Human Antibody Libraries Displayed on the Surface of Filamentous Phages: A Plentiful Source of Antibodies for Diverse Purposes.

Applications of phage display have been very abundant and successful over the last decade. Phage display links protein or peptide function to genetic-coding information in individual bacteriophage particles, allowing the high-throughput screening of libraries of 106-1012 sequences for suitable properties. Protein and peptide display on the surface of filamentous phages and their selection on specific targets is a robust methodology with a wide variety of applications Display and selection of antibody fragments, which include basically the variable region, comprising only the antigen binding site, resemble immune selection, allowing the selection of specific antibody fragments from non-immune libraries. The affinity of selected antibodies keeps a relationship with the size of the library, in such a way that high-affinity antibodies can be selected from highly diverse libraries. The required diversity for the selection of high-affinity antibodies from an immune library is much lower than the one required for such a selection from a non-immune library. In this work, general aspects on the generation and screening of a phage display library will be discussed, including the characterization of the selected antibody fragments. Some ideas about the possible application of this methodology for the isolation and characterization of neutralizing antibodies of venom from venomous animals will also be addressed.
