Abstract The SARS-CoV-2 coronavirus pandemic calls for coordinated efforts by the scientific community for the development of vaccines. The most advanced strategies have focused on modifications of technologies that were already under development for other viruses, such as SARS, MERS, and even Influenza. Classic and new technologies, such as inactivated and attenuated viruses (non-replicative and replicative), DNA and mRNA vaccines, and nanoparticles containing SARS-CoV-2 antigens, are some of the strategies currently investigated. Although there is a very high expectation for the effectiveness of the most advanced vaccine candidates, there are still no established correlates of protection. Previous experience in vaccine development for other pathogens shows that differences in vaccine formulation can result in diverse immune responses and consequently, different protective properties. Therefore the importance of continuing investigations on a broad range of strategies. Expertise in vaccine development in Brazil was refocused to the new coronavirus. Impressive collaboration between institutions will support further developments until we have available a safe, effective, and economically viable vaccine. Established competence and collaborations will allow preparedness for future challenges and can also be used to address local issues as neglected infectious diseases.

Abstract: The systemic effect of chronic periodontitis (CP) has been suggested by several studies as an etiologic factor and modulator of diseases based on the changes in the inflammatory marker levels. This study aimed to investigate the relationship between the changes in clinical periodontal outcomes and serum biomarkers (CRP, IL-6, albumin and percentage of leukocytes) after non-surgical periodontal therapy in systemically healthy adults. An interventional study was conducted with a sample of 29 individuals without CP (control group) and 33 with CP (CP group). Periodontal clinical variables were recorded, and the serum levels of inflammatory markers were measured. Statistical analysis included the chi-square and Student’s t-tests and Pearson’s correlation analysis. After 90 days of non-surgical periodontal treatment, a reduction of periodontal parameters and IL-6 in both groups could be observed (P < 0.001). The correlation analysis revealed a directly proportional correlation between changes in the probing depth (r = 0.349, P = 0.049) and clinical attachment level (r = 0.374, P = 0.034) with CRP in the CP group. The findings suggest a reduction of IL-6 serum concentration and periodontal clinical measures 90 days after periodontal therapy in both groups.

Hypertension is characterized by a pro-inflammatory status, including redox imbalance and increased levels of pro-inflammatory cytokines, which may be exacerbated after heat exposure. However, the effects of heat exposure, specifically in individuals with inflammatory chronic diseases such as hypertension, are complex and not well understood. This study compared the effects of heat exposure on plasma cytokine levels and redox status parameters in 8 hypertensive (H) and 8 normotensive (N) subjects (age: 46.5±1.3 and 45.6±1.4 years old, body mass index: 25.8±0.8 and 25.6±0.6 kg/m2, mean arterial pressure: 98.0±2.8 and 86.0±2.3 mmHg, respectively). They remained at rest in a sitting position for 10 min in a thermoneutral environment (22°C) followed by 30 min in a heated environmental chamber (38°C and 60% relative humidity). Blood samples were collected before and after heat exposure. Plasma cytokine levels were measured using sandwich ELISA kits. Plasma redox status was determined by thiobarbituric acid reactive substances (TBARS) levels and ferric reducing ability of plasma (FRAP). Hypertensive subjects showed higher plasma levels of IL-10 at baseline (P<0.05), although levels of this cytokine were similar between groups after heat exposure. Moreover, after heat exposure, hypertensive individuals showed higher plasma levels of soluble TNF receptor (sTNFR1) and lower TBARS (P<0.01) and FRAP (P<0.05) levels. Controlled hypertensive subjects, who use angiotensin-converting-enzyme inhibitor (ACE inhibitors), present an anti-inflammatory status and balanced redox status. Nevertheless, exposure to a heat stress condition seems to cause an imbalance in the redox status and an unregulated inflammatory response.

Determinou-se a dose inseminante para fertilização artificial e descreveu-se o desenvolvimento embrionário de tambaqui (Colossoma macropomum). Os gametas foram coletados de reprodutores induzidos hormonalmente. Foi realizada fertilização artificial nas proporções de espermatozoides/ovócito de D1-50.666; D2-75.999; D3-101.332; D4-126.665; D5-151.998. O desenvolvimento embrionário foi acompanhado por meio de observações periódicas em estereoscópio até a eclosão dos ovos. Na fase de fechamento do blastóporo foi calculada a taxa de fertilização nas diferentes doses inseminantes. A porcentagem de fertilização aumentou de forma linear segundo a equação Ŷ =0,050 + 0,00000773X (R²=97,5), atingindo um platô em 84% na proporção de 102.486 espermatozoides/ovócito. Os embriões apresentaram segmentação meroblástica discoidal, típica de ovos telolécitos, com eclosão ocorrendo aos 357 horas-grau após a fertilização. Conclui-se que o desenvolvimento embrionário de tambaqui obedece ao esperado para peixes com ovos telolécitos e recomenda-se o uso da dose inseminante de aproximadamente 100.000 espermatozoides/ovócito na rotina de fertilização artificial dessa espécie.

The objective of this research was to determine the insemination dose for artificial fertilization and describe the embryonic development of tambaqui (Colossoma macropomun). The gametes were collected from induced breeding hormonally. An artificial fertilization was performed with different sperm/oocyte ratios of D1-50666, D2-75999, D3-101 332, 126 665-D4, D5-151 998 sperm/oocyte. Embryonic development was monitored through periodic stereoscopic observations until hatching. When embryos reached the blastopore closure stage, the rate of fertilization in different insemination doses was calculated. A regression equation was estimated to determine the ideal proportion of the gametes. The fertilization rate increased linearly according to the equation Ŷ = 0.050 + 0.00000773 X (R² = 97.5), up to the proportion of 102.486 spermatozoa/oocyte, and, from this point, the fertilization rate was maintained at 84%. The embryonic development of tambaqui was meroblastic discoidal, as expected from telolecithal eggs and we recommend the use of the insemination dose of approximately 100.000 sperm/oocyte in the artificial fertilization of tambaqui.

Acreditou-se durante muito tempo que a espécie caprina era resistente à infecção por Mycobacterium bovis, porém tal hipótese foi desconsiderada quando relatos da enfermidade surgiram em vários países. No entanto, ainda permanecem desconhecidas certas características da tuberculose em caprinos e suas implicações na saúde pública e caprinocultura nacional. Objetivou-se com este trabalho descrever os aspectos nosológicos, radiológicos, anátomo-histopatológicos, baciloscópicos e biomoleculares da tuberculose em caprinos leiteiros com doença respiratória, naturalmente infectados e procedentes do estado de Pernambuco. Para isso foram tuberculinizadas 442 cabras com sintomas respiratórios e, destas, 3,4% (15/442) foram consideradas positivas ao teste. Dos animais positivos, sete foram monitorados clinicamente durante 12 meses, descrevendo-se os achados obtidos. O agente etiológico foi identificado através da reação em cadeia da polimerase, por amplificação de sequências genômicas do Complexo Mycobacterium tuberculosis e posteriormente de Mycobacterium bovis. Este é o primeiro diagnóstico molecular com caracterização do envolvimento do Mycobacterium bovis na tuberculose caprina no Brasil.

For a long time, it was believed that the goat species is resistant to infection by Mycobacterium bovis; however, this hypothesis changed when reports of the disease became apparent in various countries. Nevertheless, certain characteristics of tuberculosis in goats and its impact on public health are still unknown. The objective of this study was to describe nosologic, radiologic, anatomo-histopathological, bacilloscopic and biomolecular aspects of tuberculosis in dairy goats with respiratory disease naturally infected, from the state of Pernambuco, Brazil. When we tuberculinized 442 goats with respiratory symptoms, 3.4% (15/442) of these were considered positive in the test. From the positive goats, seven were monitored clinically for 12 months. The etiological agent was identified through the polymerase chain reaction, by amplification of genomic sequences of the Mycobacterium tuberculosis complex and later Mycobacterium bovis. This is the first molecular diagnosis which characterizes the involvement of Mycobacterium bovis in goat tuberculosis in Brazil.

O presente trabalho foi desenvolvido para verificar a ocorrência e distribuição de Cymbasoma longispinosum Bourne, 1890 em um estuário tropical amazônico da região norte do Brasil. As coletas foram realizadas bimestralmente de julho/2003 a julho/2004 em dois diferentes transectos (rios Muriá e Curuçá) situados ao longo do estuário do Curuçá (Pará, Norte do Brasil). Amostras foram coletadas durante marés de quadratura por intermédio de arrastos com redes de plâncton de 200μm de abertura de malha, através de um pequeno barco a motor (1 a 1,5 knots). Amostras adicionais de água subsuperficial foram coletadas para determinação dos parâmetros ambientais. Machos e fêmeas de C. longispinosum foram observados apenas durante os meses de setembro e novembro/2003. O maior número de organismos foi encontrado em setembro/2003, no transecto do rio Muriá. A presença de C. longispinosum nas amostras obtidas durante setembro e novembro/2003 poderia estar provavelmente relacionada ao período reprodutivo desta espécie no estuário estudado, o qual está diretamente relacionado ao período seco na região. Os mais elevados valores de salinidade, bem como os elevados números de indivíduos observados no mês de setembro/2003 confirmam a suposição anterior, visto que nenhum indivíduo de C. longispinosum foi registrado durante os meses que incluem o período chuvoso (janeiro a junho).

The present work was carried out to verify the occurrence and distribution of Cymbasoma longispinosum Bourne, 1890 in a tropical Amazon estuary from North Brazil. Samplings were performed bimonthly from July/2003 to July/2004 at two different transects (Muriá and Curuçá rivers) situated along the Curuçá estuary (Pará, North Brazil). Samples were collected during neap tides via gentle (1 to 1.5 knots) 200 μm-mesh net tows from a small boat. Additional subsurface water samples were collected for the determination of environmental parameters. Males and females of Cymbasoma longispinosum were only observed during September and November/2003. The highest number of organisms was found in September/2003 at the Muriá River transect. The presence of C. longispinosum in samples obtained during September and November/2003 could probably be related to the reproductive period of this species in the studied estuary, which is directly related to the dry period in the region. The highest salinity values and the highest number of individuals observed in September/2003 corroborate with the previous assumption, since no C. longispinosum was found during the months comprising the rainy period (January to June).

O presente estudo teve como objetivo principal avaliar a variação sazonal na estrutura da comunidade dos copépodos durante os meses de julho, setembro e novembro de 2003 (período seco) e janeiro, março e maio de 2004 (período chuvoso) no estuário do Curuçá, Norte do Brasil. As amostras foram coletadas nas marés de quadratura com auxílio de uma rede deplâncton com 200µm de abertura de malha, rebocada por meio de uma pequena embarcação a motor. As medidas de condutividade da água foram realizadas in situ utilizando-se um condutivímetro eletrônico e a salinidade foi posteriormente obtida através da transformação dos valores de condutividade. Os valores de salinidade variaram sazonalmente de 7, 2 ± 0, 1a 39, 2 ± 1, 8 (média ± desvio padrão), tendo sido principalmente influenciados pelas diferenças nas taxas de precipitação entre os períodos de amostragem estudados. Foram identificados no total 30 táxons, com Acartia tonsa constituindo a espécie mais representativa durante todo o período de estudo,seguida por Acartia lilljeborgii, Subeucalanus pileatus e Paracalanus quasimodo. Durante este trabalho, os valores de densidade, índices ecológicos e dominância das espécies de copépodos apresentaram um padrão sazonal claro, mostrando que a área estudada pode ser considerada sazonalmente heterogênea em relação a estes parâmetros investigados.

The main purpose of this study was to investigate the seasonal variation of copepod community structure during the months of July, September and November 2003 (dry season) and January, March and May 2004 (rainy season) in the Curuçá estuary, northern Brazil. Samples were collected during neap tides via gentle 200µm mesh net tows from a small powerboat. Measurements of surface water conductivity were accomplished in situ using an electronic conductivimeter and salinity was later obtained through the transformation of the conductivity values. Salinity varied seasonally from 7.2 ± 0.1 to 39.2 ± 1.8 (mean ± standard deviation) and was influenced mainly by differences in the amount of rainfall between the studied sampling seasons. In total, 30 Copepoda taxa were identified and Acartia tonsa comprised the most representative species throughout the entire studied period followed by Acartia lilljeborgii, Subeucalanus pileatus and Paracalanus quasimodo. In the present study, the density values, ecological indexes and copepod species dominance presented a clear seasonal pattern, showing that the studied area may be considered seasonally heterogeneous in relation to the investigated parameters.

Happy emotional states have not been extensively explored in functional magnetic resonance imaging studies using autobiographic recall paradigms. We investigated the brain circuitry engaged during induction of happiness by standardized script-driven autobiographical recall in 11 healthy subjects (6 males), aged 32.4 ± 7.2 years, without physical or psychiatric disorders, selected according to their ability to vividly recall personal experiences. Blood oxygen level-dependent (BOLD) changes were recorded during auditory presentation of personal scripts of happiness, neutral content and negative emotional content (irritability). The same uniform structure was used for the cueing narratives of both emotionally salient and neutral conditions, in order to decrease the variability of findings. In the happiness relative to the neutral condition, there was an increased BOLD signal in the left dorsal prefrontal cortex and anterior insula, thalamus bilaterally, left hypothalamus, left anterior cingulate gyrus, and midportions of the left middle temporal gyrus (P < 0.05, corrected for multiple comparisons). Relative to the irritability condition, the happiness condition showed increased activity in the left insula, thalamus and hypothalamus, and in anterior and midportions of the inferior and middle temporal gyri bilaterally (P < 0.05, corrected), varying in size between 13 and 64 voxels. Findings of happiness-related increased activity in prefrontal and subcortical regions extend the results of previous functional imaging studies of autobiographical recall. The BOLD signal changes identified reflect general aspects of emotional processing, emotional control, and the processing of sensory and bodily signals associated with internally generated feelings of happiness. These results reinforce the notion that happiness induction engages a wide network of brain regions.

The research was accomplished in eight dairy water buffalo herds, randomically choosen in Região do Alto São Francisco, State of Minas Gerais, Brazil. Information was collected from March to November, 2003 during 270 days of observation. In order to determine the somatic cell count (SCC) in presence or absence of microbial isolation, 1,393 samples were collected from 285 lactating females and microbiological exams and SCC were done. Samples obtained from udders without evidence of clinical or subclinical inflammation showed infection for a great variety of microbial mastitis pathogens. The low SCC did not necessarily indicate the absence of intramammary infection, suggesting that SCC patterns used for bovine cannot be appropriate in order to control mastitis in buffalo herds.

O diabetes mellitus (DM) é uma doença de alta prevalência nas sociedades modernas, na maioria das vezes com tratamento inadequado ou ausente. Apesar de geralmente considerado como fator de risco independente para ocorrência e gravidade de infecções em geral, o DM não apresenta evidência clínica forte de sua relação com infecção. Observa-se, porém, uma maior ocorrência de certas infecções em pacientes com DM, com curso menos favorável para algumas delas. Há também tipos de infecção quase exclusivos de pacientes com DM. Experimentalmente, observa-se depressão da atividade dos neutrófilos, menor eficiência da imunidade celular, alteração dos sistemas antioxidantes e menor produção de interleucinas. Com relação às infecções comuns, as que envolvem o trato respiratório não têm comprovadamente maior gravidade em pacientes com DM, exceção feita ao pneumococo - por isso a recomendação para sua vacinação contra S. pneumoniae e influenza. Quanto ao trato urinário, há maior ocorrência de bacteriúria assintomática em mulheres com DM, com maiores índices de pielonefrite, necrose papilar, abscesso perinéfrico, pielonefrite xantogranulomatosa, e cistite e pielonefrite gangrenosas. Periodontite e infecções de partes moles são também mais comuns no DM. Cada tipo de infecção é associado a germes típicos, e seu conhecimento é fundamental para um tratamento inicial adequado. As infecções quase exclusivas de pacientes com DM incluem otite externa maligna, mucormicose rinocerebral, colecistite gangrenosa e o somatório de alterações que caracterizam o pé diabético. O conhecimento destas infecções assume maior importância por requererem freqüentemente uma abordagem multidisciplinar, envolvendo endocrinologistas, infectologistas, cirurgiões vasculares e nefrologistas, dentre outros.

Diabetes mellitus (DM) is a highly prevalent disease in modern societies, most of the times being either inappropriately treated or not treated at all. Though generally considered to be an independent risk factor for the occurrence and severity of infections in general, DM has not been clinically proven to have any strong association with infection. There are, however, certain infections that are more common in DM, sometimes with a less favorable course than in non-diabetics. Other types of infection affect almost exclusively patients with DM. Experimentally, DM patients have a depressed neutrophil activity, a less effective cell mediated immunity, altered antioxidant systems and diminished interleukin synthesis. Regarding common infections, those affecting the respiratory tract have not been proven to be more serious in DM patients, with the exception of S. pneumoniae infections - thus the recommendation to vaccinate them against pneumococcus and influenza. As for urinary tract infections, bacteriuria takes place more commonly in women with DM, more frequently complicated by pyelonephritis, papillary necrosis, xanthogranulomatous pyelonephritis and gangrenous cystitis/ pyelonephritis. Periodontitis and soft-tissue infections are also more common in DM. Each specific infection is associated with typical microbes, and their knowledge is essential for an appropriate initial treatment. The nearly exclusively DM-related infections include malignant external otitis, rhinocerebral mucormycosis, gangrenous cholecystitis, and the sum of alterations referred to as "diabetic foot". The general knowledge of these infections assumes great importance since they often require a multidisciplinary approach, involving endocrinologists, infectologists, nephrologists and vascular surgeons, among others.
