A New Molecularly Imprinted Polymer for In-Tube SPME/UHPLC-MS/MS of Anandamide in Plasma Samples

Parkinson’s disease is a neurodegenerative disorder characterized by progressive loss of dopaminergic neurons. There is substantial evidence that the endocannabinoid system modulates the dopaminergic activity in the basal ganglia, a forebrain system that integrates cortical information to coordinate motor activity regulating signals. In this article, a fused-silica capillary with a molecularly imprinted polymer was developed for in-tube solid-phase microextraction of the endocannabinoid anandamide in plasma samples from Parkinson’s disease patients and further analysis by ultra-high-performance liquid chromatography with tandem mass spectrometry. The molecularly imprinted polymer capillary presented recognition sites with complementary shape, size, and functionality to anandamide. Scanning electron micrographs and Fourier transform infrared spectra illustrated the physical and chemical modification of the printed and non-printed capillary surface. The in-tube solid-phase microextraction ultra-high-performance liquid chromatography with tandem mass spectrometry method presented a linear range from 0.1 to 20 ng mL-1, precision with coefficient of variation values ranging from 1.2 to 13%, and relative standard deviation accuracy ranging from -3.6 to 7.5%. The method developed herein can adequately determine anandamide in plasma samples from Parkinson’s disease patients. By applying the standard addition approach, the anandamide plasmatic concentration in these samples was found to range from 0.2 to 0.4 ng mL-1.

Abstract The iAtlantic Project has established an international collaborative strategy to improve mapping and characterization of deep and open ocean ecosystems in understudied regions of the Atlantic and evaluate their health. In December 2022, the first iAtlantic expedition in the South Atlantic set off to map and explore seafloor ecosystems in the Santos Basin slope (200-1,000 m depths) in collaboration with the Petrobras ‘Santos Basin - Regional Characterization Project.’ The 17-day ‘iAtlantic_BR10-Petrobras’ cruise was conducted on board the research vessel NPqHOc Vital de Oliveira (Brazilian Navy) and performed (a) water column structure characterization, (b) seafloor morphology mapping, (c) description of benthic habitats and communities by seafloor imagery and biological/ geological sampling, and (d) ex-situ experiments to assess the functioning of sedimentary ecosystems and their responses to climate-related environmental changes. This study describes the rationale behind the iAtlantic_BR10-Petrobras cruise science plan, reports its sampling strategy and methods, and summarizes its collected data and preliminary results.

This guideline aimed to update the concepts and formulate the standards of conduct and scientific evidence that support them, regarding the diagnosis and treatment of the Cardiomyopathy of Chagas disease, with special emphasis on the rationality base that supported it.nbsp;
Chagas disease in the 21st century maintains an epidemiological pattern of endemicity in 21 Latin American countries. Researchers and managers from endemic and non-endemic countries point to the need to adopt comprehensive public health policies to effectively control the interhuman transmission of T. cruzi infection, and to obtain an optimized level of care for already infected individuals, focusing on diagnostic and therapeutic opportunistic opportunities.

Pathogenic and pathophysiological mechanisms of the Cardiomyopathy of Chagas disease were revisited after in-depth updating and the notion that necrosis and fibrosis are stimulated by tissue parasitic persistence and adverse immune reaction, as fundamental mechanisms, assisted by autonomic and microvascular disorders, was well established. Some of them have recently formed potential targets of therapies.nbsp;
The natural history of the acute and chronic phases was reviewed, with enhancement for oral transmission, indeterminate form and chronic syndromes. Recent meta-analyses of observational studies have estimated the risk of evolution from acute and indeterminate forms and mortality after chronic cardiomyopathy. Therapeutic approaches applicable to individuals with Indeterminate form of Chagas disease were specifically addressed. All methods to detect structural and/or functional alterations with various cardiac imaging techniques were also reviewed, with recommendations for use in various clinical scenarios. Mortality risk stratification based on the Rassi score, with recent studies of its application, was complemented by methods that detect myocardial fibrosis.nbsp;
The current methodology for etiological diagnosis and the consequent implications of trypanonomic treatment deserved a comprehensive and in-depth approach. Also the treatment of patients at risk or with heart failure, arrhythmias and thromboembolic events, based on pharmacological and complementary resources, received special attention. Additional chapters supported the conducts applicable to several special contexts, including t. cruzi/HIV co-infection, risk during surgeries, in pregnant women, in the reactivation of infection after heart transplantation, and others.nbsp; nbsp;nbsp;
Finally, two chapters of great social significance, addressing the structuring of specialized services to care for individuals with the Cardiomyopathy of Chagas disease, and reviewing the concepts of severe heart disease and its medical-labor implications completed this guideline.
Esta diretriz teve como objetivo principal atualizar os conceitos e formular as normas de conduta e evidências científicas que as suportam, quanto ao diagnóstico e tratamento da CDC, com especial ênfase na base de racionalidade que a embasou.
A DC no século XXI mantém padrão epidemiológico de endemicidade em 21 países da América Latina. Investigadores e gestores de países endêmicos e não endêmicos indigitam a necessidade de se adotarem políticas abrangentes, de saúde pública, para controle eficaz da transmissão inter-humanos da infecção pelo T. cruzi, e obter-se nível otimizado de atendimento aos indivíduos já infectados, com foco em oportunização diagnóstica e terapêutica.
Mecanismos patogênicos e fisiopatológicos da CDC foram revisitados após atualização aprofundada e ficou bem consolidada a noção de que necrose e fibrose sejam estimuladas pela persistência parasitária tissular e reação imune adversa, como mecanismos fundamentais, coadjuvados por distúrbios autonômicos e microvasculares. Alguns deles recentemente constituíram alvos potenciais de terapêuticas.
A história natural das fases aguda e crônica foi revista, com realce para a transmissão oral, a forma indeterminada e as síndromes crônicas. Metanálises recentes de estudos observacionais estimaram o risco de evolução a partir das formas aguda e indeterminada e de mortalidade após instalação da cardiomiopatia crônica. Condutas terapêuticas aplicáveis aos indivíduos com a FIDC foram abordadas especificamente. Todos os métodos para detectar alterações estruturais e/ou funcionais com variadas técnicas de imageamento cardíaco também foram revisados, com recomendações de uso nos vários cenários clínicos. Estratificação de risco de mortalidade fundamentada no escore de Rassi, com estudos recentes de sua aplicação, foi complementada por métodos que detectam fibrose miocárdica.
A metodologia atual para diagnóstico etiológico e as consequentes implicações do tratamento tripanossomicida mereceram enfoque abrangente e aprofundado. Também o tratamento de pacientes em risco ou com insuficiência cardíaca, arritmias e eventos tromboembólicos, baseado em recursos farmacológicos e complementares, recebeu especial atenção. Capítulos suplementares subsidiaram as condutas aplicáveis a diversos contextos especiais, entre eles o da co-infecção por T. cruzi/HIV, risco durante cirurgias, em grávidas, na reativação da infecção após transplante cardíacos, e outros.nbsp;nbsp;nbsp;
Por fim, dois capítulos de grande significado social, abordando a estruturação de serviços especializados para atendimento aos indivíduos com a CDC, e revisando os conceitos de cardiopatia grave e suas implicações médico-trabalhistas completaram esta diretriz.nbsp;

ABSTRACT Introduction: Systemic Mastocytosis comprises a group of neoplastic diseases characterized by clonal expansion and infiltration of mast cells into several organs. The diagnosis and treatment of this disease may be challenging for non-specialists. Objective: Make suggestions or recommendations in Systemic Mastocytosis based in a panel of Brazilian specialists. Method and results: An online expert panel with 18 multidisciplinary specialists was convened to propose recommendations on the diagnosis and treatment of Systemic Mastocytosis in Brazil. Recommendations were based on discussions of topics and multiple-choice questions and were graded using the Oxford Centre for Evidence-Based Medicine 2011 Levels of Evidence Chart. Conclusion: Twenty-two recommendations or suggestions were proposed based on a literature review and graded according to the findings.

ABSTRACT The low quality of the water currently used for irrigation purposes harms the quality of the products and can lead to a reduction in production. Thus, the objective of this work was to verify the effect of saline water in irrigation water, on biometric variables of pumpkin crop using a system based on fuzzy rules. The agronomic experimental part of this work was carried out in a greenhouse. The experimental design was in randomized blocks, with 5 doses of salinity (0, 1.25, 2.5, 3.75, and 5 dS m-1) and with 5 repetitions. The salinity doses and evaluations carried out throughout the cycle (days after transplanting) were defined as input variables in the mathematical model. For the output variables, the collected biometric responses were defined: number of leaves, number of flowers, leaf fresh mass, leaf dry mass, stem fresh mass, stem length, root fresh mass, and root length. After evaluation, the mathematical model was developed and its validation was carried out using statistical methods and Receiver Operating Characteristic Curves (ROC). It was observed that salinity affects the bush pumpkin crop with a reduction in the evaluated parameters. The mathematical model proved to be efficient for this evaluation.

Resumo Objetivo: O objetivo principal deste artigo é compreender o comportamento de jovens em início de carreira no contexto da Covid-19, considerando a intenção de continuar utilizando os serviços da rede social profissional LinkedIn. Originalidade/valor: Este artigo amplia o escopo dos estudos de aceitação na intenção de continuar utilizando (uso contínuo) o LinkedIn, analisando uma série de fatores positivos e negativos em relação aos processos. Design/metodologia/abordagem: Realizou-se uma pesquisa descritiva com abordagem quantitativa. Os autores coletaram dados de um survey (n = 386) e, com o objetivo de testar as hipóteses, aplicaram a técnica de modelagem de equações estruturais (MEE), com estimação de mínimos quadrados parciais, e um efeito moderador com o construto otimismo tecnológico para as variáveis preditivas da intenção de continuar utilizando. Resultados: Os resultados indicaram que todas as relações diretas e indiretas foram aceitas, exceto uma moderação (H7d), que foi rejeitada, visto que os usuários menos otimistas com a tecnologia podem se sentir positivamente sobrecarregados quanto ao nível de utilidade percebida do serviço. A variedade de serviços do LinkedIn pode levar o usuário a uma intenção de continuidade no uso da plataforma, uma vez que o objetivo dessa rede é facilitar a autoapresentação coletiva, mantendo as relações profissionais ativas. Os resultados também indicaram que os entrevistados da amostra são mais propensos a se envolverem ativa e semipassivamente no LinkedIn.
Abstract Purpose: The primary purpose of this article is to understand the behavior of young people at the beginning of their careers, in the context of Covid-19, considering the intention to continue using the services of the professional social network LinkedIn. Originality/value: This article expands the scope of studying a user group from acceptance in the continuance intention (continuous use) of LinkedIn, analyzing a series of positive and negative factors regarding the processes. Design/methodology/approach: We carried out a descriptive quantitative approach. The authors collected data from a survey (n = 386). To test the hypotheses, the researchers applied the technique of structural equation modeling (SEM), with estimation and partial least squares, and a moderating effect with the technological optimism construct for the predictive variables of the intention to continue using. Findings: The results indicated that all direct and indirect relationships were accepted, except a moderation (H7d) which was rejected, considering that users less optimistic about the technology may feel positively overwhelmed regarding the service’s perceived usefulness. LinkedIn’s variety of services can lead users to continue using the platform. The platform’s objective is to facilitate collective self-presentation, keeping profes sional relationships active. The results also indicated that the sample respondents are more likely to be actively and semi-passively involved on LinkedIn.

Local muscle oxygenation during different cuff-pressures intervention: a punctual near-infrared spectroscopy measurement

Abstract Aim: To verify the response of tissue saturation index (TSI) during ischemia-reperfusion (IR) interventions with different cuff-pressures. Methods: Twenty-nine healthy men experienced in resistance training were recruited. Each one has undergone a control condition (no cuff) and one of the three IR interventions: 1) 190 mmHg (CP-190, 22.7 ± 3.0 years; 176.6 ± 3.9 cm; 77.3 ± 9.5 kg; 2) 100 mmHg (CP-100, 22.9 ± 6.3 years; 180.5 ± 4.0 cm; 85.2 ± 14.1 kg) and 3) 20 mmHg (CP-20, 20.3 ± 2.4 years; 171.8 ± 5.2 cm; 72.4 ± 6.0 kg). Cuffs were placed on the proximal region of the thighs. IR interventions consisted of three cycles of 2-min occlusion-reperfusion. TSI was measured using near-infrared spectroscopy (NIRS), positioned on the middle portion of the vastus lateralis of the dominant leg. The oxygenation was measured at the control conditions (no cuff) and during cuff interventions. Results: While TSI values of CP-20 did not change compared to control (p > 0.05), the TSI in CP-190 was lower in the ischemia (p < 0.05), and CP-100 was lower in the second and third ischemia (p < 0.05). However, the TSI value increased during reperfusion but did not return to control levels (p < 0.05). Conclusion: TSI of the CP-190 significantly decreased during ischemia. However, these values increased by about 16% in the reperfusion period. Thus, our results show that the RI intervention may have caused an increase in metabolic demand, as even with the release of blood flow, the TSI values were below those of the other interventions.

Abstract Background Eragrostis plana is the main invasive plant in the Pampa Biome of southern Brazil. This plant is highly competitive, tolerant to abiotic stresses, and very difficult to manage. Plants exposed to abiotic stresses and herbicides develop mechanisms that help alleviate or reduce damage caused by stressors and transmit this ability to the progenies. Objective The study’s objectives were to ascertain whether acclimatization to stress due to drought and sub-lethal doses of glyphosate may decrease plant sensitivity to glyphosate and investigate the possible memory mechanisms involved in this process. Methods A population of E. plana was submitted to drought, glyphosate, or a combination of drought plus glyphosate for two generations. The progenies were analyzed for sensitivity to the herbicide glyphosate and for biochemical, metabolites, and molecular responses. Results When testing sensitivity to glyphosate in the G 2 generations, the DRYxGLY (plants stressed with drought plus glyphosate) was the least sensitive (ED50 443.0 g a.e. ha-1), while CHK (without stresses) was the most sensitive (ED50 278.0 g a.e. ha-1). Evaluating the effects of glyphosate, the CHK population showed a greater accumulation of oxidative damage and shikimic acid. While DRYxGLY had greater antioxidant activity and higher expressions of the EPSPS and ABC-carrier MRP10 genes. Conclusions Recurrent selection with drought stress and sub-lethal rate of glyphosate (DRYxGLY) showed reduced sensitivity to glyphosate in the second generation ( G 2 ). The conjunction of factors, including the upregulation of EPSPS and the ABC MRP10 transporter, antioxidant enzymes seem to decrease the sensitivity of the DRYxGLY population to glyphosate.

ABSTRACT Nitrogen fertilization in corn is a factor of great impact on production costs, due to its importance in crop yield. In this way, poultry litter and nitrogen-fixing bacteria can be alternative management strategies to supply the demand. Therefore, the objective of this work was to evaluate the impacts of the inoculation of the bacterium A. brasilense associated with poultry litter and mineral fertilization on some plant parameters and on the final productivity of corn grains. The experiment was conducted in a randomized block design, with 8 treatments and 8 repetitions, the treatments being constituted as follows: T1 - Control, T2 - A. brasilense, T3 - 100% N-mineral, T4 - 100% N-mineral + A. brasilense, T5 - 50% N-mineral + 50% N-organic, T6 - 50% N-mineral + 50% N-organic + A. brasilense, T7 - 100% N-organic, T8 - 100% N-organic + A. brasilense. For the plant parameters and final productivity of corn grains, the use of the mineral source and mineral and organic association resulted in greater increments when compared to the organic source alone. The inoculation with A. brasilense did not influence the analyzed parameters.
RESUMO A adubação nitrogenada na cultura do milho é um fator de grande impacto nos custos de produção, devido sua importância no rendimento da cultura. Desta forma, cama de aves e bactérias fixadoras de nitrogénio podem ser alternativas de estratégias de gestão para suprir a demanda. Portanto, o objetivo deste trabalho foi avaliar os impactos da inoculação da bactéria A. brasilense associada à cama de aves e a adubação mineral sobre alguns parâmetros de planta e na produtividade final de grãos de milho. O delineamento experimental utilizado foi o de blocos ao acaso, com 8 tratamentos e 8 repetições, sendo os tratamentos assim constituídos: T1 - Testemunha, T2 - A. brasilense, T3 - 100% N-mineral, T4 - 100% N-mineral + A. brasilense, T5 - 50% N-mineral + 50% N-orgânico, T6 - 50% N-mineral + 50% N-orgânico + A. brasilense, T7 - 100% N-orgânico, T8 - 100% N-orgânico + A. brasilense. Para os parâmetros de planta e produtividade final de grãos de milho, a utilização da fonte mineral e associação mineral e orgânica potencializou maiores incrementos quando comparada à fonte somente via orgânica. A inoculação com A. brasilense não influenciou nos parâmetros analisados.

The Amazonian Rubiaceae species Uncaria guianensis (UG) is locally used as anti-inflammatory, antitumor, antidiabetic, anti-ulcers, and others. The phenolic content of its leaves is characterized by the great predominance of the flavonoid kaempferol-3,7-O-(α)-L-dirhamnoside (kaempferitrin). The present study quantitatively evaluates the kaempferitrin content in the leaves and branches of cultivated and wild UG specimens collected in different locations of the Brazilian Amazon rainforest by employing high-performance liquid chromatography coupled to a diode array detector (HPLC-DAD). Besides, the understanding of the polyphenol profile performed by electron spray ionization is deepened by tandem mass spectrometry analysis (ESI-MS/MS), using a previously approached leaf UG extract, and the flavonoid quercetin-3,7-O-(α)-L-dirhamnoside was first isolated from UG. All samples showed quite similar qualitative polyphenol profiles. Kaempferitrin in UG ranged from 1.1 to 1.9 mg 100 mg-1 for dry leaves of adult wild plants, 0.3 to 0.7 mg 100 mg-1 for dry leaves of cultivated young plants and 0.00 to 0.04 mg 100 mg-1 for dry branches of adult wild plants. Besides suggesting the distribution of kaempferitrin in the species, these results reinforce this flavonol as a suitable chemical marker for UG leaves and the products derived from them.

ABSTRACT The present work aimed to develop mathematical fuzzy models to evaluate the effects of different doses of sewage sludge and irrigation with wastewater and potable water. Such models were elaborated from an experiment carried out at the Faculty of Agronomic Sciences, in the Department of Soil and Environmental Resources, from the Sao Paulo State University, in Brazil. The experiment was carried out in a randomized block design, in a 6 × 2 factorial scheme, with 6 doses of sewage sludge (0, 25, 50, 75, 100 and 125 of the recommended dose of N), and in the presence and absence of wastewater. In the development of the fuzzy model, the Mamdani method was used for the defuzzification. As input variables, the doses of sewage sludge and the types of water were used. For the output variables, it was sought to evaluate the biometric and developmental components of the culture. It can be inferred that the model developed presented a good fit when compared to the regression model, and that the use of sewage sludge may prove to be a potential future replacement of mineral nitrogen.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.
