Chemical Constituents and Cytotoxic Activity of Rhinella jimi (Anura: Bufonidae)

Rhinella jimi toads (Stevaux, 2002) belong to the Bufonidae family, are endemic in the Brazilian Northeast and are commonly found during rainy periods. In general, amphibians of this family have in their poisons different metabolites that show a diversity of pharmacological activities. The isolation and identification of these compounds are of great importance, and techniques such as high-performance liquid chromatography coupled to mass spectrometry are widely used for the discovery of novel and known compounds in these poisons. For R. jimi poison, the ethyl acetate and methanolic extracts were obtained and thirty compounds were identified by combining ultra-performance liquid chromatography coupled to mass spectrometry (UPLC-MS) with direct infusion atmospheric pressure chemical ionization mass spectrometry (DI-APCI-MS/MS) and direct infusion electrospray mass spectrometry (DI-ESI-MS/MS) for each extract, respectively. Marinobufagin (2) and marinobufotoxin (19) were the majorities of each extract, respectively. In addition, other bufadienolides mainly present in the ethyl acetate extract, such other bufotoxins, alkaloids and arginine diacid derivatives were identified in the methanol extract. In a cytotoxic assay by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), the extracts and compound 2 demonstrated half-maximal inhibitory concentration (IC50) values better than the positive control doxorubicin, evidencing excellent cytotoxic. This is the most complete study of the chemical composition of R. jimi toad poison and its respective cytotoxic activity, promoting the enrichment of knowledge about this family and species.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

INTRODUÇÃO: Neste estudo, buscamos avaliar os resultados clínicos de pacientes com lesões coronárias em vasos de fino calibre, numa coorte de pacientes do mundo real submetidos a intervenção coronária percutânea (ICP) com stents farmacológicos. MÉTODOS: Entre maio de 2002 e dezembro de 2009, 1.380 pacientes consecutivos do Registro DESIRE (Drug Eluting Stents In the Real World), com 1.683 lesões em vasos de fino calibre (< 2,5 mm de diâmetro), foram submetidos a ICP, eletiva ou de urgência, com implante de 1.818 stents farmacológicos (CypherTM, 89%; TaxusTM, 7,5%; Xience V TM/PromusTM, 3%; EndeavorTM, 0,33%; BiomatrixTM, 0,2%) e incluídos neste estudo. O seguimento clínico de até 7 anos (mediana, 2,8 anos) foi completo em 98%, sendo obtido com 1, 6 e 12 meses e, então, anualmente. Tivemos como objetivo determinar as taxas de eventos cardíacos adversos maiores (ECAM) ao longo do período do acompanhamento clínico. RESULTADOS: A média de idade foi de 64,5 ± 11,7 anos, com predomínio de pacientes do sexo masculino (76,7%). Diabetes melito foi encontrado em 31,6% dos casos e 9,8% eram portadores de insuficiência renal crônica. A artéria descendente anterior foi o vaso mais frequentemente tratado (43,4%) e dois terços das lesões eram de alta complexidade (B2/C). Sucesso angiográfico do procedimento foi obtido em 98,8% dos casos. A taxa de ECAM foi de 12,8% durante o seguimento, com 4,5% de óbito cardíaco, 4,2% de infarto agudo do miocárdio, 4,5% de nova revascularização da lesão-alvo e 1,5% de trombose do stent. CONCLUSÕES: No Registro DESIRE, o tratamento de vasos de pequeno calibre com stents farmacológicos, em pacientes não-selecionados, associou-se a excelentes resultados agudos e tardios, e a baixas taxas de trombose ao longo da evolução.
BACKGROUND: In this study, we sought to evaluate the clinical outcomes of patients with coronary lesions in small vessels in a cohort of real world patients undergoing percutaneous coronary intervention (PCI) with drug-eluting stents (DES). METHODS: Between May 2002 and December 2009, 1,380 consecutive patients from the DESIRE Registry (Drug Eluting Stents In The Real World), with 1,683 lesions in small vessels (< 2.5 mm in diameter) were consecutively submitted to elective or emergency PCI, with 1,818 DES (CypherTM, 89%; TaxusTM, 7.5%; Xience V TM/PromusTM, 3%; Endeavor, 0.33%; BiomatrixTM, 0.2%) and included in this study. The clinical follow-up of up to 7 years (median, 2.8 years) was completed for 98%, and was obtained at 1, 6, 12 months and then annually. Our objective was to determine the rates of major cardiac events (MACE) during clinical follow-up. RESULTS: Mean age was 64.5 ± 11.7 years, with a prevalence of male patients (76.7%). Diabetes mellitus was observed in 31.6% of the cases and 9.8% had chronic renal failure. Left anterior descending artery was the most frequent vessel treated (43.4%) and two-thirds of the lesions were complex lesions (B2/C). Angiographic success was obtained in 98.8% of the cases. The rate of MACE was 12.8% during follow-up with 4.5% of cardiac death, 4.2% of acute myocardial infarction, 4.5% of repeat target lesion revascularization and 1.5% of stent thrombosis. CONCLUSIONS: In the DESIRE Registry, the use of DES to treat small vessels in non-selected patients was associated with excellent early and late outcomes and low thrombosis rates.

Neste artigo são apresentadas a síntese e a caracterização de dois compostos binucleares de ferro, os quais contêm uma ponte μ-oxo. Os compostos [(SO4)(L5)Fe(μ-O)Fe(L5)(SO4 )]•6H2O, 1, e [Cl(L5)Fe(μ-O)Fe(L5)Cl]Cl2• 2H2O, 2, foram obtidos nas reações entre 1-(bis-piridin-2-ilmetil-amino)-3-cloropropan-2-ol (L5) e os sais FeSO4•7H2 O e FeCl3•6H2 O, respectivamente. Os espectros eletrônicos dos complexos apresentam absorções somente na região do ultravioleta, sendo que a análise eletroquímica revelou que, após a formação da espécie FeIII FeII, a unidade binuclear do composto 1 é mais estável do que a do composto 2. Os ligantes monodentados (sulfato e cloreto) exercem influência sobre os parâmetros Mössbauer determinados para 1 e 2, particularmente sobre os desdobramentos de quadrupolo. Os compostos foram empregados como catalisadores em reações de oxidação do cicloexano, usando H2O2 e t-BuOOH como oxidantes em uma razão substrato:oxidante:catalisador de 1000:1000:1. Os resultados indicam que o composto 2 é um catalisador mais eficiente que o composto 1.
We report herein the synthesis and characterization of two dinuclear μ-oxo iron compounds obtained through the reactions of FeSO4•7H2 O and FeCl3•6H2 O with 1-(bis-pyridin-2-ylmethyl-amino)-3-chloropropan-2-ol (L5), which resulted in the compounds [(SO4)(L5)Fe(μ-O)Fe(L5)(SO4 )]•6H2O, 1, and [Cl(L5)Fe(μ-O)Fe(L5)Cl]Cl2• 2H2O, 2. The electronic spectra of both compounds show absorption bands only in the UV range. The electrochemical analysis showed that the dinuclear unit is more stable under reduction in compound 1 than in compound 2, while the Mössbauer spectroscopy revealed that the monodentate ligands (sulfate and chloride) have a significant influence on the Mössbauer parameters determined for 1 and 2, particularly on the quadrupole splitting values. Both compounds were studied as catalysts in reactions of cyclohexane oxidation, using H2O2 and t-BuOOH as oxidants, in a substrate:oxidant:catalyst ratio of 1000:1000:1. Cyclohexanol, cyclohexanone, cyclohexyl hydroperoxide, t-butyl cyclohexyl peroxide and adipic acid were formed during the process. The experiments revealed that compound 2 is, in general, more active than compound 1 in promoting cyclohexane oxidation.

Devido à crescente expansão da leishmaniose visceral americana (LVA) no Brasil, o presente estudo teve como objetivo identificar as espécies de flebotomíneos em áreas vulneráveis à transmissão dessa parasitose, bem como em outras sem qualquer informação sobre a presença desses dípteros no Paraná. As coletas de flebotomíneos foram realizadas em 46 localidades distribuídas em 37 municípios do Paraná, no período de março de 2004 a novembro de 2005. Em cada uma das localidades foram instaladas armadilhas de Falcão, durante três noites consecutivas, em vegetação natural e ambientes antrópicos (intra e peridomicílio). Ocasionalmente, foram instalas armadilhas de Shannon e feitas inspeções de paredes e aspiração em domicílio, peridomicílio e extradomicílio. O tratamento dos dados baseou-se na estimativa das freqüências e abundância das espécies, segundo cinco regiões de distintas paisagens originais. Coletaram-se 38.662 flebotomíneos de 23 espécies. Predominaram Nyssomyia neivai (Pinto) (75.6%), Ny. whitmani (Antunes & Coutinho) (10.1%), Migonemyia migonei (França) (7.8%), Expapillata firmatoi (Barreto et al.) (2.1%) and Pintomyia fischeri (Pinto) (1,6%); representando juntas 97,2% dos flebotomíneos coletados. Lutzomyia longipalpis (Lutz & Neiva) o principal vetor da LVA não foi encontrado. No entanto, capturou-se Lu. gaminarai (Cordero et al.), cujas fêmeas são morfologicamente semelhantes às de Lu. longipalpis. As espécies mais freqüentes e abundantes têm sido apontadas como vetores da leishmaniose tegumentar no Paraná e em outras áreas das Regiões Sudeste e Sul do Brasil. A presença de Lu. gaminarai no Paraná suscita a necessidade de estudos do seu comportamento, inclusive em relação à sua competência vetorial do agente da leishmaniose visceral.
Due to the spread of the Visceral leishmaniasis in Brazilian territory, this research was undertaken with the objective to identify the sandfly fauna and aspects of the species’ behavior in forest and anthropic environments in Paraná State, Brazil. The sandfly captures were sporadically carried out in 37 municipalities of the Paraná state, during the period from March 2004 to November 2005. Each municipality was once visited, when Falcão traps were installed during three consecutive nights in forested fragments and anthropic environments; captures with Shannon traps installed in forests and edge of forests, inspection of walls and electric aspiration in domiciles and forested areas were sporadically undertaken. The data analysis was based on estimates of frequency and abundance of the species, by five native landscapes. A total of 38,662 specimens belonging to 23 species were captured. Nyssomyia neivai (Pinto) (75.6%), Ny. whitmani (Antunes & Coutinho) (10.1%), Migonemyia migonei (França) (7.8%), Expapillata firmatoi (Barreto et al.) (2.1%) and Pintomyia fischeri (Pinto) (1.6%) together constituted 97.2% of the phlebotomine colletcted. High frequencies of sand flies were found in forests, henhouses, pigpens and domiciles. The three most frequent species have been implicated in transmission of tegumentary leishmaniasis in Paraná and in the Brazilian Southeastern and Southern Regions. Lutzomyia longipalpis (Lutz & Neiva), the visceral leishmaniasis vector was no found, however Lutzomyia gaminariai (Cordero et al.), female of which is similar to that of Lu. longipalpis was captured, raising the need to investigate its behavior, including as regard to the vectorial competence.

Materiais cerâmicos à base de óxidos de manganês, de níquel e de cobalto foram preparados por meio de mistura de óxidos seguida de homogeneização e sinterização a 1250 ºC. Os compostos foram caracterizados por meio das técnicas de difração de raios X e microscopia eletrônica de varredura. Análise termogravimétrica foi feita sob atmosferas oxidante e redutora. O comportamento termistor foi estudado por meio de medidas de resistividade elétrica pela técnica dc de duas pontas de prova entre temperatura ambiente e 220 °C. Os principais resultados mostram a preparação de termistores em ampla faixa de resposta elétrica por meio da variação da composição relativa dos óxidos de manganês, de níquel e de cobalto.
Ceramic thermistors based on manganese, nickel and cobalt oxides were prepared by mixing these oxides followed by pressing and sintering at 1250 °C. The oxides were characterized by X-ray diffraction and scanning electron microscopy. The thermistor behavior was studied by two-probe dc electrical resistivity measurements in the room temperature-220 °C range. The main results show the possibility of preparing thermistors in a wide range of electrical response by suitable choice of the relative concentrations of the precursor oxides.

This paper reports the construction of an axisymmetric nonpremixed piloted jet burner, with well-defined initial and boundary conditions, known as the Delft burner, to assess turbulence-chemistry interaction in non-premixed turbulent flames. Detailed experimental information is described, involving hot-wire anemometry, thin-wire thermocouples and chemiluminescence visualization measurements. Radial profile of the axial mean velocity indicates excellent agreement between flow patterns developed within Delft installation and the one described herein. Chemiluminescence emissions from CH and C2 free-radicals were acquired with a CCD camera. Tomography reconstruction analysis was utilised to compare radical emissions and temperature spatial distributions. There was a strong dependence between temperature and CH/C2 emissions. This is an indication that these radicals can be used in flame front studies.

This work analyzes an active fuzzy logic control system in a Rijke type pulse combustor. During the system development, a study of the existing types of control for pulse combustion was carried out and a simulation model was implemented to be used with the package Matlab and Simulink. Blocks which were not available in the simulator library were developed. A fuzzy controller was developed and its membership functions and inference rules were established. The obtained simulation showed that fuzzy logic is viable in the control of combustion instabilities. The obtained results indicated that the control system responded to pulses in an efficient and desirable way. It was verified that the system needed approximately 0.2 s to increase the tube internal pressure from 30 to 90 mbar, with an assumed total delay of 2 ms. The effects of delay variation were studied. Convergence was always obtained and general performance was not affected by the delay. The controller sends a pressure signal in phase with the Rijke tube internal pressure signal, through the speakers, when an increase the oscillations pressure amplitude is desired. On the other hand, when a decrease of the tube internal pressure amplitude is desired, the controller sends a signal 180º out of phase.

Os Autores descrevem a reação de IgA ELISA na leishmaniose mucocutânea. Títulos acima do normal foram encontrados no primeiro e segundo anos de infecção e em soros de micoses profundas acompanhados por envolvimento mucoso ou doença disseminada.
The Authors describe an anti-Leishmania IgA-ELISA assay in mucocutaneous leishmaniasis. Increased titers were found in leishmaniasis patients, mainly in the first and second year of infection and in deep mycoses patients showing either mucosal involvement or widespread disease.
