Toads of the Rhinella genus have a pair of paratoid glands that store biological secretions of high toxicity and varied chemical composition, rich in biologically active compounds. The present work aimed to carry out the investigation of the metabolomic profile and evaluation of the biological potential of the secretion paratoid glands (PGS) from Rhinella granulosa. The paratoid secretion was collected in the Piauí state (Brazil), extracted with methanol and the extract was analyzed by ultra-performance liquid chromatography with quadrupole time-of-flight mass spectrometry. Fifty chemical constituents were identified. The extract showed cytotoxicity against tumor cell lines of the central nervous system (half maximal inhibitory concentration (IC50) = 1.9 µg mL-1) and prostate (IC50 = 1.6 µg mL-1), unsatisfactory antimicrobial potential (minimal inhibitory concentration (MIC) > 312 µg mL-1) and inhibited the enzyme acetylcholinesterase (IC50 = 5.119 mg mL-1). The results presented relevant information about the PGS and contributed to the understanding of the metabolomic and biological potential of R. granulosa.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Espécies vegetais hospedeiras de begomovírus isolados de tomateiro em Goiás e no Distrito Federal

A determinação da gama de hospedeiros de begomovírus isolados de tomateiro (Lycopersicon esculentum) é de elevada importância nos estudos conduzidos com esses patógenos, uma vez que contribui para o entendimento da larga disseminação dos vírus em condições de campo e oferece subsídios para o estudo da variabilidade genética de espécies e estirpes identificadas em diversas regiões do país. Visando a determinação e a comparação da gama de hospedeiros de dois isolados de begomovírus de tomateiro obtidos de lavouras da região de Anápolis-GO (GO-ANPL) e do Distrito Federal (DF-BR2) inocularam-se 31 espécies vegetais pertencentes a oito famílias botânicas, sob duas modalidades de inoculação: mecânica e com a mosca branca, Bemisia tabaci biótipo B. Constatou-se que o GO-ANPL e o DF-BR2 infetaram exclusivamente plantas da família Solanaceae como Datura stramonium, Nicandra physalodes e Nicotiana benthamiana. Para o GO-ANPL, o número de espécies vegetais infetadas com o emprego do inseto vetor foi superior ao obtido pela inoculação mecânica, diferindo dos resultados obtidos para o DF-BR2 em que as plantas hospedeiras foram igualmente infetadas em ambos os métodos de inoculação. A comparação entre as hospedeiras dos dois isolados e destes com as de outros begomovírus de tomateiro da região Nordeste revelaram que há variação tanto nas espécies hospedeiras como na sintomatologia exibida pelas plantas infetadas. Os testes foram todos confirmados com hibridização com sondas moleculares, em "dot blot".
Determination of the begomovirus host range determination in tomato (Lycopersicon esculentum) is an important feature in unveiling the genetic variability of species and strains of the virus and how it spreads under field conditions. In order to determine the host range of two begomoviruses isolated from tomato, denoted GO-ANPL and DF-BR2, 31 plant species of eight botanical families were tested via mechanical and vector inoculation. Both isolates only infected plants of the family Solanaceae as Datura stramonium, Nicandra physalodes e Nicotiana benthamiana. The GO-ANPL isolate infected a higher number of plants when they were inoculated by the insect vector. Results were distinct from those obtained with the DF-BR2 isolate, where both inoculation methods were equally effective. Comparison between these two isolates with other begomoviruses reported in Northeast Brazil revealed different patterns in host range and symptomss. Virus infection was confirmed by dot blot hybridization using specific molecular probes to begomovirus isolates.

No estudo da interação de um begomovírus isolado de tomateiro (Lycopersicon esculentum) em Anápolis-GO, denominado GO-ANPL (taxonomicamente relacionado ao Tomato rugose mosaic virus, ToRMV) com o vetor Bemisia tabaci biótipo B, determinaram-se o período de acesso de aquisição do vírus (PAA), o período de acesso de inoculação (PAI), o período de latência (PL), a sua retenção e transmissão à progênie. Nos experimentos empregaram-se plantas de tomateiro 'Santa Clara' e cinco insetos por planta. Constatou-se um PAA mínimo de 15 min com o qual 6% dos tomateiros foram infetados. Este percentual aumentou para 65% quando o PAA foi estendido para 24 h. O PAI mínimo foi de 30 min registrando-se 18% de infecção, o qual foi elevado para 67% com 24 h de PAI. Observou-se o término do PL 16 h após o vetor adquirir o vírus. Na detecção do GO-ANPL no vetor via PCR, testes com mais de 2.500 espécimens constataram o vírus em ninfas desenvolvidas em tomateiro infetado, em adultos com diferentes PAAs, mas não em ovos de fêmeas avirulíferas ovipositados em planta infetada. Observou-se a passagem transestadial em 100% dos adultos testados que transmitiram o vírus em 33% dos casos. Evidenciou-se a transmissão à progênie pela detecção do vírus em ovos, ninfas e adultos provenientes de fêmeas virulíferas, contudo, a transmissão do vírus pelos adultos não foi observada. Os resultados obtidos indicam que a interação vírus-vetor é estabelecida desde a fase inicial do desenvolvimento do inseto e podem ser considerados relevantes para os estudos epidemiológicos dos begomovírus associados ao biótipo B de B. tabaci no país.
A begomovirus named GO-ANPL (taxonomically related to the Tomato rugose mosaic virus, ToRMV), obtained from tomato (Lycopesricon esculentum) plants in Anápolis, state of Goiás, was used to study virus/vector (Bemisia tabaci biotype B) interaction. The acquisition access period (AAP), the inoculation access period (IAP), and the latent period (LP) were determined by transferring five whiteflies per tomato seedling cv. Santa Clara. The minimum AAP was 15 min resulting in 6% infected plants, which reached 65% as the length of AAP increased to 24 h. At a minimum IAP of 30 min, 18% of plants were infected; this increased to 67% after an IAP of 24 h. The end of the LP in the vector occurred 16 h after virus acquisition. To detect the GO-ANPL in the vector by PCR, more than 2,500 specimens were tested. The virus was found in adults under different AAPs from the 1st to the 4th instar grown on infected plants. No virus was found in the eggs that were laid on infected plants by aviruliferous females. The GO-ANPL isolate was transmitted to the progeny of viruliferous females, since the virus was detected in eggs, all instars and adults. However, no virus transmission was observed from these adults. A high frequency of viral detection was observed in newly emerged adults from immature forms reared on virus-infected plants. These adults infected 33% of tomato plants in virus transmission assays. The results indicate that the interaction between begomovirus and B. tabaci biótipo B starts at early stages of insect development. This is important information for epidemiological studies of begomovirus in this country.
