Abstract

Aiming to increase the efficiency of in vitro establishment of grapevine rootstocks `Jales' and `Campinas', some experiments were carried out with shoot tips and nodal segments, taken from shoots of hardwood cuttings. Several 6-benzylaminopurine (BAP) concentrations were tested in MS medium with half salt strength. This cytokinin had a markedly effect on the explant growth. The increase of BAP concentration stimulated growth and foliar expansion of shoot tips and axillary buds up to 10µM. The use of 20µM, BAP led to a reduced growth of nodal segments. It was concluded that rootstock establishment can be done in an half strength MS medium supplemented with 10µM of BAP.
