Abstract Transposable elements are abundant and dynamic part of the genome, influencing organisms in different ways through their presence or mobilization, or by acting directly on pre- and post-transcriptional regulatory regions. We compared and evaluated the presence, structure, and copy number of three hAT superfamily transposons (hobo, BuT2, and mar) in five strains of Drosophila willistoni species. These D. willistoni strains are of different geographical origins, sampled across the north-south occurrence of this species. We used sequenced clones of the hAT elements in fluorescence in-situ hybridizations in the polytene chromosomes of three strains of D. willistoni. We also analyzed the structural characteristics and number of copies of these hAT elements in the 10 currently available sequenced genomes of the willistoni group. We found that hobo, BuT2, and mar were widely distributed in D. willistoni polytene chromosomes and sequenced genomes of the willistoni group, except for mar, which is restricted to the subgroup willistoni. Furthermore, the elements hobo, BuT2, and mar have different evolutionary histories. The transposon differences among D. willistoni strains, such as variation in the number, structure, and chromosomal distribution of hAT transposons, could reflect the genomic and chromosomal plasticity of D. willistoni species in adapting to highly variable environments.

Abstract Hypermutable strains of Drosophila simulans have been studied for 20 years. Several mutants were isolated and characterized, some of which had phenotypes associated with alteration in development; for example, showing ectopic legs with eyes being expressed in place of antennae. The causal agent of this hypermutability is a non-autonomous hobo-related sequence (hoboVA). Around 100 mobilizable copies of this element are present in the D. simulans genome, and these are likely mobilized by the autonomous and canonical hobo element. We have shown that hoboVA has transcription factor binding sites for the developmental genes, hunchback and even-skipped, and that this transposon is expressed in embryos, following the patterns of these genes. We suggest that hobo and hobo-related elements can be material for the emergence of new regulatory networks.

Drosophila willistoni (Insecta, Diptera) is considered a paradigm for evolutionary studies. Their chromosomes are characterized by multiple paracentric inversions that make it hard to identify and describe chromosomal poly-morphisms. In the present report we attempted to systematize the description of all the 50 inversions found in the last three decades, since we have been studying the chromosomes of several individuals of 30 different populations, including the one used in the genome sequencing project (Gd-H4-1). We present the photographic register of 11 arrangements in the left arm of the X chromosome (XL), eight in the right arm (XR), 10 in the left arm of chromosome II (IIL), eight in its right arm (IIR) and 13 in chromosome III. This information also includes their breakpoints on the reference photomap. A clear geographic difference was detected in XL and XR, with different fixed arrangements depending on the origin of the population studied. Through the comparison of all X arrangements it was possible to infer the putative ancestral arrangements, i.e., those related to all the remaining arrangements through the small number of inversions that occurred in the past, which we will call XL-A and XR-A. In the autosomes (IIL/IIR and III), fixed inversions were detected, but most are segregating in different frequencies along the geographical distribution of the D. willistoni populations.

O grupo de espécies Drosophila peruensis foi proposto recentemente e inclui quatro táxons: D. atalaia Vilela & Sene, 1982, D. boraceia Vilela & Val, 2004, D. pauliceia Ratcov & Vilela, 2007, D. peruensis Wheeler, 1959. Todas possuem a maioria das cerdas do mesonoto emergindo de manchas escuras, veias transversais esfumaçadas (com exceção de D. atalaia) e hipândrio com formato quadrado e fortemente fusionado aos gonópodes. Neste artigo são descritas duas novas espécies, Drosophila itacorubi sp. nov. e D. paraitacorubi sp. nov., pertencentes a este grupo de espécies. A genitália masculina destas espécies são ilustradas. Uma chave de identificação para as espécies do grupo D. peruensis é fornecida.

The Drosophila peruensis species group was recently proposed and includes four taxa: D. atalaia Vilela & Sene, 1982, D. boraceia Vilela & Val, 2004, D. pauliceia Ratcov & Vilela, 2007, and D. peruensis Wheeler, 1959. All these species have most of setae or setulae of mesonotum arinsing from dark spots, wings with crossveins darker (except in D. atalaia) and hypandrium squared-shaped mostly fused to gonopods. Here, we describe two new species, Drosophila itacorubi sp. nov. and Drosophila paraitacorubi sp. nov., belonging to this species group. The male genitalia of these species are figured. An identification key to the D. peruensis species group is provided.

Drosophila willistoni (Sturtevant, 1916) é uma espécie do grupo willistoni de Drosophila que apresenta ampla distribuição geográfica desde o sul dos Estados Unidos (Flórida) e México até o norte da Argentina. Esta espécie tem sido alvo de muitos estudos evolutivos dentro do grupo devido à sua considerável capacidade de explorar de forma bem sucedida diversos tipos de ambientes e também por sua grande variabilidade genética expressa através de diferentes marcadores. A linhagem 17A2 de D. willistoni foi coletada em 1991 no Estado do Rio Grande do Sul, Brasil (30°05'S, 51°39'W), e vem sendo mantida desde então no laboratório de Drosophila da Universidade Federal do Rio Grande do Sul. Diferentemente das demais linhagens de D. willistoni mantidas no laboratório, a linhagem 17A2 já produziu espontaneamente machos do tipo white (olhos brancos) e do tipo sepia (olhos marrons) em estoques mantidos a 17ºC. A fim de avaliar se esta linhagem é potencialmente hipermutável, nós submetemo-la a experimentos de estresse por temperatura. Dezoito isolinhagens foram utilizadas e, após a primeira geração, todos os indivíduos de cada isolinhagem foram mantidos a 29°C. Diferentes alterações foram observadas nas gerações seguintes, similares com mutações já bem caracterizadas em D. melanogaster (white, sépia, blistered e curly). Além destas, uma alteração com aparente fusão nas antenas foi encontrada, mas somente em indivíduos da isolinhagem nº 31. Esta última alteração não foi previamente descrita como uma mutação na espécie D. melanogaster. Nossos resultados indicam que a linhagem 17A2 de D. willistoni é uma candidata a hipermutabilidade, além de apresentar uma variabilidade genética importante. Diferentes fatores podem estar atuando para a formação deste panorama observado, tais como mobilização de elementos transponíveis, seleção por endocruzamento e alteração na função de proteínas de choque térmico (heat-shock proteins).

Drosophila willistoni (Sturtevant, 1916) is a species of the willistoni group of Drosophila having wide distribution from the South of USA (Florida) and Mexico to the North of Argentina. It has been subject of many evolutionary studies within the group, due to its considerable ability to successfully occupy a wide range of environments and also because of its great genetic variability expressed by different markers. The D. willistoni 17A2 strain was collected in 1991 in the state of Rio Grande do Sul, Brazil (30°05'S, 51°39'W), and has been maintained since then at the Drosophila laboratory of UFRGS. Different to the other D. willistoni strains maintained in the laboratory, the 17A2 strain spontaneously produced mutant males white-like (white eyes) and sepia-like (brown eyes) in stocks held at 17°C. In order to discover if this strain is potentially hypermutable, we submitted it to temperature stress tests. Eighteen isofemale strains were used in our tests and, after the first generation, all the individuals produced in each strain were maintained at 29°C. Different phenotype alterations were observed in subsequent generations, similar to mutations already well characterized in D. melanogaster (white, sepia, blistered and curly). In addition, an uncommon phenotype alteration with an apparent fusion of the antennae was observed, but only in the isofemale line nº 31. This last alteration has not been previously described as a mutation in the D. melanogaster species. Our results indicate that the D. willistoni 17A2 strain is a candidate for hypermutability, which presents considerable cryptic genetic variability. Different factors may be operating for the formation of this effect, such as the mobilization of transposable elements, effect of inbreeding and alteration of the heat-shock proteins functions.

Drosophila paulistorum Dobzhansky & Pavan, 1949 era considerada inicialmente ausente em ambientes com influência antrópica, mas a detecção dessa espécie na cidade de Porto Alegre, sul do Brasil, em 1985, sugeriu seu potencial de colonização de novos ambientes e levou a fundação de estudos ecológicos com populações desta espécie. Este estudo acompanhou as variações populacionais de D. paulistorum nesta cidade quase 20 anos depois de seu primeiro registro neste local. Coletas de Drosophilidae foram realizadas em pontos com diferentes graus de urbanização e os resultados apontaram para uma expressiva diminuição dos tamanhos populacionais de D. paulistorum em Porto Alegre. Este declínio pode estar relacionado com o crescimento da cidade, assim como com retrações naturais no tamanho das populações devido a mudanças climáticas, tais como, variações nas temperaturas mínimas e máximas como conseqüência do aquecimento global. A introdução recente de Zaprionus indianus Gupta, 1970 também parece contribuir com esse quadro, uma vez que poderia estar modificando as interações entre as espécies nativas.

Drosophila paulistorum Dobzhansky & Pavan, 1949 had initially been considered absent in anthropogenically disturbed environments, but in 1985 the detection of the species in Porto Alegre city, southern Brazil, suggested its potential to colonize new habitats and laid the foundations for ecologic studies on this species' populations. This study followed the variations in D. paulistorum populations in this town almost 20 years after its first local record. Drosophilid specimens were collected in sites with different urbanization grades and the results point to the expressive decline in D. paulistorum populations in Porto Alegre. This decline may be linked to urban growth and to naturally driven population decline, as imputed to climatic changes like variations in maximum and minimum temperatures as a consequence of a global climate warming. Also, the recent introduction of exotic species Zaprionus indianus Gupta, 1970 seems to play a role in this scenario, changing the interactions between native species.

In an attempt to understand the dynamics of transposable elements (T'S) in the genome of host species, we investigated the distribution, representativeness and conservation of DNA sequences homologous to the Drosophila melanogaster gypsy retrotransposon in 42 drosophilid species. Our results extended the knowledge about the wide distribution of gypsy in the genus Drosophila, including several Neotropical species not previously studied. The gypsy-like sequences showed high divergence compared to the D. melanogaster gypsy element. Furthermore, the conservation of the restriction sites between gypsy sequences from phylogenetically unrelated species pointed to a more complex evolutionary picture, which includes the possibility of the horizontal transfer events already described for this retrotransposon.

Using conventional staining with acetic orcein and C-banding techniques it was investigated constitutive heterochromatin chromosomal polymorphisms and the mitotic and the meiotic behavior of male and female chromosomes of Boophilus microplus (Canestrini, 1887). Some differences were detected in the population of southern Brazil as compared to the data of other authors for populations in other latitudes. The differences being mainly concerned with the distribution of constitutive centromeric heterochromatin and variation in the length of heterochromatic blocks in the pericentromeric regions of some chromosome pairs.

Disgenesia gonadal dependente de temperatura foi encontrada na prole tanto de cruzamentos intra como interlinhagens, envolvendo duas populações de Drosophila willistoni. Uma delas é derivada de um velho estoque de laboratório e a outra, de uma amostra recentemente coletada de uma população natural. Tal fenômeno não havia ainda sido descrito em D. willistoni e nós sugerimos que a disgenesia gonadal encontrada seja causada por elementos transponíveis, como ocorre em muitas outras espécies de Drosophila.

Temperature-dependent gonadal dysgenesis was shown to occur in the progeny of both inter- and intrastrain crosses involving two populations of Drosophila willistoni, one of which was an old laboratory stock, and the other, freshly collected from a natural population. We propose that the phenomenon observed was caused by the mobilization of transposable elements, as occurs in several other Drosophila species.

Populações de Drosophila paulistorum colonizadoras da área urbana de Porto Alegre, no sul do Brasil, foram estudadas com o objetivo de caracterizar o seu polimorfismo cromossômico neste novo ambiente. A despeito de serem geograficamente periféricas e ecologicamente marginais, e do processo de colonização ser um evento recente, as populações apresentaram um grande número de inversões em todos os braços cromossômicos. Diferenças nas freqüências de algumas inversões e na porcentagem de heterozigose foram encontradas quando se compararam as amostras quanto aos aspectos geográfico, microambiental e temporal. Tais diferenças, entretanto, podem ser atribuídas tanto a fatores seletivos como estocásticos.

Drosophila paulistorum populations colonizing the urban area of Porto Alegre, southern Brazil, were studied with the objective of characterizing their chromosomal polymorphism in this new environment. Despite being geographically and ecologically marginal and the fact that the colonization of the urban area seems to be a recent event, the populations showed a large number of inversions on all chromosome arms. Differences regarding inversion frequencies and percentage of heterozygosis were found when we compared the samples with respect to geographical, microenvironmental and temporal aspects. Such differences, however, could be attributed to both selective and stochastic factors
