Abstract Hereditary transthyretin amyloidosis with peripheral neuropathy (ATTRv-PN) is an autosomal dominant inherited sensorimotor and autonomic polyneuropathy with over 130 pathogenic variants identified in the TTR gene. Hereditary transthyretin amyloidosis with peripheral neuropathy is a disabling, progressive and life-threatening genetic condition that leads to death in ~ 10 years if untreated. The prospects for ATTRv-PN have changed in the last decades, as it has become a treatable neuropathy. In addition to liver transplantation, initiated in 1990, there are now at least 3 drugs approved in many countries, including Brazil, and many more are being developed. The first Brazilian consensus on ATTRv-PN was held in the city of Fortaleza, Brazil, in June 2017. Given the new advances in the area over the last 5 years, the Peripheral Neuropathy Scientific Department of the Brazilian Academy of Neurology organized a second edition of the consensus. Each panelist was responsible for reviewing the literature and updating a section of the previous paper. Thereafter, the 18 panelists got together virtually after careful review of the draft, discussed each section of the text, and reached a consensus for the final version of the manuscript.

Abstract Given the ecological and socioeconomic importance of the mangroves of the Brazilian Amazon coast, RapidEye satellite images were analyzed to recognize mangrove forest and salt flat changes to different land use through human activities. Results show that mangroves are still very well preserved, with less than 1% of the total converted to other uses, primarily urban areas and roads. These human activities have been the principal causes of use in the mangrove forest, driven by local anthropogenic pressures resulting from human settlements in the transition zone between the mainland and tidal flats. In contrast, aquaculture, the principal driver of the loss of mangroves in other regions of South America and in Asian countries, plays only a secondary role in habitat conversion on the Amazon coast. However, these human activities demand more attention and policies need to be supported by Brazilian legislation.

Abstract Mangroves occur in the tropics and subtropics. This region is constantly covered by clouds and therefore highly challenging to map and monitor. Technological advances in remote sensing have increased the flexibility of performing such analyses. In this study, mapping and change detection were carried out for mangrove areas of the South and Southeast regions of Brazil between 2008 and 2016 using multisensor data and geographical object-based image analysis (GEOBIA). The 823.03 km² mangrove areas in study site in 2008 were reduced to 789.00 km² in 2016, representing a net loss of ~34 km². A change detection analysis of the mangrove areas showed a total gain of 138.21 km², a total loss of 172.24 km² and no change for 650.79 km². The GEOBIA classification accuracy was assessed by performing a statistical analysis of confusion matrix: (2008): global accuracy = 0.92, Kappa index = 0.84 and Tau index = 0.84; and (2016): global accuracy = 0.93, Kappa index = 0.86 and Tau index = 0.86. These results demonstrate the effectiveness of the GEOBIA to map and analyze mangrove dynamics. The results exhibit an excellent accuracy. Furthermore, mangrove areas in the south and southeast Brazil were mapped from the same methodological approach.

This guideline aimed to update the concepts and formulate the standards of conduct and scientific evidence that support them, regarding the diagnosis and treatment of the Cardiomyopathy of Chagas disease, with special emphasis on the rationality base that supported it.nbsp;
Chagas disease in the 21st century maintains an epidemiological pattern of endemicity in 21 Latin American countries. Researchers and managers from endemic and non-endemic countries point to the need to adopt comprehensive public health policies to effectively control the interhuman transmission of T. cruzi infection, and to obtain an optimized level of care for already infected individuals, focusing on diagnostic and therapeutic opportunistic opportunities.
nbsp;
Pathogenic and pathophysiological mechanisms of the Cardiomyopathy of Chagas disease were revisited after in-depth updating and the notion that necrosis and fibrosis are stimulated by tissue parasitic persistence and adverse immune reaction, as fundamental mechanisms, assisted by autonomic and microvascular disorders, was well established. Some of them have recently formed potential targets of therapies.nbsp;
The natural history of the acute and chronic phases was reviewed, with enhancement for oral transmission, indeterminate form and chronic syndromes. Recent meta-analyses of observational studies have estimated the risk of evolution from acute and indeterminate forms and mortality after chronic cardiomyopathy. Therapeutic approaches applicable to individuals with Indeterminate form of Chagas disease were specifically addressed. All methods to detect structural and/or functional alterations with various cardiac imaging techniques were also reviewed, with recommendations for use in various clinical scenarios. Mortality risk stratification based on the Rassi score, with recent studies of its application, was complemented by methods that detect myocardial fibrosis.nbsp;
The current methodology for etiological diagnosis and the consequent implications of trypanonomic treatment deserved a comprehensive and in-depth approach. Also the treatment of patients at risk or with heart failure, arrhythmias and thromboembolic events, based on pharmacological and complementary resources, received special attention. Additional chapters supported the conducts applicable to several special contexts, including t. cruzi/HIV co-infection, risk during surgeries, in pregnant women, in the reactivation of infection after heart transplantation, and others.nbsp; nbsp;nbsp;
Finally, two chapters of great social significance, addressing the structuring of specialized services to care for individuals with the Cardiomyopathy of Chagas disease, and reviewing the concepts of severe heart disease and its medical-labor implications completed this guideline.
A metodologia atual para diagnóstico etiológico e as consequentes implicações do tratamento tripanossomicida mereceram enfoque abrangente e aprofundado. Também o tratamento de pacientes em risco ou com insuficiência cardíaca, arritmias e eventos tromboembólicos, baseado em recursos farmacológicos e complementares, recebeu especial atenção. Capítulos suplementares subsidiaram as condutas aplicáveis a diversos contextos especiais, entre eles o da co-infecção por T. cruzi/HIV, risco durante cirurgias, em grávidas, na reativação da infecção após transplante cardíacos, e outros.nbsp;nbsp;nbsp;
Por fim, dois capítulos de grande significado social, abordando a estruturação de serviços especializados para atendimento aos indivíduos com a CDC, e revisando os conceitos de cardiopatia grave e suas implicações médico-trabalhistas completaram esta diretriz.nbsp;

Abstract Background Endogenous phospholipases A2 (PLA2) play a fundamental role in inflammation, neurodegenerative diseases, apoptosis and cellular senescence. Neurotoxins with PLA2 activity are found in snake venoms from the Elapidae and Viperidae families. The mechanism of action of these neurotoxins have been studied using hippocampal and cerebellar neuronal cultures showing [Ca2+]i increase, mitochondrial depolarization and cell death. Astrocytes are rarely used as a model, despite being modulators at the synapses and responsible for homeostasis and defense in the central nervous system. Preserving the cell division ability, they can be utilized to study the cell proliferation process. In the present work cultured astrocytes and glioblastoma cells were employed to characterize the action of β-micrustoxin (previously named Mlx-9), a PLA2 isolated from Micrurus lemniscatus snake venom. The β-micrustoxin structure was determined and the cell proliferation, cell cycle phases and the regulatory proteins p53, p21 and p27 were investigated. Methods β-micrustoxin was characterized biochemically by a proteomic approach. Astrocytes were obtained by dissociation of pineal glands from Wistar rats; glioblastoma tumor cells were purchased from ATCC and Sigma and cultured in DMEM medium. Cell viability was evaluated by MTT assay; cell proliferation and cell cycle phases were analyzed by flow cytometry; p53, p21 and p27 proteins were studied by western blotting and immunocytochemistry. Results Proteomic analysis revealed fragments on β-micrustoxin that aligned with a PLA2 from Micrurus lemniscatus lemniscatus previously identified as transcript ID DN112835_C3_g9_i1/m.9019. β-micrustoxin impaired the viability of astrocytes and glioblastoma tumor cells. There was a reduction in cell proliferation, an increase in G2/M phase and activation of p53, p21 and p27 proteins in astrocytes. Conclusion These findings indicate that β-micrustoxin from Micrurus lemniscatus venom could inhibit cell proliferation through p53, p21 and p27 activation thus imposing cell cycle arrest at the checkpoint G2/M.

ABSTRACT The fire frequency in the Amazon increased rapidly after the 1990s due to deforestation and forest degradation, and it is expected to increase in response to climate change. We analyzed the fire occurrence and assessed seven fire hazard indices in the municipality of Canaã dos Carajás, in the eastern Amazon, for different land use and land cover (LULC) types. We used data from three weather stations located at different heights to compare the performance of the indices using skill scores and success percentages for each LULC. Overall most hotspots occurred in deforested areas and native forests, which were the main LULC types, while few were observed in rupestrian fields, urban areas, and mining areas. However, forests presented the lowest number of hotspots per unit area, especially inside protected areas, and all hotspots in forest areas were observed after a severe drought in 2015. The performance of the fire indices varied as a function of the LULC class and the weather station considered, which indicates the importance of choosing the most appropriate location of the station according to the purpose of the monitoring. The Keetch-Byram Drought Index showed the best performance for predicting fire occurrence for all LULC classes, and forests and deforested areas individually. Despite its simplicity, the Angstrom index stood out due to its good performance in the prediction of days with more than six hotspots.

Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

ABSTRACT Sweet corn (Zea mays L.) local varieties are important gene sources for breeding programs, mainly in participatory research models. This study aimed to evaluate the agronomic performance of seven sweet corn local varieties (2255A, 2029A, 2276A, 2514A, 3000A, 741B and 319A) carrying the sugary1 gene under homozygotic condition and two controls, at four environments [Anchieta (altitudes: 422 and 717 m), Guaraciaba (altitude: 624 m) and Florianópolis (altitude: 5 m)], in the Santa Catarina state, Brazil. The varieties 3000A and 319A showed a good performance in the different altitudes for ear and grain length and ear yield. At intermediate altitudes (422 and 624 m), the mean husked ear yield (milky grain) of these varieties were close to 12.5 t ha-1 (3000A) and 10.0 t ha-1 (319A). The varieties 2029A, 741B, 2514A, 2276A and 2255A stood out for ear length and diameter, number of ear rows and grain length. The best performance of the sweet corn local varieties was observed in their region of origin, mainly at altitudes ranging from 400 to 650 m. These varieties are adapted to the region and are promising alternatives to be used in breeding programs focused on finding a greater potential variability for selection purposes.

The arrowroot is considered a non-conventional vegetable which has aroused interest in the food industry because of its starch content with differentiated characteristics and high commercial value. However, agronomic management information is scarce, making it difficult to compete with other species of similar purpose, and more studies are needed regarding the crop. The aim of this study was to define adequate agronomic practices in crop aiming the productivity the rhizome, yield the starch of arrowroot. The factors evaluated were the different levels of irrigation, the planting and the plant densities. The randomized block design was used in the field trial while the totally randomized block design was used in the laboratory tests. Through the treatments, crop yield ranged from 0.68 (seedlings; without irrigation; 31250 plants ha-1) to 60.09 t ha-1 (seedlings; irrigated; 31250 plants ha-1), while starch production ranged from 50 to 6350 kg ha-1. The highest yield of rhizomes was obtained in plants produced by seedlings along with the highest density of plants, and the highest production of arrowroot starch was extracted from rhizomes from plants produced by seedlings without irrigation and a density of 50000 plants ha-1.

ABSTRACT Identification of common bean genotypes resistant to the root-knot nematodes may be useful for bean breeding programs. The objective of this study was to evaluate the reaction of bean genotypes to M. incognita race 1 and M. javanica. Two independent trials to assess resistance to both root-knot nematodes were carried out with fifty-eight common bean genotypes and six snap bean genotypes. The experiments were arranged in a randomized block design, with three replications and two plants per plot. A total of 10,000 nematode eggs were inoculated per plant 15 days after germination. At forty-five days after inoculation, the root system of each plant was harvested and the nematode eggs were extracted. The number of eggs per gram of root was counted and the Reproduction Factor and the Reduction of the Reproduction Factor were calculated. The performance of the genotypes differed between the trials, indicating different resistance reactions to the nematode species evaluated. The genotypes VP-25 and BRS Valente were resistant to M. incognita race 1. The genotypes Aporé, Ouro Vermelho, Radiante, and CNFP 10793 showed good resistance to both M. incognita race 1 and M. javanica, with potential as source of resistance in breeding programs. There was a significant correlation between root mass and number of eggs. The correlation between nematode reproduction and shoot mass was null.

ABSTRACT High-resolution satellite images, digital elevation models, bathymetric and sedimentological surveys coupled with statistical analysis were used to understand the physical environment and discuss their influence on water quality of the five upland lakes of Serra Sul dos Carajás, southeast Amazonia. The lakes have mid-altitude ranges (elevation), very small (catchment) and shallow to very shallow (central basins). Based on the length, area and volume, Violão and TI (Três Irmãs)-3 lakes may present large vertical movements of the water due to wind action and weakly stratified waters. Trophic conditions based on depth and shore development (Ld) parameters must be used with caution, since Amendoim Lake is relatively deep, but it is oligotrophic to ultra-oligotrophic. Ld values suggest that the lakes are circular to subcircular and are likely formed by solution process, as also suggested by volume development. TI-2 Lake is only presenting convex central basin and has highest dynamic ratio (DR), thus it may have high sedimentation and erosion rates. Based on the relationship between studied parameters, morphometric index and DR likely influence temperature and dissolved oxygen of waters of TI-2 Lake due to its depth profile and wind-induced surface mixing. Nevertheless, water quality parameters are controlled by catchment characteristics of the lakes.
