Anaerobically Treated Leachate from a Composting Plant: Characterization and Evaluation as a Biofertilizer

Leachate from a composting plant was characterized and applied as a biofertilizer in lettuce crops. Characteristics of untreated (UE) and anaerobically treated (TE) leachate samples were compared. The pH of TE (8.2) increased in relation to the UE (5.2) due to an increase in ammonia nitrogen in TE (1197 mg L-1) compared to UE (859 mg L-1). Anaerobic treatment was efficient in the removal of organic matter from the leachate, evaluated by the decrease of dissolved organic carbon, biochemical oxygen demand, total organic carbon and total solids. K presented the highest concentration (1743 mg L-1) in TE, followed by Mg (135 mg L-1). Cd, Pb and Cr were present at low concentrations in the samples, 0.047, 0.206 and 0.081 mg L-1, respectively. Salmonella, thermotolerant coliforms and viable helminth eggs were not present in TE, which was applied as a biofertilizer in a lettuce crop and compared to mineral fertilization based on fresh matter and dry matter production. Lettuce production using TE was statistically equivalent to mineral fertilization. Toxic metals were not detected in the lettuce shoots. It was concluded that the anaerobically treated leachate from the composting company has the potential to be used as a biofertilizer.

Abstract The iAtlantic Project has established an international collaborative strategy to improve mapping and characterization of deep and open ocean ecosystems in understudied regions of the Atlantic and evaluate their health. In December 2022, the first iAtlantic expedition in the South Atlantic set off to map and explore seafloor ecosystems in the Santos Basin slope (200-1,000 m depths) in collaboration with the Petrobras ‘Santos Basin - Regional Characterization Project.’ The 17-day ‘iAtlantic_BR10-Petrobras’ cruise was conducted on board the research vessel NPqHOc Vital de Oliveira (Brazilian Navy) and performed (a) water column structure characterization, (b) seafloor morphology mapping, (c) description of benthic habitats and communities by seafloor imagery and biological/ geological sampling, and (d) ex-situ experiments to assess the functioning of sedimentary ecosystems and their responses to climate-related environmental changes. This study describes the rationale behind the iAtlantic_BR10-Petrobras cruise science plan, reports its sampling strategy and methods, and summarizes its collected data and preliminary results.

Abstract Fibrinolytic proteases are a promising alternative in the pharmaceutical industry, they are used in the treatment of cardiovascular diseases, especially thrombosis. Microorganisms are the most interesting source of fibrinolytic proteases. The aim of this study was the production of fibrinolytic protease from Streptomyces parvulus DPUA 1573, the recovery of the protease by aqueous two-phase system and partial biochemical characterization of the enzyme. The aqueous two-phase system was performed according to a 24-full factorial design using polyethylene glycol molar mass, polyethylene glycol concentration, citrate concentration and pH as independent variables. It was analyzed the effect of different ions, surfactants, inhibitors, pH and temperature on enzyme activity. The best conditions for purifying the enzyme were 17.5% polyethylene glycol 8,000, 15% Phosphate and pH 8.0, it was obtained a partition coefficient of 7.33, a yield of 57.49% and a purification factor of 2.10-fold. There was an increase in enzyme activity in the presence of Fe2+ and a decrease in the presence of $\beta$-Mercaptoethanol, phenylmethylsulfonyl fluoride and Iodoacetic acid. The optimum pH was 7.0 and the optimum temperature was 40 ºC. The purified protease exhibited a molecular mass of 41 kDa. The fibrinolytic protease from Streptomyces parvulus proved to be a viable option for the development of a possible drug with fibrinolytic action.

The present study investigated the activity of pyrimidine derivatives against Aedes aegypti. Two compounds, 3c and 3d showed excellent larvicide activity. Additionally, quantitative structure-activity relationship (QSAR) models were built using multiple-linear regression and partial least squares with descriptors generated from Dragon and VolSurf+ software, respectively. The best model is obtained with multiple linear regression (MLR), leading to a robust model. Moreover, the QSAR model is validated by means of some internal validation techniques in order to check its reliability, quality and robustness for predicting the larvicidal activity against A. aegypti. The models confirmed that the three-dimensional structure of molecules, steric properties, hydrophobic polar surface area, log partition (logP) and a simple pattern of substituent groups as methyl, methoxy, and succinimide in the pyrimidine derivatives are responsible for the larvicidal activity of the pyrimidine derivatives. Even more, the activity decreases by an electron-withdrawing group in R1 and increases when it is replaced by an aromatic ring activator group. These findings will aid in further studies of new pyrimidine derivatives active against Aedes aegypti.

Resumo Devido à importância ecológica de Lophiosilurus alexandri, o presente trabalho avaliou sua representatividade genética, comparando estoques selvagens com plantéis de reprodutores de três larviculturas ao longo do Rio São Francisco. Noventa e sete amostras foram genotipadas com marcadores microssatélites recém-desenvolvidos. Baixos níveis de diversidade genética (número médio de alelos de 4,2) foram detectados em todos os casos, sendo mais severo no cativeiro. Os valores de FST e DEST par a par, as análises do Structure e DAPC mostraram a estruturação dos animais selvagens em dois grupos, e um terceiro formado pelas larviculturas, evidenciando a necessidade de adoção de critérios genéticos para retenção da diversidade genética no cativeiro. Por essa razão, três famílias de irmãos completos foram construídas para selecionar o melhor estimador de parentesco para a espécie e estabelecer os valores mínimos de corte para evitar cruzamentos indesejados. Dois estimadores, Wang (RW) e Lynch & Li (RLL), foram eficientes em refletir as relações de parentesco para irmãos completos nessa espécie. Segundo eles, menos de 50% dos potenciais cruzamentos são recomendáveis nas três larviculturas. A baixa diversidade genética inerente ao L. alexandri destaca a importância de minimizar a consanguinidade e evitar perda de diversidade genética, visando a recuperação da espécie.
Abstract Due to the ecological importance of Lophiosilurus alexandri, the present work evaluated its genetic representativeness by comparing wild stocks to broodstocks that were kept at three restocking hatcheries along the São Francisco River. A total of 97 samples were genotyped for newly developed microsatellite markers. Low levels of genetic diversity (average alleles number of 4.2 alleles) were detected in all cases, being more severe in captive groups. Significant pairwise FST and DEST values, Structure, and DAPC analyses showed that wild animals were structured in two groups, and a third group was formed by captive animals, evidencing the need to adopt genetic criteria to retain genetic diversity in the hatcheries. For this reason, three full-sib families were constructed to select the best relatedness estimator for L. alexandri and establish a cut-off value aimed to avoid full-sibling matings in the hatcheries. Two estimators, Wang (RW) and Lynch & Li (RLL), were accurate in reflecting the relatedness level for full-sibs in this species. According to them, less than 50% of the potential breeding matings in the three hatcheries are advisable. The innate low diversity of L. alexandri highlights the importance of minimizing inbreeding and retaining genetic diversity towards the species recovery.

RESUMO O crescent arsenal terapêutico na esclerose múltipla (EM) tem permitido tratamentos mais efetivos e personalizados, mas a escolha e o manejo das terapias modificadoras da doença (TMDs) tem se tornado cada vez mais complexos. Neste contexto, especialistas do Comitê Brasileiro de Tratamento e Pesquisa em Esclerose Múltipla e do Departamento Científico de Neuroimunologia da Academia Brasileira de Neurologia reuniram-se para estabelecer este Consenso Brasileiro para o Tratamento da EM, baseados no entendimento de que neurologistas devem ter a possibilidade de prescrever TMDs para EM de acordo com o que é melhor para cada paciente, com base em evidências e práticas atualizadas. Por meio deste documento, propomos recomendações práticas para o tratamento da EM, com foco principal na escolha e no manejo das TMDs, e revisamos os argumentos que embasam as estratégias de tratamento na EM.
ABSTRACT The expanding therapeutic arsenal in multiple sclerosis (MS) has allowed for more effective and personalized treatment, but the choice and management of disease-modifying therapies (DMTs) is becoming increasingly complex. In this context, experts from the Brazilian Committee on Treatment and Research in Multiple Sclerosis and the Neuroimmunology Scientific Department of the Brazilian Academy of Neurology have convened to establish this Brazilian Consensus for the Treatment of MS, based on their understanding that neurologists should be able to prescribe MS DMTs according to what is better for each patient, based on up-to-date evidence and practice. We herein propose practical recommendations for the treatment of MS, with the main focus on the choice and management of DMTs, as well as present a review of the scientific rationale supporting therapeutic strategies in MS.

The removal of the beta-lactam antibiotics (ceftriaxone and cefadroxil) through zero-valent copper nanoparticle (nZVC) was studied in this work. Excellent removal degrees (> 85%) were obtained for both analytes in only 20 min of reaction. Studies were performed in both oxic and anoxic conditions, and in the presence of t-butyl alcohol (TBA), an inhibitor of radicals. The results did not show significant changes. Therefore, the hydroxyl radicals are not the main species responsible for the removal. Total organic carbon cefadroxil analysis indicated a removal of 57% after 180 min of reaction. Studies involving Cu+ indicated that probably these are the principal species responsible for the removal of antibiotics. Kinetic studies have shown that two-phase reaction occurred in the antibiotics removal process and both phases followed pseudo-first order kinetic model. The first mechanism is related to the antibiotics degradation by Cu+ species and the second mechanism is related to the antibiotics adsorption by hydroxides/oxides of Cu2+ species.

Lupeol, a triterpene frequently found in Asteraceae plant species, showed moderate to low activity in different strains of Plasmodium falciparum, the most virulent malaria etiological agents. In this work, lupeol was isolated from Parahancornia fasciculata, a plant that is used to treat malaria in the Amazonia region. In the search of more activity lupeol derivatives, five new 1,2,3-triazole hybrid molecules were synthetized by copper-catalyzed azide-alkyne cycloaddition. The antiplasmodial activity of the semi-synthetic compounds were evaluated by the lactate dehydrogenase assay; the lupeol propargyl ether was the only one to disclosing increased activity (half maximal inhibitory concentration-IC50-62.0 ± 1.92 µmol L-1) in relation to lupeol (IC50 117.00 µmol L-1). Therefore, this work revealed a new class of interesting lupeol derivatives that can be obtained by linking electron donors to the hydroxy group at C-3.

Resumo Objetivou-se verificar a qualidade higiênico-sanitária da tilápia (Oreochromis spp.), comercializada em mercados públicos do município de Mossoró-RN. Para isso, foi realizado, através de checklist, um estudo observacional, para avaliação das boas práticas de manipulação em 10 estabelecimentos de comercialização do peixe. Foram obtidas 30 amostras de tilápia avaliadas quanto aos aspectos sensoriais, microbiológicos (Salmonella spp., coliformes a 45 ºC, contagem de Staphylococcus ssp., bactérias psicrotróficas e enterobactérias) e quanto ao pH e temperatura interna e externa. Observou-se que nenhum dos estabelecimentos apresentou conformidade com os padrões recomendados pela legislação. Já em relação à avaliação sensorial, 83,3% das amostras analisadas apresentavam conformidade com as exigências da legislação. Quanto à pesquisa de coliformes a 45 ºC, os resultados variaram de 0,477 a 1,509 log10 UFC/g, na pesquisa de Staphylococcus spp., de 4,854 a 5,397 log10 UFC/g e ainda 3,884 a 4,937 e 4,707 a 5,463 log10UFC/g para enterobactérias e psicrotróficas, respectivamente. As amostras apresentaram-se dentro do exigido para a contaminação por Salmonella spp.(ausência). O pH variou de 5,99 a 6,18 e as temperaturas indicaram desconformidade com o recomendado. Assim, a qualidade higiênico-sanitária da tilápia comercializada em mercados públicos de Mossoró-RN estava comprometida, apresentando risco à saúde do consumidor pelas condições insatisfatórias.
Abstract This study aimed to verify the sanitary quality of tilapia (Oreochromis spp.) sold in public markets in the city of Mossoro-RN. For such we made use of a checklist and an observational study to evaluate good handling practices in 10 establishments that commercialize fish. We obtained 30 samples of tilapia and evaluated them as to their sensory and microbiological aspects (Salmonella spp., coliforms at 45 ºC, Staphylococcus ssp. count, psychrotrophic bacteria and enterobacteria), as well as to their pH and internal and external temperature. We observed that none of the studied establishments were in accordance with the standards recommended by the legislation. Regarding the sensory evaluation, 83.3% of the samples showed compliance with the requirements of the legislation. As for the coliforms at 45 °C essay, the results ranged from 0.477 to 1.509 log10 CFU/g; in the Staphylococcus spp. essay we found results from 4.854 to 5.397 log10 CFU/g; and further, we found values from 3.884 to 4.937 and from 4.707 to 5.463 log10 CFU/g for enterobacteria and psychrotrophic bacteria, respectively. All the samples were within the requirements for contamination by Salmonella spp. (absence). The pH ranged from 5.99 to 6.18 and temperatures indicated non-compliance with the recommendations. Thus, we can infer that the sanitary quality of tilapia sold in public markets in Mossoro-RN was impaired, presenting a risk to consumer health due to unsatisfactory handling conditions.

Resumo: As artérias mesentéricas das aves são importantes para a irrigação do aparelho digestório e encontram-se associadas ao ganho de peso e conversão alimentar. Objetivou-se descrever as origens, esqueletopias, medidas e principais ramificações das artérias mesentéricas cranial e caudal em avestruzes. Foram utilizados 41 cadáveres de filhotes de avestruzes, 23 machos e 18 fêmeas, obtidos de um criadouro após morte natural. Os cadáveres foram fixados com formaldeído a 10% e tiveram o sistema vascular preenchido com Petrolatex® S-65 colorido. As artérias mesentéricas, cranial e caudal e seus ramos proximais foram dissecados "in situ" e medidas com paquímetro digital. A artéria mesentérica cranial teve comprimento médio de 3,68 ± 1,04 cm e surgiu da aorta descendente ao nível da oitava vértebra torácica na maioria dos casos. Ramificou-se em artérias jejunal e ileocecal. A artéria jejunal ofereceu média de 14,04 ±2,08 ramos ao jejuno e a artéria ileocecal originou um ramo retal e outro que se bifurcou para derivar ramos para íleo, ceco e reto. Em um espécime macho a artéria ileocecal foi ramo da artéria celíaca. A artéria mesentérica caudal originou-se na porção terminal da aorta descendente predominantemente ao nível das 4ª e 6ª vértebras sacro-caudais. Perto da extremidade caudal do rim emitiu os ramos cranial e caudal. O primeiro irrigou o reto e anastomosou-se com ramo retal da artéria mesentérica cranial; o segundo irrigou a porção final do reto, cloaca e bolsa cloacal. Não houve diferença significativa (p<0,05) entre as medidas, esqueletopia e número de ramificações das artérias entre os sexos.
Abstract: The mesenteric arteries of birds are important for the irrigation of the digestive tract and are associated with weight gain and food conversion. This study aimed to describe the origins, skeletopy, measures and main branches of cranial and caudal mesenteric arteries in ostriches. Forty-one cadavers of ostrich chicks, 23 males and 18 females, obtained from a farmer after natural death. The cadavers were fixed with 10% formaldehyde solution and their vascular system was filled with colored Petrolatex® S-65. The cranial and caudal mesenteric arteries and its proximal branches were dissected in situ and measured with a digital caliper. The mesenteric artery had an average length of 3.68cm±1.04 and emerged from the descending aorta at the level of the eighth thoracic vertebra in most cases; it branched into jejunal and ileocecal arteries. The jejunal artery sent a mean of 14 (14.04±2.08) branches to the jejunum. The ileocecal artery sent a rectal branch and another branch that irrigated ileum, cecum and rectum. In a male specimen the ileocecal artery was originated from the celiac artery. The caudal mesenteric artery emerged in the terminal portion of the descending aorta predominantly at the level of the 4th and 6th sacrocaudal vertebrae. Near the caudal end of the kidney it issued the cranial and caudal branches. The first irrigated the rectum and anastomosed with the rectal branch of the cranial mesenteric artery; the second irrigated the final part of the rectum, cloaca and cloacal bursa. There was no significant difference (p<0.05) between measurements, skeletopy and number of branches of the arteries between genders.

We herein report the synthesis of hybrid kauranoid molecules of type 1,2,3-triazole-1,4-disubstituted aiming to improve the antimalarial activity of kaurenoic and xylopic acids. The CuI-catalyzed cycloaddition of azides and kauranoid terminal alkynes was explored as a hybridization strategy. Kauranoid terminal alkynes were prepared from kaurenoic and xylopic acids that were isolated from Wedelia paludosa D. C. (Asteraceae) and Xylopia frutescens Aubl. (Annonaceae). A total of 15 kauranoid derivatives, including nine new triazoles, were obtained and five out of these were more active than the original diterpenes. Interestingly, an increased activity was observed for a kauranoid propargyl ether. Interaction between ent-kaurane diterpene derivatives and Ca2+-ATPase (PfATP6) was investigated. Synthesis of diterpene derivatives emerges as a possible route to be explored in the quest of potentially new inhibitors of PfATP6.

RESUMO O sistema plantio direto aperfeiçoa as áreas agrícolas mantendo o aporte de palha, promovendo a rotação de culturas e a conservação dos solos. Objetivou-se, com o presente estudo, avaliar a qualidade do processo de semeadura e a produtividade de grãos de milho em função de três espécies forrageiras do gênero Urochloa associadas a duas densidades populacionais de plantas de milho. O experimento foi conduzido em área da UNESP/Jaboticabal, SP. O delineamento foi em blocos ao acaso, no esquema fatorial 2 x 3, com quatro repetições. Os tratamentos foram constituídos por duas densidades de plantas de milho (55.000 e 75.000 plantas ha-1) consorciado com três espécies forrageiras (Urochloa brizantha, Urochloa decumbens e Urochloa ruziziensis) semeadas na entrelinha no estádio V4 do milho. Foram analisados, na cultura do milho, os números médios de dias de emergência, a distribuição longitudinal, a produtividade de grãos e a população inicial e final de plantas. Houve diferença entre as populações de plantas de milho (P ≤ 0,1) em que o consórcio de milho com as espécies U. brizantha e U. ruziziensis proporcionou melhores resultados permitindo concluir que o cultivo milho na densidade populacional de 75.000 plantas ha-1 em consórcio com U. brizantha e U. ruziziensis promoveu uma qualidade melhor no processo de semeadura e, em contrapartida, maiores produtividades de grãos.
ABSTRACT The no-tillage system optimizes agricultural areas, maintaining the supply of straw and promoting crop rotation and soil conservation. The aim of the present study was to evaluate sowing quality and grain yield of corn intercropped with three forage species of the Urochloa genus associated with two corn population densities. The experiment was conducted at the São Paulo State University (UNESP), in Jaboticabal-SP, Brazil. The experimental design was randomized blocks in a 2 x 3 factorial scheme with four replicates. The treatments consisted of two corn densities (55,000 and 75,000 plants ha-1) intercropped with three forages (Urochloa brizantha, Urochloa decumbens and Urochloa ruziziensis) sown between rows of corn in the V4 stage. The following corn variables were analysed: mean number of days for emergence, longitudinal distribution, grain yield, initial population and final population. There were differences between corn populations (p < 0.1) and the intercropping of corn with the species U. brizantha and U. ruziziensis promoted the best results, which permitted concluding that the cultivation of corn at the population density of 75,000 plants ha-1 intercropped U. brizantha and U. ruziziensis promoted better sowing quality and, consequently, higher grain yields.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Objective: To investigate the influence of hand dominance on the maintenance of gains after home-based modified constraint-induced movement therapy (mCIMT). Method: Aprevious randomized controlled trial was conducted to examine the addition of trunk restraint to the mCIMT. Twenty-two chronic stroke survivors with mild to moderate motor impairments received individual home-based mCIMT with or without trunk restraints, five times per week, three hours daily over two weeks. In this study, the participants were separated into dominant group, which had their paretic upper limb as dominant before the stroke (n=8), and non-dominant group (n=14) for analyses. The ability to perform unimanual tasks was measured by the Wolf Motor Function Test (WMFT) and the Motor Activity Log (MAL), whereas the capacity to perform bimanual tasks was measured using the Bilateral Activity Assessment Scale (BAAS). Results: Analysis revealed significant positive effects on the MAL amount of use and quality of the movement scales, as well as on the BAAS scores after intervention, with no differences between groups. Both groups maintained the bimanual improvements during follow-ups (BAAS-seconds 0.1, 95% CI -10.0 to 10.0), however only the dominant group maintained the unilateral improvements (MAL-amount of use: 1.5, 95% CI 0.7 to 2.3; MAL-quality: 1.3, 95% CI 0.5 to 2.1). Conclusions: Upper limb dominance did not interfere with the acquisition of upper limb skills after mCIMT. However, the participants whose paretic upper limb was dominant demonstrated better abilities to maintain the unilateral gains. The bilateral improvements were maintained, regardless of upper limb dominance.
