Effect of β-mercaptoetanol and cysteine on post-thawing quality and oxidative activity of ram sperm and on the viability of vitrified sheep embryos

RESUMO Foram avaliados os efeitos do β-mercaptoetanol (BME) e da cisteína sobre a viabilidade e a atividade oxidativa após o descongelamento do sêmen ovino e sobre o desenvolvimento in vitro e a viabilidade de embriões ovinos vitrificados. Ejaculados de quatro carneiros foram agrupados e diluídos, compondo seis tratamentos: sem antioxidantes; com BME 2mM; com BME 5mM; com BME 2mM e cisteína 5mM; com BME 5mM e cisteína 5mM; e com cisteína 5mM. Motilidade, integridade da membrana e do acrossoma, função mitocondrial, produção de espécies reativas de oxigênio e capacidade antioxidante total foram semelhantes entre os tratamentos (P>0,05). Em um meio sem antioxidantes, as taxas de clivagem e de desenvolvimento embrionário até blastocisto (60,3%, e 33,6%, respectivamente) foram semelhantes (P>0,05) às obtidas em um meio comBME 50μM e cisteína 600μM (64,3% e 36,6%, respectivamente). A viabilidade pós-descongelamento dos embriões vitrificados não diferiu entre os meios (P>0,05). O BME e a cisteína não influenciaram a viabilidade e a atividade oxidativa do sêmen ovino após o descongelamento e a viabilidade de embriões ovinos vitrificados.
ABSTRACT The effects of β-mercaptoethanol (BME) and cysteine on the viability and oxidative activity of ram sperm after thawing and on development in vitro and viability of vitrified sheep embryos were evaluated. Ejaculates from four rams were pooled and extended, composing six treatments: no antioxidants; 2mM BME; 5mM BME; 2mM BME and 5mM cysteine; 5mM BME and 5mM cysteine; and 5mM cysteine. Sperm motility, membrane and acrosome integrity, mitochondrial functionality, production of reactive oxygen species and total antioxidant capacity were similar across treatments (P>0.05). A medium with no antioxidant presented cleavage and blastocyst development rates (60.3% and 33.6%, respectively) similar (P>0.05) to those of a medium with 50μM BME and 600μM cysteine (64.3% and 36.6%, respectively). Post-thawing viability of vitrified embryos was similar between media (P>0.05). Cysteine and BME had no influence on the post-thawing viability and oxidative activity of ram sperm and on the viability of vitrified sheep embryos.

RESUMO Estudos sistemáticos que envolvem uso de tecnologias, como a eletromiografia de superfície (EMGs), podem fornecer dados importantes que capacitam os clínicos a diferenciar as alterações musculoesqueléticas, ligamentares ou neurológicas. Objetiva-se com este trabalho descrever as fases da marcha canina e o momento de ativação dos músculos de cães saudáveis da raça Boxer durante a marcha, por meio da eletromiografia de superfície. Avaliou-se a marcha de sete cães da raça Boxer, e o sinal elétrico da eletromiografia de superfície foi coletado do bíceps braquial, do tríceps braquial, do braquiocefálico, do reto femoral, do semitendinoso/semimembranoso e do glúteo superficial do hemicorpo direito. Os eletrodos monopolares circulares de prata com cloreto de prata (Ag/AgCl) foram posicionados no ponto médio entre a placa motora e a inserção desses músculos. Os sinais eletromiográficos foram capturados e adquiridos por uma interface ativa com ganho de 20x em configuração diferencial bipolar e por um condicionador de sinais (EMG Systems Brasil) de 16 canais, enquanto os cães caminharam em esteira com velocidade de 2m/s. Para análise estatística, foi aplicado teste de correlação de Pearson. Verificou-se correlação positiva entre os músculos retofemoral e bíceps braquial (r= 0,81); glúteo superficial e tríceps braquial (r= 0,69); semitendinoso/semimembranoso e bíceps braquial (r= 0,76); e entre o músculo reto femoral e o grupo muscular semitendinoso e semimembranoso (r=0,99). Os músculos bíceps braquial e braquiocefálico do mesmo membro trabalham em sinergia para posicionar o membro torácico durante o ciclo de marcha, enquanto o grupo muscular semitendinoso/semimembranoso flexiona o joelho e, simultaneamente com o quadríceps, que flexiona o quadril, impede que o membro pélvico entre em contato com o solo no balanço. Para impulsionar o corpo à frente, o músculo tríceps braquial estende o cotovelo e flexiona o ombro no contato final e o glúteo superficial estende o quadril.
ABSTRACT Systematic studies involving technologies such as surface electromyography (sEMG) may provide important data that enable veterinarians to recognize musculoskeletal, ligamentous, and neurological alterations. The aim of this study was to describe the gait cycle phases and the timing of muscle activation in healthy Boxer dogs during gait by means of sEMG. The gait cycle of seven Boxer dogs was evaluated and sEMG was recorded from the biceps brachii, triceps brachii, brachiocephalic, rectus femoris, semitendinosus, semimembranosus, and superficial gluteal muscles of the right hemibody. Circular monopolar Ag/AgCl electrodes were attached to the mean point between the motor point and the muscle insertion. The electromyographic signals were collected by an active interface with 20-fold gain in a bipolar differential configuration using a 16-channel signal conditioner (EMG Systems Brasil), while the dogs walked on a treadmill at a speed of 2m/s. Pearson's correlation was used for the statistical analysis. A positive correlation was found between the rectus femoris and biceps brachii (r= 0.81); superficial gluteal and triceps brachii (r= 0.69); semitendinosus-semimembranosus and biceps brachii (r= 0.76); and rectus femoris and semitendinosus and semimembranosus muscle groups (r=0.99). The biceps brachii and brachiocephalic muscles work in tandem to position the thoracic limb during the gait cycle, while the semitendinosus-semimembranosus group flexes the knee and, simultaneously with the quadriceps that flexes the hip, prevents the contact of the pelvic limb with the ground during the swing phase. The body is propelled forward by the triceps brachii muscle, which extends the elbow and flexes the shoulder at the final contact, while the superficial gluteal muscle extends the hip.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

O objetivo deste trabalho foi avaliar o efeito da aplicação de diferentes doses de nitrogênio no solo e de épocas de coleta sobre a eficiência nutricional do nitrogênio e a produção de biomassa em Calendula officinalis L. (Asteraceae) em condições de casa de vegetação. O substrato utilizado foi a camada de 0 - 20 cm de Latossolo Vermelho Distroférrico. O delineamento experimental foi inteiramente casualizado, com quatro repetições. Os tratamentos foram constituídos de quatro doses de N (0, 21, 42 e 84mg dm-3) e cinco épocas de colheita (15, 30, 45, 60 e 90 dias após o transplante). O teor de N na inflorescência de calêndula reduziu com as épocas de coleta, redução essa que ocorreu possivelmente devido à menor eficiência da absorção desse elemento pela planta e menor translocação do N para as inflorescências. Pode se concluir que as épocas de avaliação influenciaram na resposta da planta à eficiência de absorção (EFA), sendo a máxima EFA de N aos 51 dias após o transplante das mudas. Observou-se aumento na translocação de N da parte aérea (folhas e caules) para as inflorescências, tendo como consequência a redução do N na parte aérea. Houve incremento na produção de biomassa de calêndula em função das épocas de coleta , e do aumento das doses de N aplicadas no solo. A eficiência nutricional do N foi influenciada pelas épocas de avaliação e pelo teor de N presente no solo.
The aim of this study was to evaluate the effect of both the application of different nitrogen levels on the soil and the harvest periods on nitrogen nutritional efficiency and biomass production in Calendula officinalis L. (Asteraceae) under greenhouse conditions. The employed substrate was 0-20cm layer of Oxisol. Experimental design was completely randomized, with four replicates. Treatments consisted of four N levels (0, 21, 42 and 84 mg dm-3) and five harvest periods (15, 30, 45, 60 and 90 days after transplanting). N levels in Calendula officinalis inflorescences reduced with the harvest periods, and such a reduction was probably due to the lower efficiency of absorption of this element by the plant and to the lower N translocation to inflorescences. We can conclude that the evaluation periods influenced the response of plants to uptake efficiency (UE), and the maximal UE for N was found at 51 days after transplanting of seedlings. There was an increase in N translocation from the shoot (leaves and stems) to inflorescences, which has as consequence N reduction in the shoot. Calendula officinalis biomass production increased with the harvest periods and with the increase in the N levels applied on the soil. N nutritional efficiency was influenced by both the evaluation periods and the N levels present on the soil.

Tendo como objetivo avaliar a eficiência de absorção, translocação e uso de nitrogênio (N) e fósforo (P) pela Pfaffia glomerata, foi conduzido o presente trabalho em casa de vegetação no delineamento inteiramente casualizado com três repetições. Os tratamentos foram dispostos em esquema fatorial 5 x 5, sendo cinco doses de N (0, 125, 250, 375 e 500 mg vaso-1) e cinco doses de P (P2O5) (0, 125, 250, 375 e 500 mg vaso-1). Os resultados evidenciaram que a eficiência de absorção de N e P foi influenciada pelas doses de N e P utilizadas no experimento apresentando interação entre os fatores em estudo. A eficiência de translocação do N foi consideravelmente menor que a de P, provavelmente pela maior concentração de compostos nitrogenados nas raízes em detrimento da parte aérea. Observou-se maior eficiência de uso do P devido à menor concentração desse elemento na planta. A absorção de N e de P foi influenciada pela dose desses elementos no solo, sendo possível a estimativa das doses desses nutrientes que proporcionem maiores eficiências de absorção, translocação, uso e teor total.
Aimed at evaluating the efficiency of nitrogen (N) and phosphorus (P) uptake, translocation and use by Pfaffia glomerata, the present study was conducted in greenhouse, in completely randomized design with three replicates. Treatments were displayed in 5 x 5 factorial design, with five N levels (0, 125, 250, 375 and 500 mg pot-1) and five P (P2O5) levels (0, 125, 250, 375 and 500 mg pot-1). Results evidenced that N and P uptake efficiency was influenced by the N and P levels used in the experiment, showing interaction among the studied factors. N translocation efficiency was considerably lower than that of P, probably due to the lower concentration of nitrogenous compounds in the roots than in the shoot. P use efficiency was higher due to the lower concentration of this element in the plant. N and P uptake was influenced by the level of these elements in the soil, being possible to estimate the levels of these nutrients which provide higher uptake, translocation and use efficiency besides total content.

We encapsulated cisplatin into stealth pH-sensitive liposomes and studied their stability, cytotoxicity and accumulation in a human small-cell lung carcinoma cell line (GLC4) and its resistant subline (GLC4/CDDP). Since reduced cellular drug accumulation has been shown to be the main mechanism responsible for resistance in the GLC4/CDDP subline, we evaluated the ability of this new delivery system to improve cellular uptake. The liposomes were composed of dioleoylphosphatidylethanolamine (DOPE), cholesteryl hemisuccinate (CHEMS), and distearoylphosphatidylethanolamine-polyethyleneglycol 2000 (DSPE-PEG2000) and were characterized by determining the encapsulation percentage as a function of lipid concentration. Among the different formulations, DOPE/CHEMS/DSPE-PEG liposomes (lipid concentration equal to 40 mM) encapsulated cisplatin more efficiently than other concentrations of liposomes (about 20.0%, mean diameter of 174 nm). These liposomes presented good stability in mouse plasma which was obtained using a 0.24-M EDTA solution (70% cisplatin was retained inside the liposomes after 30 min of incubation). Concerning cytotoxic effects, they are more effective (1.34-fold) than free cisplatin for growth inhibition of the human lung cancer cell line A549. The study of cytotoxicity to GLC4 and GLC4/CDDP cell lines showed similar IC50 values (approximately 1.4 µM), i.e., cisplatin-resistant cells were sensitive to this cisplatin formulation. Platinum accumulation in both sensitive and resistant cell lines followed the same pattern, i.e., approximately the same intracellular platinum concentration (4.0 x 10-17 mol/cell) yielded the same cytotoxic effect. These results indicate that long-circulating pH-sensitive liposomes, also termed as stealth pH-sensitive liposomes, may present a promising delivery system for cisplatin-based cancer treatment. This liposome system proved to be able to circumvent the cisplatin resistance, whereas it was not observed when using non-long-circulating liposomes composed of phosphatidylcholine, phosphatidylserine, and cholesterol.

O experimento foi realizado com o objetivo de estudar características estruturais e morfológicas de cinco genótipos de Pennisetum sp. (CE 08 A.D., Venezuela, HV-241, Elefante B e Hexaplóide) sob pastejo no período de seca na Zona da Mata de Pernambuco. Os genótipos foram manejados sob lotação rotacionada (44 dias de descanso e quatro dias de pastejo) ao longo de dois ciclos de pastejo. Os dados foram analisados em delineamento experimental de blocos ao acaso com parcelas subdivididas (os genótipos representaram as parcelas e os ciclos de pastejo, as subparcelas) e quatro repetições. Houve variação nas características estruturais e morfológicas de genótipos de Pennisetum sp. Os genótipos CE 08 A.D. e Elefante B apresentaram maior densidade de lâmina foliar verde, menor porcentagem de material morto e maior densidade de perfilhos basais remanescentes e aéreos novos, com médias de 5,0±0,4 kg de MS/ha/cm, 33,0±0,2%, 22,0±0,2 e 146,0±0,4 perfilhos/m², respectivamente. O HV-241 apresentou alta participação de material morto em sua biomassa aérea (55,6±0,3%), o que pode ser atribuído à alta mortalidade de perfilhos (19,0±0,3 e 114,0±0,4 perfilhos basais e aéreos mortos/m², respectivamente). Os genótipos CE 08 A.D. e Elefante B destacaram-se como promissores para utilização sob pastejo no período de seca. O genótipo HV-241, híbrido de capim-elefante com o milheto, foi mais afetado pelos efeitos do estresse hídrico no período seco do ano.
The experiment was carried out to study structural and morphologic characteristics of five Pennisetum sp. genotypes (CE 08 A.D., Venezuela, HV-241, Elephant B and Hexaplóide) under grazing during the dry period in Pernambuco Forest Zone. Genotypes were managed under rotational stocking (44 days of resting and four days of grazing period). The experimental design was randomized blocks in a split plot arrangement, and four replications. The genotypes represented the plots, and grazing cycles, the subplots. There was variation in the structural and morphologic characteristics of the Pennisetum sp. genotypes. The CE 08 A.D. and Elephant B presented high density of green leaf blade, low percentage of dead material and high density of remaining basal tillers and new aerial tillers, with averages of 5.0±0.4 kg DM/ha/cm; 33±0.2%; 22±0.2 and 146±0.4 tillers/m², respectively. The HV-241 presented high participation of dead material in its aerial biomass (55.6±0.3%) due to high tiller mortality (19±0.3 and 114±0.4 basal and aerial dead tillers/m², respectively). The genotypes CE 08 A.D. and Elephant B had been distinguished as promising to be used under grazing in the dry period. The genotype HV-241, elephantgrass hybrid with pearl millet, was more affected by water stress during the dry period of the year.

(CE 08 A.D., Venezuela, HV-241, Elefante B e Hexaplóide) no período seco do ano na Zona da Mata de Pernambuco. Os genótipos foram manejados sob lotação rotacionada (44 dias de descanso e quatro dias de pastejo) ao longo de dois ciclos de pastejo. Adotou-se delineamento experimental em blocos ao acaso, com parcelas subdivididas (os genótipos representaram as parcelas e os ciclos de pastejo, as subparcelas) e quatro repetições. As massas de lâmina foliar total, verde e senescente pré-pastejo foram analisadas em seis repetições. O acúmulo e a eficiência de pastejo de lâmina foliar verde foram avaliados em delineamento em blocos casualizados, com quatro repetições. Os genótipos CE 08 A.D. e Elefante B apresentaram maiores massas de lâmina foliar total (1.374 kg de MS/ha) e verde pré-pastejo (737 kg de MS/ha) e maior acúmulo (654 kg de MS/ha/44 dias) e eficiência de pastejo de lâmina foliar verde (80%). No HV-241, a alta eficiência de pastejo (100% do acúmulo lâmina foliar verde) esteve associada ao baixo acúmulo de lâmina foliar verde (155 kg de MS/ha/44 dias). A menor eficiência de pastejo foi observada no Hexaplóide (59% do acúmulo de lâmina foliar verde), possivelmente em virtude das altas perdas sob pastejo (30% da massa de lâmina foliar total pré-pastejo). A massa de lâmina foliar verde diminuiu mais acentuadamente até o segundo dia de pastejo, enquanto as perdas de lâmina foliar total foram maiores no 2º e 3º dias de pastejo. Os genótipos CE 08 A.D. e Elefante B têm potencial para serem utilizados em sistemas de lotação rotacionada na Zona da Mata pernambucana.
The experiment was carried out to evaluate factors related to the grazing efficiency of five Pennisetum sp. genotypes (CE 08 A.D., Venezuela, HV-241, Elephant B and Hexaplóide) during the dry period in the Forest Zone of Pernambuco. Genotypes were managed under rotational stocking (44 days of resting and four days of grazing period). It was used a split-plot arrangement in a complete randomized block design (genotypes represented the plots; grazing cycles, the subplots). Six replications were used to determine pre-grazing total leaf blade mass and green and senescent leaf blade mass. Four replications were used for leaf blade accumulation and grazing efficiency and, for these variables, it was used a complete randomized blocks design. The CE 08 A.D. and Elephant B presented greater pre-graze total leaf blade mass, green leaf blade, green leaf blade accumulation, and grazing efficiency, with averages of 1,374 kg DM/ha, 737 kg DM/ha, 654 kg DM/ha/44 days and 80% of the green leaf blade accumulation, respectively. The highest grazing efficiency (100% of the green leaf blade accumulation) occurred at the HV-241 pastures. This high grazing efficiency was associated to the low green leaf blade accumulation (155 kg of MS/ha/44 days). The lowest grazing efficiency was observed for the Hexaplóide (59% of the green leaf blade accumulation), possibly due to high losses under grazing (30% of the pre-grazing total leaf blade mass). The green leaf blade mass decreased mostly until the second grazing day, while the losses of total leaf blade were higher in the second and third grazing days. The genotypes CE 08 A.D. and Elephant B presented potential to be used under rotational stocking, in the Pernambuco Forest Zone.

Objetivou-se com este trabalho avaliar a influência da relação folha/colmo (F/C) na degradabilidade in situ da MS (DISMS) de clones de capim-elefante. Foram estudados dois grupos de clones de capim-elefante: GRUPO I - Cameroon, Mercker México, Três Rios, Elefante de Pinda e Guaçu/IZ, de alta relação F/C; e GRUPO II - CE 08 A.D., Taiwan A-25, Goiano, CE 03 A.D. e Merc., de baixa relação F/C. Amostras pré-secas e moídas da planta inteira, colhidas aos 60 dias de crescimento no período seco, foram incubadas no rúmen de dois bovinos mestiços nos períodos de 0, 6, 12, 24, 48, 72 e 96 horas, para determinação da DISMS e de suas características. O delineamento experimental utilizado foi em blocos casualizados, em parcelas subdivididas. Não houve diferença significativa entre os grupos de capim-elefante em relação à DISMS. Foram obtidas as seguintes médias para as caratcerísticas de degradação ruminal: 79,40; 69,10; 26,90; 52,60, 20,60% e 5,50%/h para degradabilidade potencial, degradabilidade efetiva, fração solúvel, fração potencialmente degradável, fração não degradada e taxa de degradação, respectivamente. A relação folha/colmo não influenciou a DISMS de clones de capim-elefante.
The objective of this work was to evaluate the influence of leaf/stem ratio (L/S) in the in situ dry matter degradability (ISDMD) of elephantgrass clones. The experiment was carried out at IPA Experimental Station, in Vitória de Stº. Antão, PE. Two groups of elephantgrass clones were studied: GROUP I (Cameroon, Mercker México, Três Rios, Elefante de Pinda and Guaçu/IZ), characterized by its high L/S ratio, and GROUP II (CE 08 A.D., Taiwan A-25, Goiano, CE 03 A.D. and Merc.), with low L/S ratio. Samples of whole plant harvested in the 60th day of growth in the dry period were dried and milled and incubated in the rumen of the two crossbred bovines for 0, 6, 12, 24, 48, 72 e 96 hours to determine the ISDMD and ruminal fermentation parameters. The experimental design was randomized blocks in split-plot arrangement. There wasn't any significant difference between groups of elephantgrass as defined by their ISDMD. The degradation characteristics averaged 79.40, 69.10, 26.90, 52.60, 20.60%, and 5.50%/h for potential degradability, effective degradability, soluble fraction, potentially degradable fraction, no degraded fraction, and degradation rate, respectively. The leaf/stem ratio did not influence the ISDMD of elephantgrass clones.
