Extinction and emergence of genomic haplotypes during the evolution of Avian coronavirus in chicken embryos

Abstract Avian coronavirus (AvCoV) is ubiquitously present on poultry as a multitude of virus lineages. Studies on AvCoV phenotypic traits are dependent on the isolation of field strains in chicken embryonated eggs, but the mutant spectrum on each isolate is not considered. This manuscript reports the previously unknown HTS (high throughput sequencing)-based complete genome haplotyping of AvCoV isolates after passages of two field strains in chicken embryonated eggs. For the first and third passages of strain 23/2013, virus loads were 6.699 log copies/ μL and 6 log copies/ μL and, for 38/2013, 5.699 log copies/μL and 2.699 log copies/μL of reaction, respectively. The first passage of strain 23/2013 contained no variant haplotype, while, for the third passage, five putative variant haplotypes were found, with > 99.9% full genome identity with each other and with the dominant genome. Regarding strain 38/2013, five variant haplotypes were found for the first passage, with > 99.9% full genome identity with each other and with the dominant genome, and a single variant haplotype was found. Extinction and emergence of haplotypes with polymorphisms in genes involved in receptor binding and regulation of RNA synthesis were observed, suggesting that phenotypic traits of AvCoV isolates are a result of their mutant spectrum.

New Synthesis Method for a Core-Shell Composite Based on α-Bi2O3 @PPy and its Electrochemical Behavior as Supercapacitor Electrode

New composite based on polypyrrole (PPy) and bismuth oxide (α-Bi2O3) was investigated as supercapacitor electrode. The α-Bi2O3 was obtained by hydrothermal route at 500 ºC for 2 h. Cyclic voltammetry was used to electropolymerize PPy on graphite electrode (GE) or on GE/α-Bi2O3. The X-ray diffraction profile of Bi2O3 revealed the α-Bi2O3 monoclinic structure with space group P21/c.The scanning and transmission electron microscopy images showed that PPy coated α-Bi2O3. Raman spectra showed that PPy inhomogeneously coated α-Bi2O3, but it was still possible to obtain a α-Bi2O3 @PPy core-shell hybrid composite with highly ordered α-Bi2O3 with electronic interaction between the oxide and the polymer chain of the polymer. The GE/α-Bi2O3@PPy composite electrode (type I supercapacitor) displayed a predominantly capacitive profile with low impedance values and good electrochemical stability after 50 charge and discharge cycles. The specific capacitance of α-Bi2O3 @PPy composite was found in the range of 634 to 301 F g-1 to gravimetric current of 3 and 10 A g-1, respectively.

Abstract Coronary artery bypass grafting reduces plasma L-carnitine and may impair the production of myocardial energy. L-carnitine supplementation may elevate plasma L-carnitine and increase cardiac mechanical efficiency. The objective of this study was to verify the recovery of preoperative plasma LC in patients with heart failure undergoing coronary artery bypass grafting supplemented with a daily oral dose of 50 mg / kg. Volunteers with ischemic heart failure who underwent surgery were randomized into a supplemented group (A-received 50 mg / kg L-carnitine) or placebo group (B) for 60 days. Supplementation was started on the third postoperative day. The spectrophotometric enzymatic method was used to quantify plasma L-carnitine. In the preoperative period, both groups had plasma L-carnitine adequate to the reference range (18.9-71.1 μM). On the second postoperative day, there was a reduction in plasma L-carnitine in groups A (17.4%) and B (14.4%). In the comparison between the groups, plasma L-carnitine was higher in group A than B in 10º (p = 0.024), 30º (p = 0.001), and 60º postoperative day (p = 0.008). Supplementation of L-carnitine at a daily oral dose of 50 mg / kg in patients with heart failure undergoing coronary artery bypass grafting may recover preoperative plasma L-carnitine within 10 days.

Fotossensibilização primária em bovinos leiteiros causada por Froelichia humboldtiana

RESUMO: O presente trabalho foi conduzido com o objetivo de relatar um surto de fotossensibilização causado por Froelichia humboldtiana em bovinos leiteiros no Estado do Rio Grande do Norte, Brasil. Foram examinados animais de uma propriedade rural que apresentavam sintomatologia compatível com fotodermatite. Procedeu-se a coleta de amostras de sangue periférico de cinco bovinos para análise das atividades das enzimas hepáticas gamaglutamiltransferase e aspartatoaminotransferase, além da concentração de bilirrubina total, direta e indireta. Das áreas de pele com lesões de dois animais foram realizadas biópsias. Constatou-se que 15 animais de um rebanho composto por 40 animais apresentaram fotossemsibilização. Os animais tinham histórico de apresentar lesões de fotodermatite aproximadamente 10 dias após pastarem em áreas invadidas por F. humboldtiana. Ao exame clínico dos bovinos leiteiros notou-se que inicialmente apresentavam prurido e hiperemia nas áreas de pele despigmentadas do dorso e úbere, também havia alterações do comportamento. Posteriormente, as áreas hiperêmicas se apresentavam com edema que evoluíam para dermatite ulcerativa, necrotizante e exudativa, com perda de extensas áreas da epiderme. As úlceras eram mais graves nos quatro bovinos que apresentavam automutilação por lambedura. Esses quatro animais foram retirados do pasto e abrigados em local sombreado. Uma semana após, o prurido regrediu e as fissuras da pele passaram a cicatrizar. Porém, as lesões reapareceram logo após os bovinos serem reintroduzidas no pasto infestado por F. humbolditiana. Percebeu-se ainda queda na produção leiteira (redução de 50-60%) das vacas após a instalação de fotodermatite. Porém, os bezerros que ainda eram lactantes e ingeriam o leite nas vacas acometidas por fotossensibilização, não apresentaram sinais de fotodermatite. A histopatologia de biópsias de pele revelou inflamação na derme superficial constituída por mastócitos, linfócitos, e alguns plasmócitos. Na epiderme haviam extensas úlceras, recobertas por crostas, associada a infiltrado neutrofílico. As atividades séricas de AST, GGT e as concentrações de bilirrubina estavam dentro dos valores de referência normais para a espécie bovina. O diagnóstico de fotossensibilização primária associada à ingestão de F. humboldtiana foi baseado na epidemiologia, sinais clínicos, bioquímica sérica, biópsia de pele e reocorrência das lesões após os animais serem reintroduzidos no pasto invadido pela planta. Conclui-se que a F. humboldtiana é uma importante causa de fotossensibilização primária em bovinos leiteiros no semiárido brasileiro e que sua toxina provavelmente não é excretada pelo leite bovino.
ABSTRACT: The present study was conducted with the objective to report an outbreak of photosensitization caused by Froelichia humboldtiana in dairy cattle in the State of Rio Grande do Norte, Brazil. Animals from a rural property with symptoms compatible with photodermatitis were examined. Peripheral blood samples from five cattle were collected for the analysis of the activities of hepatic enzymes gammaglutamyltransferase and aspartate aminotransferase, in addition were also analysed the concentration of total, direct and indirect bilirubin. From the areas of skin with lesions of two animals, biopsies were performed. It was verified that 15 animals from a herd composed by 40 animals presented photosensitization. The animals had a history of photodermatitis lesions approximately 10 days after grazing in areas invaded by F. humboldtiana. Clinical examination of dairy cattle showed that they initially had pruritus and hyperemia in the depigmented areas of the dorsum and udder, and there were also behavioral changes. Subsequently, the hyperemic areas presented edema that evolved to ulcerative, necrotizing and exudative dermatitis, with loss of extensive areas of the epidermis. The ulcers were more severe in four bovines that had self-mutilation by licking. These four animals were removed from the pasture and sheltered in a shady location. A week later, the pruritus regressed and the fissures of the skin began to heal. However, the lesions reappeared after the cattle were reintroduced in the grass infested by F. humbolditiana. There was also a decrease in milk production (reduction of 50-60%) of cows after the installation of photodermatitis. However, calves that were still lactating and ingested the milk in photosensitized cows, showed no signs of photodermatitis. Histopathology of skin biopsies revealed inflammation in the superficial dermis consisting of mast cells, lymphocytes, and some plasma cells. In the epidermis there were extensive ulcers, covered by crusts, associated with neutrophilic infiltrate. Serum activities of AST, GGT and bilirubin concentrations were within normal reference values for the bovine species. The diagnosis of primary photosensitization associated with F. humboldtiana ingestion was based on epidemiology, clinical signs, serum biochemistry, skin biopsy and lesion reoccurrence after the animals were reintroduced in the pasture invaded by the plant. It is concluded that F. humboldtiana is an important cause of primary photosensitization in dairy cattle in the Brazilian semi-arid region and that its toxin is probably not excreted by bovine milk.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Espessura íntima-média carotídea como marcador de risco cardiovascular em pacientes com hipotireoidismo subclínico

O hipotireoidismo subclínico (HS) já foi associado a aumento do risco cardiovascular. Na avaliação desse risco, a medida da espessura íntima-média (EIM) carotídea por ultra-sonografia é capaz de detectar alterações iniciais da aterosclerose. O objetivo deste estudo foi avaliar a EIM carotídea em pacientes com HS e sua associação com um provável aumento do risco cardiovascular. Não foi encontrada diferença significativa nas medidas da EIM das pacientes com HS e das controles. Os resultados encontrados nos dois grupos estudados foram, respectivamente: 0,573 ± 0,070 mm vs. 0,576 ± 0,068 mm para as carótidas comuns (p= 0,904) e 0,602 ± 0,079 mm vs. 0,617 ± 0,102 mm para as bifurcações (p= 0,714). Mesmo após estratificação das pacientes de acordo com o TSH e com a presença ou não de auto-imunidade, a diferença entre os sub-grupos permaneceu sem significância estatística. As medidas da EIM nesses grupos nos sítios avaliados foram: TSH 4-8 mUI/L: 0,579 ± 0,070 mm e 0,586 ± 0,063 mm; TSH > 8 mUI/L: 0,569 ± 0,073 mm e 0,616 ± 0,091 mm; anti-TPO+: 0,585 ± 0,070 mm e 0,621 ± 0,085 mm; anti-TPO-: 0,554 ± 0,072 mm e 0,571 ± 0,066 mm. Também não houve diferença no lipidograma e nas dosagens de apoproteína B e de lipoproteína (a). Este fato sugere que o HS, quando leve, sem alterações metabólicas associadas, não promove aumento do risco cardiovascular.
Subclinical hypothyroidism (SH) has been associated with an increased risk for coronary disease. Carotid intima-media thickness (IMT), as assessed by ultrasonography, is a precise marker of atherosclerotic changes and can be used as an endpoint for cardiovascular events. Aims of this study were to determine carotid IMT in a group of patients with SH and its possible association with an increase in cardiovascular risk. There were no significant differences in mean carotid IMT between patients and controls. Results of both groups were, respectively: common carotid arteries, 0.573 ± 0.070 mm and 0.576 ± 0.068 mm (p= 0.904); carotid bifurcation, 0.602 ± 0.079 mm and 0.617 ± 0.102 mm (p= 0.714). Similar results were obtained when analyzing subgroups with serum TSH < or > 8 mIU/L and with positive or negative titers of TPOAb. The mean carotid IMT in these subgroups were: TSH 4-8 mIU/L: 0.579 ± 0.070 mm and 0.586 ± 0.063 mm; TSH > 8 mIU/L: 0.569 ± 0.073 mm and 0.616 ± 0.091 mm; TPOAb+: 0.585 ± 0.070 mm and 0.621 ± 0.085 mm; TPOAb-: 0.554 ± 0.072 mm and 0.571 ± 0.066 mm. No differences in the lipid profile and in the apoprotein B and lipoprotein (a) levels between the groups were found. These findings suggest that mild SH with no related metabolic changes is not associated with an increase in cardiovascular risk, as assessed by carotid IMT.
