EVALUATION OF THE CLOTTING TIME OF BOVINE THROMBIN, REPTILASE® AND THROMBIN-LIKE FRACTION OF Crotalus durissus terrificus SNAKES USING CRYOPRECIPITATE FROM HUMAN BEINGS AND DIFFERENT ANIMALS

ABSTRACT. In the present work, the author evaluated the effects of thrombin-like fraction of Crotalus durissus terrificus venom, of Reptilase® and of bovine thrombin on fibrinogen present in the cryoprecipitate from human beings as well as that of different animals such as bovine, equine, ovine and bubaline. The author also verified which of the cryoprecipitates studied presented behavior similar to that extracted from human beings for possible use in medical practice. Fibrinogen levels in the cryoprecipitate were studied in 48 blood samples obtained, as follows: 12 samples from humans, 9 from bovine, 10 from equine, 10 from ovine and 7 from bubaline. The results showed average levels of 375.50 mg% for humans, 218.33 mg% for bovine, 240.80 mg% for equine, 267.70 mg% for ovine and 664.00 mg% for bubaline. Upon forming pools of human fibrinogen and of each species of animal, the following results were obtained: 435 mg% for humans, 444 mg% for bovine, 337 mg% for equine, 390 mg% for ovine and 530 mg% for bubaline. Statistical analysis using the analysis of variance for entirely randomized experiment for the calculation of F statistics demonstrated that the fibrinogen level of bubaline was higher than that of humans which was higher than that of ovine, equine and bovine. Evaluation of clotting time with the cryoprecipitate pool studied and using different dilutions of bovine thrombin, thrombin-like fraction of Crotalus durissus terrificus venom and Reptilase® was as follows: the clotting time obtained with bovine thrombin, Reptilase® and thrombin-like fraction was similar for humans and bovine. Upon comparing the clotting times obtained with thrombin-like fraction of Crotalus durissus terrificus venom and Reptilase® using cryoprecipitate from humans and different animals, it was verified that for humans and bovine these results were similar. The other cryoprecipitates (from equine, ovine and bubaline) presented differences from dilution 1:3. The results obtained permitted the following conclusions to be drawn: bovine thrombin presented better interactivity with fibrinogen extracted both from human and from bovine cryoprecipitates; similar behavior was obtained when bovine thrombin was substituted for both Reptilase® and for thrombin-like fraction of Crotalus durissus terrificus venom; cryoprecipitate extracted from bovine can, in special circumstances substitute human cryoprecipitate in medical practice; human and bovine cryoprecipitate can be used with both Reptilase® and Crotalus durissus terrificus fractions in dilutions up to 1:5; use of bovine cryoprecipitate can be indicated using either bovine thrombin, Reptilase® or thrombin-like fraction of Crotalus durissus terrificus venom.
