Hematological problems are associated with Coronavirus disease 2019 (COVID-19). Respiratory impairment is the higher point studied, although without experimental studies related to the oxygen transport performed by erythrocytes. Therefore, we decided to investigate if erythrocytes from COVID-19 patients have their functionality changed. The case-control study included hospitalized patients with a positive real-time polymerase chain reaction (RT-PCR) result admitted to University Hospital. Volunteers (negative RT-PCR results) were recruited as a control group. Thus, we assessed different erythrocytes parameters, oxidative stress markers, and biophysical studies using whole blood and isolated hemoglobin. We found a decrease of 51% in oxygen uptake and reduced antioxidant enzyme activity in COVID-19 patients compared to controls. Raman spectrometry showed structural changes in the hemoglobin and lipids of the erythrocytes from COVID-19 patients; thus, these results were consolidated with an increase in Young’s modulus in erythrocytes followed by morphology changes. Besides, isolated hemoglobin from COVID-19 patients has a distinct interaction profile using a ligand model compared to the control. COVID-19 leads to structural, functional, and morphological damage to human erythrocytes. Our data showed structural and molecular changes and induction of oxidative stress in erythrocytes by COVID-19, a new perspective on the contribution of erythrocytes to a respiratory commitment in COVID-19.

RESUMEN La toxina iota (ITX) es una enterotoxina binaria producida como protoxina por Clostridium perfringens (C. perfringens) tipo E que es activada por enzimas proteolíticas en el intestino delgado de los animales infectados. Mediante la despolimerización de los filamentos de actina, la ITX provoca la desorganización del citoesqueleto de las células promoviendo el aumento de la permeabilidad celular. Aquí, realizamos esta revisión con el objetivo de avanzar en la comprensión de las enfermedades entéricas clostridiales causadas por las toxinas de C. perfringens y la especificidad de ITX en las lesiones de la mucosa intestinal. La ITX consta de un componente enzimático (Ia) y un componente de unión (Ib). Examinamos los hallazgos histológicos recientemente publicados sobre los efectos de la ITX y su relación con las enfermedades entéricas intestinales. Histológicamente, se ha observado necrosis hemorrágica y hemorragia multifocal en la mucosa del yeyuno-ileo, el intestino delgado y el abomaso. Aunque el diagnóstico sigue basándose en la presencia de toxinas en el contenido intestinal y en la historia clínica y/o histológica, es importante desarrollar nuevos indicadores enterotóxicos capaces de establecer métodos precisos para diferenciar las acciones de la ITX y de otras toxinas implicadas en el proceso infeccioso de C. perfringens tipo E.
ABSTRACT The iota toxin (ITX) is a binary enterotoxin produced as a protoxin by Clostridium perfringens (C. perfringens) type E that is activated by proteolytic enzymes in the small intestine of infected animals. By depolymerization of the actin filaments, ITX causes cytoskeleton disorganization of cells promoting the increase of the cell permeability. Here, we conducted this review aiming to advance the understanding of enteric clostridial diseases caused by C. perfringens toxins and the specificity of ITX in the intestinal mucosa lesions. ITX consists of an enzymatic component (Ia) and a binding component (Ib). We screened the recently published histological findings of the ITX effects and its relationship with intestinal enteric diseases. Histologically, hemorrhagic necrosis and multifocal hemorrhage have been observed in the jejunum-ileum mucosa, the small intestine, and the abomasum. Although the diagnosis is still based on the presence of toxins in the intestinal contents and the clinical and/or histological history, it is important to develop novel enterotoxemic indicators capable of establishing precise methods for differentiate the actions of ITX and other toxins involved in the infectious process of C. perfringens type E.

ABSTRACT Objectives: To evaluate the impact of COVID-19 on clinical and surgical practice, educational activities, health and lifestyle behavior of Brazilian urology residents. Materials and Methods: A web-based survey was sent to 468 Brazilian urology residents from postgraduate years (PGY) 3 to 5 to collect data on clinical practice and training after 4 months of COVID-19. We also assessed health-related and behavior changes, rate of infection by SARS-CoV-2, deployment to the front line of COVID-19, residents’ concerns, and access to personal protective equipment (PPE). Results: Massive reductions in elective and emergency patient consultations, diagnostic procedures and surgeries were reported across the country, affecting PGY 3 to 5 alike. Most in-person educational activities were abolished. The median damage to the urological training expected for 2020 was 6.0 [3.4 – 7.7], on a scale from 0 to 10, with senior residents estimating a greater damage (P< 0.001). Educational interventions developed included online case-based discussions, subspeciality conferences and lectures, and grand rounds. Most senior residents favored extending residency to compensate for training loss and most younger residents favored no additional training (p< 0.001). Modifications in health and lifestyle included weight gain (43.8%), reduced physical activity (68.6%), increased alcoholic intake (44.9%) and cigarette consumption (53.6%), worsening of sexual life (25.2%) and feelings of sadness or depression (48,2%). Almost half were summoned to work on the COVID-19 front-line and 24.4% had COVID-19. Most residents had inadequate training to deal with COVID-19 patients and most reported a shortage of PPE. Residents’ concerns included the risk of contaminating family members, being away from residency program, developing severe COVID-19 and overloading colleagues. Conclusions: COVID-19 had a massive impact in Brazilian urology residents´ training, health and lifestyle behavior, which may reflect what happened in other medical specialties. Studies should confirm these findings to help developing strategies to mitigate residents’ losses.

The key step in the chemoenzymatic synthesis of apremilast was to produce the chiral alcohol (R)-1-(3-ethoxy-4-methoxyphenyl)-2-(methylsulfonyl)ethanol, (R)-3. Two enzymatic approaches were evaluated to obtain (R)-3, one using ketoreductases and the other lipases. Bioreduction of 1-(3-ethoxy-4-methoxyphenyl)-2-(methylsulfonyl)ethanone (2), using ketoreductase KRED-P2-D12, led to (R)-3 with 48% conversion and 93% enantiomeric excess (ee). Kinetic resolution of rac-1-(3-ethoxy-4-methoxyphenyl)-2-(methylsulfonyl)ethyl acetate (rac-4), via hydrolysis reaction, with 20% of n-butanol, catalyzed by lipase from Aspergillus niger yielded (R)-3 with > 99% ee, 50% conversion and E-value (enantiomeric ratio) > 200. The reaction between enantiomerically pure (R)-3 and 4-acetylamino-isoindol-1,3-dione (8) afforded apremilast in 65% yield and 67% ee.

The flavonoid rutin presents several pharmacological effects, despite this, its application in the pharmaceutical industry can be significantly limited by its low bioavailability. The development of lipophilic derivatives by esterification of hydroxyl groups with fatty acid chains may be an effective strategy to change their physicochemical properties. This work aims to use high-speed countercurrent chromatography (HSCCC) to isolate rutin esters produced by esterification reactions catalyzed by the immobilized lipase from Candida antarctica (Novozyme 435®). The lipase-catalyzed synthesis of rutin esters (R3-R18:2) in 2-methyl-2-butanol exhibited conversions that ranged from 16 to 40% and highest conversion for short chain fatty acids (R4-R12). After initial partitioning tests of the reaction mixture in different solvent proportions followed by thin layer chromatography (TLC) analysis, the biphasic solvent systems consisting in HEMWat (hexane/ethyl acetate/methanol/water) in different proportions were chosen to separate rutin esters. These esters were separated for the first time to the reaction mixture via HSCCC. This technique proved to be more advantageous than the traditionally one since it allowed quick isolation and high purity (≥ 90%) of the products. It also permitted a substrate recovery for reuse in other enzymatic reactions. Furthermore, the results add valuable information, specially concerning the structures elucidation of different rutin esters not previously described.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

We present an electroanalytical method using square-wave anodic stripping voltammetry on electrochemically-treated screen-printed gold electrodes to the simultaneous determination of lead(II), copper(II) and mercury(II) in struvite (natural fertilizer produced from urine). Electrochemical treatment via cyclic voltammetry of the working electrodes increased surface rugosity that improved the electroanalytical responses for the three metals. Under optimized conditions and using a deposition time of 90 s, the proposed method presented the following limit of detection values: 1.3 µg L-1 for lead(II), 2.0 µg L-1 for copper(II) and 0.9 µg L-1 for mercury(II). Struvite samples were easily dissolved in supporting electrolyte solution (0.05 mol L-1 HCl). Satisfactory recovery values between 91 and 111% were obtained for the analysis of spiked samples. Hence, we can infer that the proposed electroanalytical method is an effective alternative for the determination of trace metals, since it provides precise and accurate direct analyses, with rapid responses without interference from sample matrix and enabling in field analysis.

Abstract Osteogenesis Imperfecta (OI) is a heterogeneous genetic disorder characterized by bone fragility and fracture. Mutations in 20 distinct genes can cause OI, and therefore, the genetic diagnosis of OI is frequently difficult to obtain because of the great number of genes that can be related with this disease. Studies that report the most frequently mutated genes in OI patients can help to improve molecular strategies for diagnosis of the disease. In order to characterize the mutation profile of OI in Brazilian patients, we analyzed 30 unrelated patients through SSCP screening, NGS gene panel, and/or Sanger sequencing for the 11 most frequently mutated genes in the database of mutations, including COL1A1, COL1A2, P3H1, CRTAP, PPIB, SERPINH1, SERPINF1, FKBP10, SP7, WNT1 and IFITM5. Disease-causing variants were identified in COL1A1, COL1A2, FKBP10, P3H1, and IFITM5. A total of 28 distinct mutations were identified, including seven novel changes. Our data show that the analysis of these five genes is able to detect at least 95% of causative mutations in OI disorder from Brazilian population. However, it has to be taken into considerations that distinct populations can have different frequencies of disease-causing variants. Hence, it is important to replicate this study in other groups.

RESUMO O uso do regulador de crescimento em aveia pode reduzir o acamamento de plantas com reflexos na produtividade de biomassa e grãos. Objetivou-se, no estudo, determinar a viabilidade e a eficiência do uso do regulador de crescimento Trinexapac-Ethyl em aveia branca em diferentes condições de uso de N-fertilizante e de ano favorável e desfavorável ao cultivo. Neste estudo, dois experimentos foram conduzidos nos anos de 2011, 2012 e 2013, um para quantificar a taxa de produção de biomassa e outro para determinação da produtividade de grãos e o acamamento. O delineamento experimental foi de blocos ao acaso com quatro repetições em esquema fatorial 4 x 3 para doses de regulador de crescimento (0, 200, 400 e 600 mL ha-1) e doses de nitrogênio (30, 90 e 150 kg ha-1) respectivamente. Há redução linear da taxa de biomassa com o incremento da dose de regulador de crescimento em aveia independente da condição de ano e uso de N-fertilizante. No uso reduzido alto e muito alto de N-fertilizante, a dose de 495 mL ha-1 de regulador reduz com eficiência o acamamento sem prejuízos na produtividade de grãos de aveia, seja ano favorável, intermediário ou desfavorável ao cultivo.
ABSTRACT The use of growth regulator in oats can reduce plant lodging with reflections in biomass and grain yield. The objective of the study was to determine the feasibility and efficiency of using Trinexapac-Ethyl regulator in the growth of white oat under different conditions of N-fertilizer and years favorable and unfavorable for cultivation. In this study, two experiments were conducted in the years 2011, 2012 and 2013, one for quantifying biomass production rate and the other for the determination of grain yield and lodging. The experimental design was randomized blocks with four replicates, in a 4 x 3 factorial scheme, for growth regulator doses (0, 200, 400 and 600 mL ha-1) and nitrogen doses (30, 90 and 150 kg ha-1), respectively. There is a linear reduction of biomass rate with the increase in the growth regulator dose in oat, regardless of the condition of year and use of N-fertilizer. The growth regulator dose of 495 mL ha-1 efficiently reduces lodging with reduced, high and very high use of N-fertilizer, without reducing the yield of oat grains, in favorable, intermediate or unfavorable year for cultivation.

ABSTRACT The pertinent literature discloses a family of optical profile measuring techniques highlighting the moiré methods. The Moiré technique is based on the interference of two superimposed optical grids which produce fringe patterns which can be processed to analyze the DYNAMIC MEASUREMENT IN MATERIALS mechanical behavior. The goal of this research is to experimentally determine the mechanical displacement of a slim cantilever beam by means of Moiré technique with Phase Shift. Photomechanical experimental arrangement included a digital camera, a multimedia projector and a PC. The occurrence of oscillations has been adequately analyzed. The results showed data from which useful information can withdraw, being of great importance to support agricultural machinery projects and development.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Macroinvertebrates associated with bryophyta in a first-order Atlantic Forest stream

This study describes the composition and structure of the benthic community associated with bryophytes in a first-order stream, located in a biological reserve of the Atlantic Forest, during two seasons. During three months of the dry season of 2007 and three months of the rainy season of 2008, samples of bryophytes attached to stones were collected randomly, along a 100 m stream reach. The structure of the community was analyzed through the mean density of individuals, Shannon's diversity index, Pielou's evenness, family richness, dominance index, and the percentage of Ephemeroptera, Plecoptera and Trichoptera (% EPT). Chironomidae larvae were dominant in the two periods of study, followed by Ceratopogonidae in the rainy season, and Naididae in the dry season. The orders EPT contributed 14 families. The results showed that bryophytes constitute suitable habitat which is able to shelter an abundant and diversified benthic fauna in a small extension of the stream. This habitat provides refuge during spates, and thus minimizes downstream transport of the macroinvertebrate fauna.

Genetic characterization of cassava ( Manihot esculenta ) landraces in Brazil assessed with simple sequence repeats

Based on nine microsatellite loci, the aim of this study was to appraise the genetic diversity of 42 cassava ( Manihot esculenta ) landraces from selected regions in Brazil, and examine how this variety is distributed according to origin in several municipalities in the states of Minas Gerais, São Paulo, Mato Grosso do Sul, Amazonas and Mato Grosso. High diversity values were found among the five above-mentioned regions, with 3.3 alleles per locus on an average, a high percentage of polymorphic loci varying from 88.8% to 100%, an average of 0.265 for observed heterozygosity and 0.570 for gene diversity. Most genetic diversity was concentrated within the regions themselves ( H S = 0.52). Cluster analysis and principal component based scatter plotting showed greater similarity among landraces from São Paulo, Mato Grosso do Sul and Amazonas, whereas those from Minas Gerais were clustered into a sub-group within this group. The plants from Mato Grosso, mostly collected in the municipality of General Carneiro, provided the highest differentiation. The migration of human populations is one among the possible reasons for this closer resemblance or greater disparity among plants from the various regions.
