The use of cellulose acetate dissolved in acetic acid to take stomata impressions

Summary
Various substances were tried for obtaining stomata impressions from leaves. The best results were obtained with cellulose acetate dissolved in acetic acid.
The solution was prepared by dissolving 1.2 gra (about 20 cm2 of a 0.15 mm thick commercial sheet) of cellulose acetate in 8 ml of glacial acetic acid. The viscous fluid thus obtained was applied to the leaf surface and after drying produced a thin film that showed the stomata impressions. These films can be stored indefinitely and be mounted in water for microscopical examination.
This method has been used with excellent results for counting stomata of Coffca, Triticum, Sccale, Cinchona, Mamhot, Hevea, and other species.
