Rooting in vitro and acclimatization in greenhouse of japanese persimmon (Diospyrus kaki L.)

This work aimed at surpassing the difficulties of attainment of seedlings studing the rooting and acclimatization of plants from tissue culture, these steps very critic in micropropagation of plants, to be difficult the protocol ideal establishment. It were used nodes segments with one cm of height with 2 leaves from direct organogenisis though root segments. Two times of permanence were tested (5 and 10 days), in MS culture com ½ NO3 supplemented with AIB in two concentrations (50 and 100 µM). The experimental design used was entirely randomized, with 4 repetitions and 12 explants per plot. In the acclimatization the plants were conditioned in plastic glass with Plantmax® substratum, with and without covering of the plants with transparent plastic glass, in chamber mist with intermittent irrigation during 40 days after. The results obtained in the rooting did not significantly differ, obtained until 75% of rooted buds. In the acclimatization, the plants which were not coverdwith transparent plastic glass presented greater survival, reaching 47,3%.
