NMR-Based Metabolomic Screening for Metabolites Associated with Resistance to Meloidogyne javanica in Annona muricata Roots

The resistance of Annona muricata roots to the nematode Meloidogyne javanica was investigated by nuclear magnetic resonance (NMR)-based metabolomics in combination with principal component analysis (PCA). Metabolic changes in roots exposed and not exposed to the nematode were evaluated and compared. In addition, the presence of nematicidal compounds in the root extracts was investigated through in vitro assay against Meloidogyne javanica second-stage juveniles. Plants exposed to nematodes showed significant changes in their metabolism after 24 h. Several resistance-related metabolites, including dopamine, xanthine and aromatic compounds, could be identified in the roots with the joint analysis of 1D/2D NMR and the loadings of PC3 (17.8%). A.muricata root chloroform extract, containing mainly acetogenins, has shown nematostatic activity against M. javanica, suggesting that a pre-formed defense mechanism can support the reported resistance. For the first time, metabolomic studies allowed to identify induced and pre-formed defense mechanisms and their related metabolites in Annona muricata.

RESUMO Brycon nattereri é um peixe Neotropical ameaçado de extinção reportado para trechos conservados dos rios Paraná, Tocantins e São Francisco. Populações desta espécie têm sido muito raras em algumas sub-bacias do rio Paraná. Este estudo analisou a diversidade genética e a estrutura populacional de B. nattereri em uma área de ocorrência restrita recentemente identificada na bacia do alto rio Paraná. Sete locos microssatélites e 497 pb da região mitocondrial D-Loop foram examinados para 92 indivíduos de quatro pontos ao longo da área de ocorrência. Ambos os marcadores moleculares indicaram uma única população distribuída em um trecho de aproximadamente 80 km do rio. Embora alguns dados tenham sugerido um antigo gargalo genético, os atuais níveis de diversidade genética (H E = 0,574, h = 0,616) foram similares aos de outras espécies do gênero Brycon. Estes resultados sugerem que a população de B. nattereri tem mantido níveis satisfatórios de diversidade genética, apesar da pequena área de ocorrência. Estes dados destacaram uma importante área de conservação e ações podem melhorar a qualidade do ambiente, especialmente para a vida aquática e mata ciliar, se a área for eficientemente manejada e conservada.
ABSTRACT Brycon nattereri is an endangered Neotropical fish reported along conserved stretches of the upper Paraná, Tocantins and São Francisco rivers. Populations of this species have been very rare in some Paraná River sub basins. This study analyzes the genetic diversity and population structure of B. nattereri in a restricted area of occurrence recently identified in upper Paraná River basin. Seven microsatellite loci and 497 bp of D-Loop mitochondrial region were examined in 92 individuals from four points along the area of occurrence. Both molecular markers indicated a single population distributed along a stretch of the river approximately 80 km long. Although some of the data suggest an ancient bottleneck, current levels of genetic diversity (H E = 0.574 and h = 0.616) were similar to those of other species of the genus Brycon. The results suggest that the population of B. nattereri has been able to maintain satisfactory levels of genetic diversity, in spite of the small area of occurrence. These data have highlighted an important conservation area and action may prove essential to improve the quality of the environment, and especially the water and riparian plant life, if the area is to be managed and conserved efficiently.

Abstract Asymmetric alumina hollow fibres produced by the phase inversion/sintering method present advantages in that high area/volume ratios and low mass transfer resistances are achieved due to the geometric configuration and the pore size distribution, respectively. Here we characterise hollow fibres that were prepared with different internal coagulants and at different sintering temperatures. Additionally, a palladium membrane was deposited on these different hollow fibres and hydrogen permeabilities through them were compared. More fingers were obtained when a mixture of solvent with water was used as internal coagulant, instead of pure water. At the same sintering temperature, nitrogen permeance through the fibre was increased 5-fold when a mixture of solvent and water was used as internal coagulant instead of pure solvent, and the water flux was increased 7-fold. The decrease in the sintering temperature increased the water permeance through the fibre from 21.4 to 63.9 L h-1 m-2 kPa-1, but decreased its mechanical strength from 74 to 41 MPa. The hydrogen permeance at 450 °C was increased from 5.54x10-5 to 3.06 x10-3 mol m-2 s-1 kPa-1 when using a more permeable hollow fibre as substrate. These results elucidate better conditions to fabricate hollow fibres that present low mass transfer resistances.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Objective: To present the most relevant findings regarding the Brazilian Medical Association guidelines for the diagnosis and differential diagnosis of panic disorder. Methods: We used the methodology proposed by the Brazilian Medical Association for the Diretrizes Project. The MEDLINE (PubMed), Scopus, Web of Science, and LILACS online databases were queried for articles published from 1980 to 2012. Searchable questions were structured using the PICO format (acronym for “patient” [or population], “intervention” [or exposure], “comparison” [or control], and “outcome”). Results: We present data on clinical manifestations and implications of panic disorder and its association with depression, drug abuse, dependence and anxiety disorders. In addition, discussions were held on the main psychiatric and clinical differential diagnoses. Conclusions: The guidelines are proposed to serve as a reference for the general practitioner and specialist to assist in and facilitate the diagnosis of panic disorder.

Com vista ao estudo de atração e penetração de Meloidogyne javanica (Treub) Chitwood e Heterodera glycines (Ichinoe) em soja (Glycine max L.), desenvolveu-se uma técnica empregando-se segmento de raiz com 2cm de comprimento. Nos segmentos de raiz de soja infectados, observou-se que a penetração de juvenis de segundo estádio (J2) de M. javanica ocorre pela coifa seguida de migração entre os feixes vasculares do cilindro central. Juvenis de H. glycines penetraram, aproximadamente, 15mm da coifa. A região seccionada da raiz de soja atraiu três vezes mais J2 de M. javanica do que a região da coifa, mas esta não foi tão atrativa para J2 de H. glycines. A obstrução conjunta da coifa e do local seccionado reduziu (83%) a penetração de J2, tanto de M. javanica quanto de H. glycines. Quando apenas um desses locais foi obstruído, a outra extremidade livre compensou o processo atrativo. Portanto, as substâncias atrativas são liberadas por essas extremidades. A penetração de J2 de M. javanica foi maior no segmento de raiz quando comparada com a plântula intacta de soja. Entretanto, os J2 de H. glycines penetraram menos em segmentos de raiz e em plântulas sem folhas, quando comparados com plântulas intactas e com as seccionadas no colo. Portanto, na cultivar de soja "Embrapa 20", a atração e os locais de penetração de J2 de H. glycines e M. javanica são diferenciados. Esta técnica poderá ser útil nos estudos de atração e penetração de outros nematoides endoparasitas.
To study the attraction and penetration of Meloidogyne javanica (Treub) Chitwood and Heterodera glycines (Ichinoe) in soybean (Glycine max L.), a technique using 2-cm long root segments was developed. In infected soybean root segments penetration of second stage juveniles (J2) of M. javanica occured through the root cap following migration between the vascular bundles of the central cylinder. Juveniles of H. glycines penetrated about 15mm from the root cap. The cut region of the soybean root attracted three times more J2 of M. javanica when compared to the root cap, but was not as attractive to H. glycines J2. Sealing both root cap and cut region reduced (83%) the J2 penetration of M. javanica and H. glycines. When only one of these penetration sites was sealed the open site compensated attractiveness. These results show that the attractive substances are released by these two penetration sites. The M. javanica J2 penetration was higher in root segments when compared to whole soybean plants. However, H. glycines J2 penetrated less in root segments and in roots of seedlings without leaves when compared to roots of whole soybean plants and roots of plants without the aerial part. In short, attraction and infection sites of H. glycines and M. javanica J2s in soybean cultivar "Embrapa 20" are different and this technique may be useful in studies of attraction and penetration of other endoparasitic nematodes.

In this work, a controller for regulating the transients in water distribution networks is established. The control technique is the H<FONT FACE=Symbol>¥</FONT> Control. The developed controller is applied to a water distribution network and the results of this application demonstrate that the technique allowed the establishment of a robust controller, capable of attenuating the disturbances in a suitable way, being effective in controlling the oscillations of the state variables in question.

Dois modelos experimentais foram usados para estudar o efeito de uma carga hiperosmolar nos fluxos bidirecionais de sódio e cloro no intestino de animais em restrição energética. No modelo I, 1 mL de NaCl 900 mOsm foi introduzido no intestino delgado e deixado de 5 a 70 minutos, a fim de determinar o tempo para ocorrer a maior secreção de sódio, o que se observou aos 10 minutos. No modelo II, os fluxos birecionais de sódio e cloro foram determinados em ratos em restrição energética após carga hipertônica, aos 10 minutos. Foram estudados efeitos de: inibidor metabólico (2-4 dinitrofenol) e farmacológicos. Sugere-se que na má nutrição ocorre impedimento da reabsorção intestinal, favorecendo aumento resultante de secreção.
Two experimental models were tried in young malnourished rats in order to study effect of an hyperosmolar challenge in the small intestine on the bi-directional fluxes of sodium. Weanling rats were fed with energy restricted diets. In model I 1 mL of NaCl 900 mOsm/kg was introduced in the small intestine of the rats and left from 5 up to 70 min, in order to determine the moment of higher net Na+ secretion, which occurred at 10 min. In model II, the bi-directional fluxes of Na+ and Cl- were studied using NaCl or mannitol 900 mOsm/kg under the effect of mecholil, atropine or 2-4 dinitrophenol, for 10 min. Mecholil decreased the Na+ absorption enhancing the net secretion. Control rats were used as reference. In the restricted diets animals occurred an increase of the net secretion stimulated by NaCl 900 mOsm/kg, and this effect was enhanced by mecholil. It is suggested that in malnutrition there is an impairment in Na- intestinal absorption.
