Resumo Adenium obesum (Forssk.) Roem.

Abstract Adenium obesum (Forssk.) Roem. & Schult., widely known as desert rose, has attracted interest for its esthetic characteristics, which are influenced by the process of seedling production. Using two consecutive experiments installed fully at random, the study aimed at assessing the use of different substrates and hydrogel to produce desert rose seedlings in a protected environment. First, eleven substrates were tested, prepared as follows: OrganoAmazon®; PuroHumus®; soil; rice husk in natura; carbonized rice husk, sawdust and cattle manure. Then, three substrates were tested (S1 - OrganoAmazon® + PuroHumus®; S2 - OrganoAmazon® + PuroHumus® + rice husk in natura and S3 - OrganoAmazon® + PuroHumus® + carbonized rice husk) associated to four hydrogel levels: 0 (daily irrigation), 1, 2 and 3 g L−1 (irrigated on alternate days). Growth variables and quality index of seedlings were assessed. Six substrates (all of them without addition of sawdust and where used in mix) were grouped as those the promoted appropriate growth of seedlings, with height, number of leaves, collar diameter, root length and aerial part biomass superior to 6.5 cm; 13; 12 mm; 7.5 cm and 0.45 g, respectively. The substrate composed of OrganoAmazon® + PuroHumus® (1:1 v/v), when irrigated daily, was the one that favored most the production of quality seedlings. The use of hydrogel in substrates with shifts of irrigation on alternate days did not favor the production of desert rose seedlings.

Abstract Petunia is a very important ornamental plant with a broad range of flower colour and size, and most of the cultivars grown are propagated through seeds. Cytoplasmic male sterility (CMS) is a maternally inherited character determined by mitochondrial genes that results in impaired pollen development. The unique and well characterized male sterile cytoplasm in Petunia is a valuable resource for hybrids production because it prevents self-fertilization of mother plants and ensures the purity of F1s. Introgression of the male sterile cytoplasm in elite lines of Petunia is achieved following a backcross scheme and can be assisted using molecular markers associated to the trait of interest. The objective of this study was to develop a molecular marker to identify the male sterile cytoplasm of Petunia. A PCR-based marker amplifying a region of the mitochondrial CMS-associated urfS only in the male sterile plants was designed. Results showed differential PCR amplification of a ≈ 600 bp product in plants carrying male sterile cytoplasm in four Petunia species and their F1s and BC1 generations. A multiplex PCR reaction was subsequently set up, adding specific primers amplifying a ≈ 800 bp product from the conserved region trnT-trn-L of the chloroplast genome as a positive control in order to unambiguously identify the cytoplasm types as normal or sterile. A rapid, simple and precise molecular marker is now available for assisting breeding of F1 hybrids in Petunia.
(Rosa spp.) Scientific Article

Munar, Edwin Andres Villagran; Aldana, Carlos Ricardo Bojaca

Abstract In passive plastic greenhouses that do not have heating systems, the phenomenon of thermal inversion occurs, which means that the temperature inside the greenhouse is lower than the outdoor environment; therefore, the plants are exposed to thermal conditions that are unsuitable for production. The aim of this work was to evaluate passive air climatization alternatives seeking to optimize the nocturnal microclimate of a multi-tunnel Gothic greenhouse projected for rose production in Bogota savannah. The experimental approach included the use of a CFD-2D simulation model successfully validated by recording and archive of temperature data in a greenhouse. A total of 24 steady-state simulations were developed that included 3 nighttime temperature values, 2 sky conditions and 4 greenhouse configurations. The results showed that under the clear sky condition there was a thermal inversion with a value of −0.53 and −1.11 °C for the standard night-time greenhouse configuration. The installation of a thermal screen and porous mesh in the fixed roof vents allowed to control the presence of thermal inversion under clear sky conditions and optimizing the indoor temperature value in the greenhouse under cloudy sky conditions.

Abstract Propagation and seedling growth characteristics of five plant species: Duranta repens (Yellow bush), Hamelia patens (Fire bush), Ficus retusa (Yellow ficus), Buxus sempervirens (Boxwood / West indies) and Acalypha wilkesiana (Copper leaf), were examined in the nursery as indices of suitability for use as hedge plants in the landscape. The experiment was laid out in a Completely Randomized Design (CRD) with four replicates where the treatments were the plant species. Cuttings were cultivated in polyethylene pots filled with top soil medium. Data collected included: time to rooting, rooting percentage, number of leaves/plant, seedling height and stem girth. They were subjected to analysis of variance and means separated using Least Significant Difference (LSD) at 5% probability level. It took Acalypha wilkesiana, Duranta repens and Hamelia patens species shortest time (21 days) to root while Buxus sempervirens and Ficus retusa took the longest time (28 days) for rooting. Duranta repens species produced the highest rooting percentage (80.5%) at 28 days after planting and the highest number of leaves/plant (15leaves). This study revealed Duranta repens as the easiest to propagate among these species. All plant species evaluated except Ficus retusa are easily propagated in the nursery thus indicating their potential suitability for commercial production in the nursery and use as hedge plants. Duranta repens and Buxus sempervirens are recommended as hedge plant choices based on their ease of propagation and early establishment.

Abstract Integrated production systems have been used with various crops, and their use in floriculture is innovative. The effects of green fertilization in floriculture and the appropriate fertilization levels are still unknown. The aim was to identify the best dose of chemical fertilizer, with or without green fertilization, for integrated production of ‘Carola’ roses. The treatments consisted of 4 doses of the chemical fertilization recommended for rose bushes, (25%, 50%, 75%, and 100%), with or without green fertilization (calopo). Plants that were not treated with 100% (or complete) of chemical fertilization were supplemented monthly with Bokashi (16 g/plant, via the soil) and biofertilizer (5% via the leaves). The assessments were conducted 3 times per week for a year. The use of less chemical fertilizer did not affect rose production or quality, whereas the use of green fertilization did not provide a satisfactory outcome. The analyses, biometric, accumulation and nutrient content, and chemical characteristics of the soil, indicated that green fertilization with calopo was not beneficial. Moreover, with the exception of nitrogen and magnesium, there is the possibility of using 75% of the recommended chemical fertilization in rose bushes.

Abstract Zamioculcas zamiifolia (Araceae) is one of the most widely grown exotic species in Brazil as ornamental plants and in landscape design. Despite tolerating transport and being well adapted to low-light environments, this ornamental is attacked by different pathogens. Thus, the aim was to detect and identify the pathogen that causes stem rot in commercial Z. zamiifolia crops. Z. zamiifolia plants exhibiting stem rot symptoms were sent for phytosanitary diagnosis. In a culture medium, the fungal isolate obtained (SR-001) displayed the following morphological characteristics: cotton-like aerial mycelium, septate hyaline hyphae with no spore production, and the formation of small brown spherical sclerotia. To confirm pathogenicity, Z. zamiifolia plants were inoculated with the SR-001 isolate and, after fifteen days, the fungus was re-isolated when the same rot symptoms emerged. The SR-001 isolate was identified as Sclerotium rolfsii and its representative sequence was deposited in GenBank (Access MG694322). This fungal isolate has not been associated with diseases in Z. zamiifolia in Brazil, and this is the first report of the fungus infecting this ornamental plant species in a cultivated area.

Bezerra, Gabrielle Alves; Gabriel, Afra Vital Matos Dias; Mariano, Eduardo D.; Cardoso, Jean Carlos

Abstract Sustainable production of orchids may be achieved with the use of symbiotic microorganisms that improve resistance of plants to pests and diseases and reduce the need of substrate fertilization. In this study, one fungus (F1), one yeast (Y1) and one gram-positive bacteria (B+) were isolated from the roots of the orchid Oncidium varicosum and used in the cultivation of protocorms and plantlets obtained from seeds of this species. Cultivation of protocorms in Murashige and Skoog (MS) medium enriched with sucrose and inoculated with microorganisms did not result in better development of protocorms. However, the use of plantlets in the rooting phase in MS medium without sucrose and inoculated with B+ resulted in plantlets with more leaves and roots and longer shoots and roots, suggesting synergism between microorganism and plant when sucrose is absent. The major benefits of co-cultivating plantlets and these microorganisms were observed in the acclimatization in the greenhouse. Plantlets obtained with oat meal agar medium and B+ showed the highest pseudobulb diameter, number of roots, height and fresh weight among treatments. We concluded that the application of rooting-derived microorganisms in the in vitro culture and plantlet acclimatization of Oncidium varicosum is beneficial.

    Resumo em Português:

Resumo A zínia (Zinnia elegans Jacq.)

Abstract P. grandiflora is a known ornamental plant with abundant flowering. The flowers exhibit varied coloration with distinct forms and simple folded petals and/or multiple. The objective of this work was to induce regeneration via organogenesis and in vitro flowering of P. grandiflora. Nodal segments of seedlings germinated in vitro were used as explant source for regeneration. Kinetin (KIN) and 6-Benzylaminopurine (BA) were used for the induction of organogenesis. The treatments supplemented with 1.0 and 1.5 mg L−1 BA induced the highest number of adventitious shoots with an average number of 7.0 (±1.55) e 5.4 (±0.83), respectively. The microcuttings obtained from regenerated shoots produced floral buds. The floral buds were located in the axillary and terminal regions of the microcuttings and developed in approximately 10 days of cultivation until the anthesis. The highest number of flower buds was observed in the presence of 0.75 mg L−1 of gibberellic acid. This study opens new perspectives for the establishment of biotechnological tools to be applied for this important ornamental species.
  • The application of different reproduction techniques for rare species waterlily tulip (Tulipa kaufmanniana Regel.) propagation under ex situ conditions Scientific Article

    Resumo em Inglês:

Abstract T. kaufmanniana is a threatened and endemic species of the Tien Shan Mountains with a complex of valuable ornamental features. However, the commercial usage of the plant is limited due to the restricted availability of the bulbs in nature, difficulties in overcoming seed dormancy and low efficiency of the species reproduction. Results of the investigation of the reproductive biology of this species in the culture conditions allowed us to characterize T. kaufmanniana as viable with successful seedage. In the work reported here the effect of low temperature on the proper development of embryos is addressed, and an attempt is made to clarify the effect of cytokinins (TDZ, BAP) and auxin (NAA) on the regeneration capacity of isolated T. kaufmanniana embryos for the adventitious shoots and bulblets formation. Direct shoot organogenesis was induced by the combined action of chilling and BAP or TDZ treatment. Among the variants tested, TDZ promoted the formation of adventitious bulbs in 48 % of the chilled embryos, compared to non-chilled embryos (0.0%) and embryos exposed to the same concentration of BAP (39%). The best culture condition for T. kaufmanniana embryos growth and bulblets formation consisted of chilling for 10 wk in the dark at 4 °C, 4 wk at 20 °C under 16 h light photoperiod and 10 wk at 4 °C in the dark. Sufficiently effective protocol for clonal micropropagation was developed for the first time for T. kaufmanniana by the use of immature zygotic embryos as primary explants.
  • Establishment and traffic stress response of tall fescue as affected by Mycorrhiza fungi and Trinexapac-ethyl Scientific Article

    Jabbari, Alireza; Rohollahi, Iman

    Resumo em Inglês:

Abstract Heavy traffic regularly causes damage of substantial aboveground parts to turf grasses. Several factors could affect the traffic stress severity, including the number of occasions per season and the soil moisture content during the traffic stress. Mycorrhiza results in numerous advantages to host plant like improving the turfgrass establishment, root structure and also abiotic stress tolerance of plants. Festuca arundinacea remains green during the playing season and it is the most popular turf grass for football. Trinexapac-ethyl (TE) might be influence the wear as growth retardant. Accordingly, mycorrhiza infection effect on tall fescue establishment, and TE effect on traffic stress after establishment were studied. The treatments, including traffic stress (traffic and non-traffic stress), TE (0 and 200 g 100 m−2) and mycorrhiza (Control; Glomus mosseae and Glomus intraradices) 50 g m−2 were evaluated on two cultivars of tall fescue in two growing season of 2017 and 2018. Both mycorrhizas were significantly colonized on turfgrass root during establishment. G. intraradices inoculation increased the establishment by passing 60 days from seeding in F. arundinacea cv. Barvado. After establishment, traffic stress reduced the relative water content (RWC) in 2017 and 2018, and also reduced the turf quality and soluble sugar content (SSC). TE improved the RWC, SSC, turf quality and also colonization percentage. Mycorrhiza inoculation enhanced tall fescue establishment. TE application at 200 g 100 m−2 had no influence on traffic stress, however, it improved turf quality, RWC, SSC and mycorrhiza colonization.
