ABSTRACT A 6-year field monitoring study was designed to compare the presence of fall armyworm Spodoptera frugiperda (J. E. Smith), leaf injury, its parasitoids, 100 seed weight, and seed yield of Bt maize hybrids approved for commercialization in the Brazilian market. Field trials were planted in two municipalities, divided into two periods, from 2011 to 2014 (period I), and due to the approval of different Bt hybrids, from 2015 to 2016 (period II). Treatments were Bt, non-Bt maize and all non-Bt maize sprayed with insecticide methomyl. Six Bt-maize hybrids were planted, expressing the following proteins: Cry1Ab, Cry1F, Cry1A.105 (Cry1Ab +Cry1Ac +Cry1F), Cry2Ab2a and Vip3. In 2015 and 2016 Bt Powercore was planted due to its commercial approval. The number of S. frugiperda larvae for most Bt hybrids was lower than on non-Bt hybrids, except for Fórmula TL and Herculex in period II, in both municipalities. In period I, Bt hybrids VT, VT 2 and Viptera showed almost no injury on their leaves, despite the high number of fall armyworm larvae found in plants. Different parasitoids emerged from larvae from Bt and non Bt hybrids: Archytas sp., Campoletis sp., Chelonus sp., Eiphosoma sp. and Ophion luteus. The average 100 seed weight was higher in municipality from Sete Lagoas for all Bt hybrids, non-Bt hybrids and non-Bt hybrids sprayed with methomyl, when compared to Nova Porteirinha during period I. Fórmula TL, non-Bt Fórmula TL sprayed with methomyl, and non-Bt Fórmula TL showed the lowest seed yield in both municipalities and periods.

ABSTRACT Soybean looper (Chrysodeixis includens) and Spodoptera group are important defoliators insects responsible for significant yield losses in soybean, cotton and maize in Brazil. Bioinsecticides and transgenic plants expressing insecticidal proteins from Bacillus thuringiensis are among the most used sustainable control methods for pest control in agriculture, especially for several lepidopteran species. To provide new components for insect control, this study aimed to select and characterize B. thuringiensis strains toxic to C. includens, S. cosmioides, S. eridania, and S. frugiperda. We performed initial bioassays with fifty B. thuringiensis strains for C. includens and selected four strains – 1608A, 726, 773, and 775E – that caused high larval mortality (100%) to be tested against Spodoptera species. These strains harbored cry insecticidal genes, megaplasmids, absence of β-exotoxin and showed two major proteins about 65 and 130 kDa in SDS-page attributed to Cry protein classes toxic to lepidopteran species. The 1608A and 775E strains also showed high toxicity to Spodoptera species that demonstrate great potential for B. thuringiensis-based bioproduct development and as sources of new insecticidal genes for transgenic crops for multiple pest control relevant in the Brazilian agricultural system.

Abstract The main objective of this study was to characterize the toxicity and genetic divergence of 18 Bacillus thuringiensis strains in the biological control of Spodoptera eridania. Bacterial suspensions were added to the S. eridania diet. Half of the selected B. thuringiensis strains caused high mortality seven days after infection. The genetic divergence of B. thuringiensis strains was assessed based on Enterobacterial Repetitive Intergenic Consensus (ERIC) and Repetitive Extragenic Palindromic (REP) sequences, and five phylogenetic groups were formed. Despite their genetic diversity B. thuringiensis strains did not show any correlation between the collection sites and toxicity to larvae. Some B. thuringiensis strains are highly toxic to S. eridania, thus highlighting the potential of their endotoxins as biopesticides.

Resumo: O objetivo deste trabalho foi avaliar uma gama de hospedeiros, in vitro, quanto à resposta a Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV), um vírus patogênico à lagarta-do-cartucho (Spodoptera frugiperda; Lepidoptera: Noctuidae), para o posterior desenvolvimento de um biopesticida baseado em sistema de cultura de células. As linhagens celulares de Bombyx mori (BM-5), Lymantria dispar (IPLB-LD-625Y), Trichoplusia ni (BTI-Tn-5B1-4), Anticarsia gemmatalis (UFL-AG-286) e S. frugiperda (IPLB-SF-21AE e Sf9) foram testadas quanto a sua suscetibilidade a um isolado brasileiro de SfMNPV altamente virulento. Os efeitos citopáticos induzidos pelo vírus, a produção de partículas virais e a síntese de polipeptídeos virais foram examinados e comparados. Ambas as células de S. frugiperda apresentaram hipertrofia dos núcleos celulares e produção de muitos poliedros. O SDS-Page de proteínas com radiomarcação mostrou que as proteínas celulares apresentaram inibição de síntese, enquanto uma intensa banda de cerca de 30 kDa, reconhecida como poliedrina, era sintetizada. As outras linhagens celulares não apresentaram produção de poliedros, apesar de algumas terem apresentado alterações morfológicas e inibição de síntese proteica em resposta à infecção viral. As linhagens celulares SF-21 e Sf9 são recomendadas para a posterior produção in vitro de SfMNPV.

Abstract: The objective of this work was to evaluate the response of an in vitro host range to Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV), a pathogenic virus to the fall armyworm (Spodoptera frugiperda, Lepidoptera: Noctuidae), for the further development of a biopesticide based on cell culture systems. The cell lines from Bombyx mori (BM-5), Lymantria dispar (IPLB-LD-625Y), Trichoplusia ni (BTI-Tn-5B1-4), Anticarsia gemmatalis (UFL-AG-286), and S. frugiperda (IPLB-SF-21AE and Sf9) were tested for their susceptibility to a highly-virulent Brazilian isolate of SfMNPV. The cytopathic effects induced by the virus, the production of viral particles, and the synthesis of viral polypeptides were examined and compared. Both S. frugiperda cell lines showed hypertrophy of cell nuclei and production of many polyhedra. The SDS-Page of radiolabed proteins showed that the cell protein synthesis was shutoff, while an intense band of about 30 kDa, recognized as polyhedrin, was synthesized. The other cell lines did not show polyhedra production, although some of them underwent morphological changes and protein synthesis shutdown in response to virus infection. The SF-21 and Sf9 cell lines are recommended for further in vitro production of SfMNPV.

RESUMO: O entomopatógeno Bacillus thuringiensis é amplamente empregado no controle de pragas, porém estudos de seu efeito sobre organismos não alvo ainda são incipientes. Com isso, objetivou-se neste trabalho avaliar se larvas de Tenebrio molitor (Coleoptera: Tenebrionidae), tratadas com Bacillus thuringiensis, podem afetar as variáveis biológicas de Podisus nigrispinus (Hemiptera: Pentatomidae) e Palmistichus elaeisis (Hymenoptera: Eulophidae), importantes inimigos naturais de lagartas desfolhadoras na cultura do eucalipto. Larvas de T. molitor foram alimentadas com farelo de trigo contendo diferentes concentrações de Bacillus thuringiensis (0,00; 0,25; 0,50; 1,00; 2,00 e 4,00 g de Agree®/kg farelo). Quando as larvas atingiram em média 2 cm de comprimento, foram usadas como presa para P. nigrispinus (bioensaio I) e as pupas como hospedeiro para P. elaeisis (bioensaio II). As variáveis biológicas período de oviposição e número de ovos por postura do predador P. nigrispinus foram alteradas. Já as variáveis biológicas de P. elaeisis não foram modificadas, sendo possível o uso conjunto desses métodos de controle no manejo integrado de pragas.

ABSTRACT: The entomopathogen Bacillus thuringiensis (Bt) is widely used as one of the ingredients in pest control formulations, but researches conducted on its effect on non-target organisms are still in the nascent stage. This investigation aimed to uncover if Bt treated with Tenebrio molitor (Coleoptera: Tenebrionidae) larvae and pupae could affect the biological variables of Podisus nigrispinus (Hemiptera: Pentatomidae) and Palmistichus elaeisis (Hymenoptera: Eulophidae), all of which established natural enemies of leaf defoliator caterpillars in the eucalyptus culture. Larvae of T. molitor were fed on wheat bran containing different concentrations of B. thuringiensis (0.00; 0.25; 0.50; 1.00; 2.00 and 4.00 g Agree/kg bran). When the larvae attained size of about 2 cm, they were used as prey for P. nigrispinus (Bioassay I), and their pupae used as hosts for P. elaeisis (Bioassay II). Only the biological variables oviposition period and egg numbers by posture of the predator P. nigrispinus were altered. The biological variables of P. elaeisis were not altered, since it was possible to use these control methods within the integrated pest management.
