimental foi em blocos casualizados (DBC), em esquema fatorial 4 (N sementeira, N cobertura, N sementeira e cobertura e sem aplicação de N) x 2 (sem e com inoculação da estirpe CIAT 899T), com três repetições. Aos 28 dias após a sementeira, todos os tratamentos receberam aplicação via foliar de molibdénio na dose de 80,0 g ha-1. Nesse mesmo período, os tratamentos contendo N cobertura receberam aplicação de 40 kg ha-1. Na época de floração foram avaliados os parâmetros de matéria seca da parte aérea (MSPA), teor de nitrogénio da parte aérea (TNPA), número de nódulos (NN) e matéria seca dos nódulos (MSN). Aos 120 dias (época da colheita), foram avaliados o número de vagens (NVP) e de grãos por planta (NGP) e produtividade de grãos (PG - kg ha-1). A inoculação mostrou-se uma prática viável quando não foi utilizado nitrogénio mineral e o N foi utilizado somente na cobertura, sendo capaz de promover bom desenvolvimento vegetal e produtividade, no entanto, não equiparável à dose completa de N mineral.
The objective of this research was to evaluate the development and productivity of bean crop (Phaseolus vulgaris L.), when submitted to the inoculation and application of nitrogen fertilizer (N) at sowing and/or top dressing (cover). The experimental design was in randomized blocks (RBD) in 4 (N sowing, N top dressing, N sowing and top dressing and without N application) x 2 (with and without inoculation of strain CIAT 899T) factorial combination with three replications. At 28 days after sowing, all treatments received the foliar application of 80.0 g ha-1of molybdenum. In the same period, the treatments containing N top dressing received the application of 40 kg ha-1. At flowering time, the parameters, dry matter of the aerial part (DMAP), shoot nitrogen content (TNPA), number (NN) and dry matter of nodules (MSN) were evaluated. After 120 days (harvest time) the following parameters were evaluated: number of pods (NVP) and grains per plant (NGP) and grain yield (PG - kg ha-1). Inoculation proved to be a viable practice when it was not used mineral nitrogen. The use of inoculated seeds and also top dressing of nitrogen fertilization in cover promoted good development and productivity, however, not comparable to the full dose of mineral N.

This study aimed to use a differential pulse voltammetric (DPV) assay, based on the electroreduction of oxygen, to evaluate the antioxidant capacity of pequi pulp. The results were expressed in coefficient of antioxidant capacity (K), at different pH values and in trolox equivalent antioxidant capacity (TEAC), and were compared to the 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) spectrophotometric assays. The principal component analysis showed higher K for pequi pulp extract and pH tending to 7.15. The antioxidant capacity was related to the presence of gallic acid in its composition. Regarding the TEAC value, the best result was obtained by the DPV test (53.43 ± 5.14 µmol trolox g-1 pulp). The pequi pulp showed antioxidant capacity, compared to the standard antioxidant compounds evaluated. It was concluded that the voltammetric protocol was efficient to assess the total antioxidant capacity of pequi pulp and can be an alternative to conventional spectrophotometric assays, for the analysis of food matrices.

ABSTRACT Studies have shown that the benefits from music can improve the welfare conditions of laboratory, pet and production animals. The study aimed to evaluate the influence of music (Antonio Lucio Vivaldi compositions) on behavior aspects (position, activity, stereotypes, social and agonistic behavior) and physiological responses (respiratory rate and rectal temperature) in gestating sows in individual cages (stage 1), and collective stalls (stage 2). 28 animals were studied, 14 belonged to music group, and 14 to the control group (without music).The respiratory rate had the lowest averages in the group with music, for example, in stage 1, time 1, values of 19.28 and 21.50 mov.min−1. In stage 2, the values found in the same time were 20.36 and 24.16, and in time 2, values of 24.37 and 27.18 mov.min−1. These findings follow in stage 2 until time 4.Behaviors position, activity, stereotypes and agonistic interaction with the researcher were also influenced by the music. We concluded from the results that music influenced the behavior of animals, providing greater relaxing and reduce stereotypes aspects related to a better welfare.

Resumo Até 2010, o conhecimento sobre a diversidade de fungos do Brasil estava registrado em publicações esparsas de taxonomia e ecologia e em algumas poucas listas de espécies. Com a publicação do Catálogo de Plantas e Fungos do Brasil, e a disponibilização da lista online, tem sido possível agregar o conhecimento disperso. A versão ora apresentada acrescenta 2.111 nomes de espécies aos 3.608 listados em 2010. São citadas 5.719 espécies de fungos distribuídas em 1.246 gêneros, 102 ordens e 13 divisões, consistindo em considerável aumento em relação a 2010, quando estavam registrados 924 gêneros e 78 ordens. Predominam os Basidiomycota (2.741 espécies, em 22 ordens) e Ascomycota (1.881 espécies, em 41 ordens). A Mata Atlântica possui a maior quantidade de registros, com 3.017 espécies, seguido pela Amazonia (1.050), Caatinga (999), Cerrado (638) e Pampa e Pantanal com 84 e 35 espécies, respectivamente. A região Nordeste tem a maior riqueza (2.617 especies), seguida pelo Sudeste (2.252), Sul (1.995), Norte (1.301) e Centro Oeste (488 espécies). Em relação aos Estados da Federação, São Paulo (1.846 espécies), Pernambuco (1.611) e Rio Grande do Sul (1.377) são os mais diversos.
Abstract Knowledge about the Brazilian fungal diversity was, until 2010, recorded in few taxonomy and ecology publications, as well as in a handful of species lists. With the publication of the Catálogo de Plantas e Fungos do Brasil and the continued availability of an online list, it has been possible to aggregate this dispersed knowledge. The version presented here adds 2,111 species names to the 3,608 listed in 2010. A total of 5,719 species of fungi distributed in 1,246 genera, 102 orders and 13 phyla represents a considerable increase over the last five years, when only 924 genera and 78 orders were registered. Basidiomycota (2,741 species in 22 orders) and Ascomycota (1,881 species in 41 orders) predominate over other groups. The Atlantic Rainforest has the largest number of records, with 3,017 species, followed by Amazon Rainforest (1,050), Caatinga (999), Cerrado (638) and Pampa and Pantanal with 84 and 35 species, respectively. Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A dengue é uma doença negligenciada responsável pelo óbito de 22.000 pacientes por ano em áreas onde a doença é endêmica. Embora grandes esforços tenham sido investidos em pesquisas na busca de tratamentos para a dengue, não há ainda nenhuma vacina ou antiviral disponível no mercado para tratamento desta patologia. Entre as estratégias que vêm sendo utilizadas para a busca e desenvolvimento de antivirais contra a dengue, a inibição de enzimas é aquela que tem se mostrado a mais promissora. O presente artigo de revisão descreve os recentes avanços no que tange à utilização de enzimas do vírus da dengue como alvos para o desenho de antivirais.
Dengue is a neglected disease, one that is responsible for 22,000 deaths each year in areas where it is endemic. Despite the tremendous efforts invested in research for dengue treatments, no vaccine or antiviral has reached the market, and disease treatment is limited to supportive care. Among strategies that have been used in the search for and development of an antiviral against dengue, the inhibition of viral enzymes has been proven to be the most successful approach. This review covers the major progresses that have been published concerning compounds that inhibit viral enzymes.
