  • Efeitos do tamanho da semente no crescimento e produção de cultivares de feijoeiro de diferentes tamanhos de semente Research Articles

    Desenvolveu-se um experimento de campo para avaliar os efeitos da variação do tamanho da semente da mesma cultivar na acumulação de biomassa e produção de grãos de cultivares de feijoeiro (Phaseolus vulgaris L.) de diferentes tamanhos de semente. Empregou-se um esquema fatorial 6 X 2 com quatro repetições, onde seis cultivares de feijoeiro constituíram as parcelas e, dois tamanhos de semente (pequeno e grande), as subparcelas. Efetuaram-se oito amostragens semanais de biomassa e mensuraram-se os componentes de produção na maturação. Sementes grandes aumentaram o índice de área foliar e a biomassa de parte aérea e raiz das cultivares de feijoeiro, particularmente no início do ciclo, mas não afetaram a massa de vagens. O estímulo ao crescimento ocasionado pelas sementes grandes foi mais intenso nas cultivares eretas do que nas prostradas. Plantas oriundas de sementes pequenas apresentaram maiores taxas de crescimento relativo e de assimilação líquida do que as oriundas de sementes grandes. Estas não afetaram a produção de grãos, mas reduziram o número de sementes por vagem e o índice de colheita, e levaram a massa de cem sementes. Os resultados indicaram que a semeadura de sementes grandes de uma cultivar de feijoeiro pode estimular o crescimento inicial do cultivo, o que pode ser vantajoso no estabelecimento da cultura sob condições de estresse. Entretanto, algum efeito compensatório, associado a atraso na senescência foliar, maior taxa de assimilação líquida, ou maior número de sementes por vagem, permite que plantas oriundas de sementes pequenas obtenham produção de grãos similar. Efeitos pouco consistentes do tamanho da semente no rendimento de grãos indicam que os custos adicionais da utilização apenas de sementes maiores podem não ser viáveis, mas as sementes grandes produziram maiores grãos na colheita, que, geralmente, atingem melhor preço de mercado.

    A field experiment was conducted to evaluate the effects of planting different sizes of seed of the same cultivar on biomass accumulation and grain yield of common bean (Phaseolus vulgaris L.) cultivars of different seed sizes. A 6 X 2 split-plot factorial design with four replicates was employed, with six bean cultivars as main plots and two seed sizes (small and large, the latter twice as heavy as the former) of the same cultivar as subplots. Eight weekly samplings of biomass were performed, and yield components were measured at maturity. Large seeds increased the leaf area index and the shoot and root biomass of bean cultivars, particularly at the beginning of the growth cycle, but they did not affect the pod biomass. Improved growth associated with the large seed was more intense for erect than for prostrate cultivars. Plants originating from small seed presented a higher relative growth rate and net assimilation rate than plants from large seed. Large seed did not affect grain yield, but reduced the number of seeds per pod, increased the 100-seed mass, and reduced the harvest index. The results indicate that sowing larger seeds of a bean cultivar can improve early-season plant growth, which might be advantageous for crop establishment in stress environments. However, some compensatory effect, associated with delayed leaf senescence, higher net assimilation rate, or greater number of seeds per pod, allows plants originating from small seed to achieve similar grain yield. Lack of consistent effects of the seed size on grain yield indicate that the extra costs of sowing only the largest seed would not be profitable, but large seed resulted in larger grains at harvest which usually attain better market price.
  • Produção de etileno e expressão do gene de ACC-oxidase durante a maturação de frutos de Coffea arabica L. Research Articles

    O fitormônio etileno está associado a vários eventos fisiológicos e etapas de desenvolvimento da plantas. A enzima 1-aminociclopropano-1-carboxilato oxidase (ACO) catalisa o último passo da biossíntese desse fitormônio. Neste trabalho, estudou-se a produção de etileno durante a maturação de frutos de café, assim como a expressão do gene de ACC oxidase de cafeeiro durante a maturação e em diferentes órgãos da planta. Verificou-se rápido crescimento na produção de etileno em frutos verde-cana, após o final da formação do endosperma, com um decréscimo nos frutos-cereja, indicando uma fase climatérica na maturação. Usando oligonucleotídeos previamente descritos, efetuou-se a clonagem e caracterização de um gene de ACO de C. arabica (Ca-ACO). A seqüência de nucleotídeos, assim como a de aminoácidos, demonstrou alta similaridade com ACO de frutos climatéricos. O padrão de transcrição gênica de Ca-ACO em frutos em diferentes fases de maturação e em diferentes tecidos foi analisado em Northern Blots. Frutos verdes apresentaram a menor transcrição de Ca-ACO, com um aumento a partir dos frutos verde-cana, havendo pouca alteração nas etapas subseqüentes de maturação, sugerindo a atuação de mecanismos de controle pós-transcricionais. Os resultados indicam a natureza climatérica da maturação dos frutos de café.

    The phytohormone ethylene is involved in several physiological and developmental processes in higher plants, including ripening of fruits, abscission of organs and tissues, senescence, wound response as well as in other abiotic stresses. The enzyme 1-aminocyclopropane-1-carboxylate oxidase (ACO) catalyzes the last step of ethylene biosynthesis. The production of ethylene and the expression of a Coffea arabica ACO gene during the last stages of fruit maturation were investigated. A rapid increase of ethylene production at the green-yellow stage of fruit ripening, after the end of endosperm formation, and a decrease of ethylene production at the cherry stage indicates a climacteric phase during ripening. An ACC oxidase (Ca-ACO) from coffee fruit cDNA was cloned and characterized using primers previously reported. The cDNA is homologous to previously described ACC oxidase cDNA in Coffea. The nucleotide and amino acid deduced sequences of the clone showed high homology with ACO from climacteric fruits. Northern blots were performed to determine the Ca-ACO transcription pattern from different tissues and from fruits at different ripening stages. Coffee fruits at an early ripening stage (green) showed the lowest level of Ca-ACO transcript accumulation. The transcript levels of Ca-ACO did not change significantly during the later stages, suggesting the presence of post- transcriptional control mechanisms. These results, taken together, strongly suggest a climacteric nature of coffee fruit ripening.
  • Desenvolvimento da planta de soja nodulada após o alagamento do sistema radicular com diferentes fontes de nitrogênio Research Articles

    O alagamento leva à hipoxia, um estresse ao qual a fixação simbiótica de N2 é muito sensível. Estudou-se a resposta da planta de soja nodulada ao alagamento do sistema radicular por 21 dias, na presença e na ausência de NO3- e NH4+ no meio. Avaliaram-se parâmetros de crescimento da planta e componentes nitrogenados orgânicos transportados à parte aérea através do xilema. A inundação diminuiu drasticamente a fixação de N2, independentemente da sua fonte, como indicaram os níveis de ureídeos na seiva do xilema. Na ausência de fonte de N, o desenvolvimento foi fortemente reduzido durante o alagamento e seu acúmulo na parte aérea foi pequeno. O alagamento na presença de 5 mM de NO3- ou NH4+ proporcionou acumulação de N total na parte aérea, mas somente o NO3- promoveu aumento na matéria seca total, estatura e área foliar das plantas acima do tratamento inundado sem N. Entretanto, seu acúmulo na parte aérea dos tratamentos inundados com NO3- e NH4+ foi menor que a testemunha não inundada. O incremento na matéria seca total, estatura e área foliar das plantas do tratamento com NO3- foi similar ao da testemunha não inundada. Os dados demonstram claramente o efeito benéfico do NO3- durante um período prolongado de alagamento do sistema radicular nodulado da soja.

    Flooding leads to hypoxia, a stress to which symbiotic N2 fixation is especially sensitive. The response of fully nodulated soybean plants to a 21-day period of flooding was studied by measurements of growth parameters and xylem transport of organic nitrogenous components to the shoot, in the presence and absence of NO3- and NH4+ in the medium. Flooding was found to seriously impair N2 fixation, irrespective of the N source, as indicated by strongly reduced xylem ureide levels. In the absence of a source of N, growth was strongly reduced during flooding while accumulation of N in the shoot was virtually abolished. Flooding in the presence of 5 mM NO3- or NH4+ led to the accumulation of total N in the shoot but only NO3- promoted increases in total dry matter, plant height and leaf area above that found in the absence of N. The accumulation of N, however, was lower than that of the non-flooded control for both NO3- and NH4+. The increases in total dry matter, plant height and leaf area with NO3- was as high as those of the non-flooded control. These data clearly show the beneficial effects of NO3- during a prolonged period of flooding of the nodulated root system of soybean.
  • Amadurecimento de goiabas 'Pedro Sato': estudo sobre sua natureza climatérica ou não climatérica Research Articles

    A goiaba (Psidium guajava L.) é um fruto tropical que apresenta rápido amadurecimento após a colheita. Entretanto, as bases fisiológicas envolvidas no seu processo de amadurecimento ainda não estão totalmente claras, o que torna difícil o desenvolvimento de técnicas que prolonguem o armazenamento. Realizaram-se dois experimentos com o objetivo de determinar o comportamento do amadurecimento de goiabas 'Pedro Sato'. No primeiro, goiabas em três estádios de maturação (I - verde-escuro, II - verde-claro e III - verde-amarelado) foram armazenadas à temperatura ambiente (23 ± 1°C e 85 ± 5 % UR), avaliando-se taxa respiratória, a produção de etileno, a cor da casca e da polpa e a firmeza. No segundo experimento, aplicaram-se etileno e 1-metilciclopropeno (1-MCP) nas goiabas no estádio verde-claro e estudou-se o comportamento do amadurecimento durante armazenamento à temperatura ambiente. Frutos de todos os estádios de maturação mostraram aumento gradual na taxa respiratória e produção do etileno. As mudanças intensas na cor da casca e da polpa e da firmeza precederam a máxima taxa respiratória e produção de etileno. 1-MCP retardou o amadurecimento, enquanto a aplicação de etileno não promoveu esse processo. Tais resultados não permitem a classificação da goiaba 'Pedro Sato' como um tradicional fruto climatérico.

    Guava (Psidium guajava L.) is a tropical fruit exhibiting rapid post-harvest ripening. However, the physiological basis involved in the ripening process of guava is not totally clear, which makes it difficult to develop technologies to enhance fruit storability. Two experiments were carried out with the objective of determining the ripening behavior of 'Pedro Sato' guavas. In the first experiment, guava fruits at three maturity stages (I - dark green, II - light green and III - yellow-green) were stored at room temperature (23 ± 1°C and 85 ± 5 % RH). The respiratory rate, ethylene production, pulp and skin colours, and firmness were evaluated. In the second experiment, ethylene and 1-methylcyclopropene (1-MCP) were applied to guavas at the light green maturity stage and the ripening behaviour during storage at room temperature was studied. Fruits from all maturity stages showed a gradual increase in the respiratory rate and ethylene production. The intense changes in pulp and skin colours and in firmness preceded the maximum respiratory rate and ethylene production. 1-MCP reduced the rate of ripening, while the application of ethylene did not promote this process. These results do not permit the classification of 'Pedro Sato' guava as a traditional climacteric fruit.

    Neste trabalho, examinamos a hipótese de que a técnica de fluorescência da clorofila a possa ser uma ferramenta eficiente para dar suporte à seleção de espécies adaptadas à alta irradiância e, entretanto, aptas a serem utilizadas em plantios de recuperação de áreas degradadas. Realiizou-se o experimento na Base de Operações Geólogo Pedro de Moura, Amazonas, Brasil. Os transientes de fluorescência foram obtidos utilizando-se um fluorômetro portátil, em folhas de Bombacopsis macrocalyx (Bm), Eugenia cumini (Ec), Iryanthera macrophyla (Im) e Senna reticulata (Sr) submetidas a alta irradiância, no período entre 12 e 13h. A partir do teste OJIP, calcularam-se: o índice de desempenho (PI ABS), a densidade de centros de reação por seção transversal (RC/CS), a eficiência máxima do fotossistema II (fiPo) e a probabilidade da energia de excitação (psio) ou de um fóton absorvido (fiEo) mover um elétron após a quinona A. Observou-se maior valor de PI ABS em Ec (0,40), e, os menores, em Bm (0,08) e Im (0,06). Esses baixos valores resultaram dos baixos valores de RC/CS (121 para Bm; 142 para Im) e fiPo (0,50 para Bm; 0,48 para Im) quando comparados com os exibidos por Ec (RC/CS = 303; fiPo = 0,72) e Sr (RC/CS = 326; fiPo = 0,73). Observou-se, ainda, que em Sr, a diminuição de seu PI ABS quando comparada com Ec, resultou do baixo valor de psio, no qual Sr apresentou 32 % menor que Ec. Quanto ao PI ABS por planta, Ec revelou o maior número de indivíduos com alto desempenho, e Im e Bm, o maior número de indivíduos com baixo desempenho. Conclui-se, portanto, que, por meio da técnica da fluorescência da clorofila a, foi possível diagnosticar o desempenho fotoquímico das plantas no campo, selecionando as espécies mais aptas à recuperação de áreas degradadas.

    We examine the hypothesis that the chlorophyll a fluorescence technique can be an efficient tool to support the selection of species adapted to high irradiance and therefore, suitable for use in initial rehabilitation projects of degraded areas. The experiment was conducted at the Pedro de Moura Operational Base, Amazonas, Brazil. The fluorescence transients were obtained using a portable fluorometer for leaves of Bombacopsis macrocalyx (Bm), Eugenia cumini (Ec), Iryanthera macrophyla (Im) and Senna reticulata (Sr), which were subjected to high irradiance between 12:00 am and 1:00 p.m. Using the OJIP test, the following were calculated: performance index (PI ABS), density of reaction centres per cross section (RC/CS), maximum efficiency of photosystem II (phiPo), and probability of energy excitation (psio) or that an absorbed photon (phiEo) moves an electron beyond quinone A. The highest value of PI ABS was found in Ec (0.40) while the lowest values were found in Bm (0.08) and Im (0.06). These low values of PI ABS in Bm and Im were a result of the low values for RC/CS (121 for Bm; 142 for Im) and phiPo (0.50 for Bm; 0.48 for Im) when compared to the values in Ec (RC/CS=303; phiPo=0.72) and Sr (RC/CS=326; phiPo=0.73). It was also observed that in Sr the decrease in PI ABS when compared with Ec resulted from a low value of psio, which was 32 % lower than the value found in Ec. Regarding the PI ABS per plant, Ec had the largest number of individuals with high performance while Im and Bm had the largest numbers of individuals with low performance. In conclusion, the chlorophyll a fluorescence technique enabled the evaluation of the photochemical performance of plants in the field, in turn permitting the selection of species most suitable for rehabilitation of degraded areas.
  • Isolamento e caracterização de novas lectinas de sementes de Canavalia ensiformis DC e Dioclea grandiflora Mart. ex Benth. Research Articles

    Isolaram-se lectinas de sementes de Canavalia ensiformis e Dioclea grandiflora. Coluna de filtração em gel produziu uma fração correspondente Con A ou lectina de D. grandiflora, enquanto ensaios de eritroaglutinação revelaram uma fração distinta, com uma lectina que aglutinou glóbulos vermelhos humanos, mas não de coelho. Cromatografia de interação hidrofóbica mostrou que a fração contendo a última lectina rendeu uma proteína que prontamente aglutinou eritrócitos humanos. Essa lectina também foi purificada por cromatografia de afinidade em coluna Lac-Sepharose e mostrou propriedades similares àquela purificada da coluna de interação hidrofóbica. Apesar de pequenas diferenças (conteúdo de carboidratos ou A1%1cm), ambas lectinas mostraram propriedades moleculares semelhantes, consistindo em dois monômeros não-covalentemente ligados, com Mr de 29-30 kDa, e com valores de pI que as indicavam como proteínas pouco acídicas. As lectinas de C. ensiformis (CEL-II) e D. grandiflora (DGL-II) reconheceram especificamente o grupo sanguíneo "H-type 2" (alfa-L-Fuc (1-2)-beta-D-Gal (1-4)-beta-D-GlcNAc-O-R), com ligação mais fraca com os grupos "H-type 1, H-type 3, H-type 4, Leª ou Le y. Inibição de eritroaglutinação por carboidratos mostrou que açúcares simples foram fracamente reconhecidos pelas lectinas. A região N-terminal apresentou um seqüência única observada apenas em algumas lectinas de Diocleinae (denominadas tipo II). Os resultados confirmam a existência de um segundo tipo distinto de lectina, filogeneticamente próxima a espécies de Diocleinae. Os dados indicam uma similaridade funcional entre as lectinas desse tipo, que possuem características que as distinguem das clássicas lectinas Man/Glc.

    Two lectins were isolated from Canavalia ensiformis and Dioclea grandiflora seeds. Gel filtration produced a fraction corresponding to Con A or D. grandiflora lectin while erythroagglutination assays revealed a distinct fraction presenting a lectin that agglutinates human red blood cells (RBCs) but not rabbit RBCs. Hydrophobic interaction chromatography showed that the latter fraction yielded a protein that readily agglutinates human erythrocytes; the lectin was also purified by affinity chromatography on Lac-Sepharose showing similar properties to that of the Phenyl-Sepharose-purified lectin. Despite minor differences (carbohydrate content or A1%1cm), the two lectins showed similar molecular properties in that they consisted of two non-covalently linked monomers having a Mr of 29-30 kDa and their pI values indicated that both lectins were slightly acidic proteins. The C. ensiformis lectin (CEL-II) and D. grandiflora lectin (DGL-II) specifically recognised the H-type 2 blood group (alpha-L-Fuc (1-2)-beta-D-Gal (1-4)-beta-D-GlcNAc-O-R), while binding to H-type 1, H-type 3, H-type 4, Leª or Le y was weaker. Carbohydrate inhibition of erythroagglutination showed that simple sugars were weakly recognised by the lectins, if at all. The N-terminal region presented a unique sequence hitherto found only in some Diocleinae lectins (designated type II). The overall results confirmed the existence of a second distinct lectin type, phylogenetically close to Diocleinae species. The data indicate a functional similarity among lectins of this type which possesses distinctive characteristics differentiating them from "classical" Man/Glc lectins.

    Essa espécie arbórea da Amazônia, Aniba rosaeodora Ducke, pertence à família das Lauraceae. Em vista da intensa e desordenada exploração, visando à extração de óleos essenciais (linalol), o pau-rosa figura na lista de espécies ameaçadas de extinção. Poucos estudos ecofisiológicos têm sido feito no sentido de subsidiar futuros programas de plantios florestais. Dessa forma, pesquisou-se o efeito de diferentes irradiâncias sobre as características fotossintéticas e de crescimento em plantas de A. rosaeodora, na fase juvenil. Plantas com nove meses foram submetidas a quatro tratamentos de luz (T1 = 10 a 250 µmol.m-2.s-1 / controle; T2 = 500 a 800, T3 = 700 a 1.000 e T4 = 1.300 a 1.800 µmol.m-2.s-1 / pleno sol). Analisaram-se variáveis alométricas, trocas gasosas, teores de pigmentos cloroplastídicos e a fluorescência da clorofila a. Quanto ao crescimento relativo, observou-se que plantas de A. rosaeodora exibiram maior acúmulo de biomassa quando crescidas sob condições intermediárias de irradiância (T2). O melhor desempenho fotossintético foi induzido por T3. Quando se correlacionou crescimento com fotossíntese, percebeu-se que as plantas submetidas aos tratamentos T2 e T3 exibiram melhores respostas em comparação com os extremos de menor (T1) e maior irradiâncias (T4). Os maiores teores de pigmentos cloroplastídicos foram obtidos nas plantas sob sombra (T1) e os menores teores encontrados naquelas expostas a pleno sol (T4). Quanto à eficiência fotoquímica do fotossistema II (Fv/Fm) observou-se que apenas as plantas do tratamento de sombra (T1) não apresentaram estresse por alta irradiância. Os resultados sugerem que ambos os tratamentos (T1 e T4) alteraram a funcionalidade das plantas de A. rosaeodora, inibindo a fotossíntese e o crescimento. A pleno sol, plantas de A. rosaeodora desenvolveram mecanismos de fotoproteção. No entanto, a espécie apresentou melhor resposta fotossintética e ganho de biomassa em condições intermediárias de irradiância, demonstrando relativa plasticidade fisiológica, durante o estádio juvenil.

    Aniba rosaeodora is an Amazonian tree species that belongs to the family Lauraceae. Due to intense exploitation for extraction of essential oils (mainly linalol), A. rosaeodora is now considered an endangered species. On the other hand, there is little information about its ecophysiology which would be useful to support future forest planting programs. Hence, the effect of different light intensities on the growth and photosynthetic characteristics of young plants of A. rosaeodora was studied. Nine-month-old plants were subjected to four light treatments (T1= 10 a 250 µmol.m-2.s-1 / control; T2=500 to 800, T3=700 to 1000 and T4=1300 to 1800 µmol.m-2.s-1 / full sunlight). Allometric variables, gas exchange, contents of pigments and chlorophyll a fluorescence were analysed. As to the relative growth rates, it was found that plants of A. rosaeodora showed higher biomass accumulation when grown under intermediary irradiance conditions (T2). The best photosynthetic performance was achieved under conditions of T3. When growth was correlated with photosynthesis, it was found that plants under treatments T2 and T3 presented better responses in comparison with the lowest (T1) and highest (T4) light extremes. The highest pigment contents were obtained for plants in the shade (T1) and the lowest for those exposed to full sunlight (T4). The photochemical efficiency of photosystem II (Fv/Fm) was found that only plants in the shade treatment (T1) presented no stress from high irradiance. These findings suggest that both treatments (T1 and T4) altered the function of the A. rosaeodora plants, inhibiting photosynthesis and growth. Plants of A. rosaeodora developed photo-protection mechanisms under full sunlight. However, the species presented better photosynthetic response and biomass gain under intermediary irradiance conditions, displaying relative physiological plasticity, during the seedling phase.
