ABSTRACT The incidence and clinical characteristics of NTM diseases in Brazil remain relatively unknown. The present study describes the diagnosis of NTM isolates, the clinical presentation and treatment outcomes. We analyzed NTM isolates in patients of a tertiary hospital in the Southeast region of Brazil, from January 2008 to July 2019. The ATS/IDSA criteria for diagnosis and treatment of these patients was applied. Mycobacterium kansasii were identified in 13/113 (11.5%) patients. In 59/113 (52.2%) patients who met the ATS criteria for disease, 29/59 (49.1%) received treatment, and 22/29 (75.8%) were cured. The major species identified was M. kansasii. The most frequent symptoms among the treated patients were dyspnea and cough, and the proportion of cured patients was high.

Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group.

This work reports the use of the Octave, a free computational software, to optimize the experimental design in the synthesis of ethyl oleate from oleic acid and ethanol catalyzed by a native lipase from Aspergillus niger. The central composite factorial design (CCD) was performed with 2 levels and 3 variables (temperature, mass of native A. niger lipase and oleic acid:ethanol molar ratio) in conjunction with response surface methodology (RSM). The results indicate that variable 1 (oleic acid:ethanol molar ratio; X1) and 2 (mass of A. niger lipase, X2), as well as the interaction between them, are statistically significant. The optimal reaction condition was estimated using ANOVA data and the response surface. The experimental results are consistent with those estimated, and thus, ethyl oleate was obtained with conversion (> 90%) using oleic acid and ethanol in a 1:2 molar ratio, A. niger lipase (8,00 mg) and temperature of 45 °C in 6 hours of reaction.

Abstract Health professionals working to mitigate the COVID-19 pandemic are one of the main risk groups for the disease, being prioritized for vaccination. Considering this, the aim of this study was to analyze the immune response of these professionals immunized with CoronaVac in the first and second doses. Blood samples were collected after the first and second doses of the vaccine (CoronaVac) and used to investigate hematological and biochemical parameters, analysis of immunoglobulin production, cytokines, and gene expression profile, as well as the identification of subsets of immune cells. Post-first dose immunological phenotypic memory (CD27+) profiles (T CD4+, TCD8+ and CD19+) showed a significant increase, as did Monocyte APCs (CD80+HLA-DR+) in relation to the second dose. The cytokines IL-2, IL-6 and IFN-° showed increased values in relation to the other analyzed cytokines. The Th2/Th17 profile in the second dose was characterized by gene expression analysis. The production of IgM and IgG after vaccination showed statistically significant values in the comparison between doses. CoronaVac showed activation of APCs monocytes, memory response of T and B lymphocytes, with immunoglobulins production. This set of responses is characterized by the Th2/Th17 immunological profile.

Methodological differences can affect sequencing depth with a possible impact on the accuracy of genetic diagnosis

Abstract For a better interpretation of variants, evidence-based databases, such as ClinVar, compile data on the presumed relationships between variants and phenotypes. In this study, we aimed to analyze the pattern of sequencing depth in variants from whole-exome sequencing data in the 1000 Genomes project phase 3, focusing on the variants present in the ClinVar database that were predicted to affect protein-coding regions. We demonstrate that the distribution of the sequencing depth varies across different sequencing centers (pair-wise comparison, p < 0.001). Most importantly, we found that the distribution pattern of sequencing depth is specific to each facility, making it possible to correctly assign 96.9% of the samples to their sequencing center. Thus, indicating the presence of a systematic bias, related to the methods used in the different facilities, which generates significant variations in breadth and depth in whole-exome sequencing data in clinically relevant regions. Our results show that methodological differences, leading to significant heterogeneity in sequencing depth, may potentially influence the accuracy of genetic diagnosis. Furthermore, our findings highlight how it is still challenging to integrate results from different sequencing centers, which may also have an impact on genomic research.

RESUMO: A displasia coxofemoral (DCF) é afecção comum na clínica cirúrgica de pequenos animais. Entre as técnicas cirúrgicas para controle da DCF, a osteotomia pélvica tripla (OPT) e a cunha sacroilíaca (CSI), promovem rotação lateral acetabular no eixo axial (ventroversão), aumentando a cobertura acetabular e a estabilidade da articulação. Desta forma, o presente estudo objetivou avaliar radiograficamente, por meio da aferição do ângulo de Norberg (NA) e da porcentagem de cobertura acetabular (PCA), a ventroversão acetabular induzida pela técnica da cunha sacroilíaca, associada ou não às osteotomias pélvicas, além de verificar a exequibilidade das cunhas confeccionadas de poliamida com angulação de 20ο e 30ο. O software utilizado para aferir o AN e o PCA foi o AutoCAD® 2009. Dez pelves de cadáveres caninos foram avaliadas radiograficamente em quatro momentos: MO (Grupo Controle), M1 (cunhas de 20ο e 30ο), M2 e M3 (cunhas associadas à osteotomia bilateral do púbis e ísquio, respectivamente). Não houve aumento significativo da ventroversão em M1, M2 e M3. A técnica de cunha sacroilíaca de poliamida mostrou-se exequível, estável e de fácil aplicação. Não obstante, o software utilizado mostrou-se eficiente e de fácil utilização nas aferições do AN e PCA. Neste estudo, mesmo tratando-se de cadáveres de cães adultos e de maioria não displásicos, concluiu-se que a utilização da técnica de cunha sacroilíaca não necessita de associação à ostectomia púbica e a osteotomia do ísquio por não promoverem aumento significativo do AN e da PCA.
ABSTRACT: Canine hip dysplasia (CHD) is a common condition observed in the surgical clinics for small animals. Among the surgical techniques for management of CHD, triple pelvic osteotomy and sacroiliac wedge promote acetabular lateral axial rotation (ventroversion), increasing acetabular coverage and joint stability. The present study aimed to evaluate radiographically, by measuring the Norberg angle (NA) and the acetabular coverage percentage (ACP), the acetabular ventroversion induced by the sacroiliac wedge technique, with or without pelvic osteotomies; we also checked the feasibility of wedges made of polyamide with an angulation of 20° and 30°. The software used to measure NA and ACP was AutoCAD® 2009. Pelves from 10 canine corpses were evaluated radiographically at four time-points: M0 (Control Group), M1 (wedges of 20° and 30°), M2 and M3 (wedges associated with bilateral pubis and ischium osteotomies, respectively). There was no significant increase in the acetabular ventroversion at M1, M2, and M3. The polyamide sacroiliac wedge technique proved to be feasible, stable, and easy to apply. Further, the software proved to be efficient and easy to use for NA and ACP measurements. In the present study, even in the cases of non-dysplasic adult canine corpses, it was concluded that the sacroiliac wedge technique does not require to be accompanied by pubis and ischial osteotomies because they did not significantly increase the NA and ACP.

ABSTRACT The hydrodynamic behavior of an anaerobic fixed bed reactor (AFBR) was evaluated in the treatment of cattle slaughterhouse wastewater. The AFBR was operated at hydraulic retention time (HRT) of 14, 11 and 8 h. Stimulus-response assays were carried out with Eosin Y and the experimental data were adjusted to the single-parameter theoretical models of dispersion and N-continuous stirred tank reactors in series (N-CSTR). The experimental results of the residence time distribution curves showed that at lower flow rate, the reactor showed plug flow behavior with correlation coefficient (r) of 0.88 and number of dispersion of 0.2 for high dispersion (HD). However, at higher and intermediate flow rates, the AFBR behave as a complete mixture flow, (r) of 0.94 and 0.96, respectively. Residence time distribution curves in the AFBR showed a good approximation of the complete mixing model at hydraulic residence time of 11 and 8 h, with 5 and 2 N-CSTR reactors in series, respectively. The volume of dead zones corresponding to 43.0, 37.4 and 11.2% of the volume of the reactor for HRT of 14, 11 and 8 h, respectively, was noted, and hydraulic short circuiting were not confirmed.

Resultados: os trabalhos foram realizados em 427 municípios (99,5% dos 429 selecionados); foi registrada presença de moluscos em 300 (70,2%) municípios e a ocorrência de Biomphalaria glabrata em 62 (21%) municípios, B. straminea em 181 (60%), B. tenagophila em três (1%); associação de B. glabrata/B. straminea foi encontrada em 53 (18%), e de B. glabrata/B. tenagophila em um (0,3%). Conclusão: os registros de B. glabrata, B. straminea e B. tenagophila estão de acordo com a distribuição conhecida.

Results: the work was carried out in 427 municipalities (99.5% of the 429 selected); the presence of mollusks was registered in 300 (70.2%) municipalities; Biomphalaria glabrata were found in 62 (21%) municipalities, B. straminea in 181 (60%), B. tenagophila in three (1%); B. glabrata/B. straminea association was found in 53 municipalities (18%) and B. glabrata/B. tenagophila association in one (0.3%) municipality. Conclusion: B. glabrata, B. straminea and B. tenagophila distribution records obtained in this study are consistent with previously known distribution.

ABSTRACT: The objective of this research was to characterize the efficiency and convenience of sheep identification system and its implementation costs. Therefore we quantified the time for implementation, reading and data transfer to a management program in a manual system (earrings) and electronic system (subcutaneous transponders). Furthermore, it was evaluated the implementation costs of the traceability systems by analysis of the absorption costs and comparison between systems. It was observed that the implementation time for the manual system was shorter (2.2 s animal−1) than the electronic system (4.67 s animal−1). As for reading time, the electronic system presented shorter time (2.89 s animal−1) than the manual system (4.67 s animal−1). As for the data transfer time, the electronic system also presented shorter time (16 s) than manual system (6 min and 42 s). In terms of economic costs, the manual system presented lower implementation costs and lower values per tracked animal to ensure the economic viability. In conclusion, the producer must analyze the context around the production unit to choose the best traceability system.

Abstract The objective of this study was to evaluate the effects of Cymbopogon citratus essential oil and its main compound (citral) against primary dental colonizers and caries-related species. Chemical characterization of the essential oil was performed by gas chromatography/mass spectroscopy (GC/MS), and the main compound was determined. Antimicrobial activity was tested against Actinomyces naeslundii, Lactobacillus acidophilus, S. gordonii, S. mitis, S. mutans, S. sanguinis and S. sobrinus. Minimum inhibitory and bactericide concentrations were determined by broth microdilution assay for streptococci and lactobacilli reference, and for clinical strains. The effect of the essential oil on bacterial adhesion and biofilm formation/disruption was investigated. Negative (without treatment) and positive controls (chlorhexidine) were used. The effect of citral on preformed biofilm was also tested using the same methodology. Monospecies and microcosm biofilms were tested. ANOVA or Kruskal-Wallis tests were used (α=0.05). Cytotoxicity of the essential oil to human keratinocytes was performed by MTT assay. GC/MS demonstrated one major component (citral). The essential oil showed an inhibitory effect on all tested bacterial species, including S. mutans and L. acidophilus. Essential oil of C. citratus (10X MIC) reduced the number of viable cells of lactobacilli and streptococci biofilms (p < 0.05). The essential oil inhibited adhesion of caries-related polymicrobial biofilm to dental enamel (p < 0.01). Citral significantly reduced the number of viable cells of streptococci biofilm (p < 0.001). The essential oil showed low cytotoxicity to human keratinocytes. Based on these findings, this study can contribute to the development of new formulations for products like mouthwash, against dental biofilms.

O estudo verificou a relação das variáveis físicas e de percepção de qualidade de vida de estudantes com idades de 14 a 16 anos da cidade de Quixadá, Ceará, Brasil. A amostra foi constituída por 48 sujeitos (24 homens e 24 mulheres). O questionário WHOQOL-BREF foi utilizado para identificar a qualidade de vida e domínios dos participantes juntamente com as mensurações de Índice de Massa Corporal - IMC, Índice de Relação Cintura-Quadril - IRCQ, flexibilidade e aplicação do Questionário Internacional de Atividade Física - IPAQ. Testada a normalidades das variáveis, realizou-se comparação dos sujeitos em relação ao sexo por meio do Teste U de Mann-Whitney e temporalmente pelo teste de t pareado ou Wilcoxon. Na análise pré e pós em relação questões 1 e 2 percebe-se que a auto avaliação da qualidade vida passou de 8.3 para 12.5 no nível “muito boa”, no entanto, não se obteve diferença significativa. Em conclusão, as relações de domínio físico e de percepção de QV foram satisfatórios. Portanto, a QV deve ser estimulada a partir de um estilo de vida ativo juntamente com mudança de hábitos de vida.
The study investigated the relationship between physical variables and quality of life perception variables of students aged 14 to 16 years old from Quixadá city, Ceará, Brazil. Forty eight subjects (24 men and 24 women) were included in the sample. The WHOQOL-BREF questionnaire was used to identify the QoL domains of participants and along with BMI measurements, waist-to-hip ratio (WHR), flexibility and application of IPAQ. Data was analyzed by descriptive statistics (mean, standard deviation, coefficient of variation and median and interquartile range) and normality was tested. Mann-Whitney U test was used to compare subjects in relation to gender and t test or Wilcoxon was used to the group analysis in relation to time. In the pre and post analysis regarding questions 1 and 2 it can be seen that self-rated life quality increased from 8.3 to 12.5 at the "very good", however with no significant differences. In conclusion, the relationship of physical domain and QoL perception were satisfactory. Therefore, QoL should be encouraged from an active lifestyle along with changing of life habits.

A obesidade é uma doença crônica que se caracteriza pelo acúmulo excessivo de gordura no tecido adiposo. Ressaltando a importância do combate a obesidade para a Saúde Pública, e considerando o papel do profissional de Educação Física no tratamento e prevenção dessa doença estabeleceu-se o seguinte objetivo geral para este estudo: compreender a percepção do profissional de Educação Física quanto ao seu papel na dinâmica do combate à obesidade. Tratou-se de uma pesquisa descritiva, de natureza qualitativa, que foi realizada no município de Aracati, Ceará. A amostra contou com a participação de 10 profissionais de Educação Física que responderam a uma entrevista semiestruturada relacionada ao papel do profissional de Educação Física no combate à obesidade. Observou-se que os profissionais referiram que sua importância diante da assistência a pessoas com obesidade é permeada pela atuação em ações de educação em saúde, destacando as doenças que essa patologia pode provocar e apontando que é fundamental combater o sedentarismo. Pode-se concluir que o profissional de Educação Física possui uma percepção relevante do seu papel no combate à obesidade visto que suas ações permitem a realização de práticas assistenciais que promovam a prevenção e a baixa incidência dessa enfermidade.
Obesity is a chronic disease characterized by excessive accumulation of fat in adipose tissue. Currently, you can see that obesity is a disease that is becoming increasingly common in everyday life. Emphasizing the importance of combating obesity for public health, and considering the role of physical education professional in the treatment and prevention of this disease was established the following overall goal for this study: Understanding the perception of physical education professional about their role the dynamics of combating obesity. This was a descriptive, qualitative, which was held in the municipality of Aracati, Ceará. The sample included the participation of 10 professionals of Physical Education who answered a semi-structured interview consists of questions related to the role of the professional physical education in combating obesity. In conducting this research, it was observed that the professionals mentioned its importance on the care of people with obesity is permeated by the presence in health education actions providing guidance as to the severity of obesity, highlighting the diseases that can cause disease and pointing out that it is crucial to combat a sedentary lifestyle. Considering the results, it can be concluded that the professional of Physical Education has an important perception of its role in fighting obesity since their actions allow the realization of care practices that promote the prevention and lower the incidence of this disease.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Black lion tamarins (Leontopithecus chrysopygus) are endangered callithrichids. Their conservation may require future translocations or reintroductions; however these approaches involve risks of pathogen introduction in the environment and stress-related opportunistic infections in these animals. In order to screen for opportunistic and potential pathogenic bacterial and fungal microbiota, ten free-ranging and ten captive Black lion tamarins were studied and the results compared. Nasal, oral and rectal swabs were collected and cultured for aerobic and facultative anaerobic bacteria and fungi, and a total 203 bacterial and 84 fungal isolates were obtained. Overall, the most frequent organisms were Staphylococcus spp., Bacillus spp., Candida spp. and Aspergillus spp. Microbiota of free-ranging and captive animals were similar in composition.
