Abstract The family Lebiasinidae includes a number of miniature and medium-sized fish species that are endemic to the Neotropical region. Pyrrhulina is the second most speciose lebiasinid genus and it is also the one with the most taxonomic uncertainties. In this context, the present study focused on the Pyrrhulina morphospecies found in a number of different drainage basins in South America to test the alternative proposals on the arrangement of the taxonomic units found within what is assumed to be a single nominal species, Pyrrhulina australis, based on a DNA Barcoding approach. The results of the analyses indicate that Pyrrhulina australis is a species complex, with intraspecific (within-group) genetic distances of up to 3.74%, well above the Optimal Threshold of 1.79% defined in the present study. The species delimitation analyses revealed a surprising level of diversification among the morphospecies evaluated, in particular, in the clade that encompasses Pyrrhulina australis (from the Paraguay River) + P. cf. rachoviana (Lower Paraná River), P. aff. australis I (Araguaia River)/II (Paraguay River)/III (Upper Paraná River)/IV(Guaporé River),and P. marilynae (Teles Pires River), which were arranged in six distinct evolutionary lineages that align with the geographical distribution of the respective drainage basins.

Resumo A família Lebiasinidae é endêmica da região Neotropical, composta por espécies de peixes em miniaturas e de médio porte. O gênero Pyrrhulina é o segundo maior em número de espécies e o grupo com mais incertezas taxonômicas dentre os representantes dessa família. Diante disso, o objetivo deste estudo foi avaliar morfoespécies provenientes de diferentes bacias de drenagens, a fim de testar as diferentes propostas de reconhecimento de unidades taxonômicas dentro do que se acreditava ser a espécie nominal Pyrrhulina australis, utilizando o princípio do DNA Barcoding. Os resultados mostraram que Pyrrhulina australis é um complexo de espécies, com distância intraespecífica dentro do grupo de 3,74%, valor acima do Threshold Ótimo de 1,79% definido para este estudo. As análises de delimitação de espécies indicam uma surpreendente diversidade dentro das morfoespécies analisadas, especialmente no clado que engloba Pyrrhulina australis (rio Paraguai) + P. cf. rachoviana (baixo rio Paraná), P. aff. australis I (rio Araguaia), II (rio Paraguai), III (alto rio Paraná), IV(rio Guaporé)e P. marilynae (rio Teles Pires),que apresentou seis linhagens evolutivas que podem estar relacionadas ao padrão de distribuição pelas bacias de drenagens.

Abstract Arapaima is a widely-distributed fish of enormous economic importance in the Amazon region. In the present study, a total of 232 specimens were sampled, 121 from five sites in the Amazon basin and 111 from five sites in the Tocantins-Araguaia basin. The analyses investigated fragments of the Cytochrome b, Control Region, Cytochrome Oxidase I, NADH dehydrogenase subunit 2 and seven loci microsatellites. The analyses revealed the existence of two mitochondrial lineages within the general area, with no haplotypes shared between basins, and genetic variability significantly higher in the Amazon than in the Tocantins-Araguaia basin. Two divergent, but sympatric mitochondrial lineages were found in the Amazon basin, but only a single lineage in the Tocantins-Araguaia basin. The existence of these two mitochondrial lineages indicates that past events, probably occurring during the Pleistocene, resulted in the separation of the populations of this species and molded its evolutionary history, which is reflected directly in its mitochondrial DNA. The analysis of the arapaima population structure identified distinct levels of diversity within the distribution of the species, indicating specific geographic regions that will require special attention for the development of conservation and management strategies.

RESUMO Astyanax é um dos táxons mais representados e diversos na região Neotropical. Com o intuito de aumentar as informações citogenéticas para Astyanax e apresentar dados que possam subsidiar estudos taxonômicos futuros, este trabalho traz uma análise citogenética de três espécies de Astyanax: duas espécies consideradas crípticas, aqui reportadas em sintopia (A. abramis e A. lacustris) e a primeira descrição cariotípica de A. pirapuan. As análises citogenéticas revelaram 2n=50 cromossomos para as três espécies, com diferença na morfologia cariotípica de cada uma. Foram observados apenas dois sítios de rDNA 18S em A. abramis e A. lacustris e até 13 para A. pirapuan. O mapeamento físico do rDNA 5S se mostrou como um marcador efetivo para a caracterização das espécies de Astyanax abordadas neste estudo.

ABSTRACT Astyanax is one of the most abundant and diverse taxa of fishes in the Neotropical region. In order to increase the amount of cytogenetic information for Astyanax as well as to exhibit data to subsidize future taxonomic studies, this work analyzed three species of Astyanax: two species are cryptic, and are here reported to live in syntopy (A. abramis and A. lacustris); the first karyotype description for A. pirapuan is also presented. Cytogenetic analyzes reveal a diploid number of 2n=50 chromosomes for three species, yet with differences in their karyotype morphology. The physical mapping of 18S rDNA showed up to thirteen sites in A. pirapuan and two in A. abramis and A. lacustris. The physical mapping of 5S rDNA has proven to be an effective marker for the characterization of species of Astyanax studied in this work.

ABSTRACT There are two species of Neotropical Round-eared bats, Tonatia bidens Spix, 1823 and Tonatia saurophila Koopman & Williams, 1951, which present highly similar morphological characteristics that can lead to errors of identification. Specimens originally identified as T. bidens have recently been reclassified as T. saurophila, and the only karyotype documented previously for these species was 2n = 16, FN = 20. In the present study, specimens of Tonatia collected in the municipality of Barra do Garças, in the Brazilian state of Mato Grosso, were analyzed morphologically, using conventional cytogenetic techniques (C-banding, Ag-NOR, and CMA3), and through sequences of the mitochondrial cytochrome c oxidase subunit I (COI) gene. In the specimens morphologically identified as T. bidens, the diploid number (2n) was 26, and the fundamental number (FN), 38, while in T. saurophila, 2n = 16 and FN = 20, which is the karyotype also described previously for T. bidens. The dendograms obtained with sequences of the COI marker resulted in the formation of two distinct groups between T. bidens and T. saurophila, consistent with the two species, with a high sequence divergence value (14.22%). Distinct clades were also observed between T. bidens and the other phyllostomines analyzed in this study, with T. bidens also close to Phyllostomus hastatus (14.18% of sequence divergence).

A diversidade genética dos espécimes de quatro populações naturais de Arapaima coletados na bacia do Araguaia-Tocantins foi avaliada com base em cinco primers para marcadores moleculares ISSR. A sequência parcial do COI (cytochrome c oxidase subunit I ) confirmou que os espécimes pertencem à espécie Arapaima gigas . Os ISSR forneceram 168 loci , dos quais 165 polimórficos. No entanto, para cada população, os valores de heterozigosidade esperada foram baixos. A AMOVA mostrou 52,63% de variação intrapopulacional e 47,37% interpopulacional. O F ST foi alto entre todas as populações (F ST ≥ 0,25); entretanto, a análise de agrupamento e a inferência Bayesiana mostraram três grandes grupos: Araguaiana-MT + São Félix do Araguaia-MT, Novo Santo Antônio-MT e Itupiranga-PA. A distância genética não teve correlação com a distância geográfica. Os ISSRs se mostraram eficientes para determinar a diversidade genética para a A. gigas , revelando que as populações da bacia Araguaia-Tocantins estão estruturadas e com baixa diversidade genética. Estes são os primeiros dados de análise populacional utilizando ISSR para A. gigas da bacia Araguaia-Tocantins e poderão ser utilizados para definir as melhores estratégias de manejo e projetos de conservação dessa espécie.

The genetic diversity of the specimens of four natural populations of Arapaima from Araguaia-Tocantins basin was assessed within and among these stocks, using five primers for ISSR. COI (cytochrome c oxidase subunit I ) partial sequences confirmed that the specimens belongs to Arapaima gigas . The ISSR provided 168 loci, of which 165 were polymorphic. However, the number of loci for each population and expected heterozygosity values were low. AMOVA showed 52.63% intra-population variation and 47.37% inter-population variation. The F ST was high among all populations (F ST ≥ 0.25), however, the cluster analysis (PCoA) and Bayesian inference showed three major groups: Araguaiana-MT + São Félix do Araguaia-MT, Novo Santo Antônio-MT and Itupiranga-PA. The genetic distance was not correlated with geographical distance. The ISSR marker revealed that the populations of the Araguaia-Tocantins are structured and have a low genetic diversity. These are the first data from a population analysis using molecular markers for A. gigas of Araguaia-Tocantins basins and may be used to define the best management strategies and conservation projects for this species.

Astyanax é um grupo bastante diverso de peixes neotropicais cujas diferentes formas ocupam distintos ambientes. Esta grande variabilidade também se reflete em aspectos citogenéticos e moleculares, que têm sido repetidamente demonstrados por meio de estudos citogenéticos. A fim de caracterizar o cariótipo de representantes deste gênero, seis espécies foram estudadas: Astyanax altiparanae, A. elachylepis, A. xavante, A. argyrimarginatus e duas espécies novas provisoriamente citadas como Astyanax sp. e A. aff. bimaculatus. Um estudo citogenético detalhado com base na coloração convencional com Giemsa, AgNORs, banda C, fluorocromos base-específicos, e FISH com sondas para genes ribossomais 18S e 5S foi realizado com o objetivo de compreender alguns dos mecanismos cromossômicos associados com a alta diversificação que caracteriza este grupo de peixes e que culminou com o estabelecimento dessas espécies. Os resultados revelaram 2n = 50 cromossomos para cinco espécies e 2n = 52 cromossomos para Astyanax sp. Pequenas variações na macroestrutura dos cariótipos foram identificadas e se mostraram relevantes quando analisadas com base nos bandamentos clássicos, coloração por fluorocromos base-específicos e FISH com sondas de DNA 18S e 5S. Esssa diversidade cariotípica detectada indica a necessidade de uma revisão taxonômica no grupo de indivíduos aqui referidos como A. aff. bimaculatus, inclusive com a descrição de Astyanax sp., incluindo a possibilidade de inserção dessa unidade em outro gênero distinto de Astyanax.

Astyanax is a diverse group of Neotropical fishes, whose different forms occupy different environments. This great diversity is also reflected on cytogenetic aspects and molecular markers, which have repeatedly been demonstrated by cytogenetic studies. In order to characterize the karyotype of species of this genus, six species were studied: Astyanax altiparanae, A.argyrimarginatus, A. elachylepis, A. xavante, and two new species provisionally called Astyanax sp. and A. aff. bimaculatus. A detailed cytogenetic study based on conventional staining with Giemsa, AgNORs, C-banding, base-specific fluorochromes, and FISH using ribosomal genes 18S and 5S was conducted, aiming to understand some of the chromosomal mechanisms associated with the high diversification that characterizes this group and culminated with the establishment of these species. The results showed 2n = 50 chromosomes for five species and a karyotype with 52 chromosomes in Astyanax sp. Small variations in the macrostructure of the karyotypes were identified, which were quite relevant when analyzed by classical banding, fluorochromes, and FISH methods. These differences among Astyanax spp. (2n = 50) are largely due to changes in the amount and types of heterochromatic blocks. Astyanax sp (2n = 52), in addition to variations due to heterochromatic blocks, has its origin possibly by events of centric fission in a pair of chromosomes followed by minor rearrangements.These results show an interesting karyotypic diversity in Astyanax and indicate the need of a review of the group referred as A. aff. bimaculatus and the description of Astyanax sp., including the possibility of inclusion of this unit in another genus.

Ageneiosus é o gênero da família Auchenipteridae mais amplamente distribuído em bacias da América do Sul. Apesar dos estudos cromossômicos nesta família serem escassos, este gênero tem o maior número de espécies analisadas, com número diploide variando de 54 a 56 cromossomos, o que difere do restante da família (2n = 58). Este estudo objetivou analisar Ageneiosus inermisda bacia do rio Araguaia. O número diploide encontrado foi de 56 cromossomos. A heterocromatina se mostrou localizada na região terminal da maioria dos cromossomos, além de um bloco heterocromático pericentromérico no par 1, um par facilmente distinguível no cariótipo pelo seu maior tamanho quando comparado aos outros pares do complemento. AgRONs foram detectadas em somente um par de cromossomos submetacêntricos, que foi confirmado pela FISH. 5S rDNA se mostrou presente em somente um par de cromossomos metacêntricos. A hibridização com a sequência [TTAGGG]n marcou os telômeros de todos os cromossomos, além de um ITS (sequência telomérica intersticial) na região proximal do braço curto do par 1. A sequência repetitiva [GATA]n se mostrou dispersa, com localização preferencial na região terminal dos cromossomos. Ageneiosus apresenta uma organização genômica um pouco diferente quando comparada a outras espécies de Auchenipteridae. As evidências indicam que uma fusão cromossômica originou o primeiro par de cromossomos metacêntricos de A. inermis, rearranjo que parece ser um evento basal para o gênero.

Ageneiosus is the most widely distributed genus of the family Auchenipteridae among South American river basins. Although chromosome studies in the family are scarce, this genus has the largest number of analyzed species, with 2n = 54 to 56 chromosomes, differing from the rest of the family (2n = 58). This study aimed to analyze Ageneiosus inermis from the Araguaia River basin. The diploid number found was of 56 chromosomes. Heterochromatin was allocated in terminal region of most chromosomes, plus a pericentromeric heterochromatic block in pair 1, a pair distinguished by size in relation to other chromosomes pairs. AgNORs were detected in only one submetacentric chromosome pair, which was confirmed by FISH. 5S rDNA was present in only one metacentric chromosome pair. Hybridization with [TTAGGG]n sequence marked the telomeres of all chromosomes, in addition to an ITS in the proximal region of the short arm of pair 1. The repetitive [GATA]n sequence was dispersed, with preferential location in terminal region of the chromosomes. Ageneiosus has a genomic organization somewhat different when compared to other Auchenipteridae species. Evidences indicate that a chromosomal fusion originated the first metacentric chromosome pair in A. inermis, rearrangement which may be a basal event for the genus

No estado de Mato Grosso, os estudos visando investigar a diversidade e riqueza de morcegos ainda são escassos. Neste estudo, a quiropterofauna da região leste mato-grossense foi investigada por meio de capturas em quatro áreas remanescentes de Cerrado com diferentes índices de conservação, no município de Nova Xavantina. As capturas ocorreram de janeiro a dezembro de 2010, totalizando 48 noites de amostragem. Foram capturados 423 indivíduos, distribuídos em 25 espécies e cinco famílias. Os quatro ambientes apresentaram maior abundância na estação chuvosa e, ao longo dos meses e entre as estações do ano a composição, riqueza e abundância de morcegos variaram. As áreas mais preservadas apresentaram maior riqueza e maior abundância de morcegos em relação às áreas degradadas. Foi possível verificar que o horário de forrageamento variou entre os espécimes capturados, ocorrendo um pico crescente de atividade até às 19:30h e uma redução acentuada por volta das 22:30h. A diversidade calculada pelo índice de Shannon-Wiener (H′) para as áreas foi de H′=2,37.

In the state of Mato Grosso, studies aiming to investigate the diversity and richness of bats are still scarce. In the present study, the chiropteran fauna of eastern Mato Grosso (Brazil) was investigated at four sites representing the Cerrado savanna biome with different degrees of anthropogenic impact, in the municipality of Nova Xavantina. Surveys were conducted between January and December, 2010, with a total of 48 nights of mist-netting. A total of 423 bats were captured, representing 25 species distributed in five families. A greater abundance of individuals was recorded at all sites during the rainy season, with considerable variation being observed over the course of the year in species richness and composition, and the abundance of bats. Species richness and bat abundance were higher in the better preserved habitats in comparison with the impacted areas. The timing of foraging activities appeared to vary among the specimens and there was a progressive increase in activity until 19:30 h, with a marked decline after 22:30 h. The ecological diversity (Shannon-Wiener index) of the bat community within the study area was H′ = 2.37.

Astyanax xavante é descrita de um córrego da drenagem do médio rio Araguaia, localizado em área de Cerrado do planalto do Brasil Central. A nova espécie se distingue das demais do gênero pela combinação de caracteres pigmentares, morfométricos, merísticos, morfológicos e dentários. Notas ecológicas para A. xavante são incluídas.

Astyanax xavante is described from a creek of the middle rio Araguaia drainage located in the Cerrado region of the Brazilian Central Plateau. The new species is distinguished from congeners by the combination of pigmentation, morphometric, meristic, morphologic, and tooth features. Ecological notes for A. xavante are included.

A biologia reprodutiva de Triportheus trifurcatus, do médio rio Araguaia, é estudada com base em 275 exemplares obtidos em sete coletas, realizadas no período de Novembro/2003 a Janeiro/2005. Nas classes de comprimento intermediário e na amostragem geral, as fêmeas predominam, mas nas classes menores prevalecem os machos. As fêmeas se apresentam, na média, maiores que os machos. Quatro estádios de maturação gonadal são identificados, macro e microscopicamente: B- maturação, C- maduro, D-esgotado/ espermiado e E- repouso. O estádio A (imaturo) não foi registrado nos biótopos amostrados. O menor exemplar macho maduro apresenta 110 mm e a menor fêmea 119 mm de comprimento padrão. O período de desova ocorre de Novembro a Janeiro, com pico reprodutivo em Dezembro/Janeiro coincidindo com os níveis mais elevados das águas. A fecundidade absoluta é considerada baixa e há correlação positiva entre fecundidade e peso das gônadas, peso corporal e comprimento padrão. A ingestão de alimento não sofre interferência durante o período reprodutivo e é sugerida desova total para a espécie em estudo.

The study of the reproductive biology of Triportheus trifurcatus of the middle rio Araguaia was carried out using 275 specimens obtained in seven collections conducted in the period between November 2003 and January 2005. Females prevailed among the classes of intermediate length, as well as in the total number of samples. On the other hand, males prevailed in the smaller classes. The average length of females was larger than that of males. Four stages of gonadal maturation were macro- and microscopically identified: B - maturation, C - mature, D - spent and E - resting. Stage A (immature) was not found in the habitats sampled. The smallest-length male was 110 mm in standard length, and the smallest female, 119 mm. The spawning period occurred from November to January, with reproductive peak in December/January, coinciding with the highest water levels. The absolute fecundity is considered low, and there is a positive correlation between fecundity and gonad weight, body weight and standard length. Food ingestion during the reproductive period did not suffer any interference, and it is suggested complete spawning for this species.

Estudamos os cromossomos mitóticos do pirarucu Arapaima gigas, proveniente do médio rio Araguaia, município de Araguaiana (MT, Brasil). As preparações mitóticas foram analisadas através de coloração com Giemsa, bandas-C e nitrato de prata (Ag-NORs). Também foram realizadas análises através de hibridização in situ utilizando sondas de DNAr 18S. O cariótipo mostrou 2n=56 cromossomos, sendo 14 pares meta-submetacêntricos e 14 subtelo-acrocêntricos. Não foram observadas diferenças cromossômicas associadas ao sexo. Foi detectado apenas um par de cromossomos portador de Ag-NORs. Essa observação foi confirmada pelo FISH do DNAr 18S. Pequenos blocos de heterocromatina foram observados na região centromérica de alguns pares cromossômicos.

The mitotic chromosomes of the pirarucu Arapaima gigas inhabiting the middle Araguaia River and collected in the municipality of Araguaiana (MT, Brazil) were studied. The chromosomes were analyzed through Giemsa staining, C-banding, Ag-NOR staining and in situ hybridization using an 18S rRNA gene probe. The karyotype had 2n=56 comprising 14 biarmed and 14 uniarmed chromosome pairs in both sexes. No cytologically distinguishable sex chromosome was identified. A single NOR-bearing chromosome pair was detected by Ag-NOR staining and confirmed by 18S rDNA- FISH. Faint constitutive heterochromatin was C-banded in the centromeric region of some chromosomes.

Os sistemas deágua doce neotropicais possuem um alto número de espécies de peixes Siluriformes, muitas das quais popularmente conhecidas como cascudos no Brasil. Dados citogenéticos sobre três espécies de cascudos, capturadas na bacia do rio Araguaia são descritos no presente trabalho. Pterygoplichthys joselimaianus possui 2n=52, com 28 cromossomos metacêntricos (M), 16 submetacêntricos (SM) e 8 subtelecêntricos/acrocêntricos (ST/A) em ambos os sexos. Hemiancistrus spinosissimus revelou 2n=52, com fórmula cariotípica 26M+22SM+4ST, nos dois sexos. H. spilomma também possui 2n=52, porém, nesta espécie foi observado um sistema de cromossomos sexuais do tipo ZZ/ZW (25M+21SM+6ST para fêmeas e 24M+22SM+6ST para machos). As células de H. spinosissimus e P. joselimaianus mostraram um par de cromossomos portadores de Ag-RONs. Em H. spilomma, três pares de cromossomos portadores de Ag-RONs foram observados. Os dados apresentados neste trabalho revelam características cromossômicas particulares, importantes para um bom reconhecimento de ambas as espécies de Hemiancistrus e mostram a importância da inserção dos dados citogenéticos nos estudos taxonômicos e filogenéticos.

The neotropical freshwater systems have a high number of catfish species (Siluriformes), and many of those are denominated "cascudos" in Brazil. Cytogenetic data about three "cascudos" species fished in the rio Araguaia are described in the present study. The Pterygoplichthys joselimaianus showed 2n=52, with 28 metacentrics (M) chromosomes, 16 submetacentrics (SM) and 8 subtelocentrics/acrocentrics (ST/A) in both sexes. Hemiancistrus spinosissimus showed 2n=52, with karyotype formulae 26M+22SM+4ST, in both sexes. Hemiancistrusspilomma also showed 2n=52, but in this species a ZZ/ZW sex chromosome system (25M+21SM+6ST in females and 24M+22SM+6ST in males) was observed. The cells from H. spinosissimus and P. joselimaianus showed one chromosome pair bearing Ag-NORs, while in the H. spilomma three chromosome pairs bearing Ag-NORs were detected. The data showed in this work reveal particular chromosomal characteristics, important for a good recognition of both Hemincistrus species, and also show the importance of the insertion of cytogenetic data on taxonomic phylogenetic studies.

Analisou-se no presente estudo, o conteúdo estomacal de juvenis de Arapaima gigas com a finalidade de se ampliar informações sobre sua alimentação em ambiente natural. Esta espécie ocorre na bacia Amazônica, com predominância em ambientes de planície. Este é o caso do vale do médio rio Araguaia, onde se situa o lago Quatro Bocas. Juvenis de A. gigas apresentaram preferência por insetos, microcrustáceos e gastrópodes, sendo sua maioria de origem autóctone. Todos os estômagos examinados continham pelo menos um item alimentar.

The stomach content of samples of juvenile Arapaima gigas was analized to obtain information about feeding in natural environments. This species occurs in the Amazonian basin, predominantly in floodplain environment. This is the case of the valley of the middle rio Araguaia, where the lago Quatro Bocas is situated. Juveniles A. gigas prefered insects, microcrustaceans and gastropods, most of autochthonous origin. All the stomachs examined contained at least one food item.

São descritos os cromossomos supranumerários observados em cinco espécies de peixes pertencentes a quatro famílias distintas de caraciformes neotropicais. Esses cromossomos mostraram-se pequenos, totalmente heterocromáticos e acrocêntricos em Leporinus friderici e Leporinus sp. e metacêntricos e totalmente heterocromáticos em Cyphocharax modesta e Prochilodus nigricans. Em Characidium cf. zebra um pequeno extra acrocêntrico é visto totalmente eucromático. Um pequeno segmento rico em pares de bases GC pôde ser observado no cromossomo extra de Leporinus sp. após a coloração com cromomicina A3. Alguns aspectos relacionados à origem desses cromossomos extras entre os caraciformes são discutidos.

Supernumerary chromosomes were described for five species of Neotropical characiform fishes. These extra chromosomes were small, acrocentric and fully heterochromatic in Leporinus friderici from two different localities as well as in Leporinus sp., but metacentric and fully heterochromatic in Cyphocharax modesta and Prochilodus nigricans. In Characidium cf. zebra, this element was small, acrocentric and euchromatic. GC-rich DNA blocks were observed in the supernumerary chromosome of Leporinus sp. using chromomycin A3. The widespread occurrence of these extra chromosomal elements suggests their independent origins.
