Synthesis, in silico Study and Antileishmanial Evaluation of New Selenides Derived from 7-Chloro-quinoline and N-Phenylacetamides

This study describes a virtual screening performed for two series of selenides (28 compounds), derived from N-phenylacetamides chlorides and 7-chloro-quinoline, to determine their potential for leishmanicidal activity against Leishmania amazonensis and Leishmania donovani. Seven compounds were predicted as potential leishmanicides; therefore, they were synthesized from elemental selenium, as a precursor for the production of NaHSe, and subsequent reactions with 4,7-dichloro-quinoline and N-phenylacetamides chlorides were performed. The compounds were characterized by infrared (IR), 1H and 13C nuclear magnetic resonance (NMR), and sent for in vitro cytotoxicity tests against L. amazonensis and were found to be active and selective, and two compounds presented half-maximal inhibitory concentrations (IC50) of 5.67 and 10.81 µg mL-1. They also presented good interaction energies in the docking study, suggesting that may exert their effects by inhibiting the N-myristoyltransferase and O-acetylserine sulfhydrylase enzymes in parasites.

Nrf2, NF-κB and PPARβ/δ mRNA Expression Profile in Patients with Coronary Artery Disease

Resumo Fundamentos: O estresse oxidativo e a inflamação estão presentes na doença arterial coronariana (DAC) e estão ligados à ativação do fator de transcrição nuclear kappa B (NF-κB). Para atenuar essas complicações, fatores de transcrição como o fator nuclear eritroide 2-relacionado ao fator 2 (Nrf2) e o receptor ativado por proliferador de peroxissoma β/δ (PPARβ/δ) podem ser ativados para inibir o NF-κB. No entanto, os dados disponíveis sobre a expressão de NF-κB, Nrf2 e PPARβ/δ em pacientes com DAC são limitados. Objetivo: Avaliar a expressão dos fatores transcricionais NF-κB e Nrf2 e o PPARβ/δ em pacientes com DAC. Métodos: Trinta e cinco pacientes (17 homens, idade média de 62,4 ± 7,55 anos) com DAC e doze pacientes (5 homens, com idade média de 63,50 ± 11,46 anos) sem DAC foram incluídos. Células mononucleares do sangue periférico (PBMCs) foram isoladas e processadas para a expressão de mRNA do Nrf2, NF-κB, NADPH: quinona oxidoredutase 1 (NQO1) e mRNAs do PPARβ/δ por meio de reação em cadeia da polimerase quantitativa em tempo real (qPCR). Valores de p < 0,05 foram considerados como estatisticamente significativos. Resultados: Não houve diferença nas expressões de mRNA do Nrf2 (1,35 ± 0,57), NF-κB (1,08 ± 0,50) ou na enzima antioxidante NQO1 (1,05 ± 0,88) no grupo DAC em comparação com o grupo sem DAC (1,16 ± 0,76, 0,95 ± 0,33, 0,81 ± 0,55, respectivamente). Entretanto, o PPARβ/δ apresentou maior expressão no grupo com DAC (1,17 ± 0,86 vs. 0,56 ± 0,34, p = 0,008). Conclusão: O principal achado do presente estudo foi o PPARβ/δ apresentar maior expressão nas PBMCs de pacientes com DAC comparados ao grupo controle, ao passo que não foram observadas diferenças nas expressões de mRNA do Nrf2 ou NF-κB.
Abstract Background: Oxidative stress and inflammation are present in coronary artery disease (CAD) and are linked to the activation of the transcription nuclear factor kappa B (NF-κB). To attenuate these complications, transcription factors like nuclear factor erythroid 2-related factor 2 (Nrf2) and peroxisome proliferator-activated receptor-β/δ (PPARβ/δ) can be activated to inhibit NF-κB. However, the available data on expression of NF-κB, Nrf2 and PPARβ/δ in CAD patients are limited. Objective: To evaluate the expression of the transcription factors NF-κB and Nrf2 and PPAR��/�� in CAD patients. Methods: Thirty-five patients (17 men, mean age 62.4 ? 7.55 years) with CAD and twelve patients (5 men, mean age 63.50 ? 11.46 years) without CAD were enrolled. Peripheral blood mononuclear cells (PBMCs) were isolated and processed for mRNA expression of Nrf2, NF-κB, NADPH: quinone oxidoreductase 1 (NQO1) and PPARβ/δ mRNAs using quantitative real-time polymerase chain reaction (qPCR). p < 0.05 was considered statistically significant. Results: There was no difference in the mRNA expressions of Nrf2 (1.35 ? 0.57), NF-κB (1.08 ? 0.50) or in the antioxidant enzyme NQO1 (1.05 ? 0.88) in the CAD group compared to the group without CAD (1.16 ? 0.76, 0.95 ? 0.33, 0.81 ? 0.55, respectively). However, PPARβ/δ was highest expressed in the CAD group (1.17 ? 0.86 vs. 0.56 ? 0.34, p = 0.008). Conclusion: The main finding of this study was the PPARβ/δ being more expressed in the PBMC of patients with CAD compared to the control group, whereas no differences were observed in Nrf2 or NF-κB mRNA expressions.

Desorption isotherms and isosteric heat of 'cajuzinho-do-cerrado' achenes

RESUMO Objetivou-se, com este trabalho, determinar as isotermas de dessorção de aquênios de cajuzinho-do-cerrado (Anacardium humile St. Hil.) em diversas condições de temperatura e atividades de água e selecionar aquela que melhor representa o fenômeno e determinar o calor isostérico de dessorção. Os frutos foram coletados no Parque Nacional das Emas, no município de Mineiros - GO, despolpados e em seguida submetidos à secagem em sílica gel a temperatura de 25 ± 2 °C, até atingirem os teores de água de 17,6; 13,6; 11,1; 8,7; 5,3 (% b.s). Após a secagem foram determinadas as isotermas de dessorção pelo método estático indireto. A atividade de água (Aw) foi determinada sob diferentes temperaturas sendo os aquênios acondicionados em câmara B.O.D., regulada a 10, 20, 30 e 40 °C; aos dados de teor de água de equilíbrio foram ajustados a diferentes modelos matemáticos por meio da análise de regressão não linear, pelo método Gauss-Newton. O modelo Copace foi o que melhor representou a higroscopicidade dos aquênios de cajuzinho-do-cerrado enquanto os valores de calor isostérico integral de dessorção dos aquênios de cajuzinho-do-cerrado para a faixa de teor de água de 4,51 a 13,40 (% b.s.) variaram de 2734,82 a 2548,49 kJ kg-1.
ABSTRACT The objective of this study was to determine the desorption isotherms of 'cajuzinho-do-cerrado' achenes (Anacardium humile St. Hil.) in various conditions of temperature and water activity, as well as to select the one that best represents the phenomenon and to determine the isosteric heat of desorption. The fruits were collected at the Emas National Park, in the municipality of Mineiros-GO, Brazil, pulped and then subjected to drying in silica gel at temperature of 25 ± 2 °C until the moisture contents of 17.6, 13.6, 11.1, 8.7 and 5.3 (d.b.%). After drying, the desorption isotherms were determined by the indirect static method. The water activity (Aw) was determined at different temperatures, and the achenes were placed in a B.O.D. chamber, regulated at 10, 20, 30 and 40 °C. Data of hygroscopic equilibrium moisture content were fitted to different mathematical models through non-linear regression analysis, using the Gauss-Newton method. The Copace model was the one that best represented the hygroscopicity of 'cajuzinho-do-cerrado' achenes, while the integral isosteric heat of desorption of 'cajuzinho-do-cerrado' achenes for the moisture content range of 4.51 to 13.40 (% d.b.) varied from 2,734.82 to 2,548.49 kJ kg-1.

Arsenic is considered a semimetal, and its wide distribution in the Earth's crust in different chemical forms, including organic and inorganic species, has a great deal of influence on the mechanisms of toxicity. Exposure to arsenic can be either through occupational practice (use of pesticides) or by the consumption of water and food containing the element. Rice is considered a fundamental constituent of the basic diet of Brazilians and is usually cultivated in flooded conditions. Such a plantation system results in an increased amount of As in the soil and hence a greater accumulation of As in the plant, which is highlighted by the inorganic species' classification as highly toxic. Besides the use of mitigation techniques to reduce the toxicological risk, monitoring the concentration of As and its chemical species in rice and rice products is required through the establishment of legislation in the area. Thus, some world organizations are conducting improved research to determine and establish acceptable concentrations of As and its chemical species in rice, e. g., in 2012, FDA researchers described a chemical speciation methodology for As in rice and rice products. Hence, the application of existing chemical speciation methods and the establishment of parameters for ensuring food security and exposure risk assessment deserve particular consideration.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

There is a lack of information about the level of resistance of cotton genotypes to a wider range of Ramularia areola isolates occurring across the cotton growing areas of Brazil. For this purpose, firstly it is necessary to know the existence or not of genotypic and phenotypic variability among the R. areola isolates from different geographical origins. The objective of the present investigation was to verify the existence of phenotypic variability among 23 R. areola isolates collected from six cotton growing states of Brazil. Two resistant genotypes, FMT 02102996 and CNPA BA 2003-2059, and the susceptible genotype FMT 701 were individually inoculated with 23 R. areola isolates under glasshouse conditions and the severity of infection was evaluated 30 days after inoculation. Genotypes CNPA BA 2003-2059 and FMT 02102996 were susceptible to three isolates and resistant to the rest of the isolates. Genotype FMT 701 was susceptible to all the isolates except the isolates 22.3 and 42.7. Results indicate the existence of variability among R. areola isolates and that the three genotypes are useful in distinguishing phenotypic variability within isolates of this pathogen.

O objetivo desta pesquisa foi realizar um diagnóstico qualitativo dos gêneros de parasitos encontrados em amostras fecais ambientais de jacarés (Caiman latirostris Daudin, 1802), criados comercialmente em sistema fechado, no período de 2008 a 2009, no estado do Rio de Janeiro. Um total de 300 amostras foi coletado de 150 filhotes, 80 de animais de engorda e 70 de reprodução, e submetido a análises coproparasitológicas, de flutuação (método de Willis-Mollay) e sedimentação simples (método de Lutz), de acordo com Hoffmann (1987). As amostras foram visualizadas à luz da microscopia óptica. Os resultados obtidos evidenciaram a presença de oocistos de Eimeria e Isospora, cistos de Balantidium e ovos de Acanthostomum e Dujardinascaris.
The objective of this paper was to diagnose qualitatively parasite genera found in environmental fecal samples of alligators (Caiman latirostris Daudin, 1802) commercially bred from 2008 to 2009 in a closed farming system in the state of Rio de Janeiro. A total of 300 samples were collected from 150 young, 80 fattening and 70 breeding processed by two different methods, the flotation (method of Willis-Mollay) and simple sedimentation (method of Lutz), according to Hoffmann (1987). The samples were then examined by optical microscopy. The results revealed presence of Eimeria and Isospora oocysts, Balantidium cysts, and Acanthostomum and Dujardinascaris eggs.

INTRODUÇÃO: Em pacientes infectados pelo HIV, a colonização da cavidade bucal por leveduras patogênicas pode levar ao desenvolvimento de fungemias. No presente estudo, avaliamos a prevalência de leveduras na cavidade bucal de pacientes HIV-positivos e verificamos se as espécies isoladas foram enzimaticamente ativas. Além disso, as espécies identificadas foram testadas quanto à suscetibilidade a antifúngicos. MÉTODOS: Amostras de saliva e de candidose orofaríngea foram coletadas de 60 pacientes soropositivos para HIV e identificados pelo sistema API20C. A atividade enzimática foi avaliada pela produção de proteinase e fosfolipase. A suscetibilidade a antifúngicos foi determinada utilizando o método de microdiluição em caldo. RESULTADOS: As espécies mais comumente isoladas foram C. albicans (51,56%), seguido por espécies de Candida não-albicans (43,73%), Trichosporon mucoides (3,12%) e Kodamaea ohmeri (1,56%). A colonização bucal por associação de diferentes espécies foi observada em 42% dos pacientes. A atividade enzimática foi verificada na maioria das espécies isoladas, com exceção de C. glabrata, C. lusitaniae e C. guilliermondii. Resistência ao fluconazol e anfotericina B foi observada em isolados de C. albicans, C. glabrata, C. parapsilosis, C. krusei, e K. ohmeri. CONCLUSÃO: Os pacientes HIV-positivos são colonizados por espécies únicas ou múltiplas de levedura que ocasionalmente são resistentes ao fluconazol ou anfotericina B.
INTRODUCTION: In HIV-infected patients, colonization of the oral cavity by potential pathogenic yeast may lead to development of systemic fungemia. We evaluated the prevalence of yeast in the oral cavity of Brazilian HIV-positive patients and verified whether or not the species characterized were enzymatically active. Furthermore, the species identified were tested for their susceptibility to antifungal treatment. METHODS: Patient saliva and oropharyngeal candidiasis samples were collected from 60 seropositive HIV patients and identified by the API20C system. Enzymatic activity was evaluated by the production of proteinase and phospholipase. Susceptibility to antifungal treatments were determined using the broth microdilution method. RESULTS: the most commonly isolated species were C. albicans (51.56%) followed by non-albicans Candida species (43.73%), Trichosporon mucoides (3.12%) and Kodamaea ohmeri (1.56%). Oral colonization by association of different species was observed in 42% of the patients. Enzymatic activity was verified in most of species isolated, except for C. glabrata, C. lusitaniae and C. guilliermondii. Resistance to Fluconazole and Amphotericin B was observed in isolates of C. albicans, C. glabrata, C. parapsilosis, C. krusei, and K. ohmeri. CONCLUSION: HIV-positive patients are orally colonized by single or multiple species of yeast that are occasionally resistant to Fluconazole or Amphotericin B.

Introdução: O limiar do reflexo acústico corresponde à menor intensidade de um som capaz de desencadear o mecanismo de proteção da orelha média frente a sons intensos (Metz, 1952; Jerger, 1970), sendo necessária a integridade estrutural e funcional do sistema auditivo periférico e central em nível de tronco encefálico. Algumas dessas estruturas são também responsáveis pelo processamento central da informação auditiva. Objetivo: Procuramos verificar se alterações do reflexo acústico também estariam presentes na Desordem do Processamento Auditivo Central (DPA), levando a queixas relacionadas aos distúrbios da comunicação. Forma de estudo: Prospectivo randomizado. Material e método: Foram analisados cem protocolos de avaliações do Processamento Auditivo Central (PAC), realizadas segundo Pereira (1997), de indivíduos dos sexos masculino e feminino, de 7 a 18 anos, com limiares de audibilidade normais e timpanograma tipo A. A diferença entre os limiares do reflexo acústico e os de audibilidade forneceu os níveis de reflexos acústicos (NRA), considerados normais entre 70-90dB e alterados quando acima deste intervalo ou quando ocorreu ausência em uma ou mais freqüências (Carvallo, 1996; 1997; Metz, 1952). Resultados: Das crianças avaliadas 97% apresentaram algum tipo de DPA. Destas, 62% tiveram NRA alterados, numa relação estatisticamente significante. Além disso, pacientes com DPA mostraram NRA alterados, mais freqüentemente, nas desordens de grau severo, naquelas com prejuízos gnósicos auditivos combinados e naquelas com mais de uma habilidade auditiva alterada. Conclusão: Assim, indivíduos com alterações no reflexo acústico e sem alterações audiométricas devem submeter-se a provas de PAC, já que esses sintomas podem ser manifestações patológicas do sistema nervoso central.
Introduction: The acoustic reflex threshold is defined as the lowest sound intensity capable of starting the middle ear protection mechanism due to intense sounds (Metz, 1952; Jerger, 1970), being necessary the structural and functional integrity of the periferic and central auditory system. These structures are also responsable for the central processing of auditory information. Aim: We intend to verify whether acoustic reflex abnormalities can also appear in auditory processing (AP) disorders, resulting in symptoms related to speech and language disorders. Study design: Prospective randomized. Material and method: Data were analyzed from one hundred AP assessments, using Pereira (1997) method. Patients, male and female, with ages ranging from 07 to 18 years, had normal hearing thresholds and normal tympanograms patterns. The difference between the acoustic reflex and hearing thresholds defined the acoustic reflex level (ARL), considered normal between 70-90dB and altered when above of this range or when absent in one or more frequencies (Carvallo, 1996; 1997; Metz, 1952). Results: Disorders of AP were found in 97% of the patients. In this group, 62% showed ARL abnormalities, being statistically significant. Furthermore, patients with AP disorders showed ARL alterations, more frequently on severe degree disorders, on patients with combined auditory gnosis impairments and on patients with more than one auditory ability altered. Conclusion: Patients with acoustic reflex alterations and normal audiometry should perform the AP assessment, as these symptoms could unmask pathologies of the central nervous system.
