Research to identify plant bioactive compounds led to the evolution of extraction methods. This study optimized ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE) to extract the flavonoids of Croton grewioides Baill. species contributing to characterize the polar extract profile and understand the antioxidant potential of these plant constituents. The use of experimental design and statistical treatments enabled the determination of the ideal conditions for each technique. The extracts of five accessions of C. grewioides were obtained under the optimized extraction conditions and were analyzed using ultra high-resolution mass spectrometry (FT-Orbitrap MS) operating at negative ionization mode for flavonoids detection. The accurate experimental mass obtained to the main compounds was used to attribute the molecular formula. Chemical structures of the main compounds detected were proposed using structure data bases. Chemometric analysis were performed with two FT-Orbitrap MS spectra samples using the identified metabolites and, the antioxidant activity data, showing that for this species the MAE was most effective in extracting the antioxidant compounds. Abstract Some grasslands in ecotones with forests tend to be encroached by woody species, because of changes in climate and land use. Such structural changes in vegetation can be facilitated when the grassland community presents an autochthonous arboreal component, like Butia palms. We aim to identify the responses of taxonomic and functional diversity on grassland community with the occurrence of arborescent/arboreal species (autochthonous and encroaching) to palm density and grazing intensity. The study was conducted in a Butia odorata palm grove under cattle management, in Southern Brazil. To assess the taxonomic and functional composition we performed ordinations analysis with the vegetational data and using path analysis we assessed the causal relationships between variables of interest. Density of Butia odorata and woody plants were strongly positive related, suggesting a facilitation process in the establishment of arborescent plants on the grassland matrix. The abundance of less palatable plants and grazing pressure were inversely related, indicating a selection process induced by higher grazing intensity. We suggest that the grazing intensity management must be based on the autochthonous tree density, applying higher grazing intensity in areas with higher density of encroaching plants, in addition to maintaining other regions conducive to Butia palm regeneration through fallows. use component palms arborescentarboreal Brazil interest related matrix fallows

Water-soluble organic carbon (WSOC) makes up a large mass fraction of the organic carbon in the aerosol and can influence important cloud processes in the atmosphere. The capacity of WSOC to form metallic complexes with transition metals is well known; however, its influence on the aerosol of urban areas in the Amazon region is not very well known. In this study, we investigated the relationship between WSOC, black carbon (BC), levoglucosan (LEV) and transition metals (Fe, Cu and Mn) present in the PM2.5 (particles with a diameter smaller than 2.5 µm) of an urban environment during the dry season in the central Amazon. Oxalic acid (C2) was used to identify the influence of transition metals on WSOC. The mean mass concentration value of the PM2.5 was 14.72 μg m-3 (2.11-31.68 μg m-3). The WSOC made up 58.34% of the PM2.5 mass, followed by BC (20.28%), and LEV (2.62%). The WSOC showed significant correlation with the transition metals analyzed (> 0.56), especially Mn with C2 (linear coefficient (R2) = 0.74). A multiple linear regression with WSOC, BC and LEV showed a strong linear correlation between them (R2 = 0.86), indicating the influence of biomass burning and vehicle traffic on the organic aerosol.

The ofloxacin (OFLOX) is a second-generation synthetic antibiotic that can be classified as a multifunctional drug, but is a poorly soluble drug, which influences its efficiency. The inclusion complexes of OFLOX with β-cyclodextrin (βCD) or hydroxypropyl-β-cyclodextrin (HPβCD) can improve the chemical characteristics of the drug; however, studies showing the biological activity of these inclusion complexes are still scarce. The present work aimed to investigate the multifunctionality of the OFLOX and their inclusion complexes. Thus, the 1:1 βCD/OFLOX or HPβCD/OFLOX were prepared and analyzed by Fourier transform infrared spectroscopy (FTIR), thermogravimetric and differential thermal analysis (TGA and DTA), 1H nuclear magnetic resonance (NMR) and isothermal titration calorimetry (ITC). The antitumor and antibacterial effects were assessed. The results confirm the formation of the inclusion complexes, which had lower minimum inhibitory concentration (MIC) values, higher cytotoxicity and promoted the apoptosis. The present study showed, for the first time, the promising effects of the inclusion complexes as antitumor, improving the biological activities of the uncomplexed ofloxacin.

Abstract Rabies is a fatal zoonotic infection of the central nervous system of mammals and has been known to humans for millennia. The etiological agent, is a neurotropic RNA virus in the order Mononegavirales, family Rhabdoviridae, genus Lyssavirus. There are currently accepted to be two cycles for rabies transmission: the urban cycle and the sylvatic cycle. The fact that both cycles originated from a common RABV or lyssavirus ancestor and the adaptive divergence that occurred since then as this ancestor virus adapted to a wide range of fitness landscapes represented by reservoir species in the orders Carnivora and Chiroptera led to the emergence of the diverse RABV lineages currently found in the sylvatic and urban cycles. Here we study full genome phylogenies and the time to the most recent common ancestor (TMRCA) of the RABVs in the sylvatic and urban cycles. Results show that there were differences between the nucleotide substitution rates per site per year for the same RABV genes maintained independently in the urban and sylvatic cycles. The results identify the most suitable gene for phylogenetic analysis, heterotachy among RABV genes and the TMRCA for the two cycles.

Hydrocarbons present in saturated fractions of crude oils can be assessed by atmospheric pressure chemical ionization (APCI) using small hydrocarbons as ionizing reagents in a Fourier transform ion cyclotron resonance mass spectrometer (FT-ICR MS). In this work, five paraffin standards of different average molar mass distributions (Mw) were easily ionized by APCI(+)FT-ICR MS using isooctane as the reagent gas. Data of Mw, carbon number and double bond equivalent (DBE) distributions corresponding to linear and cyclic hydrocarbons (HCs) were compared to results obtained from analysis of high temperature gas chromatography with a flame ionization detector (HTGC-FID), comprehensive two-dimensional gas chromatography coupled to mass spectrometry (GC×GC-MS) and 1H and 13C nuclear magnetic resonance (NMR) spectroscopy. APCI(+)FT-ICR MS data showed good agreement with those of analytical techniques. Furthermore, the ability of APCI(+) to assess n-paraffin, even in blends with polyaromatic hydrocarbon molecules such as coronene (Mw = 301 Da) at concentrations from 2.5 to 25 µg mL-1, was demonstrated. The typical MS paraffin profile (containing repeating mass units of 14 Da) was clearly confirmed, being totally suppressed when a concentration of 25 µg mL-1 of coronene was used. This phenomenon was also evidenced in one of two saturated fractions produced using saturates, aromatics and polar (SAP) compound fractionation methodology.

Abstract The essential oil of Chenopodium ambrosioides L., Amaranthaceae, was obtained by steam distillation in a Clevenger apparatus and characterization was performed using chromatographic and spectroscopic assays (GC-FID, GC/MS, 1H NMR). Two major compounds were identified: p-cymene (42.32%) and ascaridole (49.77%). The ethanolic extract and hydrolate were fractionated by liquid–liquid partitioning and the compounds were characterized by GC/MS. The essential oil, ethanol extract and fractions by partitioning with dicloromethane, ethyl acetate and butanol were tested in tumor cell lines (K562, NALM6, B15, and RAJI). Significant cytotoxic activity was found for essential oil (IC50 = 1.0 µg/ml) for RAJI cells and fraction dicloromethane (IC50 = 34.0 µg/ml) and ethanol extract (IC50 = 47.0 µg/ml) for K562 cells. The activity of the essential oil of C. ambrosioides is probably related to the large amount of ascaridol, since the other major compound, p-cymene, is recognized as a potent anti-inflammatory and has low cytotoxic activity.

Predição da composição do clínquer industrial utilizando minimização da energia livre de Gibbs

A existência de processos químicos industriais, com diferentes fases em meio reacional, e a necessidade de conhecer a composição do sistema de interesse é um desafio comumente enfrentado pelos profissionais da indústria. Desta forma, a consideração do equilíbrio químico é uma metodologia comumente empregada por esses profissionais. Uma das principais etapas do processo de obtenção do cimento é a etapa de clinquerização, que ocorre dentro do forno rotativo. Neste estudo, é proposto o uso da técnica de minimização da energia livre de Gibbs para descrever o processo de produção de clínquer. O combustível considerado para o forno é o carvão mineral. Como resultado do estudo, objetiva-se a obtenção de uma rotina computacional que simule a condição de equilíbrio termodinâmico do sistema. Para isto, um problema de otimização não linear restrito é resolvido.
The existence of industrial chemical processes, with different phases in the reaction system, and the need to know the composition of these system is a challenge commonly faced by industry professionals. Thus, consideration of the chemical equilibrium is usually adopted. One of the main steps of the cement production is the clinker production in the rotary kiln. In this study, it has been proposed the use of the minimization of Gibbs free energy to describe the clinker production process. The fuel considered is the coal. The major objective of this study is to obtain a computational routine that simulates the condition of thermodynamic equilibrium of the system, composed by different phases. For this, a nonlinear constrained optimization problem is solved.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The aim of this study was to determine if bone marrow mononuclear cell (BMMC) transplantation is safe for moderate to severe idiopathic dilated cardiomyopathy (IDC). Clinical trials have shown that this procedure is safe and effective for ischemic patients, but little information is available regarding non-ischemic patients. Twenty-four patients with IDC, optimized therapy, age 46 ± 11.6 years, 17 males, NYHA classes II-IV, and left ventricular ejection fraction <35% were enrolled in the study. Clinical evaluation at baseline and 6 months after stem cell therapy to assess heart function included echocardiogram, magnetic resonance imaging, cardiopulmonary test, Minnesota Quality of Life Questionnaire, and NYHA classification. After cell transplantation 1 patient showed a transient increase in enzyme levels and 2 patients presented arrhythmias that were reversed within 72 h. Four patients died during follow-up, between 6 and 12 weeks after therapy. Clinical evaluation showed improvement in most patients as reflected by statistically significant decreases in Minnesota Quality of Life Questionnaire (63 ± 17.9 baseline vs 28.8 ± 16.75 at 6 months) and in class III-IV NYHA patients (18/24 baseline vs 2/20 at 6 months). Cardiopulmonary exercise tests demonstrated increased peak oxygen consumption (12.2 ± 2.4 at baseline vs 15.8 ± 7.1 mL·kg-1·min-1 at 6 months) and walked distance (377.2 ± 85.4 vs 444.1 ± 77.9 m at 6 months) in the 6-min walk test, which was not accompanied by increased left ventricular ejection fraction. Our findings indicate that BMMC therapy in IDC patients with severe ventricular dysfunction is feasible and that larger, randomized and placebo-controlled trials are warranted.

Os sinais clínicos das infecções por Ehrlichia canis e Anaplasma platys são similares, e o diagnóstico desses patógenos feito por esfregaços sanguíneos corados é difícil devido à sensibilidade e especificidade. Por outro lado, os diagnósticos moleculares são altamente sensíveis e específicos, e nested-PCRs têm sido otimizadas para o diagnóstico preciso desses patógenos em cães. Em um Hospital Veterinário Escola, amostras de sangue total com EDTA foram obtidas de 100 cães, e esfregaços foram feitos das amostras de sangue para busca dos parasitos intracelulares. Para cada amostra, DNA foi extraído e submetido à nPCR para detecção de E. canis e A. platys. Os resultados dos esfregaços sanguíneos mostraram que 9% dos animais foram positivos para E. canis e 21% para A. platys. Com relação à nPCR, 57 e 55% dos cães foram positivos para E. canis e A. platys, respectivamente. Quando comparados com a nPCR, os esfregaços sanguíneos corados revelaram resultados falso-negativos para E. canis e A. platys. Os resultados indicam que a nPCR é altamente sensível e específica para detecção de ambos os patógenos, e os diagnósticos moleculares podem ser mais úteis nos Hospitais Veterinários.
The clinical signs of Ehrlichia canis and Anaplasma platys infection are similar, and the diagnosis of these pathogens made by stained blood smears is poor due sensibility and specificity. On the other hand, the molecular diagnosis is highly sensitive and specific and nested-PCR have been optimized for accurate diagnosis these pathogens in dogs. At the veterinary teaching hospital, whole-blood samples with EDTA were obtained from 100 dogs and smears were made from blood samples for evaluation for intracellular parasites. For each sample, DNA was extracted and submitted to nPCR analysis for detection of E. canis and A. platys. The results of stained blood smears showed 9% of the animals were positive for E. canis and 21% for A. platys. Regarding of nPCR analysis, 57 and 55% of dogs were positive for E. canis and A. platys respectively. As compared to a nested PCR, the stained blood smears revealed false-negative results for both E. canis and A. platys. The results indicate that the nPCR is highly sensitive and specific for detection of both pathogens and the molecular diagnosis could be more useful at veterinary hospital.

This work focuses on the adsorption of Bromelain in expanded bed conditions, such as the adsorption kinetics parameters. The adsorption kinetics parameters showed that after 40 minutes equilibrium was achieved and maximum adsorption capacity was 6.11 U per resin mL. However, the maximum adsorption capacity was only determined by measuring the adsorption isotherm. Only by the Langmuir model the maximum adsorption capacity, Qm, and dissociation constant, kd, values could be estimated as 9.18 U/mL and 0.591, respectively, at 25 ºC and 0.1 mol/L phosphate buffer pH 7.5. A column made of glass with an inner diameter of 1 cm was used for the expanded bed adsorption (EBA). The residence time was reduced 10 fold by increasing the expansion degree 2.5 times; nonetheless, the plate number (N) value was reduced only 2 fold. After adsorption, the bromelain was eluted in packed bed mode, with a downward flow. The purification factor was about 13 fold and the total protein was reduced 4 fold. EBA showed to be feasible for purification of bromelain.

A hybrid neural model for the production of sorbitol and gluconic acid using immobilized Zymomonas mobilis cells

Only ten years were enough for hybrid neural network-first principle models (HNM) reach a status of a standard industrial tool. This modeling strategy is employed here to represent the production of sorbitol and gluconic acid from glucose and fructose, using permeabilized and immobilized Zymomonas mobilis cells. Mass component balances are derived for the substrate concentrations. A multilayered neural network is used to represent the reaction rate. Experimental results were used to develop and validate the model. The HNM allows the elucidation of the phenomena involved in the process. It is observed from the results that the resistance for mass transfer from the liquid to the particles is increased at higher substrate concentrations and that the reaction rate depends on the concentrations of substrate and product in the particles. Additionally, it may be stated that the flexibility of the HNM allows the development of a model that would otherwise be difficult, if based solely on phenomenological principles.

Além de influenciar o crescimento corpóreo, o hormônio do crescimento, ou somatotrófico, desempenha importante papel no metabolismo, composição corporal, perfil lipídico, estado cardiovascular e longevidade. Seu controle é multi-regulado por hormônios, metabólitos e peptídeos hipotalâmicos. Dados sobre a Deficiência Isolada de GH (DIGH) obtidos a partir da descrição da mutação IVS1+1G®A no gene do receptor do hormônio liberador do GH (GHRH-R) em indivíduos da cidade de Itabaianinha, SE, são revisados. São abordadas novas perspectivas sobre o modelo de resistência ao GHRH, a importância do GHRH no controle da secreção de GH, a freqüência das mutações do gene do GHRH-R, a relevância diagnóstica do IGF-I e os achados metabólicos, cardiovasculares e de qualidade de vida nestes indivíduos.
In addition to stimulating body growth, growth or somatotrophic hormone plays an important role in metabolism, body composition, lipid profile, cardiovascular status and longevity. Its control is multiregulated by hormones, metabolites and hypothalamic peptides. Obtained data of the isolated growth hormone deficiency (IGHD) after the description of the IVS1+1G®A GHRH receptor gene mutation in individuals of Itabaianinha County are reviewed. New perspectives about the growth hormone resistance model, the importance of GHRH in the control of GH secretion, the frequency of GHRH-R gene mutations, the diagnostic relevance of IGF-I and the metabolic, cardiovascular and quality of life findings are approached.

We investigated kidney and lung alterations caused by intercellular adhesion molecule type 1 (ICAM-1) blockade after ischemia and reperfusion of hind limb skeletal muscles. Rats were submitted to ligature of the infrarenal aorta for 6 h. The animals were randomized into three groups of 6 rats each: group I, sacrificed after ischemia; group II, reperfusion for 24 h, and group III, reperfusion for 24 h after receiving monoclonal anti-ICAM-1 antibodies. At the end of the experiment, blood samples were collected for creatinine, lactate dehydrogenase, creatine phosphokinase, potassium, pH and leukocyte counts. Samples were taken from the muscles of the hind limbs and from the kidneys and lungs for histological analysis and measurement of the neutrophil infiltrate by myeloperoxidase staining. The groups did not differ significantly with regard to the laboratory tests. There were no major histological alterations in the kidneys. An intense neutrophil infiltrate in the lungs, similar in all groups, was detected. Myeloperoxidase determination showed that after reperfusion there was significantly less retention of polymorphonuclear neutrophils in the muscles (352 ± 70 vs 1451 ± 235 × 10² neutrophils/mg; P<0.01) and in the kidneys (526 ± 89 vs 852 ± 73 × 10² neutrophils/mg; P<0.01) of the animals that received anti-ICAM-1 before perfusion compared to the group that did not. The use of anti-ICAM-1 antibodies in this experimental model minimized neutrophil influx, thus reducing the inflammatory process, in the muscles and kidneys after ischemia and reperfusion of the hind limbs.
