Abstract Bacteria responsible for causing infections are common in hospital environments, water, soil, and food products. The infection risk is intensified by the absence of public sanitation, poor quality of life, and food scarcity. These external factors promote the dissemination of pathogens by direct contamination or biofilm formation. In this work, we identified bacterial isolates obtained from intensive care units in the southern region of Tocantins, Brazil. We compared matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) techniques and 16S ribosomal ribonucleic acid (rRNA) molecular analysis; we also performed phenotypic characterization. Fifty-six isolates characterized using morphotinctorial tests were classified as gram-positive (80.4%; n = 45) and gram-negative (19.6%; n = 11) and were resistant to several antibiotic classes; notably, we identified the blaOXA-23 resistance gene in the ILH10 isolate. Microbial identification using MALDI-TOF MS resulted in the identification of Sphingomonas paucimobilis and Bacillus circulans. 16S rRNA sequencing revealed four isolates belonging to the genera Bacillus and Acinetobacter. The similarity was superior to 99% for Acinetobacter schindleri in the Basic Local Alignment Search Tool (BLAST), grouped in the clade superior to 90%. Several strains isolated from intensive care units (ICU) were resistant to various antibiotic classes. These techniques allowed for the identification of several microorganisms of importance in public health, enabling improvements in human infection control and proving the quality of inputs, food, and water.
Resumo As bactérias responsáveis por causar infecções são comuns em ambientes hospitalares, água, solo e produtos alimentícios. O risco de infecção é intensificado pela ausência de saneamento público, má qualidade de vida e escassez de alimentos. Esses fatores externos promovem a disseminação de patógenos por contaminação direta ou formação de biofilme. Neste trabalho, identificamos isolados bacterianos obtidos de unidades de terapia intensiva na região sul do Tocantins, Brasil. Comparamos técnicas de espectrometria de massa de tempo de voo com ionização por dessorção a laser assistida por matriz (MALDI-TOF MS) e análise molecular de ácido ribonucleico ribossômico 16S (rRNA); também realizamos caracterização fenotípica. Cinquenta e seis isolados caracterizados por testes morfotintoriais foram classificados como gram-positivos (80,3%; n = 45) e gram-negativos (19,6%; n = 11) e foram resistentes a várias classes de antibióticos; notavelmente, identificamos o gene de resistência blaOXA-23 no isolado de ILH10. A identificação microbiana usando MALDI-TOF MS resultou na identificação de Sphingomonas paucimobilis e Bacillus circulans. O sequenciamento do 16S rRNA revelou quatro isolados pertencentes aos gêneros Bacillus e Acinetobacter. A similaridade foi superior a 99% para Acinetobacter schindleri no BLAST, agrupado no clado superior a 90%. Várias cepas isoladas de ICU foram resistentes a várias classes de antibióticos. Essas técnicas permitiram a identificação de diversos microrganismos de importância em saúde pública, possibilitando melhorias no controle de infecções humanas e comprovando a qualidade dos insumos, alimentos e água.

RESUMO A região semiárida apresenta o problema inerente de acúmulo de sais no solo devido ao uso de irrigação com água salobra, e o déficit hídrico que compromete o crescimento e os índices fisiológicos das culturas. Objetivou-se com este trabalho avaliar o efeito do estresse salino e hídrico no crescimento e nas trocas gasosas da cultura da abobrinha cv. Caserta. O trabalho foi realizado em área pertencente à Universidade da Integração Internacional da Lusofonia Afro-Brasileira, Redenção, Ceará, Brasil. O delineamento experimental utilizado foi inteiramente casualizado, em esquema fatorial 5 × 2 com cinco níveis de condutividade elétrica da água de irrigação - CEa (0,5, 1,0, 1,5, 2,0 e 2,5 dS m-1), e dois regimes hídricos (50 e 100% da evapotranspiração potencial da cultura - ETc) com cinco repetições. Aos 36 dias após a semeadura foram avaliadas as variaveis:taxa de assimilação de CO2, transpiração, condutância estomática, concentração interna de carbono, temperatura foliar, índice relativo de clorofila e a eficiência do uso da água. Aos 45 dias após a semeadura, foram avaliadas as seguintes variáveis: altura da planta, diâmetro do caule, número de folhas e área foliar. O uso de água salobra (1 a 2,5 dS m-1) reduz os parâmetros de crescimento da abobrinha cv. Caserta. O aumento da CEa ocasionou redução nas variáveis fisiológicas. A lâmina de 100% proporcionou maiores valores para as variáveis taxa de assimilação de CO2, transpiração, e eficiência instantânea no uso da água, e a lâmina de 50% para concentração interna de carbono, com o aumento da CEa.
ABSTRACT Semi-arid regions present the inherent problem of accumulation of salts in the soil due to the use of brackish water for irrigation, and water deficit compromises the growth and physiological indices of crops. This study evaluated the effect of salt and water stress on growth and gas exchange in the zucchini cv. Caserta crop. The study was conducted at the University of International Integration of Afro-Brazilian Lusophony, Redenção, Ceará State, Brazil. The experimental design was completely randomized, in a 5 × 2 factorial scheme with five levels of electrical conductivity of the irrigation water - ECw (0.5, 1.0, 1.5, 2.0, and 2.5 dS m-1) and two water regimes (50 and 100% of the potential crop evapotranspiration - ETc), with five replicates. At 36 days after sowing, the following traits were evaluated: CO2 assimilation rate, transpiration, stomatal conductance, internal carbon concentration, leaf temperature, relative chlorophyll index, and water use efficiency. At 45 days after sowing, the plant height, stem diameter, number of leaves, and leaf area were also evaluated. The use of brackish water (1 to 2.5 dS m-1) reduced the growth parameters of zucchini cv. Caserta. The increase in ECw caused a decline in the physiological traits. Under 100% ETc, higher values of CO2 assimilation rate, transpiration, and instantaneous water use efficiency were recorded, and there was 50% ETc for internal carbon concentration, even with the increase in ECw.

The antifungal potential of volatile organic compounds (VOCs) produced by actinobacterial strains Streptomyces sp. (ACTB-77) and Amycolatopsis sp. (ACTB-290) from the rhizosphere of Caatinga plants against Colletotrichum musae was investigated. VOCs produced by these microorganisms (axenic and co-culture) were investigated using headspace-solid phase micro-extraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS). Although no exclusive VOC peaks were observed in the co-culture with ACTB-77, the same experiment involving ACTB-290 yielded five new peaks, including two identified alcohols, suggested as bioreductive products of the corresponding ketones by the fungus. Statistical analysis revealed that co-culture ACTB-77/C. musae has a closer similarity to the fungus than to the actinobacteria, while the co culture ACTB-290/C. musae showed closer similarity to the actinobacteria. These confirmed the more pronounced antifungal activity of the ACTB-290 strain, as observed in the fungus growth inhibition experiments. The antifungal activity of ACTB-290 was associated to its sulfur-containing metabolites, while linalool was suggested as responsible for the ACTB-77 activity.

RESUMO A cultura do amendoim apresenta grande interesse econômico, em decorrência de suas sementes, constituírem importante fonte de proteína vegetal e de óleo comestível. O objetivo do presente trabalho foi avaliar a fisiologia e crescimento da cultura do amendoim submetido a estresse salino e hídrico. O experimento foi conduzido em Auroras, Redenção, Ceará, Brasil. O delineamento experimental foi inteiramente casualizado em parcelas subdivididas, cujas parcelas referem-se aos níveis de condutividade elétrica da água de irrigação - CEa (1,0; 2,0; 3,0; 4,0 e 5,0 dS m-1) e as subparcelas aos regimes hídricos de 50 e 100% da evapotranspiração da cultura. O estresse salino afeta negativamente a altura de planta, número de folhas e massa seca da parte aérea, índices relativos de clorofila, transpiração e a temperatura foliar da cultura do amendoim. O regime hídrico de 100% da ETc proporcionou maiores desempenhos para as variáveis diâmetro do caule, número de folhas e massa seca da parte aérea da cultura do amendoim.
ABSTRACT Peanut crops are of great economic interest because their seeds are used as an important source of vegetable protein and edible oil. The objective of this study was to evaluate the physiological indices and growth of peanut crops subjected to saline and water stress. The experiment was conducted in Auroras, Redenção, Ceará, Brazil using a completely randomized split plot statistical design. The plots were composed of irrigation water treatments with varying levels of electrical conductivity - ECw (1.0, 2.0, 3.0, 4.0 and 5.0 dS m-1), while the subplots were formed by the water regimes of 50% and 100% crop evapotranspiration. Plant height, leaf number, shoot dry matter, relative chlorophyll level, transpiration, and leaf temperature were affected by salt stress. The water regime of 100% ETc provided better performance for the variables stem diameter, leaf number, and shoot dry mass of peanuts.

Abstract Candida spp. is considered an important cause of healthcare-associated infections worldwide. Currently, the emergence and spread of resistant Candida isolates are being increasingly reported, making the development of new agents urgently needed. In this study, we showed the in vitro anti-Candida activity of seven synthetic 3-alkylpyridine alkaloid analogs. Among them, alkaloid 1 presented a potent antifungal effect, which was independent of its capacity of binding with the fungal membrane ergosterol or cell wall. Analog 1 showed fungistatic and fungicidal effects against C. albicans (MIC 7.8 μg/mL and MFC 62.5 μg/mL), C. glabrata, C. krusei (MIC and MFC 31.2 μg/mL), and C. tropicalis (MIC 31.2 μg/mL and MFC 125 μg/mL). The time kill-curve study showed that compound 1 has a potent fungicidal effect in vitro, eliminating C. albicans cells. Furthermore, an in vitro synergistic effect with ketoconazole was observed for compound 1. This compound also eliminated the yeast-to-hypha transition. However, it showed high cytotoxicity against mammalian cells. Taken together, these findings support the use of compound 1 as a prototype to develop new anti-Candida agents, but molecular modifications should be done to minimize the high cytotoxicity obtained.

OBJECTIVE: To assess the possible factors that influence sleep quality in adolescents with and without chronic immunosuppressive conditions quarantined during the coronavirus disease 2019 (COVID-19) pandemic. METHODS: This cross-sectional study included 305 adolescents with chronic immunocompromised conditions and 82 healthy adolescents. Online surveys were completed, which included questions on socio-demographic data and self-rated healthcare routine during COVID-19 quarantine and the following validated questionnaires: the Pittsburgh Sleep Quality Index (PSQI), Pediatric Quality of Life Inventory 4.0 (PedsQL4.0), and Pediatric Outcome Data Collection Instrument (PODCI). RESULTS: The median current age [14 (10-18) vs. 15 (10-18) years, p=0.847] and frequency of female sex (62% vs. 58%, p=0.571) were similar in adolescents with chronic conditions compared with healthy adolescents. The frequency of poor sleep quality was similar in both groups (38% vs. 48%, p=0.118). Logistic regression analysis, including both healthy adolescents and adolescents with chronic conditions (n=387), demonstrated that self-reported increase in screen time (odds ratio [OR] 3.0; 95% confidence interval [CI] 1.3-6.8; p=0.008) and intrafamilial violence report (OR 2.1; 95% CI 1.2-3.5; p=0.008) were independently associated with poor sleep quality in these adolescents. However, the PODCI global function score was associated with a lower OR for poor sleep quality (OR 0.97; 95% CI 0.94-0.99; p=0.001). Further logistic regression, including only adolescents with chronic conditions (n=305), demonstrated that self-reported increase in screen time (OR 3.1; 95% CI 1.4-6.8; p=0.006) and intrafamilial violence report (OR 2.0; 95% CI 1.2-3.4; p=0.011) remained independently associated with poor quality of sleep, whereas a lower PODCI global function score was associated with a lower OR for sleep quality (OR 0.96; 95% CI 0.94-0.98; p<0.001). CONCLUSION: Self-reported increases in screen time and intrafamilial violence report impacted sleep quality in both healthy adolescents and those with chronic conditions. Decreased health-related quality of life was observed in adolescents with poor sleep quality.

We report herein the design and synthesis via click chemistry of twelve novel triazole retinoid analogues of tamibarotene (AM80) and the evaluation of their anticancer activities against six cancer cell lines: HL60, K562, 786, HT29, MCF7 and PC3. Among the synthesized compounds, two were more potent than tamibarotene against solid tumor cells, and one of them had similar potency to tamibarotene against HL60 cells. The bioisosteric exchange between the amide group and the 1,2,3-triazole core in the retinoid agent tamibarotene (AM80) reported in this work is a valid strategy for the generation of useful compounds against cancer.

Extramedullary plasmacytoma in a captive collared peccary (Pecari tajacu)

Resumo: Um caso de plasmoci- toma extramedular em cateto criado em cativeiro (Pecari tajacu) está sendo relatado. O animal, uma fêmea com três anos de idade, apresentou um histórico médico de lesões de pele, envolvendo junção mucocutânea e de aumento de linfonodos. Exame clínico e exames complementares (hemograma completo, análise bioquímica, raspado de pele para pesquisa de ácaros e cultura fúngica) foram realizados. Após 30 dias, o animal morreu. Na necropsia, verificou-se a presença de múltiplos nódulos, pústulas assépticas e aumento dos linfonodos. Na pele e mucosa oral, histologicamente as massas consistiam em ninhos, lençóis e cordões de células redondas, e um estroma fibrovascular bem vascularizado. Os plasmócitos foram observados infiltrados entre as fibras e músculo liso da lâmina própria. Espaços contendo eosinófilos, fluido e células livres estavam presentes na massa. A maioria das células estava bem diferenciada, com uma zona perinuclear clara, mas algumas células demonstraram-se pleomórficas. Os plasmócitos apresentavam núcleo excêntrico, redondo, basófilo, e pontilhado, com cromatina variando de densa a grosseiramente organizada e nucléolos distintos. O citoplasma finamente granular de anfifílico para basofílico. As células foram interpretadas como plasmócitos neoplásicos. Células binucleadas foram observadas e células gigantes multinucleadas eram raras. Na imunohistoquímica de tecidos da mucosa oral e de linfonodos observou-se positividade moderada e multifocal para Mb-1. As células tumorais revelaram positividade fraca e intermitente para Bcl-2. Com base na localização, achados histopatológicos e imuno-histoquímicos, um plasmocitoma extramedular foi diagnosticado.
Abstract: An extramedullary plasmacytoma case in a captive collared peccary (Pecari tajacu) is reported. The animal, a female aging three years old, had a medical history of diffusely distributed skin and mucocutaneous junction lesions, associated with swollen lymph nodes. Clinical examination and complementary exams (complete blood count, biochemical analysis, skin scraping to search mites and fungal culture) were performed. Thirty days after examination, the animal died. At necropsy, multiple consistent nodules, aseptic pustules and swollen lymph nodes were found. On histological exams of the skin and oral mucosa, we observed a large number of round cells forming masses organized in nests, sheets, and cords of cells in a well-vascularized fibrovascular tissue. Neoplastic plasma cells infiltrated between the fibers and the lamina propria of smooth muscle. Spaces among the cell masses were filled with some eosinophil and fluid. Most of the cells were well differentiated, presenting a perinuclear clear zone. In some points, the cells were pleomorphic. The plasma cells presented eccentric, basophilic and spherical nuclei, showing a dense to organized chromatin with distinct nucleoli. Binucleate cells were observed, but multinucleated giant cells were rare. Oral mucosa and lymph nodes tested by immunohistochemical analyses were positive for Mb-1, with a multifocal distribution. In regard to Bcl-2, the neoplastic cells were intermittent weakly positive. So, an extramedullary plasmacytoma was diagnosed in the collared peccary considering the location, the histopathological and immunohistochemical findings.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Cyclosporin safety in a simplified rat brain tumor implantation model

Neoplasias encefálicas constituem a segunda causa neurológica de morte. Foi desenvolvido um modelo animal simplificado de tumor cerebral em ratos utilizando a linhagem celular W256 (carcinoma 256 de Walker) para permitir teste de novos tratamentos. Ratos Wistar foram inoculados nos gânglios da base (caudato subfrontal direito) com uma solução celular tumoral, por via estereotáxica. Este modelo demonstrou crescimento tumoral em 95% dos animais inoculados com sucesso, além de mostrar ausência de metástases extracranianas e infecção sistêmica. A mediana de sobrevida dos animais foi de 10 dias. O volume tumoral estimado foi de 17,08±6,7 mm³ no sétimo dia e de 67,25±19,8 mm³ no nono dia após a inoculação. O tempo de duplicação foi estimado em 24,25 h. O crescimento tumoral induziu a caquexia, mas não houve alterações bioquímicas ou hematológicas. Esse modelo permite fácil reprodução e comporta-se como um tumor indiferenciado, mostrando potencial para estudar migração celular tumoral no sistema nervoso central. Dexametasona 3,0 mg/kg/dia reduziu significantemente a sobrevida dos animais inoculados com tumor nesse modelo. Ciclosporina 10 mg/kg/dia não teve efeito na sobrevida, sendo sua administração bem tolerada.
Brain cancer is the second neurological cause of death. A simplified animal brain tumor model using W256 (carcinoma 256, Walker) cell line was developed to permit the testing of novel treatment modalities. Wistar rats had a cell tumor solution inoculated stereotactically in the basal ganglia (right subfrontal caudate). This model yielded tumor growth in 95% of the animals, and showed absence of extracranial metastasis and systemic infection. Survival median was 10 days. Estimated tumor volume was 17.08±6.7 mm³ on the 7th day and 67.25±19.8 mm³ on 9th day post-inoculation. Doubling time was 24.25 h. Tumor growth induced cachexia, but no hematological or biochemical alterations. This model behaved as an undifferentiated tumor and can be promising for studying tumor cell migration in the central nervous system. Dexamethasone 3.0 mg/kg/day diminished significantly survival in this model. Cyclosporine 10 mg/kg/day administration was safely tolerated.

This work studied the anti-inflammatory activities of the hydroalcoholic extracts (HAEs) from Erythrina velutina Willd. (Ev) and E. mulungu Mart. ex Benth. (Em) in the carrageenan- and dextran-induced mice hind paw edema models. These medicinal plants belonging to the Fabaceae family are used in some Brazilian communities to treat pain, inflammation, insomnia and disorders of the central nervous system. In the present work, the extracts were administered orally in male mice at the doses of 200 or 400 mg/kg. In the carrageenan-induced test, only Em showed anti-inflammatory activity, decreasing the paw edema, at the doses of 200 and 400 mg/kg. No effect was observed with Ev in this model. On the other hand, in the dextran model, Ev demonstrated anti-inflammatory effect, showing decrease of the paw edema at the 1, 2, 3, 4 and 24th h. Em (200 or 400 mg/kg) presented anti-inflammatory effect at the 2, 3 and 4th h after administration of dextran, as compared to control. In conclusion, the work showed that Ev and Em present anti-edematous actions, which possibly occurs by distinct mechanisms. While Ev seems to interfere especially in inflammatory processes in which mast cells have an important role, Em exerts greater activity in the inflammatory process that depends mainly on polymorphonuclear leucocytes. However, further studies are needed to determine the exact mechanism of action of the species investigated.

O presente trabalho teve por objetivo verificar a ocorrência de demência senil em amostra de pessoas de 65 e mais anos de idade residentes no Município de Fortaleza. Foram testados 865 idosos, a partir do teste de Informação, Memória e Concentração, IMC (teste de Hachinski, modificado), que aborda aspectos concernentes à identificação da pessoa, à memória de fatos atuais e remotos e à concentração, determinando os estados demenciais em função do número e grau de respostas numa seqüência programada e previamente testada quanto à sensibilidade e especificidade. Analisando-se os resultados quanto a idade, sexo e condição social, evidenciou-se uma prevalência de 8,4% de demência senil no conjunto geral, com diferença não-significativa na faixa de 75 e mais anos de idade (9,3%), comparada ao grupo de 65 a 74 anos (7,5%). Os percentuais foram semelhantes para homens e mulheres: 8,7% e 8,3%, respectivamente. Quanto à condição social, foi verificado um aumento progressivo nas proporções de estados demenciais, desde 4,2% para o estrato A/B (abastados), 6,9% para o grupo C (nível intermediário) e 10,3% no nível D/E correspondente ao estrato de pessoas economicamente desprivilegiadas. Estes resultados, entretanto, deverão ser vistos com cena reserva, dado o grau de recusa por ocasião das entrevistas, especialmente nos estratros A/B (19,3%) e D/E (9,3%).

The Test includes questions on personal identification, recent and remote memory and concentration. Dementia was determined as a function of the total score obtained by the person in the test previously examined as far as sensitivity and specificity are concerned. The results were analysed according to age, sex and social strata. It was demonstrated a prevalence of dementia of 8.4% in the sample studied with no significant difference in the proportions of cases among people 75 years old or older (9.3%) as compared to the group younger, 65 to 74 years old, (7.5%). The percentage of dementia was similar between men (8.7%) and women (8.3%). It was verified a progressive increase in the frequence of dementia ranging from 5.2% for the higher social classes (A/B) to 6.9% and 10.3% for medium (C) and lower (D/E) ones respectively, with a significant difference between the social classes A/B and D/E. These results, however, should be seen with caution because of the percentage of refuse especially in the higher, A/B (19.3%) and lower D/E (9.3%) social classes.
