ABSTRACT Determining the bioactivity of essential oils extracted from plants is crucial for the development of alternatives for controlling insect pests in stored grains. This study aimed to investigate the bioactivity of Piper aduncum L. essential oil for the control of Sitophilus zeamais (Coleoptera: Curculionidae), Sitophilus oryzae (Coleoptera: Curculionidae) and Cryptolestes ferrugineus (Coleoptera: Cucujidae). Bioassays were conducted to determine the oil toxicity, sublethal effects, attractiveness and flight initiation behavior. Toxicity tests were carried out to determine the lethal concentrations for 50 and 95 % of the insects (LC50 and LC95). Tests were also performed with sublethal exposures to C. ferrurineus, S. oryzae and S. zeamais, using their respective LC5 values (0.0002, 0.097 and 0.11 µL cm-2), to investigate effects on the attraction or repellency activity, as well as flight tests. C. ferrugineus showed greater LC50 (0.0005 µL cm-2) and LC95 (0.0012 µL cm-2) susceptibility, in relation to S. oryzae LC50 (0.35 µL cm-2) and LC95 (1.26 µL cm-2), and S. zeamais LC50 (0.39 µL cm-2) and LC95 (1.40 µL cm-2). The exposure to the sublethal LC5 concentration influenced the behavioral responses of attractiveness/repellency and flight activity. The oil had a neutral effect on C. ferrugineus LC5 (0.0002 µL cm-2) and an attractive effect on S. oryzae and S. zeamais LC5 (0.097 and 0.11 µL cm-2), and can be considered a potential insecticide for controlling pest weevil species of stored grains.
RESUMO Determinar a bioatividade de óleos essenciais extraídos de plantas é primordial para o desenvolvimento de alternativas de controle de insetos-praga em grãos armazenados. Objetivou-se investigar a bioatividade do óleo essencial de Piper aduncum L. para o controle de Sitophilus zeamais (Coleoptera: Curculionidae), Sitophilus oryzae (Coleoptera: Curculionidae) e Cryptolestes ferrugineus (Coleoptera: Cucujidae). Foram realizados bioensaios para determinar a toxicidade do óleo, efeitos subletais, comportamento de atratividade e iniciação de voo. Testes de toxicidade foram realizados, determinando-se as concentrações letais para 50 e 95 % dos insetos (CL50 e CL95). Também foram realizados testes com exposições subletais para C. ferrurineus, S. oryzae e S. zeamais, utilizando-se as suas respectivas CL5 (0,0002; 0,097; e 0,11 µL cm-2), para averiguar efeitos na atividade de atração ou repelência, e testes de voo. C. ferrugineus apresentou maior susceptibilidade CL50 (0,0005 µL cm-2) e CL95 (0,0012 µL cm-2), em relação a S. oryzae CL50 (0,35 µL cm-2) e CL95 (1,26 µL cm-2) e S. zeamais CL50 (0,39 µL cm-2) e CL95 (1,40 µL cm-2). A exposição à concentração subletal CL5 influenciou nas respostas comportamentais de atratividade/repelência e atividade de voo das espécies. O óleo apresentou efeito neutro para C. ferrugineus CL5 (0,0002 µL cm-2) e atrativo para S. oryzae e S. zeamais CL5 (0,097 e 0,11 µL cm-2), podendo ser considerado potencial inseticida para o controle de espécies de gorgulhos-praga de grãos armazenados.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

ASSESSMENT OF SOIL LOSS SUSCEPTIBILITY IN SANTA RITA WATERSHED IN SOUTHERN BRAZIL

ABSTRACT Estimation of soil loss susceptibility is of great importance for the management of watersheds. Thus, several models for soil loss prediction have been proposed. This study estimated the total annual soil loss for the Santa Rita watershed, located in southern Brazil, using the Revised Universal Soil Loss Equation. In addition, a classification to soil loss index (ISL) was proposed to identify regions with critical soil loss values. Altitude, slope, land use, and soil class data were applied to the model, in addition to spatial information for 78 soil samples collected within the study area. It was found that there is an average annual loss of 35.94 Mg ha-1 year-1, with the most substantial loss occurring in areas with predominantly exposed soil and annual cropping. Furthermore, the ISL values indicated that approximately 50% of the study area is experiencing erosion estimates above the tolerable limit. Results emphasize the need for changes in conservation and application practices present in the watershed, considering land use and soil bearing capacity.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Anti-Ehrlichia properties of the dichloromethane fraction of Ageratum conyzoides associated with doxycycline: In vitro study

RESUMO: O crescente número de casos de erliquiose canina por Ehrlichia canis em hospitais e clínicas veterinárias tem demonstrado a necessidade de um novo protocolo de medicamentos para essa doença. A doxiciclina é usada para tratar a erliquiose, mas a resistência do microrganismo a esse protocolo de tratamento, bem como os diversos efeitos colaterais para os animais, tornou-se uma preocupação. Vários estudos têm demonstrado interação positiva com extratos de plantas e fármacos, que permitem a redução da concentração necessária para produzir o efeito desejado, minimizando os efeitos adversos. Este estudo determinou a eficiência da combinação da fração diclorometânica (DCM) de Ageratum conyzoides L. com atividade anti-Ehrlichia canis associada com doxiciclina por meio do ensaio de Checkerboard. O material vegetal foi coletado em São Luís, Maranhão, nordeste do Brasil, seguido pela extração em MeOH:H2O (8:2) e partição da fração diclorometânica. Após a determinação da Concentração Inibitória Mínima (CIM) da fração em estudo frente às células DH82 infectadas com Ehrlichia canis, a mesma foi combinada com a doxiciclina para derivação do Índice de Concentração Inibitória da Fração (Índice CIF). Observou-se uma redução de 5,83 vezes a concentração inibitória mínima da doxiciclina mostrando que esta fração de A. conyzoides, composta predominantemente por lignanas identificadas por espectrometria de massas, notavelmente intensificou a atividade desse fármaco contra E. canis, resultando em um efeito sinérgico.
ABSTRACT: The increasing number of cases of canine ehrlichiosis caused by Ehrlichia canis in hospitals and veterinary clinics has demonstrated the need for a new drug protocol for this disease. Doxycycline is used to treat ehrlichiosis, but the resistance of the microorganism to this treatment protocol, as well as the various side effects to the animals, has become a concern. Several studies have shown a positive interaction with extracts of plants and drugs, which allow for the reduction of the concentration necessary to produce the desired effect, minimizing adverse effects. This study determined the efficiency of the combination of the dichloromethane (DCM) fraction of Ageratum conyzoides L. with anti-Ehrlichia activity and doxycycline by using the checkerboard assay. Plant material was collected in São Luís, northeastern Brazil, followed by extraction in MeOH: H2O (8:2) and partitioning of the DCM fraction. After determining the Minimum Inhibitory Concentration (MIC) of the fraction under study against DH82 cells infected with Ehrlichia canis, it was combined with doxycycline to derive the Fractional Inhibitory Concentration Index (CIF Index). A reduction of 5.83 times the doxycycline minimum inhibitory concentration was observed, showing that this fraction of A. conyzoides composed predominantly by the class of lignans, identified by mass spectrometry notably intensified the activity of doxycycline against E. canis, resulting in a synergistic effect.

ABSTRACT Objective Polycystic ovary syndrome (PCOS) is a recognized risk factor for nonalcoholic fatty liver disease (NAFLD). The aims of this study were to investigate the prevalence and factors associated with NAFLD in women with PCOS and evaluate noninvasive indices of hepatic fibrosis in patients with PCOS and NAFLD. Subjects and methods Patients with PCOS (n = 87) and women without PCOS (n = 40; controls) were included. NAFLD was diagnosed by abdominal ultrasonography after exclusion of alcohol consumption and viral or autoimmune liver disease. Anthropometric, clinical and metabolic variables, homeostasis model assessment of insulin resistance (HOMA-IR) index, lipid accumulation product (LAP), FIB-4 index, NAFLD score, and transient elastography (TE; FibroScan) were obtained in subsets of patients with PCOS and NAFLD. Results A total of 87 patients with PCOS were included (mean age: 34.4 ± 5.7 years, mean body mass index [BMI]: 34.7 ± 4.7 kg/m 2 ). NAFLD was present in 67 (77.0%) patients with PCOS versus 21 of 40 (52.5%) controls (p = 0.005). Women with PCOS and liver steatosis, compared with their NAFLD-free counterparts, had higher values of BMI, waist circumference, triglycerides, total cholesterol, alanine and aspartate aminotransferases, and γ-glutamyltransferase, along with higher frequencies of obesity, metabolic syndrome, and insulin resistance. NAFLD was independently associated with waist circumference, serum triglycerides, and alanine aminotransferase levels. The FIB-4 index was not compatible with advanced fibrosis in any of the evaluated patients, while NAFLD score and TE were compatible with advanced liver fibrosis in 1 of 26 (3.8%) and 3 of 25 (12%) patients, respectively. Conclusion Women with PCOS had a high risk of NAFLD, and a combination of both was associated with central obesity, dyslipidemia, insulin resistance, and metabolic syndrome. Noninvasive methods suggested low rates of severe hepatic fibrosis in Brazilian women with PCOS. Arch Endocrinol Metab. 2020;64(3):235-42

In this study, a novel type of tris(2-acyl-1,3-indandione)-aluminum(III) coordination compounds of the general formula [Al(acind)3]H2O, where 2-acyl-1,3-indandione (acind), 2-acetyl-1,3-indandione (aind), 2-benzoyl-1,3-indandione (bind), and 4-methyl-2-benzoyl-1,3-indandione (mbind), were synthesized and characterized by elemental analysis (CHN), Fourier transform infrared (FTIR) and nuclear magnetic resonance (1H NMR) spectroscopies, thermal analysis (TG/DTG and DTA), and optical absorption spectroscopy in the UV-Vis region. These compounds present remarkably high green luminescence in powder and in thin-film forms. However, when these compounds are applied in glass/ITO/β-NPB/spiro-2CBP/[Al(acind)3]/Al and glass/ITO/β-NPB/[Al(acind)3]/LiF/Al electroluminescent devices, where spiro-2CBP is 2,7-bis(carbazol-9-yl)-9,9-spirobifluorene and β-NPB is N,N’-bis(naphthalen-2-yl)-N,N’-bis(phenyl)-benzidine, the emission color tuned from green to red, reflecting a change from the direct charge recombination in the emitting layer of the [Al(acind)3] complexes to an exciplex-based emission in which [Al(acind)3] complexes and spiro 2-CBP acted as acceptor and donor, respectively. These results suggest that [Al(acind)3] complexes have potential applications as molecular light converter materials for fabricating new electroluminescent devices.

Resumo Três espécies de pargos de pescaria artesanal e de pequena escala foram examinadas quanto à presença de parasitas. Um total de 139 peixes lutjanídeos foram coletados: 69 espécimes de Lutjanus analis, 47 de Lutjanus vivanus e 23 de Lutjanus synagris. Cistos de plerocercóides de cestodas Trypanorhyncha encontrados durante a necrópsia nas vísceras de pargos foram amostrados para análise parasitológica. Estas larvas plerocercóides pertencem aos gêneros Floriceps, Pseudogrillotia e Oncomegas. Embora esses parasitas não tenham qualquer potencial zoonótico, peixes com elevadas cargas de plerocercóides são rejeitados durante a inspeção, pois comprometem a higiene da carne. Os achados sugerem que baixas infecções por essas formais larvais de cestodas em lutjanídeos não levariam a rejeição do consumidor quanto ao produto alimentar. A ocorrência de Floriceps sp. em L. analis e em L. synagris, de Pseudogrillotia sp. em L. vivanus e em L. syngaris, e de Oncomegas sp. em L. synagris são novas descobertas nestas espécies de peixes. Estes parasitas são novos para essas espécies de peixes e não foram relatados em pargos da costa nordeste do Brasil. Os resultados contribuem para o conhecimento existente sobre a fauna parasitária do peixe e sua distribuição ao longo do litoral do Brasil.
Abstract Three species of snappers from artisanal, small-scale fisheries were examined for the presence of parasites. A total of 139 lutjanid fish were collected: 69 specimens of Lutjanus analis, 47 specimens of Lutjanus vivanus, and 23 specimens of Lutjanus synagris. Encysted plerocercoid of trypanorhynch cestodes found at necropsy in the viscera of snappers were sampled for parasitological analysis. These plerocercoid larvae belong to the genera Floriceps, Pseudogrillotia , and Oncomegas. Although these parasites do not have any zoonotic potential, fish with heavy loads of plerocercoids are rejected during inspection as they compromise meat hygiene. Our findings suggest that light larval cestode infections in lutjanids would not lead to consumer rejection of the food product. The occurrence of Floriceps sp. in L. analis and in L. synagris, of Pseudogrillotia sp. in L. vivanus and in L. synagris , and of Oncomegas sp. in L. synagris are new findings in these fish species. These parasites are new to these fish species and have not been reported in snappers from the northeast coast of Brazil. The results contribute to the existing body of knowledge about the parasitic fauna of fish and its distribution along the coastline of Brazil.

Abstract Background: Cnidarians produce toxins, which are composed of different polypeptides that induce pharmacological effects of biotechnological interest, such as antitumor, antiophidic and anti-clotting activities. This study aimed to evaluate toxicological activities and potential as antitumor and antiophidic agents contained in total extracts from five cnidarians: Millepora alcicornis, Stichodactyla helianthus, Plexaura homomalla, Bartholomea annulata and Condylactis gigantea (total and body wall). Methods: The cnidarian extracts were evaluated by electrophoresis and for their phospholipase, proteolytic, hemorrhagic, coagulant, fibrinogenolytic, neuromuscular blocking, muscle-damaging, edema-inducing and cytotoxic activities. Results: All cnidarian extracts showed indirect hemolytic activity, but only S. helianthus induced direct hemolysis and neurotoxic effect. However, the hydrolysis of NBD-PC, a PLA2 substrate, was presented only by the C gigantea (body wall) and S. helianthus. The extracts from P. homomalla and S. helianthus induced edema, while only C gigantea and S. helianthus showed intensified myotoxic activity. The proteolytic activity upon casein and fibrinogen was presented mainly by B. annulata extract and all were unable to induce hemorrhage or fibrinogen coagulation. Cnidarian extracts were able to neutralize clotting induced by Bothrops jararacussu snake venom, except M. alcicornis. All cnidarian extracts were able to inhibit hemorrhagic activity induced by Bothrops moojeni venom. Only the C. gigantea (body wall) inhibited thrombin-induced coagulation. All cnidarian extracts showed antitumor effect against Jurkat cells, of which C. gigantea (body wall) and S. helianthus were the most active; however, only C. gigantea (body wall) and M. alcicornis were active against B16F10 cells. Conclusion: The cnidarian extracts analyzed showed relevant in vitro inhibitory potential over the activities induced by Bothrops venoms; these results may contribute to elucidate the possible mechanisms of interaction between cnidarian extracts and snake venoms.

Resumo Este estudo relata Kudoa spp. (Myxozoa, Multivalvulida) de espécies de peixes Lutjanus analis, Bagre marinus, Aspistor luniscutis e Lutjanus jocu que foram capturados na costa litorânea de Aracaju, Estado de Sergipe, Brasil. Os parasitas formavam plasmódios ovais rodeando o esófago em L. analis, enquanto que nas espécies B. marinus, A. luniscutis e L. jocu formavam plasmódios alongados no interior das fibras musculares. Fenômenos de liquefação do músculo do hospedeiro não foram observados. O presente estudo proporciona uma descrição morfológica e morfométrica de cada parasita, sendo feita uma comparação com todas as espécies conhecidas em todo mundo. A inexistência de dados moleculares prejudica a identificação específica dos parasitas. A importância destes parasitas é discutida e a necessidade de mais estudos relacionados a infecções em peixes brasileiros é enfatizada por causa do alto impacto econômico de algumas espécies de Kudoa que causam liquefação nos músculos dos hospedeiros e tornam estes peixes impróprios para consumo.
Abstract This study reports on Kudoa spp. (Myxozoa, Multivalvulida) from the fish species Lutjanus analis, Bagre marinus, Aspistor luniscutis and Lutjanus jocu, which were caught in Aracaju, state of Sergipe, Brazil. The parasites formed oval plasmodia around the esophagus of L. analis, and elongated plasmodia inside the skeletal muscle of B. marinus, A. luniscutis and L. jocu. Host myoliquefaction was not observed in all the cases studied. The current study provides a morphological and morphometric description of each parasite as well as a comparison with all the species described worldwide. Lack of molecular data impaired specific identification of the parasites. The importance of these parasites is discussed and the need for further studies on infections in Brazilian fish is emphasized because of the high economic impact of some Kudoa species which cause liquefaction in hosts’ muscles and render these fish unsuitable for consumption.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A biflorina é uma o-naftoquinona com comprovados efeitos citotóxicos frente a células tumorais além de atividades antimicrobiana, antitumoral e antimutagênica. A biflorina é um composto isolado das raízes da planta Capraria biflora L.(Schrophulariaceae), originária das Antilhas e da América do Sul, que está localizada em zonas temperadas e tropicais. Este composto mostrou uma potente atividade contra microorganismos gram-positivos e bactérias álcool-ácido resistentes. Tem mostrado eficiência em inibir a proliferação celular em linhagens tumorais CEM, HL-60, B16, HCT-8, MCF-7. Recentemente, a linhagem tumoral SK-Br3 foi tratada com biflorina e demonstrou importante efeito citotóxico. Neste artigo, relatamos informações relacionadas com os primeiros estudos de caracterização estrutural, bem como os últimos relatos sobre a atividade biológica desta molécula.

(Schrophulariaceae), indigenous of the West Indies and South America, which is located in temperate or tropical areas. This compound has shown to be strongly active against grampositive and alcohol-acid-resistant bacteria. It has been efficient in inhibiting the proliferation tumor cell lines CEM, HL-60, B16, HCT-8 and MCF-7. Recently, SK-Br3 cell line was treated with biflorin showing important cytotoxic effects. In this article, information related to the first structural characterization studies are presented, as well as the latest reports concerning the biological activity of this molecule.

O principal vetor de leishmaniose visceral (LV) no Brasil é Lutzomyia longipalpis. Entretanto, a ausência de L. longipalpis em área com casos autóctones de LV demonstra a existência de outras espécies na transmissão dessa doença. Estudo realizado na cidade de La Banda, Argentina, e São Vicente Férrer, Brasil, correlacionou a ausência de L. longipalpis e a presença de Lutzomyia migoneicom casos autóctones de LV. Em São Vicente Férrer, foi comprovada a infecção natural de L. migonei por Leishmania infantum chagasi. Dessa forma, o objetivo deste trabalho foi avaliar a ecologia dos flebotomíneos L. longipalpis e L. migonei no município de Fortaleza, área endêmica para LV. A captura de flebotomíneos foi realizada em 22 pontos de coleta distribuídos nas quatro regiões do município de Fortaleza. No total, foram capturados 32.403 flebotomíneos. Destes, 18.166 (56%) eram da espécie L. longipalpis e 14.237 (44%) eram L. migonei. Houve diferença significativa de densidade entre os vetores em cada local de captura (intra e peri) (p<0,0001). Esses achados confirmam que, na cidade de Fortaleza, L. migonei e L. longipalpis estão bem distribuídos, bem como adaptados ao ambiente intradomiciliar e que L. migonei, possivelmente, compartilhe com L. longipalpis o papel de vetor da LV em Fortaleza.
The main vector for visceral leishmaniasis (VL) in Brazil is Lutzomyia longipalpis. However, the absence of L. longipalpis in a region of autochthonous VL demonstrates the participation of other species in the transmission of the parasite. Studies conducted in La Banda, Argentina, and São Vicente Férrer, Pernambuco State, Brazil, have correlated the absence of L. longipalpisand the presence of L. migonei with autochthonous cases of VL. In São Vicente Férrer, Pernambuco, there was evidence for the natural infection of L. migonei with Leishmania infantum chagasi. Thus, the objective of this work was to assess the ecology of the sand flies L. longipalpis and L. migonei in Fortaleza, an endemic area for VL. Insect capture was conducted at 22 sampling points distributed across four regions of Fortaleza. In total, 32,403 sand flies were captured; of these, 18,166 (56%) were identified as L. longipalpis and 14,237 (44%) as L. migonei. There were significant density differences found between the vectors at each sampling site (indoors and outdoors) (p <0.0001). These findings confirm that L. migonei and L. longipalpis are distributed throughout Fortaleza, where they have adapted to an indoor environment, and suggest that L. migonei may share the role as a vector with L. longipalpis in the transmission of VL in Fortaleza.

O objetivo foi realizar um estudo epidemiológico para determinar as áreas de risco de transmissão de leishmaniose visceral pela detecção e quantificação de infecções naturais por Leishmania infantum em Lutzomyia longipalpis. As coletas foram realizadas entre fevereiro de 2009 e janeiro de 2010 em 21 locais, distribuídos em quatro regiões do município de Fortaleza. As amostras foram testadas quanto à presença de DNA de Leishmania por PCR em tempo real (qPCR). Dos 123 pools de flebotomíneos investigados, 45 foram positivos para L. infantum, e a taxa de infecção mínima foi de 3,7%. Nas regiões Norte, Sul, Leste e Oeste, a prevalência de flebotomíneos infectados foi de 3,4%, 4,7%, 4,9% e 8,4%, respectivamente. A carga de parasitas nos pools variou de 2,45 ± 0,96 a 2.820.246 ± 106.072. Não foram observadas diferenças significativas na frequência de flebotomíneos infectados entre as regiões (P = 0,3014), estação do ano (P = 0,3906) ou localização da armadilha (P = 0,8486). Foram observadas diferenças significativas na frequência de flebotomíneos somente na região oeste durante a estação chuvosa (P = 0,0152). A qPCR foi capaz de detectar e quantificar L. infantum em L. longipalpis, identificando as áreas de maior risco de transmissão de leishmaniose visceral.
The objective of this study was to perform an epidemiological survey to determine the areas at risk of visceral leishmaniasis through the detection and quantification of natural infection by Leishmania infantumin Lutzomyia longipalpis. The sandflies were captured between February 2009 and January 2010, at 21 sites in four regions of the Fortaleza municipality. Samples were screened for the presence of Leishmania DNA by Real Time PCR (qPCR), amplification of kDNA minicircle sequence. Out of the 123 pools of analyzed sandflies, 45 were positive for L.infantum, and the minimum infection rate was 3.7%. In the north, south, east and west regions, the pool screen assay predicted sand-fly infection prevalence of 3.4%, 4.7%, 4.9% and 8.4%, respectively. The parasite load ranged from 2.45 ± 0.96 to 2,820,246 ± 106,072. No statistical differences were found with respect to the frequency of sand-fly infection between the regions (P=0.3014), seasons (P = 0.3906) or trap locations (P = 0.8486). Statistical differences were found with respect to the frequency of sand-fly infection between the two seasons only in the west region (P=0.0152). The qPCR was able to detect and quantify L. infantum in L. longipalpis, therefore succeeding in identifying the areas of greatest risk of VL transmission.

INTRODUÇÃO: Nos últimos anos, a leishmaniose visceral, um importante problema de saúde pública, vem apresentando expansão das áreas rurais para as urbanas de muitas regiões do Brasil, incluindo-se Aracaju, capital do Estado de Sergipe. No entanto, não existem estudos sobre a fauna de flebotomíneos presente nesse município ou de sua distribuição ao longo do ano. MÉTODOS: As coletas de flebotomíneos foram realizadas em uma área rural de Aracaju, capital do Estado de Sergipe no periodo de setembro de 2007 a julho de 2009. Armadilhas CDC modificadas acopladas com luz ultravioleta (UV) foram utilizadas para avaliar a distribuição mensal e a presença de flebotomíneos nos ambientes doméstico e peridoméstico. RESULTADOS: Lutzomyia longipalpis foi a espécie mais abundante (90,4%), seguida por Evandromyia lenti (9,6%). Os locais com as maiores quantidades de L. longipalpis (51,1%) foram um galinheiro e as casas mais próximas a ele. Houve uma correlação positiva entre a precipitação mensal e a abundância de L. longipalpis. CONCLUSÕES: Lutzomyia longipalpis é a espécie mais abundante e, provavelmente, o principal vetor do agente da leishmaniose visceral na área rural de Aracaju. Foi observado um aumento na frequência de L. longipalpis nos meses chuvosos. As coletas realizadas no ambiente externo e interno dos domicílios reforçam a relevância dos galinheiros para a presença de L. longipalpis no ambiente peridoméstico. O grande número de L. longipalpis dentro das casas confirma o comportamento endofílico dessa espécie e a possibilidade de transmissão intradomiciliar da leishmaniose visceral.
INTRODUCTION: In recent years, visceral leishmaniasis, a major public health problem, has been spreading from the rural to urban areas in many areas of Brazil, including Aracaju, the capital of the State of Sergipe. However, there are no studies of the sandfly fauna in this municipality or its variation over the year. METHODS: Phlebotomine sandflies were collected from a rural area of Aracaju from September 2007 to July 2009. Modified CDC ultra-violet (UV) light traps were used to evaluate sandfly monthly distribution and their presence in the domestic and peridomestic environments. RESULTS: The most abundant species was Lutzomyia longipalpis (90.4%) followed by Evandromyia lenti (9.6%). A chicken shed trap site had the highest proportion of L. longipalpis (51.1%) and large numbers of L. longipalpis were also collected in the houses closest to the chicken shed. There was a positive correlation between monthly rainfall and L. longipalpis abundance. CONCLUSIONS: Lutzomyia longipalpis is the most abundant species and is probably the main vector of the visceral leishmaniasis agent in the rural area of Aracaju. An increase in L. longipalpis frequency was observed during the rainy season. The peridomicile-intradomicile observations corroborate the importance of chicken sheds for the presence of L. longipalpis in the peridomestic environment. The great numbers of L. longipalpis inside the houses confirm the endophilic behaviour of this species and the possibility of visceral transmission in the intradomicile.
