Resumo O tilirosídeo é um flavonóide glicosídico presente em muitas espécies de plantas, incluindo Helicteres velutina K. Schum (Malvaceae sensu lato), conhecida no Brasil como “pitó”. Esta molécula mostrou ter muitas atividades biológicas, porém nenhum estudo foi realizado para investigar a toxicidade dessa substância. O presente trabalho teve como objetivo avaliar a possível toxicidade celular in silico, in vitro e ex-vivo do kaempferol-3-O-β-D- (6 ”-Ep-coumaroil) glucopiranosídeo (tilirosídeo), por meio de análises de estrutura química, toxicidade avaliação e propriedades bioativas preditivas, utilizando amostras humanas para testes in vitro e ex-vivo. A análise in silico sugere que o tilirosídeo exibe bom índice de absorção para penetrar nas membranas biológicas. Além disso, apresentou considerável potencial de proteção celular contra os radicais livres e com atividades anticarcinogênica, antioxidante, antineoplásica, antiinflamatória, anti-hemorrágica e antitrombótica. A avaliação dos efeitos hemolíticos e genotóxicos do tilirosídeo mostrou baixas taxas de hemólise nas hemácias e ausência de toxicidade em células da mucosa oral. Os dados obtidos indicam que esta molécula pode possuir uma abordagem terapêutica promissora como uma possível nova droga com potencial biotecnológico.
Abstract Tiliroside is a glycosidic flavonoid present in many plants species including Helicteres velutina K. Schum (Malvaceae sensu lato), commonly known in Brazil as “pitó”. This molecule has been shown to have many biological activities, however no study has been carried out to investigate the toxicity of this substance. The present work aimed to evaluate the possible cellular toxicity in silico, in vitro and ex-vivo of the kaempferol-3-O-β-D-(6”-E-p-coumaroyl) glucopyranoside (tiliroside), through chemical structure analysis, toxicity assessment and predictive bioactive properties, using human samples for in vitro and ex-vivo tests. The in silico analysis suggests that tiliroside exhibited great absorption index when penetrating biological membranes. In addition, it also displayed considerable potential for cellular protection against free radicals, and anticarcinogenic, antioxidant, antineoplastic, anti-inflammatory, anti-hemorrhagic and antithrombotic activities. The assessment of the hemolytic and genotoxic effects of tiliroside showed low hemolysis rates in red blood cells and absence of cellular toxicity in the oral mucosa cells. The data obtained indicate that this molecule could be a promising therapeutic approach as a possible new drug with biotechnological potential.

Production of mycelial biomass, proteases and protease inhibitors by Ganoderma lucidum under different submerged fermentation conditions

Abstract Ganoderma lucidum is a medicinal mushroom widely recognized as a source of biomolecules with pharmacological properties, however, little is known about the factors that influence the synthesis of bioactive proteins by this fungus when cultivated under submerged fermentation. The objective of this work was to evaluate the production of mycelial biomass and intracellular proteases and protease inhibitors by G. lucidum cultivated under different submerged fermentation conditions. The cultivation was carried out in a medium composed of glucose (10 or 20 g.L-1), soy peptone (2.5 or 5 g.L-1) and yeast extract (5 g.L-1), with incubation under agitation (120 rpm) and non-agitation, totaling 8 experimental conditions. Biomass production was determined from the dry weight, while glucose consumption was estimated by quantification of reducing sugars. The proteins were extracted in NaCl (0.15 M), and the protein extracts were submitted to protein quantification by the Bradford method, total proteolytic activity using azocasein, caseinolytic and fibrinolytic activity in Petri dishes, activity of serine (trypsin and chymotrypsin) and cysteine (papain) protease inhibitors. Cultivation in agitated condition showed higher biomass production with a maximum value of 7 g.L-1, in addition to higher activities of trypsin, chymotrypsin and papain inhibitors, with 154 IU.mg-1, 153 IU.mg-1 e 343 IU.mg-1 of protein, respectively. The non-agitated condition showed a greater potential for obtaining proteins, total proteases, caseinolytic and fibrinolytic enzymes, with maximum values of 433 mg.g-1 of extract, 71 U.mL-1 of extract, 63.62 mm2 and 50.27 mm2, respectively. Thus, a medium composed of soy peptone, yest extract and glucose in a 1:2:4 proportion is recommended, under agitation to produce protease inhibitors, and the non-agitated condition when the target is, mainly caseinolytic and fibrinolytic enzymes.
Resumo Ganoderma lucidum é um cogumelo medicinal amplamente reconhecido como fonte de biomoléculas com propriedades farmacológicas, entretanto, pouco se conhece acerca dos fatores que influenciam a síntese de proteínas bioativas por esse fungo, quando cultivado sob fermentação submersa. O objetivo deste trabalho foi avaliar a produção de biomassa micelial e de proteases e inibidores de proteases intracelulares por G. lucidum cultivado em diferentes condições de fermentação submersa. O cultivo foi realizado em meio contendo glicose (10 ou 20 g.L-1), peptona de soja (2,5 ou 5 g.L-1) e extrato de levedura (5 g.L-1), com incubação sob agitação (120 rpm) ou não-agitado, totalizando 8 condições experimentais. A produção de biomassa foi determinada a partir do peso seco e o consumo de glicose a partir da quantificação de açúcares redutores. As proteínas foram extraídas em NaCl (0,15 M) e os extratos proteicos foram submetidos à quantificação de proteínas pelo método de Bradford, atividade proteolítica total usando azocaseína, atividade caseinolítica e fibrinolítica em placa de Petri, atividade de inibidores de serino-proteases (tripsina e quimotripsina) e cisteíno-protease (papaína). O cultivo em condição agitada apresentou maior produção de biomassa com valor máximo de 7g.L-1, além de maiores atividades de inibidores de tripsina, quimotripsina e papaína, com 154 UI.mg-1, 153 UI.mg-1 e 343 UI.mg-1 de proteína, respectivamente. A condição não-agitada demonstrou maior potencial para a obtenção de proteínas, proteases totais, enzimas caseinolíticas e fibrinolíticas, com valores máximos de 433 mg.g-1 de extrato, 71 U.mL-1 de extrato, 63,62 mm2 e 50,27 mm2, respectivamente. Assim, recomenda-se o meio composto de peptona de soja, extrato de levedura e glicose na proporção 1:2:4, em condição agitada para a produção de inibidores de proteases, e a condição não-agitada para a síntese de proteases, principalmente enzimas caseinolíticas e fibrinolíticas.

Abstract Sugarcane cultivation stands out in Brazilian agribusiness, covering more than eight million hectares for the production of sugar, ethanol, and by-products. Fertilization is one of the limiting factors in sugarcane yield, for which filter cake is a viable solution to meet plant nutritional needs. This study aimed to assess the effect of enriched filter cake on gas exchange and yield in RB041443 sugarcane, cultivated in soils of the coastal tablelands of Paraíba, Brazil. The experiment was conducted in the Monte Alegre S/A sugarcane mill, in the municipality of Mamanguape, using a randomized blocks experimental design, with 12 treatments (T1- cake, T2- cake + MAP, T3- cake + gypsum, T4 - cake + phosphate, T5- cake + bagasse, T6- cake + MAP + gypsum, T7- cake + MAP + phosphate, T8- cake + MAP + bagasse, T9- cake + gypsum + phosphate, T10- cake + gypsum + bagasse, T11- cake + phosphate + bagasse, and T12- control (only MAP)), and 4 replications, totaling 48 plots. A significant effect (5% probability) was also observed for the variables number of leaves and tons of stem per hectare (TSH). T1- cake, T4- cake + phosphate, T6- cake + MAP + gypsum and T10- cake + gypsum + bagasse, had the best results for TSH, with yields greater than 140 t ha-1. Regarding stomatal conductance, the highest values were obtained in T6 and T8, which, together with T11, had the highest gs values. Concerning the internal carbon concentration, T1, T2, T6, and T8 stood out. T6 also had a significant effect on transpiration. From this study, it was concluded that the use of enriched filter cake as a base fertilizer in sugarcane culture contributes to increasing the yield of the RB041443 variety, generating positive responses for plant gas exchange, being T1 and T10 indicated to increase the production in the sugar-energy sector.
Resumo A cultura da cana-de-açúcar ocupa uma posição de destaque para o agronegócio brasileiro, abrangendo mais de oito milhões de hectares na produção de açúcar, etanol e subprodutos. Um dos fatores limitantes na produtividade da cana-de-açúcar é a questão da adubação, sendo a torta de filtro uma solução viável para suprir as necessidades nutricionais da planta. Com esse estudo, objetivou-se avaliar o efeito do uso da torta de filtro enriquecida sob as trocas gasosas e produtividade da variedade de cana-de-açúcar RB041443 cultivada em solos dos tabuleiros costeiros da Paraíba. O experimento foi conduzido na Usina Monte Alegre S/A, no município de Mamanguape-PB, utilizando o delineamento experimental em blocos casualizados com 12 tratamentos (T1- Torta, T2- Torta + MAP, T3- Torta + Gesso, T4 - Torta + Fosfato, T5- Torta + Bagaço, T6- Torta + MAP + Gesso, T7- Torta + MAP + Fosfato, T8- Torta + MAP + Bagaço, T9- Torta + Gesso + Fosfato, T10- Torta + Gesso + Bagaço, T11 - Torta + Fosfato + Bagaço, e T12 - Testemunha (Apenas MAP)), 4 repetições, totalizando 48 parcelas. Efeito significativo a 5% também foi observado para a variável número de folhas e tonelada de colmo por hectare. Para a variável TCH, os melhores resultados foram obtidos nos tratamentos T1- Torta, T4-Torta + Fosfato, T6- Torta + MAP + Gesso e T10- Torta + Gesso + Bagaço, com produtividades superiores a 140 t ha-1. Para a condutância estomática, resultados superiores foram obtidos no T6 e T8, que, junto ao T11, agruparam-se com os maiores valores de gs. Em relação a concentração interna de carbono, destacaram-se os tratamentos T1, T2, T6 e T8. Para a variável transpiração, o T6 apresentou influência significativa da torta de filtro. Com o referido estudo, concluiu-se que a utilização da torta de filtro enriquecida como adubação de fundação na cultura da cana-de-açúcar possibilita aumento no rendimento produtivo da variedade RB 041443, bem como respostas positivas para as trocas gasosas da planta, sendo os tratamentos T1 e T10 indicados para aumentar a produção do setor sucroenergético.

Characterization of edaphic fauna in different monocultures in Savanna of Piauí

Resumo A cobertura vegetal pode mostrar padrões de diversidade e composição da comunidade de invertebrados do solo, em função da quantidade e qualidade da serapilheira em determinado habitat específico. O objetivo deste trabalho foi caracterizar a distribuição da fauna edáfica em diferentes monocultivos. O estudo foi realizado na fazenda Chapada Grande no município de Regeneração, PI. Foram escolhidas quatro áreas de monocultivos: soja em sistema de plantio direto, eucalipto, pastagem, além de uma mata nativa de cerrado preservada. Foram realizadas coletas da fauna do solo, em período seco e úmido, por meio de armadilhas do tipo pitfall contendo 4% de formol. A fauna edáfica foi avaliada pelo número de indivíduos por armadilha por dia, riqueza e riqueza média, índice de diversidade de Shannon e índice de uniformidade de Pielou. Os dados foram submetidos à análise de variância e à análise multivariada de Componentes Principais (ACP). O maior número de indivíduos por armadilha dia e de riqueza média foram registrados nas áreas pastagem, eucalipto e mata nos dois períodos, enquanto que a soja mostrou valores inferiores com predomínio dos grupos Coleoptera e Formicidae. As áreas de pastagem e mata mostraram maiores valores de índice de Shannon nas duas épocas avaliadas provavelmente em função de maiores aportes de resíduos orgânicos no solo que favorece as condições de abrigo, alimentação e reprodução. Em relação ao índice de Pielou o sistema com soja mostrou maiores valores nessa variável. Os grupos Araneae, Coleoptera, Formicidae e Diptera predominaram no período úmido, enquanto Coleoptera e Formicidae se destacaram no período seco. Sistemas que geram maior acúmulo de resíduos abrigam uma maior diversidade de invertebrados da fauna edáfica. A sazonalidade apresentou efeito sobre todas as variáveis analisadas sendo que o período úmido mostrou valores mais expressivos.
Abstract Vegetation cover may show diversity and composition patterns of the soil invertebrate community, as a function of litter quantity and quality in a specific habitat. The objective of this work was to characterize the distribution of edaphic fauna in different monocultures. The study was carried out at Chapada Grande farm in Regeneração, PI. Four monoculture areas were chosen: no-tillage soybean, eucalyptus, pasture, and a preserved native cerrado forest. Soil fauna was collected in a dry and wet period by pitfall traps containing 4% formaldehyde. The edaphic fauna was evaluated by the number of individuals per trap per day, average richness and richness, Shannon diversity index and Pielou uniformity index. Data were submitted to analysis of variance and multivariate Principal Component Analysis (PCA). The highest number of individuals per day trap and of average richness were registered in the pasture, eucalyptus and forest areas in both periods, while soybean showed lower values with predominance of Coleoptera and Formicidae groups. The pasture and forest areas showed of higher Shannon index values in the two evaluated seasons, probably due to higher contributions organic residues in the soil that favors the shelter, feeding and reproduction conditions. Regarding the Pielou index, the soybean system showed higher values in this variable. The Aranae, Coleoptera, Formicidae and Diptera groups predominated in the humid period, while Coleoptera and Formicidae predominated in the dry period. Systems that generate greater accumulation of residues harbor a greater diversity of invertebrates of the edaphic fauna. Seasonality had an effect on all variables analyzed and the wet period showed more expressive values.

Early liver function improvement following successful treatment of chronic hepatitis C in patients with decompensated cirrhosis: a real-life study

OBJECTIVES: Despite higher rates of sustained virologic response (SVR), important concerns remain when patients with decompensated cirrhosis due to hepatitis C virus (HCV) are treated with direct-acting antiviral agents (DAA). Questions include efficacy, safety, and the magnitude of liver function improvement. Here, we aimed to evaluate HCV treatment data in this specific population in Brazil. METHODS: We included 85 patients with decompensated cirrhosis submitted to HCV therapy with DAA followed at two academic tertiary centers in the southeastern region of Brazil. RESULTS: Seventy-nine patients (92.9%) were Child-Pugh (CP) score B, and six (7.1%) were CP score C. The mean MELD score was 12.86. The most common treatment was sofosbuvir plus daclatasvir±ribavirin for 24 weeks. The overall intention-to-treat (ITT) SVR rate was 87.4% (74/85) and modified-ITT 96.1% (74/77). ITT SVR was associated with lower baseline INR values (p=0.029). Adverse events (AE) occurred in 57.9% (44/76) of patients. Serious AE were reported in 12.8% (10/78), and were related to the presence of hepatic encephalopathy (p=0.027). SVR was associated with improvement in CP (p<0.0001) and MELD scores (p=0.021). Among baseline CP score B patients with SVR, 46% (29/63) regressed to CP score A. Ascites was independently associated with no improvement in liver function in patients who achieved SVR (p=0.001; OR:39.285; 95% CI:4.301-258.832). CONCLUSIONS: Patients with decompensated HCV cirrhosis showed a high SVR rate with interferon-free therapy. Early liver function improvement occurred after successful HCV eradication. However, long-term follow-up of these patients after SVR remains strongly advised.

This work focuses on the validation and application of solid phase extraction followed by high-performance liquid chromatography (SPE-HPLC) analysis of important endocrine disruptors compounds (EDC) from different classes in highly complex wastewater matrix. The endocrine disruptors investigated included three different categories: pharmaceuticals (sulfamethoxazole (SMZ), trimethoprim (TMP) and diclofenac (DCF)), hormones (estrone (E1), 17α-ethinylestradiol (EE2) and 17β-estradiol (E2)) and plastic materials (bisphenol A (BPA)). The method involves pre-concentration by SPE using Strata-X extraction cartridges followed by HPLC coupled with diode array detector (DAD). As the assumption of homoscedasticity was met for analytical data, ordinary linear regression procedure was applied to the data. The method was considered validated for 7 EDC after consistent evaluation of the key analytical parameters. Recoveries were ranged from 52.3 to 179.6%. Limits of quantification were in the range 6.0-0.4 µg L−1. The described method was applied to evaluate the occurrence and removal efficiency of EDC in two biological sewage treatment plants (STPs). The influent mean concentrations of E1, E2 and EE2 hormones were 79.54; 175.09 and 102.19 µg L−1, respectively, while for SMZ, TMP, DCF and BPA were 215.17; 187.01; 218.97 and 87.12 µg L−1 for STP A, respectively. The efficiencies for the removal of EDCs ranged from 0 to 79.4%.

Agroindustrial residues, such as leaves of fruit plants, can be sources of bioactive molecules, thus adding value to co-products that are rarely explored in agroindustry. In this context, this study aimed to look into the phytochemical profiles in detail and to explore the antitumor potential of S. tuberosa and S. mombin leaves. We observed that, S. tuberosa leaf extract was cytotoxic in both tumor and healthy cells. S. mombin extracts selectively inhibited cell proliferation in the tumor cell line for prostate cancer (PC3) and did not significantly affect healthy cells. The metabolic profiles of the extracts were evaluated by gas chromatography coupled with mass spectrometry and twenty-three metabolites were identified. The correlation of metabolic profiles with cytotoxic tests indicated possible chemical markers that may be responsible for the inhibition of cell proliferation. This study revealed that the unexplored co-products in agroindustry may have great therapeutic potential, and therefore should be screened for biologically active compounds.

Electrochemical methods are powerful in the characterization and design of redox-modulating agents. We, herein, report the electrochemical investigation, in aprotic medium, of eleven synthetic 3-thio-substituted-nor-beta-lapachones, along with the determination of cytotoxic activity and correspondent selectivity index, against several cancer cell lines and one normal cell. Four of the quinones are novel compounds. The redox behavior is representative of two independent systems: the easy reduction of the quinone moiety and, at far more negative potential, the reductive cleavage of the C-S-C bonding; and the anodic part controlled by the oxidation of the sulfur moiety. The compounds have shown relevant cytotoxic activity, with emphasis on 3-phenyl-thio-2,2-dimethyl-2,3-dihydronaphtho[1,2-b]furan-4,5-dione (compound 2), which mechanism of molecular action was shown to be related to reactive oxygen species (ROS) release. Despite the absence of a linear correlation, there is a trend: the majority of the thionaphthoquinones, with values of first wave reduction potential, less negative than -0.65 V, were active. The less electrophilic compound (3-(cyclohexylthio)-2,2-dimethyl-2,3-dihydronaphtho[1,2-b]furan-4,5-dione, cyclohexyl derivative) is also the less cytotoxic toward cancer cells. Agents containing chalcogens and quinones can be used to attack entities with a disturbed redox balance.

O presente trabalho teve por objetivo avaliar o efeito do gradiente vegetacional formado por Campo Graminóide (CGR), Cerrado Sensu Stricto (CSS), Cerradão (CRD) e Floresta Estacional Semidecídua (FES) sobre a densidade, diversidade e composição da comunidade da fauna edáfica em duas épocas do ano do Parque Nacional de Sete Cidades (PI). Para amostragem da fauna foram colocadas um total de oito armadilhas do tipo Pit-fall, distanciadas por 10 m, em cada área na parte central de cada sistema, onde permaneceram por sete dias. No período úmido houve uma tendência a aumento do número de indivíduos conforme a complexidade da formação vegetal e o inverso ocorreu no período seco. Verificou-se uma variação ambiental dos fatores climáticos temperatura e umidade conforme a formação vegetal contribuindo para uma distribuição heterogênea da fauna. Apenas no período seco a formação CGR apresentou valor significativamente menor de riqueza média. Em relação às variáveis de diversidade e uniformidade não apresentaram variações drásticas em relação ao gradiente vegetacional estudado. Os grupos dominantes no gradiente vegetacional foram Formicidae, Coleoptera, Aranae, Acari e Collembola, com redução do número de Coleoptera na época seca. Pela análise de componentes principais (ACP) constatou-se maiores diferenças na composição das comunidades entre as formações vegetais para o período chuvoso. Nesta época, as formações vegetais os estádios FES e CRD estiveram associados a uma maior diversidade de invertebrados do que CSS e CGR, demonstrando influência da complexidade vegetacional sobre a comunidade da fauna do solo.
Abstract The vegetation physionomy and cover can show patterns of diversity and composition of the edaphic community, depending on the quantity and quality of litter in a specific habitat. The objective of this study was to evaluate the effect of the vegetation gradient formed by Graminoid Field (GRF), Cerrado Sensu Stricto (CSS), Cerradão (CRD) and Semideciduous Seasonal Forest (SSF) on density, diversity and composition of the edaphic fauna community in two seasons of the year, in the Sete Cidades National Park (Piauí state). For fauna sampling, a total of eight pitfall traps, distanced 10 m, were placed in each area in the central part of each system, where they remained for seven days. In the wet period, there was a tendency to increase the number of individuals as a function of the complexity of the vegetation formation, with the inverse occurring in the dry period. It was verified an environmental variation of the climatic factors temperature and humidity according to the vegetal formation, contributing to a heterogeneous distribution of the fauna. The GRF formation presented a significantly lower value of average richness only in the dry period. Regarding the variables of diversity and uniformity, they did not show drastic variations in relation to the vegetation gradient studied. The dominant groups in the vegetation gradient were Formicidae, Coleoptera, Aranae, Acari and Collembola, with reduction of the number of Coleoptera in the dry season. Principal component analysis (PCA) revealed greater differences in the composition of the communities between the vegetation formations for the rainy season. At this time, the formations SSF and CRD were associated to a greater diversity of invertebrates than CSS and GRF, demonstrating the influence of the vegetation complexity on the soil fauna community.

Este artigo descreve o desenho, os métodos e resultados preliminares deste estudo de coorte. Trezentas e cinquenta e quatro crianças foram acompanhadas clínico e laboratorialmente por dois anos, com 15% de perdas de seguimento. A taxa global de novas infecções foi de aproximadamente 10% na coorte de crianças no primeiro ano de seguimento. Informações sobre a força de infecção sorotipo-específica do dengue nos primeiros anos de vida, bem como a avaliação da cinética de anticorpos materno-transferidos poderão contribuir para a compreensão da etiopatogenia da doença.

The diterpene ent-18,19-dihydroxytrachylobane was biotransformed for the first time by Rhizopus stolonifer, and yielded the new ent-11β,18,19-trihydroxytrachylobane derivative besides the new ent-kaur-11-ene diterpenes ent-16α,18,19-trihydroxykaur-11-ene and ent-18,19-dihydroxy-16α-methoxykaur-11-ene. Their structures were determined by spectrometric methods.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Validity of the six-minute step test of free cadence in patients with chronic obstructive pulmonary disease

Objetivos: Avaliar a validade concorrente do desempenho no teste de degrau de seis minutos (TD6) em analisar a capacidade física da DPOC, relacionando-o com o teste de caminhada de seis minutos (TC6), bem como verificar a presença de validade de critério preditiva do TD6, determinando um valor de corte para identificar baixa capacidade física. Método: Trinta e dois pacientes com DPOC estágios leve-muito grave, de ambos os sexos, entre 50-87 anos, realizaram dois TD6 e dois TC6. Resultados: Validade concorrente: observou-se correlação forte positiva (Pearson) entre o número de subidas do primeiro (T1), segundo (T2) e do melhor dos dois (T1 ou T2) TD6 com a distância percorrida no TC6. Embora constatamos correlações negativas entre os desempenhos no TD6 com o índice BODE e VEF1, essas não foram suficientes para caracterizar validade, não sendo traçadas as curvas ROC. A validade preditiva (curvas ROC) do TD6 para identificar baixa capacidade física (comparação com o TC6), utilizando o desempenho do T1 ou T2 ou somente do T1, foi considerada acurada para alguns propósitos, sendo a área abaixo da curva de 0,8 (IC95% 0,62-0,98) e 0,85 (IC95% 0,70-0,99), respectivamente. Para classificar os pacientes, escolhemos o ponto de corte de 86 e 78 degraus, apresentando sensibilidade de 90% e 90% e especificidade de 64% e 68%, para T1 ou T2 e somente T1, respectivamente. Conclusão: O desempenho no TD6 apresentou-se válido para verificar capacidade física na DPOC e identificou pacientes com baixa capacidade física, utilizando como ponto de corte 78 degraus, em que valores abaixo disso representaram pior prognóstico.
Objectives: to evaluate the concurrent validity of the six-minute step test (6MST) in assessing exercise capacity of COPD patients using the six-minute walk test (6MWT) as a gold-standard. The predictive validity of the 6MST was assessed to determine a cut-off point for identification of low exercise capacity. Method: thirty-two COPD patients (50-87 years old) with mild to very severe obstruction performed the 6MST and 6MWT twice. Results: Concurrent validity: a strong positive correlation (Pearson) between the number of ascents on the first (T1), second (T2) and the best of both (T1 or T2) tests during the 6MWT was observed. Although a moderate negative correlation with BODE index and FEV1 was found, it was considered insufficient to test the validity, therefore ROC curves were not applied. The predictive validity (ROC) of the 6MST to identify low physical capacity (compared with the 6MWT) using the performance of T1 or T2, or solely T1 was considered accurate, and the area under the curve was 0.8 (IC95% 0.62-0.98) and 0.85 (IC95% 0.70-0.99), respectively. To classify patients, the cut-off points of 86 and 78 steps were chosen, with both values showing 90% of sensitivity and specificity of 64% and 68% for T1 or T2, or solely T1, respectively. Conclusion: The number of steps on the 6MST was valid to verify exercise capacity in COPD patients and the cut-off point of 78 steps was able to identify patients with poor exercise tolerance. Values under this cut-off point are considered to identify patients with a poorer prognosis.

Potent antileukemic action of naphthoquinoidal compounds: evidence for an intrinsic death mechanism based on oxidative stress and inhibition of DNA repair

O presente estudo descreve a acentuada atividade citotóxica da nor-β-lapachona, seus derivados arilamino substituídos, naftoquinonas iodadas e metilada, além de nor-β-lapachonas 1,2,3-triazólicas, contra quatro linhagens de células de leucemia humana (HL-60, K562, Molt-4 e Jurkat). Nor-β-lapachonas arilamino substituídas foram identificadas com potente atividade, revelando-se como potenciais protótipos contra as linhagens tumorais descritas. Estudos utilizando o ensaio cometa evidenciaram danos ao ácido desoxirribonucleico (ADN) causado pelos derivados arilamino substituídos devido o aumento dos níveis intracelulares de espécies reativas de oxigênio (ERO's). Células de HL-60 foram selecionadas para a continuidade dos estudos de mecanismos moleculares subjacentes e apoptose induzida pelos derivados quinoidais foi observada por análise de citometria de fluxo. Cepas de Saccharomyces cerevisiae foram utilizadas para uma investigação preliminar sobre o mecanismo de ação em topoisomerases de ADN. Os estudos sugerem que, aparentemente, a citotoxidade dos compostos não envolve a inibição de topoisomerases, mas que o tratamento prejudica a atividade de reparação do ADN, provocando assim a morte celular. A capacidade em induzir apoptose e aberrações cromossômicas em fibroblastos de pulmão de hamster chinês (células V79) também foi investigada. Núcleos apoptóticos foram observados e nossos estudos indicam uma correlação entre dano ao ADN e apoptose.
The current study describes that nor-β-lapachone and its arylamino derivatives, iodinated and methylated naphthoquinones and nor-β-lapachone-based 1,2,3-triazoles exhibited pronounced cytotoxic effects against four human leukemia cell lines (HL-60, K562, Molt-4 and Jurkat). Nor-β-lapachones arylamino substituted with potent activity were identified, revealing themselves as potential prototypes against tumor cell lines. Moreover, cells treated with these compounds showed DNA damage according to the standard comet assay, a finding that was, at least in part, due to increased intracellular levels of ROS. HL-60 cells were chosen to study the underlying molecular mechanisms of cytotoxicity. Drug-induced apoptosis in HL-60 cells was observed by flow cytometry analyses. Strains of Saccharomyces cerevisiae were used for a preliminary investigation into the mechanism of drug action on DNA topoisomerases. These results suggested that the cytotoxicity of these compounds apparently does not involve topoisomerase inhibition, but that treatment impairs DNA repair activity, thus triggering cell death. Considering their pro-oxidant properties, we investigated the ability of these compounds to induce apoptosis and chromosomal aberrations as micronuclei in Chinese hamster lung fibroblasts (V79 cells). Morphological apoptotic nuclei and micronuclei induction following drug treatment were observed, suggesting a correlation between DNA damage and apoptosis.
