    Hydrogen peroxide bleaching of sodium alginate from seaweeds oh the Sargassum genus was studied. The influence of H2O2 concentration (percentage of H2O2 on a dry weight alginate basis, w/w) and NaOH/H2O2 ratio (% NaOH/% H2O2, both referred to a dry weight alginate basis, w/w) on the molecular weight, color removal and content of Fe3+ ions of bleached alginate samples was investigated by UV and IR spectroscopies, colorimetric determination of Fe3+ ions and vapor pressure osmometry. Higher yield, purity and molecular weight of alginate were obtained using 3% (or less) of hydrogen peroxide and a NaOH/H2O2 ratio of 1.2 for bleaching.
  • Biossensor enzimático para detecção de fungicidas ditiocarbamatos: estudo cinético da enzima aldeído desidrogenase e otimização do biossensor Artigo

    Initially, all major factors that affect the rate of the AldH-catalyzed reaction (enzyme concentration, substrate concentration, temperature and pH) were investigated. Optimal activity was observed between pH values of 7.5 and 9.5 in the temperature range of 25 to 50 ºC. Kinetic parameters, such as Km (2.92 µmol L-1) and Vmax (1.33 10-2 µmol min-1) demonstrate a strong enzyme-substrate affinity. The sensors were based on screen-printed electrodes modified with the Meldola Blue-Reinecke salt (MBRS) combination. Operational conditions (NAD+ and substrate contents, enzyme loading and response time) were optimized. Also, two enzyme immobilization procedures were tested: entrapment in poly(vinyl alcohol) bearing styrylpyridinium groups (PVA-SbQ) and crosslinking with glutaraldehyde. Chronoamperometry was employed to observe the biosensor responses during enzymatic hydrolysis of propionaldehyde and also to construct inhibition curves with maneb and zineb fungicides. Best results were found with the following conditions: [NAD+] = 0.25 mmol L-1; [propionaldehyde] = 80 µmol L-1; enzyme loading = 0.8 U per electrode; response time = 10 min, and inhibition time = 10 min. Current intensities around 103 ± 13 nA with the sensors and good stability was obtained for both immobilization procedures. Detection limits, calculated using 10% inhibition were 31.5 µg L-1 and 35 µg L-1 for maneb and zineb, respectively. Results obtained with other MBRS-modified electrodes consisting of mono and bi-enzymic sensors were compared. The ability to catalyze NADH oxidation by MB was also highlighted.
  • Desenvolvimento e validação de um método analítico para a determinação de histamina em vinhos utilizando cromatografia líquida de alta eficiência com detecção por fluorescência Artigo

    A rapid and efficient method for the analysis of histamine in wines using HPLC with fluorescence detection after derivatization was developed and validated. The method LOD and LOQ values were 0.25 and 0.50 mg L-1 respectively. The repeatability and intermediary precision for the instrument and for the method presented RSD values of 3.7 and 2.9%, and 6.0 and 5.6%, respectively. The recoveries were 95.5 and 89.9% for the fortification levels of 2 and 10 mg L-1. The method was applied to determine the histamine content in Cabernet Sauvignon wines, which presented values between 1.2 and 5.7 mg L-1.

    The study of fossils has made considerable progress over the last years as a result of the use of new experimental techniques. This paper describes the chemical composition of a fossilized fish of the Cretaceous period, from a 100 million-year-old, material originated from the Araripe Basin (northeastern Brazil). The chemical composition of the fossilized fish was analyzed by means of X-ray powder diffraction and Fourier transform infrared spectroscopy (FT-IR). The spectroscopic study has proven that the main substances found in the fossilized fish are CaCO3 and Ca5(PO4)3(OH). A tentative mechanism to explain the fossilization process is also given.

    The purpose of this work is to study theoretically stereoelectronic aspects of the interaction between heme and artemisinin in the transitional heme-artemisinin complex. Through semi-empirical calculations using the PM3 method, the potential energy barrier of artemisinin rotation relative to heme in the heme-artemisinin complex was studied in vacuum and in the partially solvated state. The minimum heat of formation obtained for the complex with and without water molecules is -702.39 and -100.86 kcal mol-1, respectively, which corresponds to the dihedral angle C-Fe-O1-O2 of 43.93º and 51.90º around the iron-oxygen O1 bond, respectively. The water molecules bind to heme via 13 hydrogen bonds and O-H•••O and 6 C-H•••O interactions, which accounts for -67.23 kcal mol-1. It is observed that the inclusion of water molecules does not affect significantly the stability of the heme-artemisinin complex.

    A proficiency assay of the determination of dithiocarbamate pesticide residues in banana was carried out. Fourteen laboratories participated in this study. Homogeneity and stability testing were performed by INCQS on the samples sent to the laboratories. Analytical results supplied by the pesticide residues laboratory of the VWA/KvW, Amsterdam, Holland, were used to define the designated value for the thiram concentration in the study samples. RESULTS: Fifty percent of the participating laboratories had satisfactory results. Efforts are needed to improve the precision of the analytical results and to decrease the number of false negative results observed.
  • Vivianite and siderite in lateritic iron crust: an example of bioreduction

    Lateritic iron crust (LIC) samples from Padauari (AM) were analysed by XRD, optical microscopy and SEM-EDS. The equilibrium of iron minerals (IM) was studied using Eh-pH diagram. It was shown that the minerals of the LIC are goethite (alpha-FeOOH), vivianite [Fe3(PO4)2.8H2O] and siderite (FeCO3). Carbonate grains are a solid solution of FeCO3-MnCO3. The LIC presents textures and structures of dissolution of IM. The siderite and vivianite are stable from Eh =-0.3 to 0.0 V and pH=5.0-7.5. These results indicate that vivianite and siderite are products of bioreduction through biogenic dissolution of IM, the new conditions of ecosystems of the Amazon region.

    The chemical composition of the ethanolic extract of the leaves and roots from Philodendron imbe Schott was investigated. The main constituents isolated the leaves were beta-sitosterol one polyprenoid hexaprenol, and 6beta-hydroxy-stigmast-4-en-3-one, a steroid, not yet reported in the Philodendron gender. A mixture of constituents, namely, ethyl myristoleate, alpha-bisabolol, ethyl isopalmitate, 3-octadecenyl-phenol and the major component ethyl palmitate, were isolated from the roots. Structure elucidation of these secondary metabolites was accomplished by spectrometric analysis, including 1D and 2D NMR experiments such as ¹H, ¹H and ¹H,13C-COSY.
  • Estudo da síntese da zeólita ZSM-5 a partir de argilas naturais Artigo

    The synthesis of ZSM-5 zeolite was studied by three synthesis methods: standard synthesis, three day synthesis and synthesis employing clays (kaolin and montmorillonite). The raw materials and prepared materials were characterized by Chemical Analysis by X-Ray Fluorescence (XRF) and X-Ray Diffraction (XRD). The results show that it is possible to synthesized ZSM-5 zeolite using clays as raw materials. Kaolin gives phases of higher crystalinity than montmorillonite.
  • Chemical composition of umbu (Spondias tuberosa Arr. Cam) seeds

    The umbu tree (Spondias tuberosa Arr. Cam) is an important fruit tree the economy of the semi-arid northeastern region of Brazil. With the objective of finding use for the seeds, physical and chemical characterizations of the seeds from 2 cultivars in 2 maturation stages were carried out and their fatty acid and mineral profiles determined. The results showed no differences between the seeds analyzed. The yield was about 10% and the dimensions as follows: length from 1.48 to 2.11 cm and width from 0.76 to 1.16 cm. The average lipid content was 55% of which 69% was unsaturated and the average protein content was 24%. The seeds were a good source of the following minerals: P, K, Mg, Fe and Cu. The overall results indicated that the oil or the seeds could be used for food stuffs if no toxic agents were found.

    Manipulation of the flux of substrates was utilized to control the amount of 3-hydroxyvalerate and 3-hydroxy-4-pentenoate produced by Burkohlderia sp. The 3HV production rate was directly proportional to the propionic acid uptake rate with the last one completely directed to 3HV biosynthesis. The 3HPE production rate was inversely proportional to the sucrose uptake rate probably due to operation of the glyoxylate cycle.

    In an attempt to reduce toxicity and widen the spectrum of activity of cisplatin and its analogues, much attention has been focused on designing new platinum complexes. This work reports the synthesis and characterization of novel compounds of the platinum (II) and platinum (IV) containing 2-furoic hydrazide acid and iodide as ligands. Although the prepared compounds do not present the classical structure of biologically active platinum analogues, they could be potentially active or useful as precursors to prepare antitumor platinum complexes. The reported compounds were characterized by ¹H NMR, 13C NMR, 195Pt NMR, IR and elemental analyses.

    In this work commercial filters papers were organomodified with tetraethylorthosilicate (TEOS) and 3-aminopropyltriethoxysilane (3-APTS), aiming at the development of a new analytical procedure for in-situ speciation of labile and inert metal species in aquatic systems. Parameters that exert influence on the metal lability such as pH, chelating time, concentration and characteristics of the organic matter were studied in the laboratory using tests for metal recuperation. The results showed slower kinetics for Cu ion than for Ni, Mn and Cd in the absence of aquatic humic substances (AHS). The relative lability observed for complexed metals in aquatic humic substances using organomodified filter papers was Cu>>Cd>Ni>Mn. The pH values, structural characteristics and concentration of AHS exert strong influence on the lability of the metals. The results obtained showed that the utilization of organomodified filter papers can be an interesting and promising alternative for in situ characterization of metal lability in aquatic systems.

    Iron, copper and lead distribution was evaluated in sediment cores from a disturbed mangrove area in Guanabara Bay: a core from a seaward site where mangrove vegetation was removed ~20 yr before sampling (MD); a core from an intermediate site with dead vegetation, apparently due to insect attack (MP), and a core from a landward site with living vegetation (MV). Metal concentrations showed increasing values seaward while organic matter content showed an inverse trend, displaying a negative correlation with metals. This unusual correlation indicates opposite sources, since metals come from the bay and the main OM origin is probably degraded mangrove vegetation. Plant cover loss seems to be a critical factor affecting metal accumulation, particularly due to changes in OM input.

    In this work, aqueous suspensions of aluminas with different particle sizes were evaluated. The effect of pH on the electrosteric stabilization using PMAA-NH4 (ammonium polymethacrylate) as deflocculant was studied. The amount of deflocculant was optimized and rheologic properties were determined at four different pH values. Sedimentation was also evaluated. For suspensions with pH 4, an electrostatic mechanism of stabilization was observed, probably due to a flat adsorption of PMMA- on the alumina surface, leading to a small efficiency in relation to steric stabilization. For a suspension with pH 12, the steric mechanism of stabilization prevails. Suspensions with pH 7 and 9 present a higher flocculation degree. In relation to particle size, A-1000 samples present a smaller particle size, leading to a smaller interparticle distance (IPS), making stabilization more difficult.

    A simple method was proposed for determination of paracetamol and ibuprofen in tablets, based on UV measurements and partial least squares. The procedure was performed at pH 10.5, in the concentration ranges 3.00-15.00 µg ml-1 (paracetamol) and 2.40-12.00 µg ml-1 (ibuprofen). The model was able to predict paracetamol and ibuprofen in synthetic mixtures with root mean squares errors of prediction of 0.12 and 0.17 µg ml-1, respectively. Figures of merit (sensitivity, limit of detection and precision) were also estimated. The results achieved for the determination of these drugs in pharmaceutical formulations were in agreement with label claims and verified by HPLC.

    A method for HPLC determination of sulfadimethoxyne in milk is presented. The analyte isolation and concentration were performed by solid-phase extraction through a C-8 cartridge, pre-conditioned with hexane, methanol and water and eluted with MeOH. The recovery determination was done with a spiked solution of 20, 50 or 100 µg L-1. In this concentration range, the recovery was 83.2% with a RDS of 15.4%. For quantification, a Zorbax Eclipse XDB-C8 (4.6 mm x 150 mm, 5 µm), a mobile phase of MeCN: 0.01 mol L-1 KH2PO4 aq. (1:4), and a variable wavelength detector (275 nm) were used.

    The first days of radioactivity, the discoveries of X-rays, radioactivity, of alpha- and beta- particles and gamma- radiation, of new radioactive elements, of artificial radioactivity, the neutron and positron and nuclear fission are reviewed as well as several adverse historical marks, such as the Manhattan project and some nuclear and radiological accidents. Nuclear energy generation in Brazil and the world, as an alternative to minimize environmental problems, is discussed, as are the medicinal, industrial and food applications of ionizing radiation. The text leads the reader to reflect on the subject and to consider its various aspects with scientific and technological maturity.

    During the last decades several aporphinoid alkaloids of the Ocotea species have been isolated. This review describes the occurrence of the fifty four aporphinoids in seventeen different species of Ocotea: thirty nine (39) aporphine sensu stricto, four (4) oxoaporphine, five (5) 6a,7-dehydroaporphine, one (1) didehydroaporphine, one (1) C-3-O-aporphine, one (1) C-4-O-aporphine, two (2) phenanthrene, one (1) proaporphine and their 13C NMR spectral data.

    Gravimetric and Bailey-Andrew methods are tedious and provide inflated results. Spectrofotometry is adequate for caffeine analysis but is lengthy. Gas chromatography also is applied to the caffeine analysis but derivatization is needed. High performance liquid chromatography with ultraviolet detection (HPLC-UV) and reversed phase is simple and rapid for xanthine multianalysis. In HPLC-UV-gel permeation, organic solvents are not used. HPLC-mass spectrometry provides an unequivocal structural identification of xanthines. Capillary electrophoresis is fast and the solvent consumption is smaller than in HPLC. Chemometric methods offer an effective means for chemical data handling in multivariate analysis. Infrared spectroscopy alone or associated with chemometries could predict the caffeine content in a very accurate form. Electroanalytical methods are considered of low cost and easy application in caffeine analysis.

    This article surveys a selection of the most recent advances in aziridine synthesis. Novel synthetic methods and new insights into existing methodologies for the selective construction of the title compounds reported in the past decade are discussed in terms of synthetic applicability and environmentally benign conditions. Mechanisms involving stereoselective preparation of structurally diverse aziridines are also presented in order to highlight the most important issues associated with the synthesis of these versatile building blocks.

    During the last five decades, as a result of an interaction between natural product chemistry, synthetic organic chemistry, molecular biology and spectroscopy, scientists reached an extraordinary level of comprehension about the natural processes by which living organisms build up complex molecules. In this context, 13C nuclear magnetic resonance spectroscopy, allied with isotopic labeling, played a determinant role. Nowadays, the widespread use of modern NMR techniques allows an even more detailed picture of the biochemical steps by accurate manipulation of the atomic nuclei. This article focuses on the development of such techniques and their impact on biosynthetic studies.

    Atropisomerism is a special kind of stereoisomeric relationship that arises from the freezing of a certain conformation of an organic molecule, associated with a high rotational barrier about a single covalent bond. Atropisomerism has been originally described in orto-functionalyzed biphenyl derivatives, but a lot of other organic functionalities can present this structural phenomenon, characterized by the presence of chiral properties in compounds that don't present classical stereogenic centers. Atropisomeric compounds, intermediates and catalysts have well-know importance in organic synthesis, but the influence of the axial chirality in substances able to modulate biological systems is still not very exploited in drug design and development. In this context, the present account describes the importance of this structural property in the medicinal chemistry of different classes of bioactive compounds or therapeutic agents, emphasizing how atropisomerism could affect the molecular recognition of a ligand or a prototype by the target bioreceptor.

    REVIEW: Living organisms encountered in hostile environments that are characterized by extreme temperatures rely on novel molecular mechanisms to enhance the thermal stability of their proteins, nucleic acids, lipids and cell membranes. Proteins isolated from thermophilic organisms usually exhibit higher intrinsic thermal stabilities than their counterparts isolated from mesophilic organisms. Although the molecular basis of protein thermostability is only partially understood, structural studies have suggested that the factors that may contribute to enhance protein thermostability mainly include hydrophobic packing, enhanced secondary structure propensity, helix dipole stabilization, absence of residues sensitive to oxidation or deamination, and increased electrostatic interactions. Thermostable enzymes such as amylases, xylanases and pectinases isolated from thermophilic organisms are potentially of interest in the optimization of industrial processes due to their enhanced stability. In the present review, an attempt is made to delineate the structural factors that increase enzyme thermostability and to document the research results in the production of these enzymes.

    Special clays are a group of clays different from the large volume of clay mineral products named "Industrial Clays": kaolins, ball clays, refractory clays, bentonites, fuller's earths, common clays. Two groups of special clays exist: rare, as in the case of hectorite and sepiolite and restricted areas, as in the case of white bentonite, halloysite and palygorskite (attapulgite). A review is given of the most important producers of the special clays and their properties in the Western World, as well as a discussion of the occurrence of these types of clays in Brazil.

    Health problems related to UV radiation can be minimized by the appropriate use of sunscreens. Different kinds of sunscreens are reported in the literature, even though there is a misleading denomination among them and few discussions are presented about how they work. This paper describes some important aspects in order to understand sunscreen phenomena such as: solar radiation effect, type of solar filters, protection mechanism, formulations and solar protection factor (SPF). Moreover the importance of Chemistry and the interdisciplinary studies related to sunscreens and cosmetic researches are emphasized.

    This paper is a review of the history, synthesis and application of organophosphorus compounds, especially of those of pentavalent phosphorus, such as phosphoramidates, phosphorothioates, phosphonates and phosphonic acids with insecticide and anticancer activities. The organophosphorus compounds with agrochemical applications show great structural variety, They include not only insecticides, but also fungicides, herbicides, and others. The large variety of commercially available organophosphorus pesticides is remarkable. Even more interesting is the high efficiency of some organophosphorus compounds as anticancer agents such as cyclophosphamide and its derivatives.

    It is widely recognized that pharmacokinetic optimization needs to be addressed early in drug discovery to reduce the high failure rate in bringing drugs to market. Poor absorption, too short duration of action due to high elimination rate, or the presence of active metabolites are examples of properties that can potentially lead to unsuccessful clinical programmes. Here I describe a brief overview of advantages and molecular strategies for improving metabolic and pharmacokinetic properties applied to the discovery of fluconazol, beta-blockers, ritonavir and ezetimibe and to the development of the prodrugs enalapril and bambuterol.

    Crystalline structures of zeolites can be studied using different representations: the internal symmetry obtained by X-Ray or neutron diffraction crystallography techniques or a systematic analysis of the basic structural units which can be arranged to build the geometries of each kind of zeolite. In this work the basic concepts of three building units, SBU (Secondary Building Units), SSU (Structural SubUnits) and PBU (Periodic Building Units) are presented. The properties of the resulting crystalline structures are discussed (pores, cavities, channels), describing the influence of each one of these properties in processes of physical-chemical interest. Representative case studies of known zeolite crystalline structures are also discussed in terms of their space group classification.

    In this paper the current status of fuel cells is described with particular emphasis on high (T > 800 ºC) and intermediate (T < 800 ºC) temperature solid oxide fuel cells. Also the importance of the fuel cell technology is shown. Reviewed are the fundamental features, the basic principles, types of fuel cell, fabrication methods, cell configurations and the development of components (cathodes, anodes, electrolytes, interconnect) and materials.
  • Métodos emergentes para aumentar a eficiência do ozônio no tratamento de águas contaminadas Divulgação

    Many industrial processes produce effluents with a wide variety of xenobiotic organic pollutants, which cannot be efficiently degraded by conventional biological treatments. Thus, the development of new technologies to eliminate these refractory compounds in water has become very imperative in order to assure the quality of this important resource. Ozonation is a very promising process for the treatment of wastewaters containing non-easily removable organic compounds. The present work aims at highlighting new methods of enhancing the efficiency of ozone towards the removal organic pollutants in aqueous solution. Special attention is given to catalytic ozonation processes contemplating homo- and heterogeneous catalysis, their activity and mechanisms. Recent results and future prospects about the application of these processes to real effluents are also evaluated.
  • Antioxidantes na manutenção do equilíbrio redox cutâneo: uso e avaliação de sua eficácia Divulgação

    The skin is a very complex organ, continuously exposed to physical, chemical and microbiological agents. Enzymes as well as low-molecular weight antioxidants are present in the cutaneous tissue to counterbalance the deleterious effect caused by an oxidative stress and thus maintain homeostasis. Antioxidants such as vitamins C and E, carotenoids, and extracts with these properties have been extensively used for treatment of pathologies and skin aging prevention. We review here different mechanisms that can interfere in the redox equilibrium of the skin, as well as the chemical reactions involved in these processes. Moreover, we discuss the importance of endogenous or exogenous antioxidants that can be acquired from the diet or from oral or topical administration, and methodologies that have been developed to evaluate their efficacy.
  • Photomultiplier nonlinear response in time-domain laser-induced luminescence spectroscopy

    A new procedure to find the limiting range of the photomultiplier linear response of a low-cost, digital oscilloscope-based time-resolved laser-induced luminescence spectrometer (TRLS), is presented. A systematic investigation on the instrument response function with different signal input terminations, and the relationship between the luminescence intensity reaching the photomultiplier and the measured decay time are described. These investigations establish that setting the maximum intensity of the luminescence signal below 0.3V guarantees, for signal input terminations equal or higher than 99.7 ohm, a linear photomultiplier response.

    The validation of analytical methods was carried out for di-(ethylhexyl) phthalate (DEHP) and adipate (DEHA) the determination of in PVC films. The level of DEHP and DEHA in samples was determined by leaving the film in contact with n-heptane during 48 hours and analysis in a gas chromatograph (GC) equipped with a flame ionization detector and fused silica column with 5% phenylmethyl silicone in the dimensions 30 m x 0.53 mm x 2.65 mm. The results for detection and the quantification limits were smaller than the restriction limits. The recovery rates of DEHP and DEHA were, respectively, 69.10 and 75.30 %.

    The aim of this paper is to present a simple way of treating the general equation for acid-base titrations based on the concept of degree of dissociation, and to propose a new spreadsheet approach for simulating the titration of mixtures of polyprotic compounds. The general expression, without any approximation, is calculated a simple iteration method, making number manipulation easy and painless. The user-friendly spreadsheet was developed by using MS-Excel and Visual-Basic-for-Excel. Several graphs are drawn for helping visualizing the titration behavior. A Monte Carlo function for error simulation was also implemented. Two examples for titration of alkalinity and McIlvaine buffer are presented.
  • Preparation, spectroscopic and acidity properties of two hydrazones: an organic lab experiment

    An undergraduate organic lab experiment is described based on the preparation of two readily accessible hydrazones. The UV-visible spectra of these N-H acids and of their conjugate bases are employed to illustrate the importance of through-conjugation in determining their acid strength and their internal charge-transfer-band transitions.
  • De Svante Arrhenius ao peagâmetro digital: 100 anos de medida de acidez Assuntos Gerais

    This work describes the establishment of the concept of pH and the evolution of its measurement. The origin of the pH definition can be found in the development of the chemistry of aqueous solutions during the XIXth century. The electrolytic dissociation theory by Svante Arrhenius played a central role. After the proposal of the pH scale by Sörensen, many years were necessary for the acceptance of this new parameter among chemists in general. Its importance was first recognized in biochemistry and related areas. Twenty years after, its importance had been recognized in many industrial and laboratorial practices. The previous methods were based on colorimetric and electrometric methods, but both suffered from many problems. Acceptance of pH in Chemistry was only possible after the development of experimental trustable measurements. The invention of the pH meter was the primordial step.

    The ammonia synthesis from its elements plays an important role in the survival of humankind. A short historical development of discoveries of the nitrogen cycle, the nitrogen sources for agriculture and the ammonia synthesis (in laboratory and industry) are presented. Some brief aspects of nitrogen chemistry are described. Short biographies of Fritz Haber and Carl Bosch, the main persons in this epopee, are also presented.
