Protein content analysis of the aqueous humor in congenital glaucoma

RESUMO

O conteúdo protéico do humor aquoso de 20 pacientes portadores de Glaucoma Congênito Primário e 18 de Catarata Congênita foi submetido a análise quantitativa e qualitativa. O humor-aquoso foi coletado através de paracentese em pré-operatório imediato.

A concentração total das proteínas do humor aquoso de todos os pacientes, foi avaliada pelo método de BRADFORD e mostrou que o conteúdo protéico total é maior nos pacientes com Glaucoma Congênito Primário quando comparado aos pacientes com Catarata Congênita, sendo a diferença, estatisticamente significante.

À análise qualitativa foi realizada pela Unidade Automatizada - Phast System, utilizando-se gel SDS-PAGE com gradiente 8 - 25%, corado pela prata. Todas as placas de gel revelaram a presença bem marcante da proteína de 67 KD (Albumina). Outras proteínas, de alto e baixo peso molecular foram reveladas, com incidência estatisticamente não significante.

SUMMARY

Quantitative and qualitative analysis of the protein content of the aqueous humor from 20 primary congenital glaucoma and 18 congenital cataract individuais was performed.

Their aqueous humor was obteined during required surgical treatment. Bradford Method showed that the mean concentration of total protein content in the aqueous humor of patients with primary congenital glaucoma, was greater and statistically significant than the patients with congenital cataract.

Qualitative analysis was performed with Phast System Unit and consisted of SDS - PAGE 8 - 25% gradient and silver staining. All gels revealed the presence of one well defined 67 KD protein band (Albumin). High and low molecular weight proteins bands, were revealed and the statistical analysis was no significant.
