Conventional diagnostic methods and immunohistochemistry in the detection of gastric Helicobacter species in dogs with chronic gastropathy

ABSTRACT An accurate diagnostic test for Helicobacter spp. infection in dogs is necessary but no gold standard diagnostic method has yet been established. The purpose of this study was to assess the sensitivity of conventional diagnostic methods as opposed to immunohistochemistry (IHC) in the detection of Helicobacter spp. in gastric samples of dogs with chronic gastropathy. Samples of gastric fundus, body and antrum were collected by gastroscopy from 13 domestic dogs presenting chronic vomit and submitted to the rapid urease test (RUT), cytopathology, histopathology with Hematoxylin-Eosin (HE) and Warthin-Starry (WS) stain, and IHC. Cohen's kappa coefficient was carried out to determine the agreement between techniques compared to IHC. Prevalence of colonization detected by IHC was 92% and was higher in the gastric fundus. The sensitivity of each technique in detecting Helicobacter spp. in the gastric fundus, body, and antrum, respectively, were as follows: RUT 50%, 70%, 17%; cytology 58%; 70%, 50%; HE 42%; 70%, 50%; WS 67%; 80%, 50%. We found that IHC was the most sensitive method for Helicobacter spp. diagnosis revealing that gastric fundus samples have higher presence of bacteria. Squash cytology enhances sensitivity and ancillary staining such as WS should be taken into consideration towards diagnosis.
RESUMO É necessário um teste diagnóstico preciso no diagnóstico de Helicobacter spp. em cães, mas nenhum método padrão-ouro foi estabelecido até o momento. O objetivo foi avaliar a sensibilidade dos métodos diagnósticos convencionais em comparação com a imuno-histoquímica (IHC) na detecção de Helicobacter spp. em amostras gástricas de cães com gastropatia crônica. Amostras de fundo, corpo e antro gástrico foram coletadas por gastroscopia de 13 cães com vômitos crônicos, e submetidas ao teste rápido da urease (RUT), citopatologia, histopatologia com coloração de hematoxilina-eosina (HE) e Warthin-Starry (WS), e IHC. O coeficiente kappa de Cohen foi realizado para determinar a concordância entre as técnicas, comparadas ao IHC. A prevalência de colonização detectada na IHC foi de 92%, sendo maior no fundo gástrico. A sensibilidade de cada técnica na detecção de Helicobacter spp. no fundo, corpo e antro gástrico, respectivamente, foram: RUT 50%, 70%, 17%; citologia 58%; 70%, 50%; HE 42%; 70%, 50%; WS 67%; 80%, 50%. Concluiu-se que a IHC foi o método mais sensível de diagnóstico de Helicobacter spp., revelando que as amostras de fundo gástrico têm maior presença de bactérias. A citologia por squash aumenta a sensibilidade, e a coloração auxiliar WS deve ser levada em consideração para o diagnóstico.

RESUMO O objetivo do presente estudo foi padronizar um protocolo de reação em cadeia da polimerase (PCR) para a autenticação de leite bovino e bubalino e a detecção da presença de Salmonella spp. e Listeria monocytogenes. Para isso, o DNA-alvo foi extraído, misturado e submetido ao ensaio de PCR. Amostras de leite foram fraudadas e contaminadas experimentalmente com os micro-organismos, para se avaliar a detecção do DNA-alvo em diferentes tempos de cultivo, os títulos bacterianos e a concentração de material genético. Além disso, o protocolo foi testado com DNA extraído diretamente do alimento, sem a etapa de pré-enriquecimento. A PCR quadriplex proposta mostrou boa precisão na identificação de sequências de DNA-alvo. Foi possível identificar simultaneamente todas as sequências de DNA no momento da inoculação (0h), quando as amostras estavam contaminadas com 2 UFC/250mL, e com seis horas de cultura, quando o inóculo inicial foi de 1 UFC/250mL. Também foi possível detectar diretamente as sequências de DNA do alimento quando este foi inoculado com 3 UFC/mL de bactérias. Dessa forma, a metodologia proposta apresentou desempenho satisfatório, otimização do tempo de análise e potencial para detecção de micro-organismos em baixos títulos, podendo ser utilizada para detecção de fraude e contaminação.
ABSTRACT The objective of the present study was to Standardize a Polymerase Chain Reaction (PCR) protocol for the authentication of bovine and buffalo milk, and to detect the presence of Salmonella spp. and Listeria monocytogenes. For this, the target DNA was extracted, mixed, and subjected to a PCR assay. Milk samples were defrauded and experimentally contaminated with microorganisms to assess the detection of target DNA at different times of cultivation, bacterial titers, and concentration of genetic material. In addition, the protocol was tested with DNA extracted directly from food, without a pre-enrichment step. The proposed quadruplex PCR showed good accuracy in identifying target DNA sequences. It was possible to simultaneously identify all DNA sequences at the time of inoculation (0h), when the samples were contaminated with 2 CFU/250mL and with 6h of culture when the initial inoculum was 1 CFU/250mL. It was also possible to directly detect DNA sequences from the food when it was inoculated with 3 CFU/mL bacteria. Thus, the proposed methodology showed satisfactory performance, optimization of the analysis time, and a potential for the detection of microorganisms at low titers, which can be used for the detection of fraud and contamination.

Abstract The influence of environmental variables on planktonic biodiversity is widely known. However, the absence of information about the cyanobacterial community in tropical estuarine regions motivated this work, whose objective was to investigate the spatio-temporal variation of cyanobacterial density related to physicochemical factors in a Brazilian Amazonian estuary. For the qualitative and quantitative study of cyanobacteria and physicochemical variables, samples were collected in April/July/2009 and April/August/2010. We identified 31 species of the orders Chroococcales, Oscillatoriales and Nostocales. Species of the genera Aphanocapsa, Dolichospermum, Komvophoron, Microcystis, Pseudanabaena and Merismopedia were frequent and abundant throughout the study period. Some of the found genera have already been described as potential toxin producers. The dynamics of cyanobacteria were temporal, the highest densities occurred in 2010 (average= 1080.86 ± 702.86 cells.mL-1) mainly influenced by the high values of pH, temperature, electrical conductivity, total dissolved solids, ammonium nitrogen which led cyanobacteria to present different responses in terms of richness, density and diversity between the years.

RESUMO O presente trabalho relata o caso de aspergilose sistêmica em um cavalo da raça Quarto de Milha. O animal apresentava quadro de emagrecimento progressivo e alopecia. Os parâmetros avaliados durante o exame clínico encontravam-se de acordo com os valores normais para a espécie. No hemograma verificou-se leucocitose por neutrofilia, sem desvio à esquerda, proteínas plasmáticas totais elevadas e aumento do fibrinogênio, indicando um processo inflamatório acompanhado de desidratação. Os exames bioquímicos séricos demonstraram aumento no valor da ureia. Apesar dos exames realizados e da terapia instituída com antibióticos de amplo espectro e suporte nutricional, após 51 dias de internamento o quadro clínico evoluiu para caquexia e decúbito permanente do paciente, o qual foi submetido à eutanásia seguida de necropsia. Com base nos achados nos exames necroscópico e histopatológico, diagnosticou-se infecção sistêmica por Aspergillus sp., acometendo pulmões, fígado, rins, peritônio parietal e encéfalo. Dentre os fatores predisponentes à relatada infecção fúngica sistêmica, destaca-se o status imunológico debilitado do paciente, possivelmente proporcionado por tratamentos prolongados com antibióticos e anti-inflamatórios esteroidais. O presente trabalho relata o primeiro caso descrito de arpergilose sistêmica no Brasil. O diagnóstico definitivo só foi possível após necropsia e histopatologia, visto que os sinais clínicos inespecíficos não foram determinantes para o diagnóstico em vida.
ABSTRACT This paper reports the case of systemic aspergillosis on a Quarter Horse. The animal had progressive weight loss and alopecia. The parameters evaluated during the clinical examination were in agreement with the normal values for the species. The hemogram showed neutrophilic leukocytosis, without left-sided, elevated total plasma proteins and an increase in fibrinogen, indicating an inflammatory process accompanied by dehydration. Serum biochemical tests showed an increase in the value of urea. Despite the performed tests and established therapy with broad spectrum antibiotics and nutritional support, after 51 days of hospitalization the clinical condition evolved into cachexia and permanent decubitus of the patient which underwent to euthanasia and necropsy. From the findings of necropsy and histopathology, systemic infection was diagnosed by Aspergillus sp., affecting the lungs, liver, kidneys, parietal peritoneum and encephalon. Among the factors predisposing to the reported systemic fungal infection, the patient's weakened immune status is highlighted, possibly due to prolonged antibiotic and steroidal anti-inflammatory treatments. This paper reports the first case described of systemic aspergillosis in Brazil. Definitive diagnosis was only possible after necropsy and histopathology, since nonspecific clinical signs were not determinant for the diagnosis in life.

RESUMO Objetivou-se, com este trabalho, avaliar a quantidade de fibra na dieta que é capaz de promover melhor uso de nutrientes na dieta de cabras leiteiras quando a palma forrageira é a principal fonte de forragem. Quinze cabras leiteiras alpinas foram distribuídas em um quadrado latino triplo 5 × 5 contendo cinco tratamentos e cinco períodos. Testaram-se cinco dietas compostas por diferentes teores de fibra em detergente neutro de feno de tifton (FDNf) na matéria seca de rações contendo palma forrageira: D1: 11,98%; D2: 18,31%; D3: 23,68%; D4: 28,76% e D5: 32,12% de FDNf. A ingestão de matéria seca, proteína bruta, fibra em detergente neutro, carboidratos não fibrosos e extrato etéreo diminuiu linearmente com o aumento do conteúdo de FDNf. Houve um aumento linear da produção de leite e da proteína do leite com o aumento da FDNf dietética. A gordura, a lactose, o extrato seco, o pH e a acidez do leite não foram afetados pelo aumento dos níveis de FDNf na dieta. Em situações em que as cabras leiteiras são alimentadas com palma forrageira e outros tipos de forragem são limitados, 11,98% de FDN de forragem diferente da palma podem ser usados como fonte efetiva de fibra.
ABSTRACT This study aimed to evaluate the amount of fiber in the diet that is able to promote better use of nutrients in the diet of dairy goats when forage palm is the main source of roughage. Fifteen Alpine dairy goats were distributed in a 5 × 5 triple Latin square containing five treatments and five periods, and each treatment contained. We tested five diets composed of different contents of neutral detergent fiber from Tifton hay (NDFf) in the dry matter of rations containing forage palm: D1: 11.98%; D2: 18.31%; D3: 23.68%; D4: 28.76% and D5: 32.12% NDFf. The intake of dry matter, crude protein, neutral detergent fiber, non-fiber carbohydrates and ether extract decreased linearly with the increase in NDFf content. There was a linear increase of milk production and milk protein with increasing dietary NDFf. The fat, dry extract, protein, pH and acidity of the evaluated milk were not affected by increasing the levels of NDFf in the diet. In situations where dairy goats are fed forage palm and other types of forage are limited, 11.98% NDF from forage other than palm can be used as an effective source of NDF.

RESUMO Objetivou-se avaliar a influência de dietas contendo silagem de diferentes cultivares de sorgo sobre as características físicas e químicas na carne de ovinos sem padrão racial definido, com idade entre cinco e sete meses e peso vivo médio de 17,7±3,7kg, alimentados com dietas contendo silagem de diferentes cultivares de sorgo. O experimento foi conduzido na Estação Experimental Pendência, pertencente à Empresa Estadual de Pesquisa Agropecuária - Emepa, tendo um período experimental de 42 dias. Utilizou-se um delineamento inteiramente ao acaso, com cinco tratamentos (silagem de sorgo BRS 610, BRS 655, BRS 800, BRS 810 e Ponta Negra) e sete repetições. Ao atingirem o peso de abate de 26,24kg, os animais foram encaminhados ao abate. Procedeu-se à análise do pH, da cor, da perda de peso por cocção, da força de cisalhamento, bem como à determinação de umidade, proteínas, cinzas e lipídeos totais no músculo Longissimus dorsi. As dietas não influenciaram os parâmetros de pH (5,64), cor (L*23,13, a* 18,00, b*15,78), perdas de peso por cocção (34,43%), umidade (72,53%), proteína (24,08%), cinzas (1,09%) e lipídeos (2,99%). A força de cisalhamento diferiu entre as dietas, com a silagem do cultivar BRS 655 propiciando uma carne mais macia (2,10kgf/cm2). As silagens de sorgo utilizadas na confecção das dietas de cordeiros em confinamento não interferem nas características de qualidade da carne de ovinos sem padrão racial definido. Dietas contendo silagem de sorgo granífero BRS 655 proporcionaram uma carne mais macia.
ABSTRACT This study aimed to evaluate the influence of diets containing silage of different sorghum cultivars on the physical and chemical characteristics in the meat of lambs. Thirty five animals of undefined breed with average age between 5 and 7 months and average body weight of 17. 7±3.7kg were used. The lambs were randomly assigned to five treatments (sorghum silage BRS 610, BRS 655, BRS 800, BRS 810 e Ponta Negra) with seven replicates. When they reach 26.24kg of weight, the animals were sent to be slaughtered. Proceeded to the analysis of pH, color, cooking losses, shear force, determination of humidity, protein, ash and total amount of lipids in the Longissimus dorsi. The diets did not influence the parameters of pH (5.64), color (L*23.13, a* 18.00, b*15.78), cooking losses (34.43%), humidity (72.53%), protein (24.08%), ash (1.09%) or lipids (2.99%). The shear force differed among the diets, with the silage of cultivar BRS 655 providing a tender meat (2.10kgf/cm2). The sorghum silages used in in the diets did not interfere in the meat quality characteristics of feedlot lambs. Diets containing sorghum grain silage BRS 655 provided a tender meat.

Recent evidence suggests that cell-derived circulating miRNAs may serve as biomarkers of cardiovascular diseases. However, a few studies have investigated the potential of circulating miRNAs as biomarkers for left ventricular hypertrophy (LVH). In this study, we aimed to characterize the miRNA profiles that could distinguish hypertensive patients with LHV, hypertensive patients without LVH and control subjects, and identify potential miRNAs as biomarkers of LVH. LVH was defined by left ventricular mass indexed to body surface area >125 g/m2 in men and >110 g/m2 in women and patients were classified as hypertensive when presenting a systolic blood pressure of 140 mmHg or more, or a diastolic blood pressure of 90 mmHg or more. We employed miRNA PCR array to screen serum miRNAs profiles of patients with LVH, essential hypertension and healthy subjects. We identified 75 differentially expressed miRNAs, including 49 upregulated miRNAs and 26 downregulated miRNAs between LVH and control patients. We chose 2 miRNAs with significant differences for further testing in 59 patients. RT-PCR analysis of serum samples confirmed that miR-7-5p and miR-26b-5p were upregulated in the serum of LVH hypertensive patients compared with healthy subjects. Our findings suggest that these miRNAs may play a role in the pathogenesis of hypertensive LVH and may represent novel biomarkers for this disease.

RESUMO Este estudo teve como objetivo avaliar a suplementação do composto antioxidante comercial EconomasE (Alltech, Brasil ( AOX) sobre o equilíbrio acidobásico, os parâmetros urinários, o hemograma completo e a fragilidade osmótica de eritrócitos (FOE) de gatos estressados. Foram utilizados 24 gatos adultos (3,49±0,87kg), distribuídos em delineamento inteiramente ao acaso, com quatro níveis (0, 250, 500, 750mg de AOX/kg de alimento na matéria seca) e seis repetições, durante o período de 80 dias. Os gatos foram induzidos ao estresse por meio da presença de cães próximo ao recinto experimental (agente estressor; AE), do 61º dia até o final do experimento. A pressão parcial de dióxido de carbono (pCO2) e o bicarbonato (HCO3 -) aumentaram linearmente (P<0,05) conforme o aumento da ingestão de AOX. Os maiores valores de dióxido de carbono total (tCO2) (P<0,05) foram observados nos gatos alimentados com 500 e 750mg de AOX/kg de dieta. As concentrações de hemoglobina foram maiores nos animais alimentados com 250 e 500mg de AOX/kg de dieta. Os parâmetros urinários e da FOE foram semelhantes nos gatos em todas as dietas. Esses dados indicam que a suplementação com AOX apresenta efeitos benéficos no equilíbrio acidobásico e na concentração de hemoglobina de gatos induzidos ao estresse.
ABSTRACT This study aimed to evaluate the supplementation of a commercial antioxidant EconomasE (Alltech, Brazil - AOX) on the acid-base balance, urinary parameters, red blood count (RBC) and erythrocyte osmotic fragility (EOF) of stressed cats. Twenty-four adult cats (3.49±0.87kg) were distributed in a completely randomized design with four levels (0, 250, 500, 750mg AOX/kg feed dry matter) and six replicates for a period of 80 days. Cats were induced to stress through the presence of dogs in the experimental environment (stressor agent; AE) from the 61th day to the end. Partial pressure of carbon dioxide (pCO2) and bicarbonate (HCO3) increased linearly (P<0.05) with increased intake of AOX. The highest values of total carbon dioxide (tCO2) (P<0.05) were observed in cats fed the 500 and 750mg AOX/kg diet. Hemoglobin concentration was higher (P<0.05) in animals fed the 250 and 500mg AOX/kg diet. The urinary parameters and EOF were similar among all diets. These data indicate that AOX supplementation has beneficial effects in acid-base balance and hemoglobin concentration of stress-induced cats.

O presente trabalho visou avaliar as temperaturas superficiais de diferentes regiões anatômicas de búfalos ao longo do tempo, por meio da termografia infravermelha, e correlacioná-las a índices bioclimatológicos de conforto térmico. O ensaio foi realizado em região de clima tropical úmido (Afi de Köppen), de abril a agosto. Dez touros (n=10) foram avaliados a cada 25 dias (manhã: seis-nove horas; tarde: 12-15h), quanto à frequência respiratória (FR), temperatura retal (TR) e imagens termográficas da órbita ocular (ORB), flanco direito (FLd), flanco esquerdo (FLe) e escroto (ESC). Os dados climatológicos foram ininterruptamente monitorados, e calculados o índice de temperatura e umidade (ITU) e o índice de conforto de Benezra (ICB). O ITU foi ≥78, com diferença entre turnos (P<0,05). Já o ICB variou de 1,96 a 2,25 e apresentou diferenças ao longo dos meses e entre turnos (P<0,05). As temperaturas observadas foram de TR=38,2±0,5ºC, ORB=36,1±0,8ºC, FLd=33,5±2,5ºC, FLe=35,4±1,7ºC e ESC=33,3±1,1ºC, as quais variaram significativamente ao longo dos meses e entre turnos (P<0,05). O ITU apresentou correlações positivas com ORB (0,72), FLd (0,77), FLe (0,75) e ESC (0,41) (P<0,0001). A temperatura máxima de ORB apresentou a maior correlação com a TR (0,58; P<0,0001). Portanto, as temperaturas superficiais dos animais sofrem interferências das variações climáticas e se elevam ao longo do dia, devido à variação nos índices de conforto térmico; a temperatura máxima de ORB foi o parâmetro mais condicionado à temperatura retal. Também, as oscilações de temperatura de superfície de ORB, FLd, FLe e ESC podem ser aferidas em bubalinos com o uso da termografia infravermelha, de modo preciso e não invasivo.
This paper aimed to evaluate the surface temperatures of buffalo bulls using infrared thermography, considering four distinct anatomical parts over time, and to correlate surface temperatures and thermal comfort indexes. The humid tropical climate (Köppen's Afi) was predominant in the research station where the experiment was performed and the trial lasted from April to August. Ten bulls (n=10) were evaluated every 25 days (morning: 6:00-9:00; afternoon: 12:00-15:00) and the parameters assessed were respiratory rate (RR), rectal temperature (RT), and the thermograms of surface temperature for orbital area (ORB), right flank (RF), left flank (LF) and scrotum (SCR). Climatological data was continuously monitored and the Temperature and Humidity Index (THI) and the Index of Comfort of Benezra (ICB) were calculated. The average values of THI were ≥78, and significant differences between shifts were observed (P<0.05). The ICB ranged from 1.96 to 2.25 and significant differences were observed for shifts and throughout the months (P<0.05). The averages of surface temperatures were RT=38.2±0.5°C, ORB=36.1±0.8°C, LF=33.5±2.5°C, RF=35.4±1.7ºC and SCR=33.3±1.1°C, which exhibited significant differences for shifts and throughout the months (P<0.05). Positive correlations were obtained between THI and ORB (0.72), RF (0.77), LF (0.75) and SCR (0.41) (P<0.0001). The maximum temperature of ORB showed the highest correlation with RT (0.58, P<0.0001). Therefore, the surface temperatures are subject to climatic variations and increase throughout the day, due to the variation in thermal comfort indexes, and the maximum ORB temperature was the parameter most related to rectal temperature. Lastly, the results indicate that IRT may be a useful non-invasive and accurate tool to detect the variations in ORB, LF, RF and SCR temperature in buffalo bulls.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The performances of the diluents TES and CEBRAN II were compared as cryopreservatives of semen from non human primates of the genus Ateles. The experiment was carried out using one Ateles marginatus and two Ateles paniscus specimens, males and adults, maintained in the same captivity conditions at the National Center of Primates (CENP-SVS/MS). The animals were subjected to clinical and andrological examinations - testicular biometry - before the semen collection by eletroejaculation. Evaluations of motility and forward movement in the fresh semen were made. Semen were made dilution was made with the diluents TES and CEBRAN II. The ejaculates were diluted with the diluents (2:1proportion), packed in 0.25mL plastic straws and cryopreserved in liquid nitrogen. After thawing, the packed ejaculates were appraised in thermo resistance test (TTR). The averages of volume and concentration were, respectively, 1.94mL (0.83) and 3,020,000 sptz/mL (275.97). The pH 8 and seminal coagulation were observed in all samples. The results suggest that the TES diluent presents better efficiency in the preservation of Ateles semen than CEBRAN II.

Plasma cytokine response, lipid peroxidation and NF-κB activation in skeletal muscle following maximum progressive swimming

Our objective was to determine lipid peroxidation and nuclear factor-κB (NF-κB) activation in skeletal muscle and the plasma cytokine profile following maximum progressive swimming. Adult male Swiss mice (N = 15) adapted to the aquatic environment were randomly divided into three groups: immediately after exercise (EX1), 3 h after exercise (EX2) and control. Animals from the exercising groups swam until exhaustion, with an initial workload of 2% of body mass attached to the tail. Control mice did not perform any exercise but were kept immersed in water for 20 min. Maximum swimming led to reactive oxygen species (ROS) generation in skeletal muscle, as indicated by increased thiobarbituric acid reactive species (TBARS) levels (4062.67 ±1487.10 vs 19,072.48 ± 8738.16 nmol malondialdehyde (MDA)/mg protein, control vs EX1). Exercise also promoted NF-κB activation in soleus muscle. Cytokine secretion following exercise was marked by increased plasma interleukin-6 (IL-6) levels 3 h post-exercise (P < 0.05). Interleukin-10 (IL-10) levels were reduced following exercise and remained reduced 3 h post-exercise (P < 0.05). Plasma levels of other cytokines investigated, monocyte chemotactic protein-1 (MCP-1), tumor necrosis factor-alpha (TNF-α), interferon-gamma (IFN-γ) and interleukin-12 (IL-12), were not altered by exercise. The present findings showed that maximum swimming, as well as other exercise models, led to lipid peroxidation and NF-κB activation in skeletal muscle and increased plasma IL-6 levels. The plasma cytokine response was also marked by reduced IL-10 levels. These results were attributed to exercise type and intensity.

Computer simulation of sympatric speciation with Penna ageing model

One species is simulated to split into two separate species via random mutations, even if both populations live together in the same environment. This speciation is achieved in the Penna bitstring model of biological ageing, with modified Verhulst factors, and in part by additional bitstrings regulating phenotype and mate selection.
