Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Physicochemical properties and biological effects of quaternary ammonium methacrylates in an experimental adhesive resin for bonding orthodontic brackets

Abstract Fixed orthodontic appliances may lead to biofilm accumulation around them that may increase caries risk. This study aimed to evaluate the influence of quaternary ammonium methacrylates (QAMs) on the physicochemical properties, cytotoxicity, and antibacterial activity of adhesive resins for orthodontic purposes. Methodology: A base resin was prepared with a comonomer blend and photoinitiator/co-initiator system. Two different QAMs were added to the base adhesive: dimethylaminododecyl methacrylate at 5 wt.% (DMADDM) or dimethylaminohexadecyl methacrylate (DMAHDM) at 10 wt.%. The base adhesive, without QAMs, (GC) and the commercial Transbond™ XT Primer 3M (GT) were used as control. The resins were tested immediately and after six months of aging in the water regarding the antibacterial activity and shear bond strength (SBS). The antibacterial activity was tested against Streptococcus mutans via metabolic activity assay (MTT test). The groups were also tested for the degree of conversion (DC) and cytotoxicity against keratinocytes. Results: The resins containing QAM showed antibacterial activity compared to the commercial material by immediately reducing the metabolic activity by about 60%. However, the antibacterial activity decreased after aging (p<0.05). None of the groups presented any differences for SBS (p>0.05) and DC (p>0.05). The incorporation of DMADDM and DMAHDM significantly reduced the keratinocyte viability compared to the GT and GC groups (p<0.05). Conclusion: Both adhesives with QAMs showed a significant reduction in bacterial metabolic activity, but this effect decreased after water aging. Lower cell viability was observed for the group with the longer alkyl chain-QAM, without significant differences for the bonding ability and degree of conversion. The addition of QAMs in adhesives may affect the keratinocytes viability, and the aging effects maybe decrease the bacterial activity of QAM-doped materials.

A rare case of PLA2R- and THSD7A-positive idiopathic membranous nephropathy

RESUMO A Nefropatia Membranosa Idiopática (NMi) é uma frequente causa de síndrome nefrótica em adultos e sua etiologia pode ser estratificada em primária/idiopática ou secundária. O conhecimento da fisiopatologia da NMi sugeriu a presença de autoanticorpos (PLA2R e a THSD7A) direcionados contra antígenos existentes nos podócitos. A detecção de anticorpos contra um domínio favorece NMi. A presença de autoanticorpos contra um desses domínios autoexcluiria a possibilidade de autoanticorpos contra o outro domínio; no entanto, recentemente foram descritos casos que apresentaram dupla positividade para PLA2R e THSD7A, comprovando que, por mecanismos fisiopatológicos ainda não conhecidos, raramente pode existir produção concomitante de anticorpos contra os dois alvos. O presente estudo tem por objetivo relatar o caso de um paciente de 46 anos de idade, do sexo masculino, que apresentou quadro de proteinúria nefrótica, hematúria, hipoalbuminemia e hipercolesterolemia submetido a biópsia e exame histopatológico (ML, IF, IHQ e ME), confirmando um caso raro de NMi com positividade dupla para os anticorpos anti-PLA2R e anti-THSD7A e associação à nefropatia por IgA, mostrando nossa experiência com a utilização de subclasses de IgG, PLA2R e THSD7A na rotina laboratorial para a investigação da GNM e enfatizando a importância de uma abordagem ampla para adequada elucidação e conhecimento dos mecanismos fisiopatológicos na NMi.
ABSTRACT Idiopathic membranous nephropathy (IMN) is a frequent cause of nephrotic syndrome in adults. In terms of etiology, the condition may be categorized as primary/idiopathic or secondary. Literature on the pathophysiology of IMN has indicated the presence of autoantibodies (PLA2R and THSD7A) directed against podocyte antigens. The detection of antibodies against a domain favors IMN. The presence of autoantibodies against one of the domains would in theory exclude the possibility of there being autoantibodies against the other domain. However, cases of patients with PLA2R- and THSD7A-positive disease have been recently reported, showing that antibodies against two targets may be concomitantly produced via yet unknown pathophysiological mechanisms. This study reports the case of a 46-year-old male patient with nephrotic-range proteinuria, hematuria, hypoalbuminemia, and hypercholesterolemia submitted to biopsy and histopathology examination (LM, IF, IHC, and EM) eventually diagnosed with PLA2R- and THSD7A-positive IMN associated with IgA nephropathy, stressing our experience with the use of IgG subclasses, PLA2R, and THSD7A in the workup for MN and the relevance of adopting a broad and adequate approach to elucidating and acquiring knowledge of the pathophysiology of IMN.

Is there a relationship between the neutrophil/lymphocyte ratio and bilaterality in patients with coxarthrosis?

RESUMO Objetivo: Avaliar a relação entre a razão neutrófilo/linfócito e a presença de sinais de artrose em ambos os quadris em pacientes acompanhados neste serviço. Métodos: Estudo transversal, retrospectivo, que usou análise de prontuários e revisão de banco de dados de pacientes maiores de 18 anos com diagnóstico de artrose de quadril acompanhados no ambulatório deste hospital. Resultado: Com relação à análise do teste de Mann-Whitney para correlacionar a razão neutrófilo/linfócito e a lateralidade, observou-se um resultado de teste bilateral de 0,036, evidenciou desse modo a diferença entre os grupos. Quando os valores absolutos de neutrófilos e linfócitos foram analisados, observaram-se p = 0,14 e p = 0,24, não foi possível observar diferenças estatisticamente significativas entre os valores absolutos nos dois grupos. Conclusões: Considerando-se as interações entre os mecanismos inflamatórios na osteoartrose e o fato de que a interação entre os neutrófilos e os linfócitos tem diferenças com relação à lateralidade da coxartrose, é possível levantar a hipótese de que a etiologia inflamatória da osteoartrose unilateral e da bilateral tem dinâmicas diferentes. Entretanto, são necessários estudos mais aprofundados, com citometria de fluxo, para avaliar o impacto com relação às diferenças nos mecanismos inflamatórios observados nesse estudo.
ABSTRACT Objective: The objective of this study was to evaluate the relationship between the neutrophil/lymphocyte ratio and the presence of signs of arthrosis in both hips in patients followed at this medical center. Methods: This was a cross-sectional, retrospective study through the analysis of medical records and database review of patients over 18 years of age with hip arthrosis, followed at the outpatient clinic of this hospital. Results: Regarding the analysis of the Mann-Whitney test to correlate the neutrophil/lymphocyte ratio and laterality, a bi-lateral test result of p = 0.036 was obtained, thus demonstrating a significant difference between the observed groups. When we analyzed the absolute values of neutrophils and lymphocytes, the authors obtained results of p = 0.14 and p = 0.24. Therefore, it was not possible to observe statistically significant differences between the absolute values in the two groups. Conclusion: Considering the interactions between the inflammatory mechanisms in osteoarthritis and the fact that the interaction between neutrophils and lymphocytes has differences in relation to the laterality of the coxarthrosis, it is hypothesized that the inflammatory etiology of unilateral and bilateral osteoarthritis could have different dynamics. However, more in-depth studies with flow cytometry are needed to assess the possible impact of these differences in the inflammatory mechanisms observed in this study.

Associação entre desmineralização óssea, atividade física e padrões antropométricos

O estudo teve por proposta, verificar a correlação entre densidade mineral óssea e nível de atividade física, bem como à ingestão alimentar e parâmetros antropométricos, além de avaliar a densidade mineral óssea (DMO) de mulheres menopausa. Foi feito o exame de densitometria óssea, da região lombar (L1 a L4), colo de fêmur, e fêmur total, como também utilizar o auto-recordatório das atividades do cotidiano de Bouchard; utilizado o registro alimentar de Buker e Stuart, para dosar e quantificar a ingestão diária de cálcio e vitamina D. Os dados foram analisados por meio do teste de Kolmogorov-Smirnov foi considerado um valor padrão de α = 0,05 para comparar as médias das DMO. Constatou-se que cem por cento dos indivíduos avaliados, apresentam um grau de DMO, abaixo da média prevista pela organização mundial de saúde (OMS), estando 14,4% no quadro de osteopenia e 85,6% com osteoporose, com uma menor DMO na região do fêmur (0,721g), e com maior perda entre as mulheres que não fazem atividade física (0,698g). Não houve correlação entre a prática de atividade física e a densidade mineral óssea, porém existe correlação quando associada a padrões antropométricos e ingestão diária de vitamina D.
This paper aimed to verify the correlation between bone mineral density and the level of physical activity, as well as the food intake and the anthropometric parameters. It intended to analyse the bone mineral density (BMD) of menopausal women through the bone densitometry test (DO) in the lumbar region (L1 to L4), femoral neck and total femur, and also use Bouchard’s self-recall of daily activities; employing the food record from Buker and Stuart to dose and quantify the daily intake of calcium and vitamin D. The data were analysed via Kolmogorov-Smirnov’s test, and default value of α = 0.05 was set to compare the BMD averages. It was observed that one hundred percent of the assessed individuals had a BMD level below the average fixed by WHO: 14.4% with osteopenia and 85.6% with osteoporosis; a lower BMD in the femoral area (0.721g) and the biggest loss among the sedentary ones (0.698g). It was noticed that there was a correlation between the physical activities and the BMD only when associated with anthropometric standards and the daily ingestion of vitamin D.

Resumo: O objetivo deste trabalho foi avaliar os aspectos biológicos de Helicoverpa zea e Helicoverpa armigera (Lepidoptera: Noctuidae) em híbridos de milho com expressão de diferentes proteínas. Foram avaliadas as proteínas Cry1F, Cry1A(b), Cry1A.105 + Cry2Ab2 e Vip3A e seus respectivos isogênicos convencionais. As larvas foram alimentadas com espiguetas de milho Bt e convencional, trocadas a cada dois dias. Os parâmetros avaliados foram: sobrevivência 48 horas após eclosão, sobrevivência larval, peso de larvas e de pupas, período de desenvolvimento larval e período pré-imaginal. Observou-se efeito significativo da interação entre evento de milho Bt e espécie de Helicoverpa para todas as variáveis biológicas avaliadas. Larvas de H. armigera apresentaram maior sobrevivência 48 horas após eclosão em milho expressando a proteína Cry1F e a Vip3 em relação a H. zea. O período letal foi maior em H. armigera do que em H. zea, mas, em milho com expressão da proteína Cry1A(b), foi quatro vezes menor. Não houve sobrevivente para ambas as espécies de Helicoverpa em nenhum dos híbridos Bt avaliados. Nos isogênicos convencionais, o índice de adaptação de H. armigera foi superior ao de H. zea, o que indica maior facilidade de adaptação ao ambiente dessa espécie.
Abstract: he objective of this work was to evaluate the biological responses of Helicoverpa zea and Helicoverpa armigera (Lepidoptera: Noctuidae) in maize hybrids expressing different proteins. The Cry1F, Cry1A(b), Cry1A.105 + Cry2Ab2, and Vip3A proteins and their isogenic conventional counterparts were evaluated. Larvae were fed on spikelets of Bt and conventional maize, which were changed every two days. The variables evaluated were: survival 48 hours after hatching, larval survival, larval and pupal weight, larval development period, and pre-imaginal period. There was a significant interaction between Bt maize event and Helicoverpa species for all evaluated biological variables. Helicoverpa armigera larvae showed higher survival 48 hours after hatching on maize expressing the Cry1F and Vip3 proteins when compared to H. zea. Lethal period was greater for H. armigera than for H. zea, but, in maize expressing Cry1A(b) protein, it was four times lower. There were no survivals for both Helicoverpa species in the evaluated Bt hybrids. In isogenic conventional counterparts, the adaptation index of H. armigera was higher than that of H. zea, which indicates greater ease of adaptation to the environment of that species.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

As aplicações de transições de fase vão muito além do contexto da termodinâmica de equilíbrio, de onde surgiu. Nós revisamos um dos principais exemplos de uma transição de fase em um sistema sem termodinâmica, o processo de contato (PC), com ênfase no método de campo médio. Apresentamos um estudo de campo médio aplicado a um modelo para a transmissão da malária em uma população. Discutimos também as novas abordagens da distribuição quase-estacionária e o processo de contato conservativo.
Phase transitions find application well beyond the context of equilibrium thermodynamics, where they arose. We review one of the prime examples of a phase transition in a system lacking thermodynamics, the contact process (PC), emphasizing mean-field theory methods. We also present mean-field results for a model of transmission of malaria in a population, and discuss the new approaches of quasi-stationary distributions, and the conserved contact process.
