RESUMO Populações do patógeno da mela da soja (Rhizoctonia solani AG-1 IA) são altamente diferenciadas geneticamente ao longo de um gradiente latitudinal nas mais importantes regiões de cultivo de soja do Brasil. Entretanto, os processos evolutivos que guiaram a adaptação regional são ainda desconhecidos. O objetivo deste trabalho foi avaliar a importância relativa da variação genética neutra e da seleção natural sobre a divergência e adaptação regional de populações de R solani AG-1 IA da soja. Para tanto, comparou-se a diferenciação fenotípica por caracteres quantitativos (QST) e a diferenciação genética neutra (baseada em dados de microssatélites) entre três pares de populações (FST). Como medidas de respostas fenotípicas do fungo (caracteres quantitativos), estimou-se a tolerância ao estresse de temperatura e a tolerância a um fungicida cúprico de amplo espectro de ação (oxicloreto de cobre) sob condições ótimas (25 °C) e de temperatura elevada (33,5 °C). No geral houve aumento da variância genética com um efeito positivo sob a herdabilidade para tolerância ao fungicida cúprico na temperatura de estresse. As diferenças genéticas entre populações foram os principais determinantes da adaptação térmica em R. solani AG-1 IA (h2 > 0.70). A análise da estrutura genética neutra (FST) indicou subdivisão entre os três pares de populações. Embora a comparação de FST com os dados QST entre pares de populações não tenha seguido um único padrão, a maioria dos valores de QST estimados não diferiram significativamente de FST, indicando que, para os caracteres quantitativos estudados, a neutralidade (ou evolução neutra) teve importante papel na adaptação regional das populações do patógeno.

ABSTRACT Populations of the soybean leaf blight pathogen (Rhizoctonia solani AG-1 IA) are highly genetically differentiated along a latitudinal gradient in the major soybean growing regions of Brazil. However, the evolutionary processes leading to regional adaptation are still unknown. The objective of this study was to evaluate the relative importance of neutral genetic variation and natural selection on the divergence and regional adaptation of populations of the soybean-infecting pathogen R. solani AG-1 IA. Therefore, we compared the phenotypic differentiation in quantitative traits (QST) and the neutral genetic differentiation (FST, based on microsatellites data) among three pairs of populations. As measures of phenotypic responses of the fungus (quantitative traits), we estimated the tolerance to temperature stress and the tolerance to a broad-spectrum fungicide (copper oxychloride) under optimal (25 °C) and high temperature conditions (33.5 °C). In general there was an increase in genetic variance with a positive effect on the heritability for tolerance to copper fungicide under temperature stress. The genetic differences among populations were the main determinants of thermal adaptation in R. solani AG-1 IA (h2 > 0.70). The analysis of neutral genetic structure (FST) indicated subdivision between the three pairs of populations. Although population pairwise comparisons between FST and QST values did not follow a single pattern, the majority of QST values did not differ significantly from FST, indicating that, for the quantitative characters studied, neutrality (or neutral evolution) had a major role in the regional adaptation of R. solani AG-1 IA populations.

ABSTRACT. Sheath blight, caused by the fungal pathogen Rhizoctonia solani AG-1 IA is one of the most important rice diseases worldwide. The objetives of this study was to determine the predominant reproductive system and the genetic structure of 18 rice-infecting populations of R. solani sampled from China, Japan and the Philippines, the most important rice production countries in Asia. Knowledge about the population genetic structure of the pathogen in Asia is useful in identifying sources of infection and formulating sustainable management strategies for rice sheath blight. From a total of 717 isolates, 423 unique multilocus genotypes were detected based on nine microsatellite loci. The three country populations of R. solani AG-1 IA exhibited a mixed reproductive system, which included both sexual and asexual components. A moderate degree of clonality indicated that the asexual sclerotia represent important source of inoculum. Population subdivision varied within and among countries, fitting the isolation by distance model. While no subdivision was found among populations within Japan or within the Philippines, subdivision was detected among populations within China. Historic migration indicated high influx of immigrants from Japan into Northern, Central and Eastern China populations. Southern China contributed a high number of immigrants to the populations from the Philippines.

RESUMO A mancha areolada de Thanatephorus é uma das doenças mais importantes da seringueira (Hevea brasiliensis) na Amazônia. Apesar disso, questão importante sobre o real posicionamento filogenético deste patógeno ainda não foi respondida. Neste estudo, nós analisamos os padrões de variação genética em seqüências da região ITS-5.8S do rDNA de uma população de T. cucumeris (fase assexuada = Rhizoctonia solani AG 2-2) associado à mancha areolada da seringueira amostrada em Belém (Pará), Manaus (Amazonas) e Xapuri/Rio Branco (Acre) na Amazônia, e comparando-a filogeneticamente com membros do complexo AG 2 descritos mundialmente. Este estudo representa um passo importante para revelar a etiologia da fase assexuada de T. cucumeris da seringueira. Filogeneticamente, por análise Bayesiana, encontramos suporte para nomear um novo grupo de anastomose de R. solani associado à mancha areolada da seringueira: o AG 2-2 Hb. Este grupo constitui-se numa unidade evolutiva independente em relação aos subgrupos mundiais do AG 2-2 analisados. Na genealogia gênica construída por análise coalescente, observou-se que a população de R. solani AG 2-2 Hb de Belém é relativamente mais velha que as demais populações analisadas. O ancestral comum de todas as três populações analisadas está associado com a mancha foliar do maracujazeiro (Passiflora edulis), em Belém e tem cerca de 0,8 unidades evolutivas coalescentes de idade. Nenhum haplótipo da região ITS-5.8S do AG 2-2 Hb de Belém foi observado em outras regiões. Entretanto, a população de Manaus compartilhou dois, de seus quatro haplótipos, com aqueles observados em Xapuri / Rio Branco, no Acre, indicando fluxo gênico.

ABSTRACT Thanatephorus target spot is one of the most important diseases of rubber tree (Hevea brasiliensis) in the Amazon. However, an important question about the actual phylogenetic placement of this pathogen has not been answered yet. In this study, we analyzed the patterns of genetic variation in sequences of the ITS-5.8S rDNA region from a population of T. cucumeris (asexual phase = Rhizoctonia solani AG 2-2) associated with the rubber tree target spot sampled in Belém (Pará), Manaus (Amazonas) and Xapuri/Rio Branco (Acre), phylogenetically comparing with worldwide described members of AG 2. This study represents an important step to reveal the etiology of the asexual phase of T. cucumeris in the rubber tree. Phylogenetically, based on Bayesian analyses, we found support to nominate a new group of R. solani anastomosis associated with rubber tree target spot: AG 2-2 Hb. This group consisted of an independent evolutionary unit in relation to the worldwide described sub-groups of AG 2-2. Considering the gene genealogy built by coalescent analysis, we observed that the population of R. solani AG 2-2 Hb from Belém was relatively older than the other two analyzed populations. The common ancestor of all three populations is associated with passion-fruit (Passiflora edulis) foliar blight in Belém and has about 0.8 coalescent evolutionary units of age. No haplotype of the ITS-5.8S region of AG 2-2 Hb from Belém was observed in any other region. However, the population from Manaus shared two of its four haplotypes with those observed in Xapuri/Rio Branco, Acre, indicating gene flow.

RESUMO Muitos fitopatógenos de ampla distribuição geográfica como os fungos fitopatogênicos do gênero Rhizoctonia, estão sujeitos a extremas flutuações de temperatura em seus habitats. Para sobreviver a tais flutuações térmicas, esses organismos desenvolveram a habilidade de regular seus valores fenotípicos para adaptações térmicas bem como a outras variações ambientais específicas. O objetivo do presente estudo foi determinar o efeito do estresse térmico na evolucionabilidade para crescimento micelial de duas populações hospedeiro-distintas de Rhizocctonia solani AG-1 IA infectando arroz (OS6) ou Urochloa (BBT1) e compará-las a uma população de R. oryzae-sativae de arroz (OS5), espécie adaptada a temperaturas mais elevadas. As populações foram submetidas a duas temperaturas de crescimento: 25°C e 35°C (temperatura ótima e de estresse para R. solani AG-1 IA). Baseando-se em medidas do crescimento micelial, estimaram-se os coeficientes de variância genotípica (IG), variância ambiental (IE) e herdabilidade no sentido amplo (h2) como medidas de evolucionabilidade. Enquanto a população OS5 de R. oryzae-sativae foi pouco influenciada pelo estresse térmico, a 35oC detectou-se redução de h2 nas populações BBT1 e OS6 de R. solani AG-1 IA. Entretanto, os valores relativamente altos de h2 a 35oC (0,63 ± 0,07) indicaram que tanto a população BBT1 quanto a OS6 de R. solani AG-1 IA têm potencial de adaptação ao estresse térmico.

ABSTRACT A large number of widely distributed plant pathogens such as the phytopathogenic fungi of the genus Rhizoctonia are subject to extreme temperature fluctuations in their habitats. To survive such thermal fluctuations, these organisms have developed the capability of regulating their phenotypic values for specific thermal adaptations and to other specific environmental variations. The objective of this study was to determine the effects of thermal stress on the evolvability for mycelial growth in two host-distinct populations of Rhizoctonia solani AG-1 IA infecting either rice (OS6) or Urochloa (BBT1), and to compare them with a population of R. oryzae-sativae from rice (OS5), a species adapted to higher temperatures. The populations were subjected to two growth temperatures: 25°C and 35°C (optimal and stress temperatures for R. solani AG-1 IA). Based on mycelial growth, estimates of genotypic variance (IG), environmental variance (IE) and broad sense heritability (h2) were determined as measures of evolvability. The population OS5 of R. oryzae-sativae was almost not influenced by temperature stress, while at 35oC there was a reduction in h2 for the populations BBT1 and OS6 of R. solani AG-1 IA. However, the relatively high h2 values for mycelial growth at 35oC (0.63 ± 0.07) indicate that both populations BBT1 and OS6 of R. solani AG-1 IA have potential for adaptation to temperature stress.

ABSTRACT. Triazole fungicides have not been effective for managing the wheat blast disease in Brazil. A broad analysis across six geographical populations of Pyricularia graminis-tritici in central-southern Brazil indicated a high level of resistance to triazole fungicides. Since P. graminis-tritici is also associated with others poaceous species, here, we analyzed whether triazole-resistant isolates of the blast pathogen could be recovered from other poaceous hosts that are invasive of sprayed wheat fields. In addition to P. graminis-tritici (Pygt), we also evaluated the levels of sensitivity of three other grass-associated blast pathogens, which included P. grisea (Pg), P. pennisetigena (Pp), and P. urashimae (Pu). Resistance to the triazole fungicides tebuconazole and epoxiconazole was assessed phenotypically based on EC50 values and molecularly by analysis of the presence of mutations in the CYP51A gene, which encodes for the target enzyme 14-alpha-demethylase. We detected triazole-resistant Pyricularia spp. (Pg, Pp, Pu and Pygt) that is associated with Avena sativa, Cenchrus echinatus, Chloris distichophylla, Cynodon sp., Digitaria horizontalis, D. sanguinalis, Panicum maximum or Urochloa spp. The major outcome from our study was the evidence that invasive poaceous species from wheat fields could be an important source of triazole resistant fungal inoculum for the initial phases of the wheat blast epidemics.

RESUMO Esta pesquisa propõe a utilização de um método baseado em leitor automatizado de microplacas para detectar resistência a fungicidas triazóis (IDM) e estrobirulinas (IQe) em populações do fungo hemibiotrófico fitopatogênico Pyricularia graminis-tritici (Pygt) do trigo. A disponibilidade de método acurado, mais rápido e eficiente para a detecção da redução da sensibilidade a fungicidas contribuiria para facilitar a tomada de decisão sobre o manejo químico da brusone do trigo no país. Há informação sobre redução da sensibilidade de triazóis no controle da brusone do trigo e evidências da ocorrência generalizada de redução da sensibilidade à estrobilurinas em populações do patógeno no Brasil. O método de microplaca foi acurado em discriminar a variação fenotípica na sensibilidade entre isolados do patógeno aos fungicidas IDM tebuconazol e epoxiconazol e ao fungicida IQe azoxistrobina. Economicamente, ambos os métodos representam alto custo, portanto, é necessário comparar a eficiência desses métodos. O método de microplacas foi mais eficiente no uso de recursos, com custo operacional total 33% inferior ao do método de placas de Petri. Em conjunto, os fatores acurácia, rapidez e eficiência no uso de recursos indicaram que o método de microplaca pode ser utilizado no monitoramento da resistência a fungicidas em populações do fungo da brusone do trigo no agroecossistema brasileiro.

ABSTRACT This study proposes the application of a method based on an automated microplate reader to detect resistance to triazole (DMI) and strobilurin (QoI) fungicides in populations of the plant pathogenic hemibiotrophic fungus Pyricularia graminis-tritici (Pygt) from wheat. Availability of an accurate, more rapid and efficient method to detect eventual reduction in sensitivity to fungicides could contribute to facilitate the decision making on chemical management of wheat blast in the country. There are reports of reduction in the sensitivity of triazoles to control wheat blast and evidence of widespread reduction in the sensitivity to strobilurins in populations of the pathogen in Brazil. The microplate method was accurate in discriminating the phenotypic variation in sensitivity among isolates of the pathogen to the DMI fungicides tebuconazole and epoxiconazole and to the QoI fungicide azoxystrobin. Economically, both methods represent high cost, therefore, the efficiency of these methods should be compared. The microplate method was more efficient in the use of resources, with a total operating cost 33% inferior to that of the Petri dish method. Together, the factors accuracy, celerity and efficiency in the use of resources indicated that the microplate method can be used to monitor resistance to fungicides among populations of the wheat blast fungus in the Brazilian agroecosystem.

RESUMO Espécies do gênero Rhizoctonia estão associadas ao complexo de manchas da bainha do arroz, sendo que R. oryzae-sativae, em particular, causa a mancha agregada da bainha. Neste estudo objetivou-se determinar o potencial adaptativo de duas populações de Rhizoctonia oryzae-sativae à cultura do milho, uma das culturas comerciais amplamente cultivadas no estado de São Paulo e que pode ser utilizada em rotação com o arroz irrigado. Não há relatos da doença em milho no Brasil. Foram inoculados, em plantas de milho, isolados de duas populações de R. oryzae-sativae e determinou-se componentes de evolucionabilidade ou do potencial adaptativo de uma população (i.e., medidas de resposta à seleção) como os coeficientes de variância genotípica (IG), variância ambiental (IE) e a herdabilidade (h2) para os níveis de agressividade da doença. Comparou-se também a diferenciação fenotípica por caracteres quantitativos (QST) e a diferenciação genética neutra (baseada em dados de microssatélites) nas duas populações (FST). Experimentos similares usando plantas de arroz inoculadas com as duas populações do patógeno foram conduzidos como controle. Os isolados das populações de R. oryzae-sativae foram patogênicos e apresentaram variação na agressividade ao milho, com predominância de baixa herdabilidade para este atributo. Os valores estimados de QST não diferiram significativamente de FST indicando que a neutralidade teve importante papel na adaptação regional de populações do patógeno.

ABSTRACT Rhizoctonia species are associated with rice sheath-blight complex, and R. oryzae-sativae, in particular, causes aggregate sheath spot. This study aimed to determine the adaptive potential of two populations of R. oryzae-sativae to maize, one of the cash crops widely grown in São Paulo State and that could be used in rotation with irrigated rice. There is no report of the disease on maize in Brazil. Isolates of two populations of R. oryzae-sativae were inoculated in maize plants and components of evolvability or adaptive potential of a population (i.e., estimates of response to selection) were determined, such as genotypic variance coefficients (IG), environmental variance (IE) and heritability (h2) for the disease aggressiveness levels. Phenotypic differentiation based on quantitative traits (QST) was also compared with neutral genetic differentiation (based on microsatellite data) in the two populations (FST). Similar experiments using rice plants inoculated with the two populations of the pathogen were used as controls. Isolates from R. oryzae-sativae populations were pathogenic and had varied aggressiveness in maize, and low heritalibity predominated for this trait. The estimated values of QST did not differ significantly from those of FST, indicating that neutrality played an important role in the regional adaptation of the pathogen populations.

RESUMO O fungo Rhizoctonia solani grupo de anastomose AG-1 IA emergiu como patógeno importante associado à queima foliar, podridão do coleto e morte de pastagens do gênero Urochloa (braquiária) na América do Sul. Neste estudo objetivou-se determinar se a adaptação de R. solani AG-1 IA à Urochloa spp. na Colômbia promoveu diferenças fenotípicas de agressividade a hospedeiros distintos. Testou-se a hipótese de que as populações do patógeno adaptadas à Urochloa não são hospedeiro-especializadas e, desta forma, não estão geneticamente isoladas, mantendo assim ampla gama de hospedeiros, incluindo as fabáceas feijão caupi e soja. Determinou-se, também, se dois grupos de isolados obtidos de populações de R. solani AG-1 IA amostradas de Urochloa na Colômbia tinham potencial adaptativo para emergir como patógeno do milho. Além do nível de agressividade da doença em diferentes hospedeiros, determinou-se os componentes de evolutibilidade como o coeficiente de variância genotípica (IG), de variância ambiental (IE) e a herdabilidade (h2). Concluiu-se que: i) A adaptação de R. solani AG-1 IA à Urochloa spp. não promoveu diferenças fenotípicas de agressividade em hospedeiros distintos e, desta forma, o patógeno mantêm ampla gama de hospedeiros; ii) A população de R. solani AG-1 IA de Urochloa híbrido Mulato da Colômbia tem potencial adaptativo para emergir como patógeno do milho.

This study aimed to determine whether the adaptation of R. solani AG-1 IA to Urochloa spp. in Colombia promoted phenotypic differences in aggressiveness to distinct hosts. Our hypothesis was that the pathogen populations adapted to Urochloa spp. are not host-specialized and hence are not genetically isolated, thereby maintaining a broad host range, including the fabaceous plants cowpea and soybeans. We also determined whether two groups of isolates from populations of R. solani AG-1 IA sampled from Urochloa in Colombia have adaptive potential to emerge as maize pathogens. Besides the levels of disease severity in different hosts, we also determined the components of evolvability, such as the coefficient of genotypic variance (IG), the environmental variance (IE) and the heritability (h2). We concluded that: i) The adaptation of R. solani AG-1 IA to Urochloa spp. did not cause any phenotypic differences in aggressiveness to distinct hosts and therefore the pathogen maintains a broad host range. ii) The population of R. solani AG-1 IA from Urochloa hybrid Mulato in Colombia has adaptive potential to emerge as a maize pathogen.

RESUMO Espécies de fungos do gênero Pyricularia estão associadas com a doença brusone, em plantas da família Poaceae, causando perdas em culturas de importância econômica como arroz, aveia, centeio, cevada, trigo e triticale. Objetivou-se caracterizar o espectro de patogenicidade de P. pennisetigena e P. zingibericola em braquiária, cevada e trigo, bem como compará-los com os das espécies P. grisea e P. oryzae patótipo Triticum, de ocorrência generalizada no agroecossistema brasileiro. Foram testados 20 isolados de Pyricularia spp. obtidos de amostras de folhas infectadas de plantas invasoras de campos de trigo. A classificação dos isolados em espécies distintas de Pyricularia foi efetuada utilizando-se filogenia molecular baseada nos genes actina e calmodulina. Pyricularia pennisetigena e P. zingibericola inoculados em folhas, à concentração ajustada para 105 conídios mL-1, foram patogênicos a braquiária, cevada e trigo, com índices variáveis de agressividade.

ABSTRACT Fungal species from the Pyricularia genus are associated with blast disease in plants from the Poaceae family, causing losses in economically important crops such as rice, oat, rye, barley, wheat and triticale. This study aimed at characterizing the pathogenicity spectrum of P. pennisetigena and P. zingibericola to signal grass, barley and wheat, as well as comparing them with those from the species P. grisea and P. oryzae pathotype Triticum, which occur widely in the Brazilian agroecosystem. Twenty isolates of Pyricularia spp. were obtained from infected leaf samples of invasive plant species from wheat fields. The isolates classification into distinct Pyricularia species was done using molecular phylogeny based on actin and calmodulin genes. Pyricularia pennisetigena and P. zingibericola inoculated on plant leaves, at a concentration adjusted to 105 conidia mL-1, were pathogenic to signal grass, barley and wheat, with varying levels of aggressiveness.

RESUMO A queima da folha e morte de pastagens do gênero Urochloa são causadas pelo fungo Rhizoctonia solani. Objetivou-se determinar que patógenos do complexo de espécies do gênero Rhizoctonia estão associados à queima da folha e bainha em Urochloa e arroz, nos Llanos Colombianos. Utilizando-se o sistema de transcetos lineares, foram amostradas áreas de pastagens de Urochloa simpátricas, adjacentes às de arroz. A identificação dos patógenos foi efetuada por caracterização morfológica, detecção molecular baseada em iniciadores específicos e sequenciamento da região ITS-5,8S do rDNA. R. solani AG-1 IA predominou como patógeno associado à queima da folha em todas as amostras de U. brizantha cv. 'Toledo' e híbrido Urochloa cv. 'Mulato'. Além de R. solani AG-1 IA (18 % das amostras), R. oryzae-sativae (71 %) e Sclerotium hydrophilum (11 %) também foram detectados. No teste de patogenicidade, o fungo R. solani AG-1 IA foi o mais agressivo a Urochloa, enquanto R. oryzae-sativae ocasionou sintomas muito leves de infecção. Este é o primeiro relato de que os fungos R. oryzae-sativae e S. hydrophilum estão associados ao complexo de doenças da bainha do arroz, na Colômbia.

ABSTRACT Foliar blight and death of signalgrass (Urochloa spp.) pastures are caused by the Rhizoctonia solani fungus. This study aimed at determining which pathogens from the Rhizoctonia species complex are associated with leaf and sheath blight in Urochloa and rice, in the Colombian Llanos. Sympatric areas of Urochloa pastures adjacent to rice cropping areas were sampled using a linear transect system. The pathogens were identified using morphological traits, molecular detection based on specific primers and sequencing of the ITS-5.8S rDNA region. R. solani AG-1 IA predominated as the pathogen associated with foliar blight in all samples from U. brizantha cv. 'Toledo' and hybrid Urochloa cv. 'Mulato'. Besides R. solani AG-1 IA (18 % of the samples), Rhizoctonia oryzae-sativae (71 %) and Sclerotium hydrophilum (11 %) were also detected. In the cross-pathogenicity test, the R. solani AG-1 IA fungus was the most aggressive to Urochloa, while R. oryzae-sativae produced very mild infection symptoms. This is the first report of R. oryzae-sativae and S. hydrophilum associated with the complex of rice sheath blight diseases in Colombia.

ResumoAs estrobilurinas estão entre os fungicidas mais comumente utilizados no controle das doenças do trigo, isoladamente ou em misturas com fungicidas inibidores da enzima succinato desidrogenase (IDHS). As estrobilurinas são conhecidas como fungicidas inibidores da quinona oxidase ou fungicidas IQo. Por ter como alvo um único sítio de ação nas células fúngicas, o gene mitocondrial cyt b, o emprego contínuo das estrobilurinas pode representar alto risco de emergência de resistência a estes fungicidas em populações de fitopatógenos. O presente trabalho teve como objetivo testar a hipótese de que a resistência a azoxistrobina no fungo Pyricularia oryzae dotrigo resultou em resistência cruzada a piraclostrobina, outro fungicida IQo. Para testar esta hipótese, foi comparado o nível de resistência à piraclostrobina apresentado por dois grupos de isolados do fungo P. oryzae do trigo: a) sensíveis à azoxistrobina e portadores do gene cyt b não mutante (haplotipo H9) e b) resistentes à azoxistrobina e portadores da mutação G143A no gene cyt b(haplotipo H1). Fungicidas pertencentes a um mesmo grupo químico apresentam resistência cruzada. Todos os isolados de P. oryzaedo trigo sensíveis à azoxistrobina foram também sensíveis à piraclostrobina. Os isolados resistentes a azoxistrobina foram, também, resistentes à piraclostrobina, indicando que há resistência cruzada para os dois fungicidas. Entre os isolados resistentes, distinguiram-se dois grupos fenotípicos: (A) altamente resistentes e (B) resistentes. Isolados de P. oryzaecom a presença da mutação G143A no gene cyt b foram 42 vezes mais resistentes à piraclostrobina. Esses altos níveis de resistência à fungicidas IQo podem ser o resultado de elevada pressão de seleção exercida pelos anos consecutivos de aplicações de estrobilurinas para o manejo de doenças do trigo no Brasil.

ABSTRACTStrobilurins are among the fungicides that are most commonly used in the control of wheat diseases, separately or in mixtures with fungicides that inhibit the enzyme succinate dehydrogenase (SDH). Strobilurins are known as quinone oxidoreductase inhibitors or QoI fungicides. As in fungal cells they have one single site of action as target, the mitochondrial cyt b gene, the continued use of strobilurins may represent a high risk of emergence of resistance to these fungicides for plant pathogens populations. This study aimed to test the hypothesis that resistance to azoxystrobin in the wheat blast fungus Pyricularia oryzae resulted in cross-resistance to pyraclostrobin, another QoI fungicide. To test this hypothesis, we compared the level of pyraclostrobin resistance presented by two groups of P. oryzae isolates from wheat: a) sensitive to azoxystrobin and carrier of the non-mutant cyt b gene (haplotype H9), and b) resistant to azoxystrobin and carrier of the G143A mutation in the cyt b gene (H1 haplotype). Fungicides belonging to the same chemical group show cross-resistance. All P.oryzae isolates sensitive to azoxystrobin were also sensitive to pyraclostrobin. Isolates that were resistant to azoxystrobin were also resistant to pyraclostrobin, indicating cross-resistance to both fungicides. Two phenotypic groups were distinguished among the resistant isolates: (A) highly resistant and (B) resistant. P. oryzae isolates with the presence of G143A mutation in the cyt b gene were 42 times more resistant to pyraclostrobin. These high levels of resistance to QoI fungicides may be the result of high selection pressure exerted by consecutive years of strobilurin application for the management of wheat diseases in Brazil.

A brusone é causada pelo fungo Ascomyceto Pyricularia oryzae, sendo a doença mundialmente mais importante do arroz. Além do arroz, P. oryzae causa a brusone em trigo no Brasil, no Paraguai e na Bolívia. A alta variabilidade genético-patotípica observada em populações locais de P. oryzae, é possivelmente responsável pela baixa durabilidade da resistência de cultivares de arroz e trigo a referida doença, e talvez também seja determinante em eventos de 'mudança de hospedeiro' pelo patógeno. Esta revisão tem por objetivo apresentar aspectos relevantes da reprodução sexuada de P. oryzae, bem como informações sobre mecanismos de regulação do ciclo reprodutivo sexual do patógeno por meio dos genes mating type e feromônios, num sistema de reconhecimento específico. O conhecimento da biologia reprodutiva e da importância da reprodução sexuada em P. oryzae é essencial para o manejo da brusone baseado em resistência durável.

Blast is caused by the Ascomycete Pyricularia oryzae and is the most important disease affecting rice in the world. Besides rice, wheat is affected by blast caused by P. oryzae in Brazil, Paraguay and Bolivia. The high genetic-pathotypic variability observed in local populations of P. oryzae is probably responsible for the low resistance durability of rice and wheat cultivarsto this disease and may also be determinant in eventsof 'host shift' by the pathogen. This review aims to present important aspects of the sexual reproduction of P. oryzae, as well as information about the regulatory mechanisms of the pathogen sexual cycle by the genes mating type and pheromones in a specific recognition system. Understanding the reproductive biology and the importance of P. oryzae sexual cycle is essential for blast disease management based on durable resistance.

Atualmente, grupamento de anastomose (AG) de Rhizoctonia sp. em crisântemo e ocorrência deste fungo em gipsófila ainda não foram relatados no Brasil. Assim, realizou-se teste de patogenicidade normal e cruzada e sequenciamento da região ITS-5.8S rDNA para identificar o AG de isolado obtido de plantas de crisântemo (Papiro Branco) e de gipsófila, ambas originárias de Holambra / São Paulo, Brasil. Após os testes, relata-se pela primeira vez a ocorrência de R. solani AG-4 HG I em crisântemo (Papiro Branco e Amarelo) e R. solani AG-4 HG III em gipsófila, no estado de São Paulo, Brasil, e, também, a sua patogenicidade cruzada.

Currently, anastomosis groups (AG) of Rhizoctonia sp. on chrysanthemum and occurrence of this fungus on gypsophila have not been reported in Brazil. However, in the present study, normal and cross pathogenicity and sequencing of ITS-5.8S rDNA regions were used to confirm the AG of isolate of Rhizoctonia sp. obtained from chrysanthemum (White Papyrus) and from gypsophila plants cultivated in Holambra / São Paulo, Brazil. After these tests, it was confirmed the report of Rhizoctonia solani AG-4 HG I on chrysanthemum (White and Yellow Papyrus) and R. solani AG-4 HG III on gypsophila in the São Paulo state, Brazil, and also their cross pathogenicity.

A mancha areolada de Thanatephorus é uma das doenças mais importantes da seringueira (Hevea brasiliensis) na Amazônia brasileira. Além da seringueira, o fungo T. cucumeris (fase anamórfica Rhizoctonia solani) causa doenças foliares em outras espécies nativas ou cultivadas na região. Baseando-se na ausência de informações sobre quais os grupos de anastomose (AG) de R. solani estão associados à seringueira e a outros hospedeiros na Amazônia, testou-se a hipótese de que estes isolados pertencem a AG distintos. Também não há informação sobre a patogenicidade cruzada, à seringueira, de isolados provenientes de outros hospedeiros. A determinação do AG foi efetuada com base em análise filogenética da região ITS do rDNA. Concluiu-se que um novo subgrupo de R. solani (o AG-2-2 Hb) é o principal agente causal da mancha areolada em seringueira. Entretanto, isolados de outros AGs (AG-1 IB e ID, AG-4 HGI) e de Ceratobasidium sp. (Rhizoctonia sp. binucleada AG-R), oriundos de hospedeiros distintos, também infectam seringueira. Isto implica diversidade de fonte de inóculo para a sobrevivência e disseminação dos patógenos. De maneira inédita se relata a ocorrência de um novo AG de R. solani infectando citros no Acre, Brasil, distinto dos demais relatados no mundo (provavelmente AG-14).

Thanatephorus target spot is one of the most important diseases of the rubber tree (Hevea brasiliensis) in the Amazon region of Brazil. In addition to rubber tree, the fungus Thanatephorus cucumeris (anamorphase = Rhizoctonia solani) causes several foliar diseases on other agricultural crops or native species in the region. Based on the lack of information on which anastomosis groups (AG) of R. solani are associated with rubber tree and other hosts in the Amazon, we tested the hypothesis that these isolates belong to distinct AGs. There is no information about the cross pathogenicity (to rubber tree) of isolates from other hosts. The AG was determined based on phylogenetic analyses of the ITS region of the rDNA. We concluded that a new subgroup of R. solani (the AG-2-2 Hb) is the main pathogen causing the rubber tree leaf spot. However, isolates from other AGs (AG-1 IB and ID, AG-4 HGI) and from Ceratobasidium sp. (binucleate Rhizoctonia sp. AG-R) from distinct hosts can also infect the rubber tree. This implicates in diversity of inoculum sources for the survival and spread of the leaf spot pathogens. For the very first time, we report the occurrence of a new AG of R. solani infecting citrus in Acre, Brazil, which is distinct from all the others already described worldwide (probably AG-14).

Os grupos 3 e 4 de anastomose (AG-3 e AG-4) do fungo Rhizoctonia solani são importantes grupos associados à batata no mundo. No Brasil, o AG-3 é relatado afetando principalmente batata e fumo. Já o AG-4 causa perdas consideráveis em culturas de importância econômica, como a soja, o feijão e o amendoim, podendo ocorrer também em hortaliças como o espinafre, o pimentão, o brócolis, o tomate, a batata e frutíferas como o melão. Recentemente foi constatada, em Brasília-DF, a associação de R. solani a plantas invasoras em áreas de cultivo de batata. Entretanto, não há informação a respeito da etiologia do patógeno bem como do papel de espécies invasoras como outras hospedeiras no ciclo do patógeno. Objetivou-se com esse estudo caracterizar isolados de R. solani obtidos de batata e de outras três espécies de plantas invasoras associadas a áreas de cultivo da cultura: juá-de-capote [Nicandra physaloides (L.) Pers., Solanaceae], beldroega (Portulaca oleracea L., Portulacaceae), e caruru (Amaranthus deflexus L., Amaranthaceae). Foi confirmada a hipótese de que os isolados obtidos de R. solani de beldroega, caruru e juá-de-capote pertencem ao grupo 4 de anastomose e são patogênicos à batata, exceto o isolado de beldroega. Estes isolados apresentaram patogenicidade cruzada às três espécies e também patogênicos à maria-pretinha (Solanum americanum Mill.), uma outra espécie de Solanaceae invasora. A classificação dos isolados no grupo AG-4 HGI ou no grupo AG-4 HGIII (isolado de caruru) foi confirmada através de características culturais e moleculares (seqüenciamento da região ITS-5.8S do rDNA). Os resultados deste trabalho trazem implicações importantes para o manejo das podridões radiculares de Rhizoctonia em batata.

The anastomosis groups 3 and 4 (AG-3 and AG-4) of the fungus Rhizoctonia solani are important groups associated with potatoes worldwide. In Brazil, the AG-3 is reported affecting mainly potatoes and tobacco. The AG-4 cause considerable losses in crops of economic importance, such as soybean, beans and peanuts and may also occur in vegetables such as spinach, pepper, broccoli, tomatoes, potatoes and fruit such as melons. The association of R. solani with invasive plants was recently established in potato production areas from Brasília, DF. However, there is no information about the etiology of the pathogen as well as the role of invasive species as alternative hosts in the life cycle of the pathogen. The objective of this study was to characterize isolates of R. solani obtained from potatoes and three other invasive plant species associated with areas of potato production: Shoo-fly plant [Nicandra physaloides (L.) Pers., Solanaceae], pigweed (Portulaca oleracea L., Portulacaceae), and low-amaranth (Amaranthus deflexus L., Amaranthaceae). It was confirmed the hypothesis that the R. solani isolates obtained from pigweed, low-amaranth and Shoo-fly plant belong to the anastomosis group 4 and, except for the isolate from pigweed, are pathogenic to potatoes. These isolates were cross pathogencic to all the three weed species tested and also to American nightshade (Solanum americanum Mill.), another Solanaceae invasive of potato fields. The placement of the isolates in the group AG-4 HGI or in the group AG-4 HGIII (isolate from caruru) was confirmed by cultural and molecular characteristics (sequencing of the ITS-5.8S region of rDNA). The results of this study provide important implications for the management of the Rhizoctonia root rot in potatoes.
