Abstract Introduction Obsessive-compulsive disorder (OCD) is the fourth most prevalent mental disorder and is a disabling condition. OCD is associated with anatomical and functional changes in the brain, in addition to dysfunctional cognitions. The treatments of choice are selective serotonin reuptake inhibitors, cognitive-behavioral therapy (CBT), and exposure and response prevention (ERP). Trial-based cognitive therapy (TBCT) is a recent and empirically validated psychotherapy with a focus on restructuring dysfunctional negative core beliefs (CBs). The objective of this study was to evaluate the efficacy of TBCT relative to ERP for treatment of OCD. Methods A randomized, single-blind clinical trial was conducted, randomizing 26 patients for individual treatment with TBCT (n = 12) or ERP (n = 14). The groups were evaluated at baseline, at the end of 3 months (12 sessions), and at 3, 6, and 12-month follow-ups. Results Both approaches reduced the severity of symptoms with large effect sizes. These results were maintained at the 12-month follow-up assessment. Conclusion TBCT may be a valid and promising treatment for this disorder.

Resumo: O óxido nítrico (ON) vem se destacando como estimulador do processo de germinação das sementes, principalmente sob condições de estresse. O objetivo deste trabalho foi avaliar os efeitos fisiológicos e bioquímicos do ON na germinação e no vigor de sementes de ervilha submetidas ao déficit hídrico. Sementes da cv. Itapuã 600 foram semeadas em substrato umedecido com água (controle), solução de PEG 6000 (-0,15 MPa) e em solução de PEG 6000 com nitroprussiato de sódio (SNP) (-0,15 MPa + 50 µM), um doador de ON. Foram avaliadas a germinação, vigor (velocidade de germinação e desenvolvimento de plântulas), atividade de enzimas antioxidantes, espécies reativas de oxigênio, peroxidação de lipídeos e atividade de amilases. A germinação e o vigor das sementes de ervilha foram reduzidos sob o déficit hídrico, mas a aplicação do SNP foi capaz de melhorar o desempenho fisiológico e bioquímico das sementes sob essa condição. O déficit hídrico promove um aumento do estresse oxidativo, mas a aplicação de ON promove maior atividade das enzimas do aparato antioxidativo e das amilases, amenizando os danos causados pelo déficit hídrico durante a germinação e crescimento de plântulas de ervilha.

Abstract: Nitric oxide (NO) has been standing out as a seed germination process stimulator, mainly under stress conditions. This work aimed to evaluate the physiological and biochemical effects of NO on germination and vigor of pea seeds submitted to water deficit. Seeds of cv. Itapuã 600 were sown in moistened substrate with water (control), PEG 6000 solution (-0.15 MPa) and PEG 6000 solution with sodium nitroprusside (SNP) (-0.15 MPa + 50 μM), an NO donor. The germination, vigor (germination speed and seedling development), activity of antioxidant enzymes, reactive oxygen species, lipid peroxidation, and amylase activity were evaluated. Germination and vigor of pea seeds were reduced under water deficit, but the application of SNP was able to improve the physiological and biochemical performance of the seeds under this condition. Water deficit promotes an increase in oxidative stress, but the application of NO promotes greater activity of antioxidative apparatus enzymes and amylases, softening the damage caused by water deficit during seed germination and growth of pea seedlings.

Resumo: O óxido nítrico (ON) pode atuar em rotas bioquímicas do processo germinativo, no entanto, são poucas informações sobre a sua ação no desempenho de sementes de ervilha. O objetivo deste trabalho foi avaliar os efeitos fisiológicos e bioquímicos do ON na germinação e no vigor de sementes de ervilha. Sementes de ervilha cv. Itapuã 600, obtidas de três lotes com diferentes níveis de qualidade fisiológica, foram semeadas em substrato umedecido com água (controle) ou solução de nitroprussiato de sódio (SNP), doadora de NO (50 μM) para avaliação da germinação, vigor, atividade de enzimas antioxidantes, espécies reativas de oxigênio, peroxidação de lipídeos e atividade de amilases. A aplicação de ON não altera a germinação de sementes de ervilha, mas aumenta o vigor, sendo mais eficaz em sementes de menor potencial fisiológico. Além disso, o ON promove redução do estresse oxidativo, favorece a translocação de reservas para o embrião e tem potencial para ser utilizado no tratamento de sementes de ervilha visando o aumento do vigor das sementes.

Abstract: Nitric oxide (NO) can act in biochemical pathways of the germination process; however, there is little information about how it acts on the performance of pea seeds. The aim of this study was to evaluate the physiological and biochemical effects of NO on pea seed germination and vigor. Pea seeds cv. Itapuã 600 obtained from three seed lots with different levels of physiological quality were sown in a substrate moistened with water (control) or sodium nitroprusside (SNP) solution, a NO donor (50 μM), to assess germination, vigor, activity of antioxidant enzymes, reactive oxygen species, lipid peroxidation, and amylase activity. NO application does not alter pea seed germination, but it increases vigor. It is more effective in seeds with lower physiological potential. In addition, NO leads to reduction in oxidative stress, favors the translocation of reserves to the embryo, and has potential for use in the treatment of pea seeds to increase seed vigor.

Resumo:A germinação é um processo complexo que envolve propriedades das moléculas que compõem a parede celular, enzimas hidrolíticas que quebram ligações entre polímeros da pectina e da hemicelulose e ação das substâncias reativas de oxigênio. Melanoxylon brauna é uma espécie florestal de alto valor econômico. Com o objetivo de avaliar alterações fisiológicas e bioquímicas no eixo embrionário durante a germinação, foram quantificados a massa fresca, o comprimento, as atividades das enzimas pectina metilesterase, poligalacturonase, superóxido dismutase, catalase, peroxidase e os teores de peróxido de hidrogênio. Foram avaliados na região micropilar a composição dos carboidratos e a resistência da micrópila seca e sem secagem. Durante a embebição, ocorrem aumentos na massa fresca e comprimento dos eixos embrionários, mesma tendência das enzimas poligalacturonase e pectina metilesterase, que participam da expansão celular. O conteúdo de peróxido de hidrogênio reduziu durante a embebição, assim como a força de ruptura da região micropilar. Conclui-se que o tegumento e os cotilédones são responsáveis por 90% da água embebida pelas sementes. Os eventos na micrópila e eixo ocorrem independentes nas primeiras 16 horas. O enfraquecimento da micrópila apresenta uma etapa elástica e outra plástica. As enzimas pectina metilesterase e a poligalacturonase atuam na expansão celular do eixo embrionário.

Abstract:Germination is a complex process that involves molecules properties that make up the cell walls, hydrolytic enzymes that break the bonds between the polymers and action of reactive oxygen substance. Melanoxylon braunais a forest species of high economic value. In order to evaluate the physiological and biochemical changes that occur in the embryonic axis during germination, fresh matter, length, activities of the enzymes pectin methylesterase, polygalacturonase, superoxide dismutase, catalase, peroxidase and hydrogen peroxide levels were quantified in the embryonic axis. Furthermore, in the micropyle area the composition of carbohydrates and micropyle physical resistance were evaluated with and without drying. During soaking, if there are increases in fresh matter and length of the embryonic axis, there is the same trend of polygalacturonase and pectin methylesterase enzymes. The hydrogen peroxide content was reduced during the soaking, as well as the puncture force of the micropylar area. It is concluded that the seed coat and the cotyledons are responsible for 90% of the water soaked by the seeds. The events in the micropyle and embryonic axis occur independently in the first 16 hours. The weakening of the micropyle features an elastic step and a plastic one. Enzymes pectin methylesterase and polygalacturonase act in cellular expansion of the embryonic axis.

A dormência de sementes é um fenômeno que influencia a distribuição das espécies de vegetais no tempo. Porém, pode retardar a germinação, e consequentemente, dificultar a produção de mudas em viveiros. Muitas sementes de espécies florestais da família Fabaceae, como Libidibia ferrea (pau-ferro), apresentam dormência física causada pela impermeabilidade do tegumento à água. Portanto, o objetivo neste trabalho foi avaliar as características fisiológicas, físicas e morfo-anatômicas das sementes de L. ferrea antes e após aplicação de diferentes tratamentos para superação de dormência. Avaliaram-se as curvas de embebição, a porcentagem de germinação, o índice de velocidade de germinação, a força requerida para ruptura da região micropilar e as alterações morfo-anatômicas por meio de imagens obtidas de microscópio eletrônico de varredura antes e após a aplicação de tratamentos para superação de dormência. Sementes de L. ferrea imersas em ácido sulfúrico apresentam padrão trifásico de embebição. A força de ruptura da região micropilar de sementes de L. ferrea é menor tanto para o tratamento com água fervente quanto para os tratamentos com ácido sulfúrico. Além disso, a força necessária para ruptura da região micropilar diminui durante a embebição das sementes após aplicação de ácido sulfúrico. O tempo de aplicação de ácido sulfúrico influencia na espessura removida da camada de macroesclereídios do tegumento das sementes.

Seed dormancy is a phenomenon that affects the distribution of plant species in time. However, it may slow germination and consequently hinder seedling production in nurseries. Many seeds of forest species of the Fabaceae family, such as Libidibia ferrea (Brazilian ironwood), have physical dormancy caused by impermeability of the seed coat to water. Therefore, the aim of this study was to evaluate the physiological, physical, and morpho-anatomical characteristics of L. ferrea seeds before and after application of different treatments for overcoming dormancy. We evaluated the imbibition curves, germination percentage, germination speed index (GSI), the force required to puncture the micropylar region, and morpho-anatomical changes through images obtained from a scanning electron microscope (SEM) before and after application of treatments for overcoming dormancy. L. ferrea seeds immersed in sulfuric acid show a triphasic pattern of imbibition. The required force to puncture the micropylar region of L. ferrea seeds is less for both the treatment with boiling water and for treatments with sulfuric acid. In addition, the required force to puncture the micropylar region decreases during imbibition of the seeds after application of sulfuric acid. The time of application of sulfuric acid influences the thickness of the material removed from the macrosclereid layer of the seed coat.

A temperatura e a umidade relativa estabelecem as condições fundamentais na conservação de sementes, quer sejam recalcitrantes, quer sejam ortodoxas. A conservação de sementes de braúna sob umidades relativas de 55, 75 e 93% e temperatura de 20ºC foi realizada com os objetivos de avaliar as respostas da germinação e do vigor, relacionando-as à permeabilidade da membrana, atividade de enzimas, teor e peroxidação de lipídios. A qualidade das sementes decresce com o aumento do teor de água do ambiente, com decréscimo também das atividades das enzimas superóxido dismutase e catalase e aumento da atividade da enzima glicose-6-fosfato desidrogenase. A permeabilidade das membranas é ampliada com o aumento da deterioração das sementes. Não há relação entre as alterações no teor e peroxidação de lipídios e a deterioração das sementes.

Temperature and relative humidity set the basic conditions for the preservation of recalcitrant and orthodox seeds. The seed conservation of braúna under relative humidities of 55, 75 and 93% at 20°C was performed with the aim of evaluating germination and vigor responses, relating them to the membrane permeability, enzyme activity, content and lipid peroxidation. Increasing environment water content decreases seed quality and also decreases the activity of the enzymes superoxide dismutase and catalase, and increases activity of glucose-6-phosphate dehydrogenase. The membrane permeability increases with increasing deterioration of the seeds. There is no relationship between the changes in lipids content and peroxidation and seed deterioration.

A germinação de sementes é dependente de fatores abióticos, sendo a temperatura um dos principais, cuja influência, em condições extremas, causa danos às sementes. Este trabalho teve por objetivo investigar o efeito das diferentes temperaturas durante a germinação de Dalbergia nigra e suas implicações na fisiologia das sementes. Avaliaram-se o percentual de germinação, o índice de velocidade de germinação (IVG) e a integridade de membranas celulares pelo teste de condutividade elétrica de sementes em diferentes tempos de exposição às temperaturas de 5, 15, 25 (controle), 35 e 45 ºC. A temperatura de 25 ºC correspondeu à temperatura ideal de germinação. Em temperaturas de 5 e 45 ºC, a germinação foi nula. Houve redução da germinação de sementes de D. nigra com o aumento do tempo de exposição das sementes às temperaturas de 5, 15, 35 e 45 ºC. Diferentemente das demais temperaturas, a semipermeabilidade da membrana não é recuperada nas temperaturas de 5 e 45 ºC. A condutividade elétrica é uma técnica eficiente para avaliar a qualidade fisiológica das sementes em diferentes temperaturas.

The seeds germination depends on abiotic factors, the temperature being one of the most important, whose influence, in extreme conditions, causes damage to the seeds. The present study aimed to investigate the effect of different temperatures during germination of Dalbergia nigra and its implications in the physiology of the seeds. We evaluated the percentage of germination, germination speed index (GSI) and integrity of cell membranes by electrical conductivity test of seeds at different times of exposure to temperatures of 5, 15, 25 (control), 35 and 45 ºC. The temperature of 25 ºC was the ideal temperature for germination. At temperatures of 5 and 45 ºC germination was null. There was a reduction of germination of D. nigra seeds with the increase of the exposure time of the seeds at temperatures of 5, 15, 35 and 45 ºC. Unlike other temperatures, the semipermeability of the membrane is not recovered at temperatures from 5 to 45 ºC. Electrical conductivity is an efficient technique to evaluate the physiological quality of the seeds at different temperatures.

O presente trabalho teve por objetivo investigar o efeito das diferentes temperaturas durante a germinação de sementes de D. nigra e suas implicações fisiológicas e bioquímicas. Avaliaram-se o porcentual de germinação e a produção de ânion superóxido (O2-) e peróxido de hidrogênio (H2O2) em sementes submetidas às temperaturas de 5, 15, 25, 35 e 45 ºC por diferentes tempos. A hidratação é estimulada a 45 ºC e inibida a 5 ºC, não havendo germinação em ambas, enquanto é mínima a 15 ºC e máxima a 25 ºC. A produção de superóxido aumenta nas temperaturas mais altas (25 e 35 ºC) após 72 horas de hidratação, coincidindo com o início da protrusão radicular. A produção de peróxido de hidrogênio decresce em todas as temperaturas, à exceção da de 5 ºC, com valores próximos entre si nas temperaturas de 15, 25 e 35 ºC, onde houve protrusão radicular.

Seed germination is dependent on abiotic factors, temperature being one of the main ones, whose influence causes seed damage under extreme conditions. The aim of the present study was to investigate the effect of different temperatures during germination of D. nigra seeds and their physiological and biochemical implications. We assessed germination percentage and production of superoxide anion (O2-) and hydrogen peroxide (H2O2) in seeds subjected to temperatures of 5, 15, 25, 35 and 45 ºC for different periods of time. Hydration is promoted at 45 ºC and inhibited at 5ºC, without germination in either, whereas it is minimal at 15 °C and at a maximum level at 25 °C. Superoxide production increases at higher temperatures (25 and 35 ºC) after 72 hours of hydration, coinciding with the beginning of radicle protrusion. Production of hydrogen peroxide decreases at all temperatures, except for 5 ºC, with values near each other at temperatures of 15, 25, and 35 ºC, where there was radicle protrusion.

The biological activity of seven extracts from leaves of different Jatropha curcas L (Euphorbiaceae) accessions was evaluated on Spodoptera frugiperda. Methanol extracts were incorporated into an artificial diet and offered to the larval stage of S. frugiperda. The parameters evaluated were length of larval and pupal stages, mortality of larval and total cycle stage, and weight of pupae. The extracts of the EMB accessions showed the best result for larval mortality at 60.00 and 56.67%, compared with the control, respectively. Hexane partition of the methanol extract of the leaves of PM-14 accessions allowed the identification of phytosterols, phytol and n-alkanols.
