Distribuição de antígenos em biópsias de pele e mucosa na paracoccidioiriomicose humana

Paracoccidioides brasiliensis antigens (strain 113) were located at ultrastructural level in both yeast and mycelial forms of the fungus. Immunofluorescence and ultrastructural immunolabeling techniques were performed using two polyclonal antisera: one against P. brasiliensis exoantigens and other against a 43-kDa glycoprotein (Gp43). The antigens deposits were seen within the cytoplasm, and over the cell wall of the fungus. Immunocytochemical techniques suggest a protein synthesis within the cytoplasm followed by excretion through the cell wall. Immunoblotting assays were employed to define reactivity of these antisera with somatic and metabolic antigens of both forms of the fungus. The anti-exoantigens serum recognized several bands in both forms of the fungus. The anti-Gp 43 serum react strongly with the 43-kDa fraction and weakly with lew other ones. The distribution of P. brasiliensis antigen(s) in human skin and mucosa of patients with paracoccidioidomycosis were studied. In this step, double immunolabeling and immunoelectro mycroscopic techniques were applied, using the same antisera. Langerhans' cells were detected by double immunolabeling using an anti-S 100 protein monoclonal antibody. Double labeling immuno-histochemistry showed that both the immune sera labeled the yeast cells in the center of the granuloma and those transmigrating through the epithelial layer equaly well. P. brasiliensis antigens, detected by antiserum against parasite exoantigens, were deposited between basal keratonocytes and permeating granulomatous cells. The P. brasiliensis antigen(s) also accumulated in the macrophages but not in the Langerhan's cells. P. brasiliensis antigens, as assessed by immunoelectro mycroscopic techniques, are presented in the cytoplasm of macrophages and in the yeast cells in the host tissues. Antigens arc transported to the cell membrane and later excreted through the cell wall. Antigenic deposits were also seen at the fungus-host interface.
